Quantity of Ascorbic Acid in the Parts of Vegetable and Its variation by Cooking Method (상용채소의 부위에 따른 환원형 Ascorbic acid량과 조리방법에 따른 C량의 변화)

  • Ascorbic acid has a strong power to reduce other materials, so by using this reducing power we can analise many materials quantitatively. Dihydro-ascorbic acid is quantitatively changed into ascorbic acid under a certain condition, if reduced by hydrogen-sulfide. Dihydro-ascorbic acid also has physiolosical effect, but less effect than ascorbic acid. The effect of dihydro-ascorbic acid is regarded as a half of that of ascorbic acid. Among the analytical methods applying this theory the method using 2.6 dichlorophenol indophenol what called the method of Indophenol is used most widely; so does Hydrazine-method. But comparing these two methods the former shows a little higher rate in analytical value. Vegetable are vital sources for vitamin C. According to the report of the commitee of FAO Korean branch the amount of daily per head average intake of vitamin C is about 70mg in raw materials. Since vitamin C is easily affected by heating or oxidation in Cooking the loss is not a little. Consequntly it is regarded that the actual amount of intake will be much less than basic amount. It is therefore very important to find out that how much percent of the loss there will be in case of cooking, in order to dicide the actual amount of in take as proper nutrition for a person. Therefore this paper intended to give some help in setting a standard amount of V.C intake, by measuring the change of the V.C amount using the general cooking method and by measuring amount of V.C contained in the part of vegetable, used in Korea abundantly.

Study of Protonation Behaviour and Distribution Ratios of Hydroxamic Acids in Hydrochloric and Perchloric Acid Solutions Through Hammett Acidity Function, Bunnett-Olsen and Excess Acidity Method

  • The protonation parameters, dissociation constants ($pK_{BH^+}$) of conjugate acid, slope values (m, ${\phi}$ and $m^*$) and correlation coefficients (r) of hydroxamic acids were determined by Hammett acidity function method, Bunnett-Olsen method and excess acidity method in hydrochloric and perchloric acid solutions. Effect of acid concentration on partition and percentage protonation was also studied. $pK_{BH^+}$ values show that hydroxamic acids do not behave as Hammett bases, but hydroxamic acids behave as weak bases in strong acidic solutions. The values of $pK_{BH^+}$ obtained through Bunnett-Olsen method and excess acidity method were compared with the Hammett acidity function. ChemAxon's MarvinSketch 6.1.5 software was also used for determining $pK_a$, pI and microspecies distribution (%) of hydroxamic acids with pH. Hydrogen donor and acceptor values and logD were also obtained. The results show that N-p-chlorophenyl-4-bromobenzohydroxamic acid has the highest $pK_a$ and lowest logD values. On the contrary, N-phenyl-3,5-dinitrobenzohydroxamic acid has lowest the $pK_a$ and highest logD values.

  • The flash point is one of the most important physical properties used to determine the fire hazard of flammable liquid mixture and defined as the lowest temperature at which a liquid produces sufficient vapor to form a combustible mixture with air. The main purpose of this paper is to measure and predict the flash point of binary flammable miscible mixtures. The flash points for n-hexanol+n-butyric acid and n-butanol+propionic acid, were measured by using Seta-flash closed cup method. The experimentally derived data were correlated with the binary interaction parameters of the van Laar and NRTL equations through the optimization method. The flash points estimated by these correlations were compared with those calculated by the method based on Raoult's law. The optimization method were found to be better than the method based on the Raoult's law.

  • An isotope dilution-liquid chromatography/tandem mass spectrometric method was developed as a candidate reference method for the accurate determination of folic acid in infant milk formula. Sample was spiked with 13C5-folic acid and then extracted with phosphate buffer (pH 6) solution. The extract was further cleaned up by deproteinization followed by a C18 solid-phase extraction cartridge. The extract was analyzed by using LC/ ESI/MS/MS with selectively monitoring the collisionally induced dissociation channels of m/z 442 → m/z 295 and m/z 447 → m/z 295, which are the neutral glutamyl loss from the [M+H]+ ions of folic acid and 13C5-folic acid, respectively. LC/MS/MS chromatograms showed substantially reduced background from chemical noises compared to LC/MS chromatograms. Repeatability and reproducibility studies showed that the LC/MS/ MS method is a reliable and reproducible method which can provide less than 1.5 relative percentage of method precision.

Influence of the Extraction Method on Quality of Citron Juice (착즙방법에 따른 유자과즙의 품질 비교)

  • This study was performed to compare quality of citron juice as affected by the extraction method. The yield of citron juice was 24.49% by method I (rotary-crushing and screening), 18.09% by method II (pressing) and 12.60% by method III (belt-pressing), respectively. Juices by methods I and II had more soluble solid contents and essential oil and pulp volume than that by method III. Method III was higher in titratable acidity than methods I and II. The contents of fructose, glucose and sucrose in method III were 0.54%, 0.37% and 1.11%, respectively, which were lower values than those in other methods. But there was no siginificant difference in the contents of total sugar by the extraction method. For fatty acids composition, the contents of oleic acid in method I, palmitic acid, linoleic acid and linolenic acid in method II, and stearic acid in method III, respectively, were highest when compared with other methods. The contents of free amino acids detected in method III were smaller than those in methods I and II. Threonin was detected only in method I, methionine and cystine were not detected in methods I. II and III. But the contents of the total amino acids in method III were $1.3{\sim}1.6$ times as large as those in methods I and II.

