Dust samples have been collected from streets, schools, subway stations and households in Daegu metropolitan city. Samples were sieved through a 100 ${\mu}m$ mesh and the concentration of 14 elements have been determined using by ICP after acid extraction. Results showed that Ca, Fe, K, Mg, Mn, Na and V were influenced by natural sources while Cd, Cr, Cu, Ni, Pb and Zn were influenced by anthropogenic sources. The measured values were remarkably higher in components from natural sources than in components from anthropogenic sources. In particular, school dust had higher levels of Ca and Pb and subway station dust had higher levels of Cu and Zn. The percentage composition of chemicals from subway stations, households, and schools were remarkably higher in components from anthropogenic sources than that from streets. It is well recognized that anthropogenic sources were affected by indoor dust. Results of pollution index of hazardous heavy metals indicated that schools, households, and subway stations were more contaminated than streets and urban areas typically had higher pollution index than rural areas. The correlation analysis among trace elements seem to suggest that there were correlations between components of soil/road dust resuspension, and components of waste incineration and fuel combustion.

In order to evaluate the degree of air pollution, soils from 125 and 60points of main streets of Seoul and Daejeon, respectively, were sampled and analyzed for the contents of lead, copper, iron and zinc by nitric acid($HNO_3$) extraction followed by atomic absorption spectrophotometry during June and September, 1990. The Pb contents of Seoul and Daejeon were 350.2$\mu$g/g and 175.9$\mu$g/g, respectively, which were 14.5times and 7.3times respectively higher than the control(24.0). In Seoul, the commercial area gave the highest value followed by resdential and industrial areas in the decreasing order, while the order in Daejeon was Industrial, commercial and residential areas. The Cu contents in Seoul and Daejeon were very similar(236.6 and 234.6, respectively) each other, and about 11times higher than the control(20.9). The concentration was highest in industrial, followed by commercial and the lowest in residential areas in Seoul, while the order in Daejeon was industrial, residential and commercial areas. The Fe contents were 25716.5 in Seoul and 22838.6 in Daejeon, both being similar to the control value(22537.0). The value was highest in industrial and next in commercial areas and the lowest in residential area both in Seoul and Daejeon. The Zn contents in Seoul and Daejeon were 728.2 and 404.0, respectively, wihch were 6.3 and 3.6times higher than the control(113.3). The value was highest in commercial and next in industrial areas and the lowest in residental area in Seoul, while the order was industrial, residential and commercial areas in Daejeon. The heavy metal content was higher in the soil of main streets than that of minor streets in Seoul. The content of lead in Seoul was higher in the streets of Chongro-ku(525.9) and Chung-ku(684.8), the central area of the city which had a heavy traffic, than those in Nowon-ku(320.3) and Kwanak-ku(312.6) which had relatively less traffic crowd. The similar pattern was observed in the content of zinc; it was higher in Chongro-ku(1305.0) and Chung-ku(1335.4) than in Nowon-ku(489.1) and Kwanak-ku(569.8). In Daejeon, the contents of lead and zinc were decreasing as the distance from Daejeon Railroad Station increased : they were respectively 157.8 and 447.7 within 2km from the station, 157.6 and 235.7 with in 2-4km, and 63.5 and 156.2 within 4-6km and 58.7 and 142.2 within 6-8km. In summary, the contents of lead, copper and zinc were generally higher in the central area of the city, areas of heavy traffic and areas of industry and commerce.

