Studies on the Processing of Crude Drugs(VIII): The Changes of Physico-chemical Parameter and Biological Activities by Processing of Evodia Fruit (한약수치에 관한 연구(제 8보): 오수유 탕포법)

  • We have studied the physico-chemical change and pharmacological transformation of traditional herbal medicines with processing. Evodia fruit (EF) has processed for the purpose of detoxification and reduction of its bitter taste. There are various methods of processing EF in Oriental medicinal reference. Among them, we processed EF according to the method of Sang-han theory, the most famous medicinal reference. We processed EF with washing in hot water, and then dried. Processed EFs(PEFs) were prepared according to the above procedure through repetition of 1,3,5 and 7 times respectively. The contents of weight loss, water extract, diluted ethanol extract, ether extract, total ash, acid insoluble ash, alkaloids and limonin in non-processed EF (NPEF) and PEFs were examined. The weight loss, and contents of water extract and dilute ethanol extract in PEF showed decrease in proportion to increase of washing time, while the contents of ether extract, total ash and acid insoluble ash showed little change as compared with those of NPEF. And the content of evodiamine and rutaecarpine was not changed distinctly. However, the content of limonin decreased in the final processed material in proportion to increase of washing times with hot water. And the intense of bitter taste in PEF was also reduced. These results were ascribed to the flowing-out of the water-soluble portion. The biological activities of NPEF and PEF were also investigated. In the test of DPPH scavenging effect, xanthine oxidase inhibition effect and TBA-Rs effect, PEF was more effective than NPEF in vitro. Also, both NPEF and PEF showed potent analgesic and anti-inflammatory effects against in mice. Especially, PEF by 3 times washing with hot water was more effective than other PEFs. From these results, it is considered that PEF should be prepared by 3 times washing with hot water.

Sulfur Dioxide, Heavy Metal and Curcumin Contents in Market-Available Turmeric (Curcuma longa L.) (유통 강황의 이산화황, 중금속 및 쿠르쿠민 함량)

  • Background: Turmeric (Curcuma longa L.) that is used as a food material has antioxidant, anticancer and anti-inflammatory properties. Recently the demand for functional foods and drugs has increased. The present study was carried out to determined of contents of residual sulfur dioxide, heavy metals, ash, acid insoluble ash and curcuminoids in turmeric from the Seoul Yak-ryeong market. Methods and Results: A total of 31 samples were obtained. Residual sulfur dioxide was not detected in any samples. Heavy metals (arsenic, cadmium, lead and mercury) were analyzed by ICP-MS (Inductively Coupled Plasma Mass Spectrometer) and a mercury analyzer and were detected in the ranges of 0.00 - 0.28, 0.00 - 0.07, 0.00 - 0.29 and 0.002 - 0.027mg/g respectively. No significant difference were observed between the average heavy metal contents of domestic and imported tumeric. However, average content of ash in domestic samples (7.8%) were significantly higher than that in imported samples (6.1%), whereas that of curcuminoids was significantly higher in imported samples (47.6mg/g) than in domestic samples (11.2mg/g). The average content of acid insoluble ash was not significantly different between two sample types (0.9% in each). Conclusions: There are no specific standards for turmeric used as food materials. Therefore, this study can be provided as basic data for the establishment of quality standards for turmeric.

Quality Improvement of Rayon Grade Bamboo Pulp by Modified Bleaching

  • The presence of high silica in bamboo hinders the use of this material for production of rayon grade pulp. Research has been carried out to overcome this deficiency and improve quality of rayon grade pulp with the modification in pulping and bleaching process. Effect of acid boosted water prehydrolysis, sulphuric acid pre-treatment of unbleached pulp, chlorination stage at lower pH and treatment of bleached pulp with $SO_2$ water were evaluated. Acid boosted water prehydrolysis of chips reduces prehydrolysis time by 50 minutes as compared to water prehydrolysis. Treatment of unbleached pulp with sulphuric acid reduces ash, acid insoluble, silica, calcium and iron contents of the pulp by 56, 31, 82, 84 and 60% respectively. The addition of acid, increase in kappa factor in $C_D$ stage and combination of both were effective in removing silica in the pulp. Treatment of final bleached pulp with $SO_2$ water removes silica to a great extent and improves optical properties of the pulp as compared to $H_2SO_4$ or PAA. Pretreatment of the pulp with acid and modification in the bleaching process can reduce silica substantially and improve the quality of rayon grade bamboo pulp.