  • A new method of synthesis of isobarbituric acid is described. With this method, isobarbituric acid can be synthesized both directly and conveniently. Yields of isobarbituric acid are up to 75 per cent.

  • To enhance the utilization of chestnut husk discarded in the processing company antioxidative activities and compounds were investigated. Antioxidative activities of solvent fractions from chestnut husk were examined by benzoic acid hydroxylation method ferric thiocyanate method and DPPH test. Ethyl acetate fraction showed strong antioxidative activities comparable to BHA. Active compounds were isolated and purified from ethyl acetate fraction by Sephadex LH-20 column chromatography and preparative HPLC. A major active compound gallic acid was identified by $^{1}$H and $^{13}$ C-NMR. The phenolic acid contents was analyzed by GC and ellagic acid (172.22 mg%) and gallic acid (107.39 mg%) were major phenolic acid of chestnut husk.

A Study on the Optimal Analytical Method for the Determination of Urinary Arsenic by Hydride Generation-Atomic Absorption Spectrometry (HG-AAS법에 의한 요중 비소의 최적 분석법에 관한 연구)

  • This study was carried out to examine the optimal analytical method for determination of urinary toxic arsenic (inorganic arsenic and its metabolites) by HG-AAS (hydride generation-atomic absorption spectrometry). In the analysis of SRMs (standard reference materials), method E (addition of 0.4% L-cysteine to pre-reductant and use 0.04M HCl as carrier acid) showed the most accurate results compared with the reference values. In the analysis of 30 urinary samples, analytical results were significantly different depend on the component of pre-reductant and the concentration of carrier acid. When the concentration of carrier acid was higher, the analytical result was lower. The recovery rates of MMA (monomethylarsonic acid) and DMA (dimethylarsenic acid) were varied by the concentration of pre-treatment acid and carrier acid and hydride generation reagents. When the concentration of carrier acid was 1.62 M (5% HCl), the recovery rates of DMA was 1%. The recovery rates of MMA and DMA in method E (=V) were 102% and 100%, respectively. The results of this study suggest that the component and concentration of pre-reductant and carrier acid must be carefully adjusted in the analysis of urinary arsenic, and method E is recommendable as the most precise analytical method for determination of urinary toxic arsenic.

Development of HPLC-UV method for detection and quantification of seven organic acids in animal feed (사료 중 유기산 7종 동시분석법 개발 및 유통 사료의 모니터링)

  • 본 연구에서는 사료 첨가제로 이용되고 있는 유기산 7종(formic acid, malic acid, lactic acid, acetic acid, citric acid, fumaric acid, propionic acid)의 동시분석법 개발을 위한 연구를 실시하였다. 7종의 화합물은 표준물질의 Retention time과 UV spectra를 통해 구별하였고, 분석법 검증은 직선성, 민감성, 선택성, 정확성, 정밀성을 통하여 검증하였다. 그 결과로 LOD와 LOQ의 범위가 각각 43~26,755 μg/kg, 12-8,026 μg/kg으로 설정하였고, 평균 회수율이 79.3~95.2%로 우수하게 보였으며, intra-day, inter-day에 대한 전반적인 상대 표준 편차(%RSD)는 3.2% 미만으로 나타났다. 이와 같이 검증된 자료를 통해 유기산의 동시분석에 대한 직선성, 민감성, 선택성, 정확성 및 정밀성을 확인하였고, 높은 수준을 나타냄을 알 수 있었다. 이를 바탕으로 유기산이 검출되는 단미사료 46 가지를 분석에 적용하여 진행하였고, 정량과 동시분석 검출을 위한 방법은 RP-HPLC/UV 검출기를 이용하여 성공적으로 개발되었다. 따라서 본 연구결과를 바탕으로 하여 사료 중의 유기산의 분석이 신속하고 정확해졌을 뿐 아니라, 다른 종류의 사료 또한 이를 적용하여 효율적으로 이용할 수 있을 것으로 판단된다.

Optimization of the Lowry Method of Protein Precipitation from the H. influenzae Type b Conjugate Vaccine Using Deoxycholic Acid and Hydrochloric Acid

  • The Lowry method was used in this study to measure protein in Haemophilus influenzae type b (Hib) conjugate vaccines (polyribosylibitol phosphate-tetanus toxoid; PRP-TT) using deoxycholic acid (DOC) to induce protein precipitation. Trichloroacetic acid (TCA) did not induce precipitation adequately from the Hib conjugate bulk and the freeze-dried Hib conjugate product. Its yield was approximately 50%. The matrix structure of Hib conjugate inhibits precipitation by TCA. Although the Lowry method can be carried out without precipitation in Hib conjugate bulk when no residual impurities (adipic acid dihydrazide [ADH], 1-ethyl-3-(3-dimethylamino-propyl) carbodiimide-HCI [EDAC], phenol and cyanogens bromide [CNBr], etc.) are present, it cannot be used for Hib conjugate products that contain sucrose 8.5%, because 8.5% concentration of sucrose enhanced the protein concentration. DOC- and HCl-induced precipitation is an alternative method for evaluating the protein content of the Hib conjugate bulk and the Hib conjugate product. The precipitation was optimal with a final concentrate of 0.1% for DOC at $4^{\circ}C$ and pH 2. This Lowry method, using DOC/HCI precipitation to induce protein precipitation, was confirmed a consistent, reproducible, and valid test for proteins in Hib conjugate bulk and its freeze-dried product.
