수분부족이 콩의 질산환원효소 활성과 Abscisic acid 함량에 미치는 영향 (Effects of Water Stress on Nitrate Reductase Activity and Abscisic Acid Content in Soybean Plants)

  • 콩의 내한발성 품종육성 및 선발에 대한 기초자료를 얻고자 팔달콩, 새알콩 및 단엽콩을 공시하여 수경재배에서는 수경액을 PEG처리로, pot 실험에서는 단수처리하여 수분부족을 유기하고 PEG 농도, 품종 및 생육시기별로 엽의 수분 potential, 질산환원효소의 활성 및 ABA 함량을 조사하였다. 1. 엽의 수분 potential은 PEG의 농도가 높을수록 낮았으며 생육시기에 따라서는 제1엽기보다 제3엽기와 제5엽기에 높았는데, 품종간에는 차이가 없었다. 엽의 수분 potential은 단수처리 3일후에 무처리에서보다 약 2.2배가 감소되었다. 2. 질산환원효소의 활성은 PEG의 농도가 높을수록 낮았고 제1엽기보다 제3엽기와 제5엽기에 높았으며, 품종간에는 팔달콩이 새알콩과 단엽콩 보다 높았다. 단수처리에서는 처리 3일후에 질산환원효소의 활성이 무처리에서보다 약 47%가 감소되었다. 3. ABA의 함량은 PEG의 농도가 높을수록 높았고 제1엽기와 제3엽기보다 제5엽기에 높았으며 품종간에는 팔달콩이 새알콩과 단엽콩보다 높았다. 단수처리에서는 처리 3일 후에 ABA의 함량이 무처리에서보다 약 1.7배가 증가하였으며 생육후기보다 생육초기에 더욱 증가하였다.

물의 종류를 달리한 동치미의 발효과정 중 비타민 C와 젖산균수의 변화 (The Changes of Vitamin C and Lactic Acid Bacteria Count in Dongchimi used Different Kinds of Water)

  • The purpose of this study was to investigate the changes of vitamin C and lactic acid bacteria count in Dongchimi used different kinds of water (distilled water, purified water, Cho Jung Carbonated Natural water). Dongchimi used different kinds of water was fermented at $10^{\circ}C$ for 46 days. The changes of pH on Dongchimi used different kinds of water decreased in all samples during fermentation period, and then showed a slow decrease after 12 days of fermentation. The total acidity of Dongchimi used Cho Jung Carbonated Natural water arrived slowly at best tasting condition(0.3~0.4 point) compared with other conditions. The changes of salt content were ranked high one by one , Cho Jung Carbonated Natural water>purified water>distilled water during fermentation period. At early stage of fermentation, the changes of turbidity of Dongchimi used Cho Jung Carbonated Natural water showed highly as compared with other test conditions for 12 days of fermentation. Vitamin C content was measured high in Dongchimi used Cho Jung Carbonated Natural water during the fermentation period. Because calcium content was high in carbonated natural water, carbonated natural water had the highest calcium content during the fermentation period. The changes of lactic acid bacteria count showed the highest price at all experimental groups in 15 days of fermentation, but those of Dongchimi used Cho Jung Carbonated Natural water showed the highest price in 19 days of fermentation.

Aspergillus awamori var. kawachii에 의한 쌀 Koji 제조시 유기산의 생산조건 (Cultural Conditions for the Production of Organic Acid During)

  • This study was carried out to investigate the influences of cultural conditions of koji on the production of organic acid during rice-koji making by Aspergillus awamori var. kawachii which is now widely used as koji-mold in brewing Takju and Yakju in Korea. The optimum temperature for the germination of the conidia of the mold was 35'8, and the time required for germination at this temperature was 8 hours. Rapid germination occurred when the water content of steamed rice was above 40%, but germination retardation occurred markedly below 35%. The optimum cultural temperature for the production of organic acid was 32$^{\circ}C$, and the production of organic acid was markedly restricted at 36$^{\circ}C$ and 4$0^{\circ}C$. It was effective for the high production of both saccharogenic amylase and organic acid to shift the cultural temperature from initial 36$^{\circ}C$ to 32$^{\circ}C$ after 20~25 hours of cultivation. Initial water content suitable to the production of organic acid was 40% in steamed rice, but its production was markedly restricted below 30% of water content. When the quantity of conidial inoculation was too small, the production of organic acid was low in initial phase, but it was retrived at later period. Acid production was markedly restricted together with the increase in koji thickness.