Effect of Tannin and Species Variation on In vitro Digestibility, Gas, and Methane Production of Tropical Browse Plants

  • Nineteen tanniferous browse plants were collected from South Africa to investigate their digestibility, gas production (GP) characteristics and methane production. Fresh samples were collected, dried in forced oven, and ground and analyzed for nutrient composition. In vitro GP and in vitro organic matter digestibility (IVOMD) were determined using rumen fluid collected, strained and anaerobically prepared. A semi-automated system was used to measure GP by incubating the sample in a shaking incubator at $39^{\circ}C$. There was significant (p<0.05) variation in chemical composition of studied browses. Crude protein (CP) content of the species ranged from 86.9 to 305.0 g/kg dry matter (DM). The neutral detergent fiber (NDF) ranged from 292.8 to 517.5 g/kg DM while acid detergent fiber (ADF) ranged from 273.3 to 495.1 g/kg DM. The ash, ether extract, non-fibrous carbohydrate, neutral detergent insoluble nitrogen, and acid detergent insoluble nitrogen and CP were negatively correlated with methane production. Methane production was positively correlated with NDF, ADF, cellulose and hemi-cellulose. Tannin decreased GP, IVOMD, total volatile fatty acid and methane production. The observed low methanogenic potential and substantial ammonia generation of some of the browses might be potentially useful as rumen manipulating agents. However, a systematic evaluation is needed to determine optimum levels of supplementation in a mixed diet in order to attain a maximal depressing effect on enteric $CH_4$ production with a minimal detrimental effect on rumen fermentation of poor quality roughage based diet.

Physicochemical and histopathological parameters of broilers with dorsal cranial myopathy

  • Objective: This study aimed to investigate the effect of dorsal cranial myopathy (DCM) on chicken meat quality. Methods: Sixty-six Ross 308 AP broilers, 47 days old, of both sexes, weighing about 3.51 kg, were slaughtered according to standard industrial practices, and evaluated for meat color, pH, chemical composition, collagen content, fatty acid profile, and histopathological parameters. Comparisons between normal and DCM-affected meat were performed using Student's t-test at the 5% significance level. Results: Histological analysis of muscle tissues affected by DCM showed myofiber degeneration, proliferation of inflammatory cells, fibroplasia, and necrosis with fibrosis. DCM samples had lower protein content and higher moisture, ash, insoluble collagen, total collagen, and pH. DCM-affected meat was redder and more yellowish. There were no differences in lipid or soluble collagen contents between groups. DCM-affected meat had higher percentages of arachidonic acid (C20:4n-6) and eicosapentaenoic acid (C20:5n-3). Conclusion: This study revealed that DCM-affected meat exhibits considerable changes in quality parameters.

Relationships between the Inorganic Constituents Contents and the Catalpol and Sugar Contents in the Rhizoma of Rehmannia glutinosa (지황(地黃)(Rehmannia glutinosa) 근경중(根莖中)의 무기성분(無機成分)과 Catalpol 및 당함량(糖含量)과의 관계(關係))

  • This study was conducted to clarify the relationships between the inorganic constituents contents and the contents of catalpol and free sugars in the rhizoma of Rehmannia glutinosa. Nitrogen contents in rhizoma was positively correlated with the contents of catalpol and ethanol extract, whereas it was negatively correlated with that of ash, galactose and fructose. Calcium contents in rhizoma was highly negatively correlated with the contents of catalpol, ethanol extract and water extract. However, the calcium contents positively correlated with that of ash, acid insoluble ash, fructose and galactose in R. glutinosa. The catalpol cotents was negatively correlated with the contents of ash, galactose and fructose, whereas positively correlated with that of ethanol extract and water extract, and the correlations between extract contents and ash contents showed highly negative significance.

