황기 메탄올 추출물이 HaCaT 세포에서 Hyaluronic acid 생성에 미치는 영향 (The effect of Astragalus membranaceus methanol extract on hyaluronic acid production in HaCaT cells)

  • Objectives: Hyaluronic acid, high molecular glycosaminoglycan, exists in extracellular matrix of tissue, especially, in skin and has been known to be deeply involved in skin hydration. In this study, we investigated the effect of methanol extract of Hwang-gi, Astragalus membranaceus root, on hyaluronic acid production in human keratinocyte HaCaT cells. Methods: We determined hyaluronic acid synthase 2 gene expression and hyaluronic acid production in HaCaT cells by using RT-PCR and ELISA, respectively. Results: Hwang-gi extract didn't show the toxicity to HaCaT cells within the treated concentration and increased the hyaluronic acid synthase 2 gene expression and hyaluronic acid production. Conclusions: Hyaluronic acid production increased by Hwang-gi could be, partially, contribute to the moisturing effect in skin by it.

Brevibacterium ammoniagenes에 의한 글루탐산 제조에 관한 연구 (Studies on the Production of L-Glutamic Acid by Brevibacterium ammoniagenes)

  • A bacterium strain (K-173-10) which was isolated from waste soil of Korea brewing factory, could be grown on acetate as the sole carbon source and accumulated a considerable amount of L-glutamic acid in the medium. This strain was identified as the new species Brevibacterium ammoniagenes. This study was concerned not only with the culture condition for the production of L-glutamic acid and the cell growth, but also with the effects on concentration of various kind of organic substances, growth factors and penicillin. The results obtained were summarized as follow; 1. It was found that the concentrations of acetate and ammonium ions affected the growth of the bacterium as well as its L-glutamate accumulation. The optimum conditions of the composition of grown media for the growth of the bacterium and its glutamic acid production was found to be 40 g/l of total acetate, $100\;{\mu}g/l$ thiamine, $0.5\;{\mu}g/l$ biotin and $1{\sim}2g/l$ corn steep liquor as the growth factors. 2. Organic acid such as succinic acid, malic acid and ${\alpha}-ketoglutaric$ acid inhibited the cell growth as well as its L-glutamic acid production. 3. The penicillin (20 units/ml) stimulated the production of glutamic acid at appropriate incubation period. 4. It was found that this strain could grow in the presence of urea and ammonium acetate but not in other nitrogen sources.

유청배지에 Lactobacillus acidophilus KCCM 32820과 Propionibacterium freudenreichii KCCM 31227로 혼합배양시 유기산 생성에 관한 연구 (Study on the Production of Organic Acid by Fermentation with Mixed Culture of Lactobacillus acidophilus KCCM 32820 and Propionibacterium freudenreichii KCCM 31227 in Whey Broth)

  • L. acidophilus KCCM 32820과 P. freudenreichii KCCM 31227의 성장곡선 및 5%와 10% 유청 배지를 고압증기멸균 및 저온살균하여 L. acidophilus KCCM 32820과 P. freudenreichii KCCM 31227을 혼합배양시 생성되는 propionic acid와 acetic acid 양을 측정하였다. L. acidophilus KCCM 32820의 대수기는 $6\sim21$시간, P. freudenreichii KCCM 31227의 대수기는 $36\sim108$시간으로 나타났다. 혼합배양시 propionic acid의 생성량은 5% 유청 배지보다는 10% 유청 배지에서, 고압증기 멸균보다는 저온살균에서 많은 양이 생성되었다. Propionic acid의 최대 생산량은 10% 유청 배지를 저온살균하였을 때 배양 120시간에 7.88 mg/mL이었다. Acetic acid도 5%보다는 10% 유청 배지에서 높은 생성량을 나타냈고 그 생성량은 배양시간이 경과함에 따라 propionic acid 생성량의 $2:1\sim3:1$이었다.

환경친화적 섬유질 배합사료의 발효와 반추위 발효특성 변화 (Fermentation of Environmental Friend Total Mixed Ration and Alteration of Rumen Fermentation Characteristics)

  • Total mixed ration (TMR) including concentrate diet and roughage together have been used for the ruminant animal. Relatively high concentrations of moisture and water soluble carbohydrate are representative feature of TMR. Those moisture and water can also provide a niche for bacterial growth. Therefore, a possible fermentation of TMR induced by micro-organism is generally accepted. The present study hypothesized that different lactic acid bacteria could alter fermentation of TMR and subsequently rumen fermentation. Three lactic acid bacteria, Lactobacillus paracasei (A), L. plantarum (B) and L. parabuchneri (C), were employed and 7 treatments under full factorial design were compared with control without inoculation. TMR for dairy cow was used. Significant alterations by treatments were detected at lactic acid and butyric acid contents in TMR (p<0.05). Treatment AC (mixture of A and C) and BC (mixture of B and C) showed great lactate production. Great butyrate production was found at treatment C. At in vitro rumen fermentation, treatments B, C and AB (mixture of A and B) showed significantly great total gas production (p<0.05). All treatments except treatments B and AB, showed less dry matter digestibility, significantly (p<0.05). Total volatile fatty acid production at treatment AC was significantly greater than others (p<0.05). In individual volatile fatty acid production, treatment AB and AC showed great acetate and propionate productions, significantly (p<0.05). This study investigated correlation between organic acid production in TMR and rumen volatile fatty acid production. And it was found that butyric acid in TMR had significant negative correlation with acetate, propionate, total volatile fatty acid, AP ratio and dry matter digestibility.