가압형 경수로 압력용기 재료인 저합금강의 동적 붕산 부식 실증 연구 (Dynamic Boric Acid Corrosion of Low Alloy Steel for Reactor Pressure Vessel of PWR using Mockup Test)

  • This work is concerned with an evaluation of dynamic boric acid corrosion (BAC) of low alloy steel for reactor pressure vessel of a pressurized water reactor (PWR). Mockup test method was newly established to investigate dynamic BAC of the low alloy steel under various conditions simulating a primary water leakage incident. The average corrosion rate was measured from the weight loss of the low alloy steel specimen, and the maximum corrosion rate was obtained by the surface profilometry after the mockup test. The corrosion rates increased with the rise of the leakage rate of the primary water containing boric acid, and the presence of oxygen dissolved in the primary water also accelerated the corrosion. From the specimen surface analysis, it was found that typical flow-accelerated corrosion and jet-impingement occurred under two-phase fluid of water droplet and steam environment. The maximum corrosion rate was determined as 5.97 mm/year at the leakage rate of 20 cc/min of the primary water with a saturated content of oxygen within the range of experimental condition of this work.

Protective Effect of Artificially Enhanced Level of L-Ascorbic Acid against Water Deficit-Induced Oxidative Stress in Rice Seedlings

  • Effects of the enhanced level of L-ascorbic acid (AA) on the water deficit-induced oxidative damage were studied in rice (Oryza sativa L.) seedlings. The seedlings sprayed with 20 to 80 mM L-galactono-${\gamma}$-lactone (GL), a putative precursor of AA, showed 2 to 5-fold higher levels of AA compared with controls. Pretreatment of the seedlings with GL prior to water stress imposition caused virtually no effect on dehydration of tissues during water deficit but substantially mitigated oxidative injury, as accessed by 2-thiobarbituric acid-reactive substances, ${\alpha}$-tocopherol, chlorophylls and ${\beta}$-carotene. Proline accumulation during water stress was also significantly lowered in the treated seedlings. In a complementary experiment, AA retarded photodegradation of ${\alpha}$-tocopherol in isolated thylakoids far more efficiently than glutathione. GL in itself did not show any noticeable reactivity toward ${\alpha}$-tocopheroxyl radical. The results demonstrate the antioxidative function of AA in rice seedlings encountering water-limited environments, suggesting a critical role of AA as a defense against oxidative stress in plants.

Recovery of Lactic Acid from Fermentation Broth by the Two-Stage Process of Nanofiltration and Water-Splitting Electrodialysis

  • A two-stage process of nanofiltration and water-splitting electrodialysis was investigated for lactic acid recovery from fermentation broth. In this process, sodium lactate is isolated from fermentation broth in the first stage of nanofiltration by using an NTR-729HF membrane, and then is converted to lactic acid in the second stage by water-splitting electrodialysis. To determine the optimal operating conditions for nanofiltration, the effects of pressure, lactate concentration, pH, and known added impurities were studied. Lactate rejection was less than 5%, magnesium rejection approximated 45%, and calcium rejection was at 40%. In subsequent water-splitting electrodialysis, both the sodium lactate conversion to lactic acid and sodium hydroxide recovery, were about 95%, with a power requirement of $0.9{\sim}1.0\; kWh$ per kg of lactate.

  • There has been many attempts to improve the membrane performance using pervaporation processes[l-3]. They are 1) blending polymer with the high flux and one with high selectivity, 2) an incorporation of functional groups interacting with permeants into a membrane through copolymerization or modification, 3) composite membrane or asymmetric membrane structure with a thin skin layer which acts as a selective layer. Among them, a polymeric membrane containing ion complex group receives an extensive attention recently because ionic complex is known to activate the water transport through ion-dipole interaction. It is especially advantageous in the separation of organic-water system. We applied the ideas of the activation of water transport through ion-dipole. We have reported on the in-sire complex membrane to separate water from aqueous aceiic acid and pyridme solution[4-5] based on the simple acid-base theory. Water transport was enhanced through in-situ complex between pyridine moiety in the membrane and the incoming acetic acid in the feed. In this case, catalytic transport mechanism was proposed. In the present study we used pyridine solution as a feed and the sulfonic acid group in the membrane.

