한약재 물 추출물의 생리활성 검색 및 MPTP-유도 신경독성에 대한 대황의 보호효과 (Screening of the Biologoical Activity from Water Extracts of the Medicinal Plants and the Protective Effect of R. palmatum on MTPT-induced Neurotoxicity)

  • This present study was designed to screen medicinal plants for the treatment of brain diseases such as Parkinson's disease or aging. We tested the effects of the water extracts from 38 species medicinal plants on antioxidant capacity, monoamine oxidase B (MAO-B) inhibitory activity, acetylcholinesterase (AChE) inhibition and antiperoxidation activity in vitro. The water extracts from 38 species were tested on their antioxidant activity using radical scavenging effects against ABTS+. The water extract of C. sappan was showed the highest antioxidant capacity, the antioxidant activity at 1 Jig of herbal extract being 0.38mM TE. Lipid peroxidation in brain homogenates induced by NADPH and ADP-Fe/sup 2+/ was strong inhibited by C. sappan and R. palmatum extracts. Among the 38 medicinal plants investigated, R. palmatum showed significant biological activity (antioxidant capacity, MAO-B inhibiory activity, and AChE inhibitory activity). The protective efficacy of R. palmatum water extract on 1-methyl-4­phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced parkinsonism and its possible mechanism were studied in C57BL/6 mice. Treatment of R. palmatum water extract protected biomacromolecules such as lipids from oxidative damage induced by MPTP. The content of MDA in brain tissue was decreased significantly by R. palmatum extract. These results suggest that R. palmatum water extract plays on effective role in attenuating MPTP-induced neurotoxicity in mice. This protective effect of R. palmatum might be estimated the result from the inhibitory activity on monoamine oxidase B and the enhancement of antioxidant activity.

효소적 가수분해를 이용한 청각으로부터 생리활성 물질의 추출 및 가수분해물의 생리활성 (Biological Compounds Extracted from Codium fragile by Enzymatic Hydrolysis and Their Biological Activities)

  • 본 연구에서는 청각으로부터 생리활성물질을 추출하기 위해서 친환경적인 효소적 방법을 이용하여, 이들 효소 가수 분해물의 TPC, TFC, 항산화 활성, acetylcholinesterase(AChE) 저해 활성 및 항염증 활성을 측정하였다. 청각의 단백질 및 탄수화물 가수분해물의 TPC는 TFC보다 높은 함량을 나타내었으며, 단백질 가수분해물이 탄수화물 가수분해물보다 높은 TPC를 나타내었다. 청각 가수분해물의 항산화 활성은 DPPH radical 소거 활성으로 측정하였고, 단백질 가수분해물에서는 Flavourzyme 가수분해물이 활성이 높았고, 탄수화물 가수분해물에서는 Promozyme 가수분해물이 높은 DPPH radical 소거 활성을 나타내었다. 따라서 두가지 가수분해물을 이용하여 hydrogen peroxide 소거능, $Fe^{2+}$ 킬레이팅 및 reducing power를 측정한 결과 두 가수분해물 모두 우수한 항산화 활성을 나타냄을 알 수 있었다. 또한, hydroxyl radical로 유도된 DNA 손상을 효과적으로 억제하였다. AChE 저해 활성에서는 Flavourzyme 및 Dextrozyme 가수분해물이 각각 우수한 AChE 저해 활성을 나타내었다. 청각 가수분해물의 RAW264.7 세포주에 대한 세포독성을 검토한 결과 세포독성을 나타내지 않았으며, lipopolysaccharide(LPS)로 유도된 nitric oxide(NO) 생성 억제능에서는 모든 가수분해물이 유의적(p<0.05)으로 NO의 생성을 억제하였다.

살충제 Carbofuran과 Phenobarbital Sodium 및 3-Methylcholanthrene이 이스라엘 잉어의 효소활성에 미치는 영향 (Effect of Insecticide Carbofuran and Phenobarbital Sodium and 3-Methylcholanthrene on Activity of Enzyme in Israeli Carp(Cyprinus israeli carpio L.))

