Measurement of Free Polysaccharide in Tetanus Toxoid-Conjugate Vaccine Using Antibody/Ammonium Sulfate Precipitation

  • A method that effectively precipitates capsular polysaccharide of Haemophilus influenzae type b (polyribosylribitol phosphate, PRP) conjugated to tetanus toxoid (TT), PRP TT in a liquid vaccine has been developed to measure free PRP present in TT-conjugate vaccine. The method involves adding anti-TT antibody and ammonium sulfate to precipitate PRP-TT conjugate and measuring free PRP in tile supernatant. This new method provides a complete precipitation of the total PRP-TT, and provides an accurate and reproducible measurements of free PRP. The accuracy of the assay was confirmed by spiking known amounts of unconjugated PRP to PRP-TT conjugate, and the new method was found to have no effect on free PRP while precipitating PRP-TT. The published acid precipitation method did not produce reproducible results due to incomplete precipitation of PRP-TT, especially when the vaccine is formulated in a salt-buffered solution.

Phase Separation of Matrix Glasses and Precipitation Characteristics of CuCl Nanocrystals in CuCl Doped Borosilicate Glasses for Nonlinear Optical Application (CuCl 미립자 분산 붕괴산염계 비선형 광학유리에서 매질유리의 상분리와 CuCl 미립자의 석출 특성)

  • To investigate an effect of phase separation on precipitation characteristics of CuCl nanocrystals in CuCl doped nonlinear optical glasses, borosilicate glass systems with 9 different compositions with ~2wt% of CuCl were selected and CuCl doped glasses were prepared by melting and precipitation method. Microstructural properties of the CuCl doped glasses were analyzed by optical absorption spectroscopy, acid elution test, TEM, and EDXS. While phase separation did not occur in Glass A~D, interconnected and droplet microstructures due to phase separation were found in Glass E, F and Glass G~I, respectively. In the particular composition of the matrix glasses in this study, the precipitation of the CuCl particles was observed in the phase separable glasses, not in phase non-separable glasses. The CuCl particles were precipitated in both silica-rich phase region and boronrich phase region of the glass matrix. In the case of 7.7Na2O-36.6B2O3-52.7SiO2(mole%) glass, the larger CuCl particles than those in the silica-rich phase region were observed in the boron-rich phase region.

  • A series of $Ce_1Mg_xZr_{1-x}O_2$ (CMZO) mixed metal oxide with different molar ratio were prepared by simple co-precipitation method. The prepared materials were tested for their catalytic activity performance using Knoevenagel condensation of various aromatic aldehydes with barbituric acid under solvent-free condition in microwave. The best catalytic activity was obtained with CMZO (1:0.6:0.4). The synthesized materials were characterized by using XRD, FT-IR, SEM-EDS techniques.

  • For volume reduction of the wastewater generated on washing the soil contaminated with cobalt, recycling and reuse experiments of the wastewater were executed. Also. the soil remediation efficiency by repetitive washing with fresh citric acid was analyzed. The soil around TRIGA was sampled for the experiment. Results of recycling experiment by replacement-precipitation method were as follows. The remediation efficiency of 1st recycling wastewater was 97% and that of 2nd recycling wastewater was 94%. Also, To obtain remediation efficiency over than 90%, the 5th repetitive washing with fresh citric acid was needed.

  • High purity alumina and amorphous silica were prepared from Ha-dong kaolin by means of appliance of sulfuric acid. The effect of sulfuric acid concentration, reaction temperature and reaction time on the formation of aluminum sulfate was investigated. The precipitation conditions ofaluminum sulfate from the sulfuric acid solution with ethanol and ammonium hydroxide were deteremined. In the optimum condition, the conversion of aluminum oxide in kaolin to aluminum oxide powder was 85.0 percent. Alumina powder was prepared by calcination of the precipitates, and its purity was 99.0 percent.

  • A one dimensional time dependent acid rain model considering size distribution of aerosols and hydrometeors is developed to predict observed chemical and physical properties of precipitation. Temporal variations of anions and cations observed are predicted fairly well with acid rain model simulations. It is found that aerosol depletion rates are highly dependent on aerosol sizes under the assumption of Marshall - Palmer raindrop size distribution. Also, the aerosol depletion during the initial rain event largely influences on ion concentrations in rainwaters.

Studies on the Present State of Acid Precipitation in Seoul Area (서울地域의 酸性强雨現像에 關한 硏究)

  • This study was carried out to investigate the phenomena of acid precipitation. The pH value, electro conductivity and major anions (sulfate, nitrate and chloride) were measured by automatic acid rain monitor and ion chromatography at 5 points in Seoul area from Jan. to Dec. 1988. 1. The acidity of rainfall was in order winter (4.31) spring (4.77) fall (4.94) summer (5.31). Rainfall with a pH of less than 5.6 was appeared 83.2 percent. 2. The range of the highest appearence frequency rate both at Hannam-dong and Songsu-dong was from pH 4.6 to 5.0 and appeared 30.7 percent and 38.3 percent respectively, Bang-i-dong was 36.3 percent in the ranged from pH 4.1 to 4.5 and Guro and Ssangmun-dong were 26 percent and 30.3 percent in the ranged from pH 5.1 to 5.5 respectively. 3. The sulfate and nitrate ion concentration in earlier rainwater ranged from 0.1 ppm to 50.2 ppm and from 0.01 ppm to 15.8 respectively. The earlier rainwaters were generally more acidic than the after rainwaters. 4. The order of the major anion concentration in rainwater was $SO_4^{2-} > Cl^- > NO_3^-$ and the acidity of it was more effective by sulfate ion than others. The correlation between pH value and anions concentration was shown positive correlationship at Guro-dong and Bang-i-dong and negative correlation at Hannam-dong but not at the other sites.

