  • Ether-soluble flavonoid and neutral fraction (Fr. C) prepared from EtOAc extract of Scutellariae Radix showed inhibitory activities against bradykinin (BK) ; inhibited BK-induced ileum or uterus contractions, antagonized BK-induced plasma extravasation and protected mice from endotoxic shock, reduced acetic acid-induced writhing in mice.

葛根湯과 加味葛根湯의 抗알레르기 및 消炎, 解熱, 鎭痛作用에 대한 實驗的 硏究 (An Experimental study on the anti-allergic effects, anti-inflammatory action, anti-pyretic action and analgesic action of Galgeun-tang, Gamigalgeun-tang and Geomahwanggalgeun-tang)

  • Allergic rhinitis and allergic dermatitis are common allergic diseases. Galgeun-tang has been used for treating various diseases, which include allergic rhinitis and allergic dermatitis. Experimental sutdies have been done to research the anti-allergic effects of Galgeun-tang, Gamigalgeun-tang and Geomahwanggalgeun-tang. We have observed: the vascular permeability response induced by serotonin and histamine, the contact dermatitis response induced by picryl chloride, the delayed type hypersensitivity response to SRC, the mice paw edema induced by carrageenin, and the rectal temperature in febrile rats induced by endotoxin, as well as the writhing syndrome induced by $0.7{\%}$ acetic acid. The results were as follows: 1. In the vascular permeability response to intradermal serotonin and histamine, Galgeun-tang and Gamigalgeun-tang showed a significant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect. 2. In the contact dermatitis response induced by picryl chloride, Galgeun-tang and Gamigalgeun-tang showed a signigicant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect. 3. In the delayed type hypersensitivity response to SRC, Galgeun-tang, Gamigalgeun-tang and Geomahwanggalgeun-tang showed a significant inhibitory effect. 4. In the mice paw edema induced by carrageenin, Galgeun-tang and Gamigalgeun-tang showed a significant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect. 5. In the rectal temperature in febrile rats induced by endotoxin, Galgeun-tang, Gamigalgeun-tang and Geomahwanggalgeun-tang showed a significant inhibitory effect. 6. In the writhing syndrome induced by a $0.7{\%}$ acetic acid solution. Galgeun-tang and Gamigalgeun-tang showed a significant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect.

산풍단(散風丹)이 생쥐의 항경련(抗痙攣), 진정(鎭靜) 및 진통작용(鎭通作用)에 미치는 영향(影響) (A study on the anti-convulsive, sedative and analgesic effects of Sanpoongdan in mouse)

  • Sanpoongdan(SPD) has been known effective on infantile convulsive disorders in clinical field of oriental medicine. The purpose of this study was to investigate the anti-convulsive, sedation and analgesic effects of SPD in mouse. The anticonvulsive effect was evaluated In mice treated with pentylenetetrazol, stychnine, and picrotoxin. For the sedative effect, observations were made on the sleeping time induced by thiopental sodium and pentobarbital sodium following oral administration of SPD. Furthermore, reduction of spontaneous movements and ataxia using rota rod method were evaluated. Analgesic effects on the writhing syndrome induced by acetic acid and on hindlimb pain induced by pressure were also observed. The findings were as follows : 1. The solid extracts of SPD revealed no effect on convulsions induced by pentylenetetrazol, strychnine, and picrotoxin. 2. Thiopental sodium-induced sleeping time was prolonged by the administration of the solid extracts of SPD, but this result was devoid of statistical significance. 3. The oral administration of SPD enhanced the sleeping induced by pentobarbital sodium. 4. Spontaneous movements were significantly depressed following the oral administration of the solid extracts of SPD. 5. Ataxia was not shown in rota rod method following the oral administration of the solid extracts of SPD. 6. The solid extracts of SPD showed positive analgesic effects on the acetic acid-induced writhing syndrome. 7. The solid extracts of SPD raised the threshold of the hindlimb pressure pain, but the result was not statistically significant. From the results, it can be concluded that SPD has sedative and analgesic effects.

Analgesic and Antipyretic Activity of Stachys schimperi Vatke

  • The analgesic and the antipyretic activity of the methanol, chloroform, hexane and acetonitrile extracts of the aerial parts of Stachys schimperi Vatke were investigated in mice. The nociceptive response was tested using acetic acid-induced writhing and tail flick method; while hypothermia affect was examined via yeast-induced fever test. The chloroform extract at 500 mg and hexane and acetonitrile extracts at 250 mg produced significant analgesic and antipyretic activity.