In this study, abiotic transformation of 1-naphthol(1-NP) via oxidative-coupling reaction and its reaction products were investigated in the presence of Mn oxides. The reaction products were characterized for their relative polarity using solvent extraction experiment and reverse-phase HPLC, and for structure using CCMS and LC/MS, and for absorption characteristics using UV-Vis spectrometry. The reaction products present in aqueous phase were more polar than parent naphthol and comprised of 1,4-naphthoquinon(1,4-NPQ) and oligomers such as dimers and trimers. Hydrophilic component present in water phase after solvent$(CH_2Cl_2)$ extractions was identified as naphthol polymerized products having molecular weight(m/z) ranging from 400 to 2,000, and showed similar UV-Vis. absorption characteristics to that of foil fulvic acid. Transformation of 1,4-NPQ, which is non-reactive to Mn oxide, to the polymerized products via cross-coupling reaction in the presence of 1-NP was also verified. In this experimental conditions(20.5 mg/L, 1-NP, 2.5 g/L $MnO_2$, pH 5), the transformation of 1-NP into the oligomers and polymerized products were about 83% of initial 1-NP concentrations, and more than 30% of the reaction products was estimated to be water insoluble fractions, not extracted by $H_2O$ methanol. Results from this study suggest that Mn oxide-mediated treatment of naphthol contaminated soils can achieve risk reduction through the formation of oligomers md polymer precipitation.

Porcine placenta was treated with alkali, acid and enzyme treatment to obtain extracts. Heavy metal contents such as Pb, As, and Hg were low enough to satisfy cosmetic agent standard. As a result of safety test(MTT assay), porcine placenta extracts showed over 80% of cell viability at $50{\mu}g/ml$, and cell toxicity was relatively lower. From antioxidation test using DPPH free radical scavenging assay, antioxidation effect was highest as 63% at $50{\mu}g/ml$ when porcine placenta was treated with alkali in pH 9. From whitening effect test using tyrosinase inhibition assay, tyrosinase inhibition effect was 30% at $50{\mu}g/ml$ concentration in alkali treated procine placenta, however, the efficiency was lower compared with arbutin or vitamin C. In anti-wrinkle effect test from elastase inhibition assay, elastase inhibition effects were 20~30% at $50{\mu}g/ml$ for 5 kinds of porcine placenta treatments, which was superior to standard, and especially, protease treated extracts showed best results. Skin formulation including 1% porcine placenta was made and the formulation was very stable for temperature and storage period. From this research, porcine placenta extract showed high potential for anti-wrinkle functional cosmetic agent.
This study was conducted to compare the antioxidative effects of Dendranthema zawadskii var. latilobum (Maxim.) Kitam and D. zawadskii var. yezoense (Maek.) Y.M. Lee & H.J. Choi due to the harvest date. Harvested samples at May 9th (early), July 17th (middle) and September 3rd (late stage) were extracted with 80% ethanol, and biological activities and antioxidant substance contents were detected. The earlier harvested samples showed the higher moisture contents and extraction yields. DPPH radical scavenging effect of early harvested D. zawadskii var. latilobum (Maxim.) Kitam ($RC_{50}$ = $0.128\;mg\;{\cdot}\;mL^{-1}$) was similar to BHT ($0.121\;mg\;{\cdot}\;mL^{-1}$). ABTS radical scavenging effects of both species harvested at early stage were higher than that of ascorbic acid which was well-known natural antioxidant. However, both species harvested at late stage showed the highest $Fe^{2+}$ chelating effect, but it was lower than that of EDTA. Total polyphenol and flavonoid contents of both species were higher when samples were harvested earlier stage. However, both species harvested at early stage contained more total polyphenols (79.93 and $75.10\;mg\;{\cdot}\;g^{-1}$) than flavonoids (57.84 and $54.91\;mg\;{\cdot}\;g^{-1}$).