Chemical Composition and Seasonal Variation of Acid Deposition in Chiang Mai, Thailand

  • This study aims to determine the chemical composition and seasonal variation of atmospheric acid deposition in order to identify possible sources contributing to precipitation. Sampling and analysis of 132 wet deposition samples were carried out from January to December 2008 at Mae Hia Research Center, Chiang Mai University, Chiang Mai Province. Total precipitation was 1,286.7 mm. Mean electro-conductivity and pH values were 0.94 mS/m and 6.27, respectively. Major cations ($Na^+$, ${NH_4}^+$, $K^+$, $Ca^{2+}$, and $Mg^{2+}$) and major anions ($HCOO^-$, $CH_3COO^-$, $Cl^-$, ${NO_3}^-$, and ${SO_4}^{2-}$) were determined by Ion Chromatography. The relative volume weight mean concentrations of anions, in descending order, were ${SO_4}^{2-}$ > ${NO_3}^-$ > $Cl^-$ > $CH_3COO^-$ > $HCOO^-$ and those of cations were $NH_4^+$ > $Ca^{2+}$ > $Mg^{2+}$ > $K^{+}$ > $Na^+$. Results of a principle component analysis highlighted the influence of various possible sources of ions such as agricultural activity, fuel combustion, marine sources, soil resuspension, and biomass burning.

A Characteristics and Distributions of Wet Deposition in Korea, 2005 (2005년 우리나라 습성강하물의 특성과 분포)

  • This study was carried out to characteristics and distributions of acid deposition in Korea. Precipitation was collected by acid deposition monitoring networks and analyzed for pH, conductivity, and following major ionic components $SO_4^{2-}$, $CI^-$, $NO_3^-$, $NH_4^+$, $Na^+$, $K^+$, $Ca^{2+}$, $Mg{2+}$. During the investigation period, Volume weighted annual mean pH of precipitation in Korea is 4.8, showing slightly acidic level. The amount of rainfall in the range of pH 5.1$\sim$5.5 charged approximately 28% of annual precipitation,23.4% in pH 4.6$\sim$5.0, and contributed 16.2% under pH 4.5. Among seasons, alkaline precipitation has occurred more often in spring, meanwhile acidic precipitation in which pH is under 4.5 has frequently occurred in autumn. Volume weighted annual mean concentrations of $SO_4^{2-}$, $NO_3^-$, $CI^-$ are 2.558 mg/L, 1.590 mg/L, 1.286 mg/L respectively, and provided that $SO_4^{2-}$, is the major contributor, followed by $NO_3^-$, $CI^-$. In case of cation, annual mean concentration for $NH_4^+$, $Na^+$, $K^+$, $Ca^{2+}$, $Mg{2+}$, $H^+$ are 0.693 mg/L, 0.528 mg/L, 0.439 mg/L, 0.455 mg/L, 0.089 mg/L,0.015 mg/L, and $NH_4$ were decided as the main contributor, followed by $Na^+$, $K^+$, $Ca^{2+}$, $Mg{2+}$, $H^+$. Annual wet deposition rate for sulfate, nitrate and ammonia are $3.316gm^{-2}yr^{-1}$, $2.057gm^{-2}yr{-1}$, $0.894gm^{-2}yr{-1}$, respectively, and it was founded that the deposition flux in summer contributes about 38.42% to 67.62% to total deposition.

Interpretation of Analytical Data of Ion Components in Precipitation, Seoul (서울 地域 降水中 이온成分 分析資料의 解析)

  • Precipitation samples were collected by the wet-only sampling method at Seoul from September 1993 to June 1995. Sample were analysed for the anions $(NO_3^-, NO_2^-, SO_4^{2-}, Cl^-, and F^-)$ and cations $(Na^+, K^+, Ca^{2+}, Mg^{2+}, and NH_4^+)$ in addition to pH and electric conductivity. In order to establish the chemical analysis data of high quality, the assurance checks for analytical data of precipitation were performed by considering the ion balance and by comparing the measured conductivity with the calculated conductivity. As we applied the various assurance checking methods by the ion balance used until recently to a data set measured in this study, the f value expressed as $\Sigma C/\Sigma A$ was found to be not appropriate for the data screening. Also, the scattering plot between cations and anions in each sample was found to show the general tendency of ion balance but was proved to not quantitate the standard of data screening at a data set of samples of various concentration levels. The h value defined as (A-C)/C for C $\geq$ A and (A-C)/A for C < A was used to check the ion balance. However, the standard of data screening by h value must very in response to total ion concentration of samples. In this study, the quality assurance of chemical analysis data was checked by considering both the ion balance of evaluating by h value and the conductivity balance. Further the quality control was achieved by these quality assurance methods. As the result, 67 samples among total 77 were obtained as valid. As the central tendency value for a statistical summary in the analytical parametr of samples, the volume-weighted mean value was found to represent more the general chemistry of precipitation rather than the arithmetic mean. The volume-weighted mean pH was 5.0 and 25% of samples was less than this mean. The concentrations of sufate and nitrate in precipitation were 90.4 ueq/L and 32.4 ueq/L which made up 59% and 21% of all anions. The raion of $SO_4^{2-}/(NO_3^- + NO_2^-)$ in precipitation was 2.7, which indicates that the contributions of $H_2SO_4$ and $HNO_3$ to the acidity of precipitation are 70% and 30%, respectively.