  • 이스라엘 잉어에 있어서 carbamate계 살충제 carbofuran의 독성에 미치는 phenobarbital sodium(PB) 또는 3-methylcholanthrene (3-MC)의 영향과 작용기작을 효소적 측면에서 구명할 목적으로 carbofuran과 PB나 3-MC를 이스라엘 잉어에 각 조합으로 처리하여 독성경감 효과를 조사하였고, 공시한 농약과 PB나 MC가 acetylcholinesterase(AChE), glutathione S-transferase(GST), UDP-glucuronosyltransferase(UDPGT) 및 cytochrome P-450-dependent monooxygenase(monooxygenase)의 효소활성에 미치는 영향을 조사하기 위하여 carbofuran과 PB나 3-MC를 각각 조합으로 처리한 후 경시적으로 이스라엘 잉어의 각 효소들의 상대활성도를 조사하였다. PB와 3-MC만 투여한 실험군에서 이스라엘 잉어의 생존수는 무처리군과 동일하였고 살충제만 처리한 실험군의 이스라엘 잉어 생존수는 처리농도가 증가하면서 감소되었으나, PB나 3-MC와 살충제를 조합처리한 실험군에서는 살충제만 처리한 실힘군에 비하여 매우 높은 생존율을 나타낸 것으로 보아 해독효과가 인정 되었다. 효소활성(in vivo)은 AChE의 경우 carbofuran 0.95 ppm만을 처리한 실험군에서는 24시간내내 각 조사시기마다 무처리군에 비해 40% 이상의 활성저해를 보였으나 carbofuran과 PB 및 3-MC를 조합처리한 실험군에서는 효소활성이 초기에 감소하다가 서서히 증가하여 24시간후에는 무처리군과 비슷한 수준을 나타냈고, GST의 경우 carbofuran만을 처리한 실험군에서는 초기에 약 20% 이상의 활성저해를 보였으나 carbofuran과 PB나 3-MC를 조합처리한 실험군에서는 약제처리 1시간 후 부터 무처리군에 비해 효소활성이 20% 이상 증가하였다. UDPGT와 monooxygenase의 효소활성은 carbofuran과 PB나 3-MC를 조합처리한 실험에서 처리 $6{\sim}12$시간 후에는 carbofuran 처리군에 비해 효소활성이 $4{\sim}8$배 이상 급격히 높아졌다. 이상의 결과에서 PB 및 3-MC처리가 이들 효소의 활성을 유도함으로써 carbofuran의 독성으로 부터 이스라엘 잉어를 보호 하는데 관여한 것으로 보인다.

산화적 스트레스와 관련하여 신경세포의 활성에 미치는 강황 열수추출물의 영향에 대한 연구 (Effect of Curcuma longa Hot Water Extract on Activity of Neuronal Cells Related to Oxidative Stress)

  • 본 연구의 목적은 인지기능을 가진 약효제를 발굴하는 것이다. 본 연구에서는 토끼의 간과 신경세포로부터 각각 유래한 angiotension converting enzyme과 acetylcholinesterase에 대한 강황 열수추출물(CLWE)의 억제효과 뿐만 아니라 항산화 효과가 조사되었다. 이리하여 먼저 산화적 스트레스와 연관이 있는 환원력과 DPPH radical, superoxide anion, hydroxyl radical, lipid peroxidation에 대한 CLEW의 소거능 및 DNA 산화에 대한 보호효과를 평가하였다. CLEW는 환원력 뿐만 아니라 본 연구에서 시험된 활성산소종 중에서 특히 superoxide anion의 소거능이 가장 우수한 것으로 나타났다. 더욱이 CLEW는 0.25% 이상의 농도에서 angiotensin converting enzyme 활성을 억제효과를 발휘하는 것으로 나타났다. 신경세포에서 CLEW는 또한 산화적 스트레스와 nitric oxide 유발을 억제하였다. 그러므로 이상의 결과는 CLEW의 항산화 효과와 신경세포 보호효과를 입증하여, 이것이 사람의 신경건강을 위한 천연생물소재로 잠재적인 가능성을 가지고 있을 것으로 시사한다.