To develop a natural antioxidative peptide, the gelatin was extracted from fish (Yellowfin sole) skin by hot $water(50^{\circ}C)$ extraction method and hydrolyzed with Alcalase, pronase and collagenase through a continuous 3-step membrane reactor. Each step enzymatic hydrolysates were determined the antioxidative activity and their synergistic effects, compared with $\alpha-tocopherol$ and butylated hydroxytoluene (BHT). Also, we tried to investigate the antioxidative disposition of peptide which was successfully separated by gel filtration, ion-exchange chromatography, and HPIC in cultured rat hepatocytes intoxicated with tert-butyl hydroperoxide (TBHP). Second step enzymatic hydrolysate (SSEH) among all hydrolysates and $\alpha-tocoperol$ was showed the strongest antioxidative activity. The optimum concentration of antioxidative activity for SSEH was $1\%(w/w)$ in linoleic acid. The synergistic effects were increased in using the hydrolysate with tocopherol and BHT. In the presence of the peptide isolated from SSEH, supplemented hepatocytes exposed to TBHP showed that delayed cell killing and decreased significantly the lipid peroxidation, compared with hepatocytes not cultured with isolated peptide.
This study was designed to determine the most effective concentration of fluoride and levels of laser irradiation for the remineralization of decayed teeth. After irradiation with a pulsed Nd:YAG laser and the topical application of fluoride, phosphate and fluoride concentration in enamel were measured. And then the changes on surface enamel using an scanning electron microscope were observed. Samples by extraction healthy, permanent, mandibular bovine teeth with no caries were obtained. Among them 371 healthy samples were selected and artificial carious lesions were made. 20 samples were assigned to each experimental group. After irradiation with a pulsed Nd:YAG laser with total energy densities of 10J/$\textrm{cm}^2$, 20J/$\textrm{cm}^2$ for each group. On the teeth, 2% NaF, 1.9% NH4F, 1.6% TiF4 Elmex gel(amine fluoride) and 1.23% APF gel were applied. After pH circulatory procedures, concentrations of fluoride with and Ionalyzer (Orion Research, Model 901, USA) and phosphates with an Uv/V is spectrophotometer (Uvikon 860, Kontrom Co, Switzerland) were measured. By etching the teeth in layers and measuring fluoride concentrations, a profile of fluoride penetration according to depth could be developed. And also the changes on the surface of the enamel using an electron scanning microscope were observed. The comparative analysis yielded the following results : 1. Phosphate concentration was low in all groups compared with the control group except for teeth treated Elmex gel, irradiated with 10J/$\textrm{cm}^2$ and 30J/$\textrm{cm}^2$ energy densities. Teeth treated with APF gel and 30J/$\textrm{cm}^2$ irradiation gad the lowest phosphate concentration. 2. Among all groups, fluoride concentrations in tooth enamel were highest in APF gel and NaF groups irradiated at 30J/$\textrm{cm}^2$. The APF gel group had the highest fluoride concentrations across all energy densities. 3. In the APF gel group, and the NaF group, the greater the energy density of the laser, the higher the fluoride concentrations in the enamel. 4. In all groups, the concentration of fluoride in the enamel by depth tended to decrease with depth. 5. Under the scanning electron microscope, under the condition of irradiation with 20J/$\textrm{cm}^2$, enamel crack was detected for the first time. In the NH4F group, spherical deposits were found on the surface of the enamel, and in the TiF4 group the surface of enamel was covered with an irregular, thin membranous mass in places. In the APF gel and NaF groups irradiated with 10J/$\textrm{cm}^2$, spherical and irregular particles covered the teeth. When these groups were irradiated at 20J/cm2, they were covered with amorphous crystals. These results suggest that one could obtain more effective anticariogenic effects without damage to teeth when less than 20J/$\textrm{cm}^2$ energy densities and APF gel are used.