A Study on Leaching Characteristics of Paraffin Waste Form Including Boric Acid

  • Preliminary experiment was peformed to investigate the leaching characteristics of paraffin waste forms that had been recently generated in large quantities at domestic nuclear power plants. At first, waste simulants whose compositions were different in mixing ratio of paraffin to boric acid were prepared. Their compressive strengths were measured and ninety-day leaching test of specimen including cobalt was carried out according to ANSI/ANS-16.1 test procedure. Water immersion test was also conducted keeping pace with leaching test and the weight change and the compressive strength of specimen were observed after ninety days. The compressive strength of waste form exhibited 666 psi (4.53 MPa) in the case where mixing ratio of boric acid to paraffin was 78/22, which was adopted in concentrate waste drying system of domestic nuclear power plants. The leaching test resulted in about 50% of the cumulative fraction leached for boric acid and cobalt, respectively. The specific gravity of waste form was 0.87 [g/g]whose value was less than that of water because the weight loss of about 39% occurred after the water immersion test of ninety days. It was also observed that the waste form which had undergone ninety-day water immersion test exhibited the compressive strength of 203 psi (1.38 MPa).

연부현상이 발생한 감귤로부터 분리한 효모에 대한 유기산의 생육 저해 효과 (Inhibitory Effects of Organic Acids against Pectinolytic Yeasts Isolated from Decayed Citrus)

  • Organic acids are known as natural sanitizers. We examined the sanitizing effects of five organic acids (acetic acid, propionic acid, citric acid, malic acid, and lactic acid) and their persistence on three pectinolytic yeast strains isolated from decayed citrus, and the persistence of their sanitizing effects was determined during storage at $4^{\circ}C$ and $16^{\circ}C$. The 7~8 log CFU/mL of the mixed three yeast mixture was exposed to various concentrations of each organic acid for 1 min. The yeast mixtures decreased under detection limit(1 log CFU/mL) in 1% of acetic acid, followed by in 3% of propionic acid with the reduction of 5 log CFU/mL. The citric acid, malic acid, and lactic acid decreased the number of yeasts under detection limit at 7.5%. When treated with deionized water and 1~5% of organic acids were treated on the surfaces of citrus contaminated by yeasts, total numbers of the yeasts decreased under detection limit(3 log CFU) at 5% of acetic acid and 4 log CFU/piece at 5% propionic acid compared with deionized water. When treated with acetic acid and propionic acid on the stem ends of the contaminated citrus, total numbers of the yeasts significantly decreased 0.5 log CFU/piece at 3% of both organic acids. During storage at $4^{\circ}C$ and $16^{\circ}C$ for 20 days, total number of yeasts significantly decreased at 2% acetic acid compared with deionized water. This study suggested that organic acids could be used to sanitize microbial contaminants from citrus for storage and transportation.

넓미역의 영양성분 및 항산화 활성 (The Nutritional Composition and Antioxidant Activity from Undariopsis peterseniana)

  • The proximate composition, free amino acid composition and mineral contents from Undariopsis peterseniana were determined, and the antioxidant activities of ethanol (EtOH) and hot water extracts of U. peterseniana were investigated using DPPH (1,1-diphenyl-1-picrylhydrazyl), ABTS (2,2'- azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)) radical scavenging effects and reducing power. The contents of moisture, ash, protein, lipid and carbohydrate were 12.5%, 23.1%, 9.7%, 0.2% and 54.5%, respectively, and alginic acid content was 12.3%. The major free amino acid contents were alanine, phenylalanine, aminoethanol, valine, glutamic acid and phosphoserine. Ca (1589.1 mg) was the largest mineral followed by Na (344.6 mg), Mg (74.3 mg), Zn (10.2 mg) and Fe (1.5 mg). The total phenolic contents of EtOH and hot water extracts were exhibited at 15.7 and 4.3 mg GAE/g sample, respectively. The antioxidant activity of EtOH extract exhibited strong ABTS and DPPH radical scavenging activities with reducing power, and hot water extract also demonstrated strong ABTS radical scavenging effects. Therefore, the results of this study suggest that U. peterseniana contained an abundance of naturally occurring nutrients (free amino acids and minerals), and the strong antioxidant activities of EtOH and hot water extracts from U. peterseniana could be good sources of natural antioxidants for healthcare products.
