Sulfonylurea Herbicide Resistance Mechanism of Some Acetohydroxy Acid Synthase Mutants and New Designed Herbicides Specific to the Mutants

  • The mutation rate of proline in the position 197 (Pro197) in acetohydroxy acid synthase (AHAS) is highest among sulfonylurea (SU) herbicide-resistance mutants. Therefore, it is significant to investigate the resistance mechanism for the mutation and to develop the herbicides specific to the mutants. SU herbicide resistance mechanism of the mutants, 197Ser, 197Thr and 197Ala, in AHAS were targeted for designing new SU-herbicide. We did molecular dynamics (MD) simulation for understanding SU herbicide-resistance mechanisms of AHAS mutants and designed new herbicides with docking and MD evaluations. We have found that mutation to 197Ala and 197Ser enlarged the entrance of the active site, while 197Thr contracted. Map of the root mean square derivation (RMSD) and radius gyrations (Rg) revealed the domain indicating the conformations for herbicide resistant. Based on the enlarging-contracting mechanism of active site entrance, we designed new herbicides with substitution at the heterocyclic moiety of a SU herbicide for the complementary binding to the changed active site entrances of mutants, and designed new herbicides. We confirmed that our screened new herbicides bonded to both AHAS wild type and mutants with higher affinity, showing more stable binding conformation than the existing herbicides.

Cyclic Dipeptides from Bacillus vallismortis BS07 Require Key Components of Plant Immunity to Induce Disease Resistance in Arabidopsis against Pseudomonas Infection

  • Cyclic dipeptides (CDPs) are one of the simplest compounds produced by living organisms. Plant-growth promoting rhizobacteria (PGPRs) also produce CDPs that can induce disease resistance. Bacillus vallismortis strain BS07 producing various CDPs has been evaluated as a potential biocontrol agent against multiple plant pathogens in chili pepper. However, plant signal pathway triggered by CDPs has not been fully elucidated yet. Here we introduce four CDPs, cyclo(Gly-L-Pro) previously identified from Aspergillus sp., and cyclo(L-Ala-L-Ile), cyclo(L-Ala-L-Leu), and cyclo(L-Leu-L-Pro) identified from B. vallismortis BS07, which induce disease resistance in Arabidopsis against Pseudomonas syringae infection. The CDPs do not directly inhibit fungal and oomycete growth in vitro. These CDPs require PHYTOALEXIN DEFICIENT4, SALICYLIC ACID INDUCTION DEFICIENT2, and NONEXPRESSOR OF PATHOGENESIS-RELATED PROTEINS1 important for salicylic acid-dependent defense to induce resistance. On the other hand, regulators involved in jasmonate-dependent event, such as ETHYLENE RECEPTOR1, JASMONATE RESPONSE1, and JASMONATE INSENSITIVE1, are necessary to the CDP-induced resistance. Furthermore, treatment of these CDPs primes Arabidopsis plants to rapidly express PATHOGENESIS-RELATED PROTEIN4 at early infection phase. Taken together, we propose that these CDPs from PGPR strains accelerate activation of jasmonate-related signaling pathway during infection.

Peptide Nucleic Acid Probe-Based Analysis as a New Detection Method for Clarithromycin Resistance in Helicobacter pylori

  • Background/Aims: Helicobacter pylori eradication rates are decreasing because of increases in clarithromycin resistance. Thus, finding an easy and accurate method of detecting clarithromycin resistance is important. Methods: We evaluated 70 H. pylori isolates from Korean patients. Dual-labeled peptide nucleic acid (PNA) probes were designed to detect resistance associated with point mutations in 23S ribosomal ribonucleic acid gene domain V (A2142G, A2143G, and T2182C). Data were analyzed by probe-based fluorescence melting curve analysis based on probe-target dissociation temperatures and compared with Sanger sequencing. Results: Among 70 H. pylori isolates, 0, 16, and 58 isolates contained A2142G, A2143G, and T2182C mutations, respectively. PNA probe-based analysis exhibited 100.0% positive predictive values for A2142G and A2143G and a 98.3% positive predictive value for T2182C. PNA probe-based analysis results correlated with 98.6% of Sanger sequencing results (${\kappa}$-value=0.990; standard error, 0.010). Conclusions: H. pylori clarithromycin resistance can be easily and accurately assessed by dual-labeled PNA probe-based melting curve analysis if probes are used based on the appropriate resistance-related mutations. This method is fast, simple, accurate, and adaptable for clinical samples. It may help clinicians choose a precise eradication regimen.