Comparative Study on the Official Compendia of Crude Drugs in Korea, China and Japan (한국·중국·일본 세 나라 생약관련 공정서 비교연구)

  • Korea, China and Japan have developed individually their own official compendia for crude drugs to tune with the times. Recent globalization has enlarged trade volume among these countries, and so the necessity for new quality standards has increased. This study was accomplished to offer basic data and appropriate suggestions to rebuilding standard principles for quality control of crude drugs in Korean official compendia. For this, seven standard items related to quality control in official compendia of these countries were compared. The items were identification, loss on drying, total ash, acid-insoluble ash, extract content, essential oil content and assay. The official compendia of three countries were the Korean Pharmacopoeia Eleventh Edition, the Korean Herbal Pharmacopoeia 2015, Chinese Pharmacopoeia 2015 and Japanese Pharmacopoeia Seventeenth Edition. Based on this basic research, new principles for each standard of crude drugs were suggested.

Quality monitoring of distributed materials from Glycyrrhizae Radix, Atractylodis Rhizoma Alba according to storage period (감초, 백출 유통품의 보관기간별 품질 모니터링)

  • This study was investigated to determine the quality monitoring of distributed materials from Glycyrrhizae Radix (26 samples), Atractylodis Rhizoma Alba (24 samples) according to storage period after $1{\sim}3$ year. We have estimated by identification, purity, loss on drying, ash, acid insoluble ash, extract content, essential oil content, assay and microbial contamination. As a result, Glycyrrhizae Radix (26 samples) were satisfied with the standard of K.P. (Korean Pharmacopoeia) and WHO's microbial contamination limit standard. In the Atractylodis Rhizoma Alba (24 samples), 2 samples were not satisfied with the standard of K.P.(Korean Pharmacopoeia) and WHO's microbial contamination limit standard. The results make practical application of the basic data for the quality control of herbal medicine in storage.

  • Quality evaluation of Magnoliae Flos and Anthrici Radix was studied to ensure their safety and efficacy, which was based on physico-chemical determination of the content of total ash, acid insoluble ash and diluted ethanol-soluble extract. In addition, quantitative analysis of magnolin and anthricin by HPLC suggested that the content of magnolin and anthricin in Magnoliae Flos and Anthrici Radix was $31.09{\pm}15.74%$ and $1.08{\pm}0.85%$.

Comparative Study of Processed (Shodhit) and Unprocessed Seeds of 'Gunja'-Abrus precatorius L.

  • 'Gunja' is attributable to the plant Abrus precatorius L. (Leguminosae). Three forms with red, brown and another with white seeds are known to occur in this species and are employed for different therapeutic uses viz. as purgative, emetic, aphrodisiac, tonic and also as an abortifacient. According to some Ayurvedic literature the seeds are poisonous and should be given to the patients after proper processing ('Shodhan'). A comparative study of various phytochemical parameters, namely, percentage of successive extractives, total proteins, tannins, total ash and acid insoluble ash of these three forms of the processed (with cow's milk and Kanji) and unprocessed seeds was done. TLC and densitometric scanning of successive extractives was also carried out to serve as markers for processed and unprocessed seeds. The percentage of proteins, tannins, alcohol and water soluble extractives decreased in the processed material. Besides, their acute toxicity, CNS activity were also studied in albino mice and it was found that white seeds are more toxic as compared to the red and brown. The toxic effect was reduced with the processing. Further, the 'Kanji' processed seeds are less toxic than the milk processed one.