산가수분해법과 반응표면분석법을 이용한 해조류 청각으로부터 레불린산의 생산 (Production of Levulinic Acid from Marine Algae Codium fragile Using Acid-Hydrolysis and Response Surface Methodology)

  • This work is focused on the possibility of marine biomass Codium fragile as renewable resources for production of levulinic acid. In an effort to optimize the reaction conditions of levulinic acid production from Codium fragile, response surface methodology was applied. A total of 18 individual experiments were designed to investigate the effect of reaction temperature, catalyst amount, and reaction time. As a result, 4.26 g/L levulinic acid from Codium fragile was produced in the condition of $160.7^{\circ}C$ of reaction temperature, 3.9% of sulfuric acid, and 39.1 min of reaction time. This result will provide the useful information for chemical production from marine resource.

단상 혐기성 소화공정에서의 동력학적 연구 (A Study on Kinetics in One-Phase Anaerobic Digestion)

  • Kinetic data for the acid phase anaerobic digestion were presented in this study and the constants were determined with acid production rate and gas production rate. Process models based on continuous culture theory were used to describe the characteristics of the acid forming microorganisms and to enable further development toward utilization of the process in a more rational manner. Acid phase digestion can be separated with appropriate manipulation of hydraulic retention time in anaerobic digestion. Kinetic analysis of data from the various hydraulic retention times using a phase specific model obtained form the acid phase indicated maximum specific growth rate of 0.40/h, saturation constant of 2,000mgCOD.$\ell$, yield coefficient of 0.35 mgVSS/msCOD utilized and decay constant of 0.04/h for the acid production rate. Similar analysis of data for the gas production rate indicated maximum specific growth rate of 0.003/h, saturation constant of 2,200mgCOD/$\ell$, yield coefficient of 0.035 mgVSS/mgCOD utilized and decay constant of 0.06/h.

배를 이용한 식포의 발효조건에 관한 연구 (A Study on Cultural Conditions for Acetic Acid Production Employing Pear Juice)

  • To produce economically important acid with pear juice, an acetic acid bacterium was selected from many isolated acetic acid bacteria. The alcohoic fermentation was conducted by inoculating pear juice with Saccharomyces cerevisiae ATCC 4124, and then the pear vinegar was prepared by batch cultivation in flaskes with the isolated Acetobacter sp. The optimum conditions for high yield of acetic acid were studied experimentally in the batch shake flask . For seed purposes the Acetobactor sp. was cultivated for 2 -days and transferred to the acid production medium . Optimum alcohol concentration, initial acidity and temperature for the acid production were 8.0% , 2.0% and 28$^{\circ}C$, respectively. Under the same conditions, the addition of yeast extract (1%) was observed to produce relatively high yield of acetic acid.

  • Structural modifications were performed on the C-3 and C-28 positions of ursolic acid and oleanolic acid and the NO production inhibitory activities of the resulting derivatives were evaluated. The SAR revealed that the ursolic acid and oleanolic acid derivatives 3a and 4a, which possessing methoxy group at C-3 position exhibit improved NO production inhibitory activity on LPS-induced RAW247 cells while showing less cytotoxicity than the parent compounds.

COMPARATIVE STUDIES ON INDIGENOUS STARTER CULTURE STRAINS FOR THEIR RATE OF ACID PRODUCTION

  • Three strains each of L. bulgaricus and S. thermophilus isolated from indigenous dahi were examined for their rate of acid production and corresponding pH values in skim milk medium incubated at $40^{\circ}C$. No significant increase in titrable acidity or decrease in pH were recorded from initial period up to 2nd hour in both the strains. Following this period, however, there was a continuous increase in titrable acidity and a decrease in pH value in the milk for all the tested strains. Further it was observed that there was a variation among these strains for their acid production rate. High rate of acid production was recorded for the L. bulgaricus as compared to S. thermophilus. The results further, suggested that efforts should be made to select a proper pair of L. bulgaricus and S. thermophilus according to their rate of acid production, at a particular temperature in order to produce a good quality product.

Alcaligenes sp. GB-77 에 의한 Polyhydroxyalkanoic Acid의 생산 (Polyhydroxyalkanoic Acid Production by Alcaligenes sp. GB-77)

  • For polyhydroxyalkanoic acid (PHA) production, several microorganisms were isolated from sewage sludge. One of them, GB-77 strain, was chosen from its PHB/HV copolymer production on only fructose without cosubstrate. The isolated strain GB-77 was identified as the genus Alcaligenes. Optimal temperature and pH for cell growth were 36C and 6.8. Optimal medium composition was 10 g/l of fructose and 5 g/l of polypeptone, 1 $\times$ 10$^{-2}$M Na$^{2}$HP0$^{4}$, 1.3 $\times$ 10$^{-2}$M KH$^{2}$PO$^{4}$. To investigate the optimal condition for polyhydroxyalkanoic acid production two-stage culture technique was used; first stage for cell growth and second stage for PHA production on unbalanced growth conditions. Optimal conditions for high PHA production were C/N ratio 50, temperature 36$\circ$C and pH 6.8. To overcome fructose inhibition on cell growth, intermittent feeding fed-batch culture technique was used. Total cell concentration was 17.4 g/l with 9.1 g/l of PHA. The purified PHA was identified PHB/HV copolymer by NMR analysis.