  • The sulphation is main deterioration of lead-acid batter, then there were many kinds of attempts to improve these problems and one of them is to add additives to the electrolyte. In this study some mixed additives such as silicate & boric acid and silicate & phosphoric acid were added to 38% $H_{2}SO_{4}$ electrolyte of lead acid battery to improve electrochemical properties of corrosion resistance, etc. of anode and cathode of lead acid battery.

  • 고기능성 폴리머 콘크리트 복합재료의 내구성을 조사하기 위하여 올소타입 불포화폴리에스테르 수지와 이소타입 불포화폴리에스테르 수지를 폴리머 결합재로 사용하였고, 탄산칼슘과 실리카 미분말을 충전재로 사용하여 공시체를 제작하고 내열수성, 내약품성, 세공분석 및 SEM 조사를 실시하였다. 이소타입 불포화폴리에스테르 수지를 사용한 공시체의 압축강도가 올소타입 불포화폴리에스테스 수지를 사용한 공시체의 압축강도보다 높은 것으로 나타났고, 탄산칼슘 충전재에 비하여 실리카 미분말을 사용한 공시체의 압축강도가 높게 나타났다. 내열수성 시험에서 이소타입 불포화폴리에스테르 수지를 사용한 공시체가 올소타입 불포화폴리에스테르 수지를 사용한 공시체보다 내열수성이 우수한 것으로 나타났고, 탄산칼슘 충전재를 사용한 공시체가 실리카 미분말을 사용한 공시체보다 내열수성이 우수한 것으로 나타났다. 내약품성 시험 후에 측정한 압축강도 감소율은 수산화나트륨 수용액에 의한 압축강도 감소율이 가장 크게 나타났고 다음으로 황산, 염산 및 염화칼슘 순으로 나타났다. 알칼리성인 수산화나트륨 수용액에서는 탄산칼슘을 충전재로 사용한 공시체가 실리카 미분말을 사용한 공시체보다 중량 감소율이 적게 나타났으나 황산과 염산의 산성시약에서는 실리카 미분말을 사용한 공시체가 탄산칼슘을 사용한 공시체보다 중량 감소율이 적게 나타났다.

Fluoroquinolone Resistance and gyrA and parC Mutations of Escherichia coli Isolated from Chicken

  • Escherichia coli is a common inhabitant of the intestinal tracts of animals and humans. The intestines of animals also represent an ideal environment for the selection and transfer of antimicrobial resistance genes. The aim of this study was to investigate the resistance of E. coli isolated from chicken fecal samples to fluoroquinolones and to analyze the characterization of mutations in its gyrA and parC gene related resistance. One hundred and twenty-eight E. coil isolates showed a high resistance to ciprofloxacin (CIP; $60.2\%$), enrofloxacin (ENO; $73.4\%$) and norfloxacin (NOR; $60.2\%$). Missense mutation in gyrA was only found in the amino acid codons of Ser-83 or Asp-87. A high percentage of isolates ($60.2\%$) showed mutations at both amino acid codons. Missense mutation in parC was found in the amino acid codon of Ser-80 or Glu-84, and seven isolates showed mutations at both amino acid codons. Isolates with a single mutation in gyrA showed minimal inhibitory concentrations (MIC) for CIP (${\le}0.5\;to\;0.75{\mu}g/ml$), ENO (1 to $4{\mu}g/ml$) and NOR (0.75 to $4{\mu}g/ml$). These MIC were level compared to isolates with two mutations, one in gyrA and one in parC, and three mutations, one in gyrA and two in parC (CIP, ${\le}0.5\;to\;3{\mu}g/ml;\;ENO,\;2\;to\;32<{\mu}g/ml;\;NOR,\;1.5\;to\;6\;{\mu}g/ml$). However, the isolates with two mutation in gyrA regardless of whether there was a mutation in parC showed high MIC for the three fluoroquinolones (CIP, 0.75 to $32{\le}{\mu}g/ml;\;ENO,\;3\;to\;32{\le}{\mu}g/ml;\;NOR,\;3\;to\;32{\le}{\mu}g/ml$). Interestingly, although the E. coil used in this study was isolated from normal flora of chicken, not clinical specimens, a high percentage of isolates showed resistance to fluoroquinolones and possessed mutations at gyrA and parC associated with fluoroquinolone resistance.