Alterations in the cellular genome affecting the expression or function of genes controlling cell growth and differentiation are considered to be the main cause of cancer. Over 30 oncogenes can be activated by insertional mutagenesis, single point mutations, chromosomal translocations and gene amplification. The ras oncogenes have been detected in $15{\sim}20%$ of human tumors that include some of the most common forms of human neoplasia and are known to acquire their transforming properties by single point mutations in two domains of their coding sequences, most commonly in codons 12 and 61. The ras gene family consists of three functional genes, N-ras, K-ras and H-ras which encode highly similar proteins of 188 or 189 amino acid residues generically known as P21. ras proteins have been shown to bind GTP and GTP, and possess intrinsic GTPase activity. Experimental study was performed to observe the mutational change of the ras gene family and apply the results to the clinical activity. 36 Golden Syrian Hamster each weighing $60{\sim}80g$ were used and painted with 0.5% DMBA by 3 times weekly on the right buccal cheek(experimental side) for 6, 8, 10, 12, 14 and 16 weeks. Left buccal cheek (control side) was treated with mineral oil as the same manner of the right side. The hamsters were sacrificed on the 6, 8, 10, 12, 14 & 16 weeks. Normal and tumor tissues from paraffin block were completely dissected by microdissection and DNA from both tissue were isolated by proteinase K/phenol/chloroform extraction. Segments of the K-ras and H-ras gene were amplified by PCR using the oligonucleotide primers corresponding to the homologous region (codon 12 and 61) of the hamster gene, and then confirmational change of ras genes was observed by SSCP and autosequencing analysis. The results were as follows : 1. Malignant lesion could be found in the experimental side from the experimental six weeks. 2. One hamster among six showed point mutation of the H-ras codon 12($G{\rightarrow}A$ transition) at the experimental 10 and 14 weeks. 3. One of six at 6 weeks, two of six at 8 weeks and one of six at 12 weeks revealed the confirmational change of the H-ras codon 61($A{\rightarrow}T$ transversion). 4. The incidence of point mutation of H-ras codon 12 and 61 were 5.5%(2 of 36) and 11%(4 of 36) respectively. 5. Point mutation of the K-ras could not be seen during the whole experimental period. Form the above results, these findings strongly support the concept that H-ras oncogenes may have the influence of the DMBA induced carcinoma of hamster buccal pouch.

Seeds of Korean pine cone (Pinus koraiensis) have long been consumed as an edible food in countries located in North-East Asia, On the other hand, Korean pine cone, containing various polyphenols, is discarded as a useless garbage after removing seeds. This study investigated the lipolytic effects of pine cone extract in differentiated 3T3-L1 adipocytes. Intracellular lipid accumulation was measured by Oil red O staining, free glycerol release by colorimetric reaction, and expression of genes related to lipid metabolism by real-time PCR. Compared to control, pine cone extract reduced intracellular lipid accumulation by 8.8% and increased free glycerol release by 8.2% a concentration of $5{\mu}g/mL$ in differentiated 3T3-L1 adipocytes. mRNA levels of fatty acid synthesis were not significantly different between control and pine cone extract, but mRNA levels of lipoprotein lipase (LPL) and hormone-sensitive lipase (HSL) significantly increased by 38.7% and 94.1% at a concentration of $5{\mu}g/mL$, respectively. Thus, pine cone extract is suggested to have lipolytic effects through induction of LPL and HSL gene expression.

Flavor components were trapped by stimultaneous steam distillation-extraction method for investigating it in the bellflower roots and fractionated into four groups such as a neutral, a basic, a phenolic and an acidic fraction. An acidic fraction methylated with diazomethane solution and three others were analysed by GC and GC-MS equipping a fused silica capillary column, and S-containing compounds in these were detected with a flame photometric detector (FPD). The total of one hundred and three compounds from the bellflower roots were identified: they were 6 aliphatic hydrocarbons, 10 aromatic hydrocarbons, 2 terpene hydrocarbons, 12 alcohols, 8 terpene alcohols, 17 aldehydes, 3 terpene aldehydes, 5 ketones, 5 esters, 3 furans, 2 thiazoles, 2 lactones, 2 sulfides, 9 phenols, l2 carboxylic acids and 5 others. The greater part of the others except carboxylic acids were identified from a neutral fraction of which was assumed to be indispensable for the reproduction of bellflower root odor in a sensory evaluation. As a result of a sensory evaluation, 1-hexanal, trans-2-hexenal, 1-hexanol, cis-3-hexenol, trans-2-hexenol, 1-octen-3-ol and so forth identified in a neutral fraction were considered to be the key compounds of grass-like odor in the bellflower roots.