황련해독탕 전탕액과 약침액이 치매유발생쥐의 인지와 기억에 미치는 영향 (Effects of Hwangryunhaedok-Tang Decoction & Hwangryunhaedok-Tang Pharmacopuncture Solution on the Cognitive & Memory Impairment Induced by Scopolamine in Mouse Model)

  • Objectives : This research was performed to investigate the effects of Hwangryunhaedok-tang decoction and Hwangryunhaedok-tang Pharmacopuncture at BL10 on cognition and memory impairment in a mouse dementia model induced by scopolamine. Methods : Fifty ICR mice were divided into 6 groups : Normal group (n=5), Control group (n=9), Positive control group for pharmacopuncture group (n=9, Donepezil 0.75 mg/kg), Positive control group for oral administration group (n=9, Donepezil 5 mg/kg), Pharmacopuncture group (n=9, Hwangryunhaedok-tang Pharmacopuncture undiluted solution 10 ml/kg), and Oral administration group (n=9, Hwangryunhaedok-tang 200 mg/kg). For a mouse dementia model, 1 mg/kg scopolamine was intraperitoneally administered to ICR mice. Hwangryunhaedok-tang pharmacopuncture was administered on BL10 for 4 weeks at intervals of 2 days. Hwangryunhaedok-tang decoction was given orally for 4 weeks every day. Morris water maze and passive avoidance test were conducted followed by measurement of acetylcholine concentration, acetylcholinesterase activity, and the amount of BDNF and p-CREB in the brain. Results : 1. In the Morris water maze test, the time spent staying around the platform significantly increased in the pharmacopuncture group and oral administration group than in the control group. 2. In the passive avoidance test, the time spent in the bright room significantly increased in the pharmacopuncture group and oral administration group than in the control group. 3. The level of acetylcholine in brains increased in the pharmacopuncture group and oral administration group than in the control group. Also, the activity of acetylcholinesterase decreased in the pharmacopuncture group and oral administration group than in the control group. 4. The expression of BDNF and p-CREB decreased in the control group, but increased in the pharmacopuncture group and oral administration group. Conclusions : These results suggest that Hwangryunhaedok-tang decoction and Hwangryunhaedok-tang pharmacopuncture at BL10 may have cognition and memory-enhancing effect in scopolamine-induced dementia in ICR mice via controlling the content of acetylcholine and the activity of acetylcholinesterase, and activating BDNF and p-CREB.

살충성(殺蟲性) O,O-Diethylphenylphosphate유도체(誘導體)들에 의(依)한 Acetylcholinesterase의 Phosphorylation에 미치는 자유(自由)에너지 관계(關係) (Linear Free Energy Relationship on the Phosphorylation of Acetylcholinesterase by Insecticidal O,O-Diethylphenylphosphate Derivatives)

  • 살충성 O,O-diethylphenylphosphate (A)와 3,5-dimethylphenyl-O,O-diethylphosphate 유도체 (B)에 의한 acetylcholinesterase(Ach E)의 phosphorylation 반응에 미치는 정량적인 자유에너지 관계(LFER)를 알아보기 위하여 Ach E의 두 작용점인 esteratic-anionic site에 대응하는 (A) 및 (B)의 P원자 및 meta 위치간의 공간직선 거리 및 알짜전하를 확장 H$\ddot{u}$ckel이론(EHT)에 의한 MO 계산 방법으로 계산하고 여러 가지 치환기상수와 $pI_{50}$ 상수를 Hammett 및 Okamoto-Brown식, Taft와 Swaln-Lupton의 DSP식에 적용한 회귀분석 결과로부터 살충작용에 미치는 정량적인 자유에너지 관계, %R과 %F를 계산하여 검토하였다. 즉, (A) 및 (B)의 살충효과는 서로 비슷하였으며 blending coefficient (${\lambda}$)는 모두 ${\lambda}$<1의 값을 나타내었다. (A) 및 (B)의 resonance effect (%R)와 field effect (%F)는 각각 %R=33.5, %F=66.5 그리고 %R=2, %F=98로 각각 계산되었는데(B)와 같이 phenyl group에 bulky한 소수성의 methyl group이 결합될수록 resonance effect($R_{\pi}$)보다는 direct through space field effect ($F_D$)가 아닌 through space field effect가 더 크게 영향을 미치게 된다는 사실을 알 수 있었다.

흰쥐의 생리활성에 미치는 송엽(松葉) 추출물(PNE)의 영향 III. 뇌세포막의 유동성 및 신경전달관련 효소의 활성에 미치는 PNE의 투여효과 (Effect of Pine Needle Extract (PNE) on Physilolgical Activity of SD Rats III. Feeding Effect of PNE on Fluidity and Neurotransmitter-Related Enztmes in Brain Membranes of SD Rats)