Effects of Salicylic Acid and Indole Acetic Acid Exogenous Applications on Induction of Faba Bean Resistance against Orobanche crenata

  • The parasitic weed, Orobanche crenata, is one of the most devastating constraint for faba bean production in Mediterranean regions. Plant host defense induction was reported as one of the most appropriate control methods in many crops. The aim of this study was to elucidate the effect of salicylic acid (SA) and indole acetic acid (IAA) on the induction of faba bean resistance to O. crenata under the field and controlled experimental conditions. Both hormones were tested on two contrasting faba bean genotypes: Giza 843 (partially resistant to O. crenata) and Lobab (susceptible) at three different application methods (seed soaking, foliar spray, and the combination of both seed soaking and foliar spray). Soaking seeds in SA or IAA provided the highest protection levels reaching ~75% compared to the untreated control plants. Both elicitors limited the chlorophyll content decrease caused by O. crenata infestation and increased phenolic compound production in host plants. Phenylalanine ammonia lyase, peroxidase, and polyphenol oxidase activities were stimulated in the host plant roots especially in the susceptible genotype Lobab. The magnitude of induction was more obvious in infested than in non-infested plants. Histological study revealed that both SA and IAA decreased the number of attached O. crenata spikes which could be related to specific defense responses in the host plant roots.

산부식과 CO2 및 Nd:YAG 레이저 조사에 따른 상아질 표면의 변화 (Effects of Acid-etching, CO2 and Nd:YAG Lasing on the Dentin Surface)

  • The purpose of this study was to examine dentin surface changes of extracted sound third molar specimens which were etched with 10% maleic acid and irradiated at $7-140J/cm^2$ with $CO_2$ and at $156-280J/cm^2$ with Nd:YAG laser. The results were as follows. 1. Dentin surfaces etched with 10% maleic acid and then irradiated at below of $42J/cm^2$ with $CO_2$ laser showed the retentive morphology for resin restoration. 2. Dentin surfaces irradiated at below of $42J/cm^2$ with $CO_2$ laser showed the increased acid-resistance. 3. Dentin surfaces irradiated at $218-280J/cm^2$ with Nd:YAG laser showed the retentive morphology. 4. Dentin surfaces irradiated at $218-280J/cm^2$ with Nd:YAG laser and etched 10% maleic acid and then $218-280J/cm^2$ with Nd:YAG laser showed the increased acid-resistance.

Application of Jasmonic Acid Followed by Salicylic Acid Inhibits Cucumber mosaic virus Replication

  • Systemic acquired resistance is a form of inducible resistance that is triggered in systemic healthy tissues of local-infected plants. Several candidate signaling molecules emerged in the past two years, including the methylated derivatives of well-known defense hormones salicylic acid (SA) and jasmonic acid (JA). In our present study, the symptom on Cucumber mosaic virus (CMV) infected Arabidopsis leaves in 0.1 mM SA or 0.06 mM JA pre-treated plants was lighter (less reactive oxygen species accumulation and less oxidative damages) than that of the control group. JA followed by SA (JA${\rightarrow}$SA) had the highest inhibitory efficiency to CMV replication, higher than JA and SA simultaneous co-pretreatment (JA+SA), and higher than a JA or a SA single pretreatment. The crosstalk between the two hormones was further investigated at the transcriptional levels of pathogenesis-related genes. The time-course measurement showed JA might play a more important role in the interaction between JA and SA.

Reliability of the Impregnated Boron Compounds, Citric Acid- and Heat-Treated Samama (Anthocephalus macrophyllus) Wood against the Fungal and Termite Attacks

  • This research aimed to evaluate the durability of Samama (Anthocephalus macrophyllus) wood treated with boron preservatives, citric acid (CA), and heating against termites. Wood samples were impregnated firstly with 5% boron solutions, such as boric acid, borax and boric acid + borax combination at 1:1 (w/w). The second impregnation used 5% CA. The impregnations were conducted in a pressure tank at 7 kg/cm2 for 4 hours. After impregnation, the samples were heat treated at 80℃ or 160℃. All the treated and control samples were exposed to decay fungi, drywood termites and subterranean termites based on SNI 7207:2014 standard. The results showed that boron preservatives reduced fungal attacks on Samama wood. The combination treatment of boric acid, CA and heat treatment at 160℃ was also effective to increase the resistance of Samama wood against white- and brown rot fungi, and drywood termites. Heat treatment consistently improved the resistance of Samama wood from decay fungi.
