  • The sulfuric acid sulfonation of 2-aminonaphthalene was preformed at $30^{\circ}C{\sim}200^{\circ}C$ for $1{\sim}15$ hours, using $4{\sim}10$ parts of 90% sulfuric acid${\sim}$20% oleum. This reaction was influenced by many factory such as reaction temperature, reaction time, strength of sulfuric acid and amount of sulfuric acid and among these factors, the effect of the reaction temperature was considerable. The main products of this reaction were Dahl's acid (2-aminonaphthalene-5-sulfonic acid, Broenner's-acid (2-aminonaphthalene-6-sulfonic acid), amnio-F-acid(2-aminonapthalene-7-sulfonic acid), and Baden acid (2-aminonaphthalene-7-sulfonic acid) and another isomers such as Tobia's acid (2-aminonaphthalene-1-sulfonic acid) and 2-aminonaphthalene-4-sulfonic acid were not formed. Of these isomeric acids, those containing the sulfonic acid group in an ${\alpha}-position$, namely, Dahl's acid and Baden acid, are simultaneously formed by sulfonation at low-temperature ($30{\sim}100^{\circ}C$), whilst those containing the sulfonic acid group in a ${\beta}$-position, namely, Broenner's acid and amino-F-acid, are formed simultaneously by sulfonation at high temperatures($150^{\circ}{\sim}200^{\circ}$).

  • The hydrolysis of ${\alpha}$-bromophenylacetamide in the acidic media has been studied. The reactions in sulfuric acid and p-toluenesulfonic acid afforded mainly mandelic acid ; on the other hand, in hydrochloric acid ${\alpha}$-chlorophenylacetic acid was the main product. In either case, it was difficult to obtain ${\alpha}$-bromophenylacetic acid without concomittant formation of mandelic acid and ${\alpha}$-chlorophenylacetic acid respectively from the hydrolysis of ${\alpha}$-bromophenylacetamide. Mandelic acid and ${\alpha}$-chlorophenylacetic acid were obtained in good yields (86, 95 %) in sulfuric and hydrochloric acid respectively; the reactions are therefore potentially useful processes for obtaining them.

  • Dietary orotic acid is known to induce the fatty liver. Fatty acid profiles in the lipid fraction of the liver and the serum in rats fed with or with orotic acid diet were analyzed. In all the hepatic lipid fraction of rats fed on the supplemented orotic acid diet, there was a significant increased in linoleic acid. In addition, linoleic acid was also increased in the triacylglycerol fraction of hepatic endoplasmic reticulum and the triacylglycerol and diacylglycerol fractions of hepatic Golgi apparatus of the orotic acid-feeding rats. In the time course study of the fatty acid profile in the hepatic triacylglycerol and diacylgycerol fractions, an increase of linoleic acid was observed similarly in the initial stages of orotic acid intake in the both fractions. However, linoleic acid in the serum triacylglycerol fraction of orotic acid-feeding rats increased from day 1, but it began to decrease the increment from day 2, resulting in the lower level of linoleic acid in the serum triacylglycerol fraction of orotic acid-feeding rats than that of rat fed a orotic acid-free diet after 10 days. Oleic acid (18:1) was increased in the only cholesteryl ester fraction of helpatic. However, oleic acid level in other fractions was not changed. The compositions of 14:0, 16:0 and 18:0 was reduced in the hepatic triacylogylcerol, diacylglycerol and cholesteryl ester fractions by orotic acid-feeding. However, these saturated fatty acids were significantly increased in the serum triacylglycerol fractions. The orotic acid indcued changes in linoleic acid level in hepatic triacylglycerol may be explained by the impaired fatty acid metabolism and limited excretion of this fatty acid from liver to serum.

  • The allelopathic effects of aqueous extracts from Trifolium repens were studied. Aqueous extracts of leaves, stems, roots and flowers of T. repens strongly inhibited the seed germination and seedling growth of Zoysia japonica. In general, the higher concentration of the extracts showed the more great inhibition effect. The phenolic compounds extracted from T repens were analysed and identified using gas chromatography. 14 phenolic compounds were isolated from the leaves, stems, roots and flowers: caffeic acid, p-hydroxy benzoic acid, ferulic acid, gallic acid, p-coumaric acid, vanillic acid, trans-cinnamic acid, 2,5 dihydroxy benzoic acid, syringic acid, 2-hydroxy cinnamic acid, benzoic acid, salicylic acid, phloroglucinol and phanylacetic acid. The seed germination and relative growth ratio of Z. japonica by 14 phenolic compounds were inhibited in whole treatment. Stronger growth inhibitor were benzoic acid, salicylic acid and trans-cinnamic acid. From these results it is suggested that the phenolic compounds from T. repens seemed to be responsible for the allelopathic potential.

  • An alkalophilic bacterium isolated from compost was selected, identified and tested for the production of glutamic acid from ammonium fumarate. The bacterium was closely related to Bacillus brevis. The conditions for glutamic acid production were pH 8.0, 2% fumaric acid, and 0.8% nutrient broth. The mechanism of glutamic acid formation in this strain was postulated as following scheme. (1) Ammonium fumarate longrightarrow Aspartic acid (2) Aspartic acid + $\alpha$-Ketoglutaric acid longrightarrow Glutamic acid + Oxaloacetic acid.

  • Acid proteinase has been discovered in Aspergillus niger (acid protease A) and Cryphonectria parasitica (acid proteinase EapC) and it plays major roles in cheese formation from milk. In this study, a partial gene encoding acid proteinase in Penicillium oxalicum HCLF-34 was cloned by using PCR with degenerate primers corresponding to highly conserved regions of the acid proteinase. The partial acid proteinase gene in P. oxalicum HCLF-34 contains an open reading frame of 438 base pairs and encodes an acid proteinase protein of 146 amino aicds. The predicted amino acid sequences showed 71 % homology with acid protease A and 67% homology with EapC.

  • This study was done to investigate whether cholic acid derivatives have anti-stress activity and what is a cause of this anti-stress effect. Seven cholic acid derivatives (cholic acid, taurocholic acid, ursodeoxycholic acid, tauroursodeoxychoic acid, chenodeoxy cholic acid, dehydrocholic acid, hyodeoxycholic acid) were used, silymarin and valproic acid were used as positive controls. Stress was induced by restraint immobilization technique plus water immersion (24hrs) and adrenal weight, spleen weight, adrenal ascorbic acid, serum cholesterol, lactate dehydrogenase (LDH), alkaline phosphatase (ALP), adrenal cholesterol, glucose and corticosterone levels were measured as stress indicators. Most cholic acid derivatives markedly decreased the adrenal weight, and TUDCA and DHCA increased the spleen weight. The restraint stress induced increments in serum LDH, ALP and cholesterol were attenuated by most cholic acid derivatives. Cholic acid, taurocholic acid and tauroursodeoxycholic acid only increased the content of adrenal ascorbate. While valproic acid showed an inhibitory effect against stress, silymarin did not. Our findings suggest that most cholic acid derivatives have anti-stress effect and that their anti-stress effect is, in part, related to choleretic activity.