  • To investigate the effect of pine needle extract (PNE) on membrane fluidirt and neurotransmiter-related enzymes in brain of Spragu-Dawley(SD), male SD rats were fed basic diets (control group), and experimantal diets (PNE group)with 0.5% and 0.1% fo PNE for 6 weeks. pine (pinus tabulaeformis C$_{ARR}$ is one of the popular plant drugs which has used as a medicine in Asia. Cholesterol levels in brain mitochondria of 0.5%-PNE and 0.1%-PNDE groups were significantly decreased in 15% and 25%, respectively, compared with control group, but cholesterol levels in brain microsomes of these PNE groups howed almost no change compared with control group. Lipofuscin accumulations in brain membranes of 0.5%-PNE and 0.1%-PNE groups were sgnificantly inhibited in 18% and 21%, respectively, compared with control group. Brain memberance fluidity was also activated in 50% and 100% by the administration of 0.5%-PNE and 0.1%-PNE. higher acetylcholinesterase(15% and25%) and lower monoamine oxidase B (25% and 15%0 activities were effectively modulated by the administration of 0.5%-PNE and 0.1%-PNDE. These results suggest that more beneficial effects such as inhibition of cholesterol and lipofuscin, increase of membrane fluidity, higher acetylcholinesterase and lower monoamone oxidase activities in brain membranes of SD rats may be effectively modulated by administration of pine needle extract (PNE).

치매동물모델 SAMP8에 있어서 기억. 학습장해에 미치는 알로에의 영향 III. SAMP8의 신경전달물질 및 그 대사산물에 미치는 알로에의 투여효과 (Effect of Aloe on Learming and Memory lmpaiments in Dementia Animal Model SAMP8)

  • Aloe(Aloe arborescens M$_{ILL}$) has been used as a home medicine for the past several thousand in the world, and has been studied on anti-bacterial and anti-fungal activities, hypotension, atherosclerosis, myocardiac infartion, apoplexy, diabetes as a chronic digenerative disease, tumors, gastrointestinal tract, liver and pancreas' diseases, and genitourinary tract etc. SAMP8 as a learing and memory impairment animal model were fed basic and/or experimental diets with 1.0% freezing dried(FD)-aloe for 8 months. The passive avoidance tests such as acqusition trial and retention test were significantly higher in aloe group than in control group. Grading score of senescence resulted in a marked decreases in aloe group compared with control group. Acetylcholinesterase(AChE) activity was remarkably increased in aloe group compared with control group. Neurotransmitters such as dopamine(DA) and serotonin(5-HT) almost did not change by the feeding of aloe-added diet, but their metabolites such as homovanillic acid(HVA) and 5-hydroxy-indole acetic acid(5-HIAA) in aloe group were significantly increased compared with control group. Therefore, the ratios of HVA/DA and 5-HIAA/5-HT as a ratio of metabolite on neurotransmitter were significantly increased by the feeding of aloe-added diet. These results suggest that aloe vara may be activated acetylcholinesterase, the metabolite of neurotransmitter, and ratios of metabolite on neurotransmitter, resulting ina greater prevention of learning and memory impairments such as Alzheimertype dementia.

Biruloquinone, an Acetylcholinesterase Inhibitor Produced by Lichen-Forming Fungus Cladonia macilenta

  • At present, acetylcholinesterase (AChE) inhibitors are the first group of drugs to treat mild to moderate Alzheimer's disease (AD). Although beneficial in improving cognitive and behavioral symptoms, the effectiveness of AChE inhibitors has been questioned since they do not delay or prevent neurodegeneration in AD patients. Therefore, in the present study, in order to develop new and effective anti-AD agents from lichen products, both the AChE inhibitory and the neuroprotective effects were evaluated. The AChE inhibitory assay was performed based on Ellman's reaction, and the neuroprotective effect was evaluated by using the MTT method on injured PC12 cells. One AChE inhibitor ($IC_{50}$ = 27.1 ${\mu}g/ml$) was isolated by means of bioactivity-guided isolation from the extract of lichen-forming fungus Cladonia macilenta, which showed the most potent AChE inhibitory activity in previous screening experiment. It was then identified as biruloquinone by MS, and $^1H$- and $^{13}C$-NMR analyses. The inhibitory kinetic assay suggested that biruloquinone is a mixed-II inhibitor on AChE. Meanwhile, biruloquinone improved the viability of the $H_2O_2$- and ${\beta}$-amyloid-injured PC12 cells at 1 to 25 ${\mu}g/ml$. The protective effects are proposed to be related to the potent antioxidant activities of biruloquinone. These results imply that biruloquinone has the potential to be developed as a multifunctional anti- AD agent.