전남지역 자생식물들의 소염활성검색 및 활성화합물 (Screening of Medicinal Plants on the Anti-inflammatory Effect and Active Component)

  • The anti-inflammatory effect of the methanolic extracts of some medicinal plants was investigated on the hydrolysis of N-benzoylarginine-p-nitroanilide by trypsin in vitro, the writhing syndrome induced by acetic acid in mice, and the carrageenin-induced paw edema in rats. Among tested medicinal plants, the methanoilc extract of Angelica keiskei, Rosa rugosa and Cudrania tricuspidata showed significant inhibitory effect. And we also found that adenosine, isolated from A. keiskei was identified as one active components.

乾癬에 活用되는 加味當歸飮子에 對한 實驗的 硏究 (Research of Experimental Kamidangkwieumja in Psoriasis)

  • The purpose of this research was to investigate the effect of Kamidangkwieumja(KDEJ) water extract on the allergy reaction in mice. The results were obtained as follows: 1. The passive cutaneous anaphylaxis induced by egg albumin in fat was not affected. 2. The lethal anaphylaxis induced by platelet activating factor in mice. was inhibited. 3. The degranulation of peritoneal mast cells induced by compound 48/80 was not affected. 4. The acute hind paw edema was inhibited after 2hours later when it was induced by histamine. 5. The permeability of evans blue into peritoneal cavity induced by acetic acid was not affected. 6. Arthus reaction in mice was not affected. 7. The delayed type hypersensitivity induced by SRBC was inhibited. 8. The contact dermatitis induced by DNFB was not affected. 9. The hemagglutination titer induced by SRBC was inhibited. 10. The writhing syndrome induced by acetic acid was inhibited. 11. The population of heper T cells in mice thymus was enhanced. 12. The phagocytic activity of peritoneal macrophages was enhanced. 13. The production of nitric oxide from peritoneal macrophages was not affected. These results suggest that the anti-allergy effect of KDEJ is caused by steroidlike and enhanced immune action. The steroidlike action of KDEJ correspond with steroid-applying-method that frequently used in clinic, so it is used io treatment of psoriasis. The research on anti-allergy of KDEJ might has to be continued.

단삼음(丹蔘飮)의 약리효능에 대한 연구 (Study of Dansameum on the Pharmacological Effect)

  • The purpose of this research was to investigate the pharmacological effects of Dansameum (DSE) water extract which is composed with Salviae Radix, Santali Lignum and Amomi Semen. DSE was administered orally at the concentartion of 100 and 500 mg/kg. DSE decreased the writhing syndrome induced by acetic acid and the permeability of evans blue into peritoneal cavity. Also, DSE inhibited the production of nitric oxide from murine macrophages. Futhermore, DSE protected the liver injury induced by galactosamine in vitro system and $CCl_4$ in vivo system. These results suggest that DSE has a diverse effects such as analgesic action, inflammatory action and liver protective action.

Antinociceptive and antidiarrhoeal activities of Sonneratia caseolaris

  • The crude ethanol extract of leaves of Sonneratia caseolaris Linn. (Sonneratiaceae) was screened for its antinociceptive and antidiarrhoeal activities. The extract produced significant writhing inhibition in acetic acid induced writhing in mice at dose of 250 and 500 mg/kg body weight (P<0.01) comparable to the standard drug diclofenac sodium at the dose of 25 mg/kg of body weight. When tested for its antidiarrhoeal effects on castor oil induced diarrhea in mice, it increased mean latent period and decreased the frequency of defecation significantly at the dose of 500 mg/kg body weight (P < 0.05) comparable to the standard drug loperamide at the dose of 50 mg/kg of body weight. The overall results tend to suggest the antinociceptive and antidiarrhoeal activities of the extract.

Synthesis, Analgesic, and Anti-Inflammatory Activities of [6-(3,5-Dimethyl-4-Chloropyrazole-1-yl)-3(2H)-Pyridazinon-2-yl]Acetamides

  • A series of structurally diverse amide derivatives of [6-(3,5-dimethyl-4- chloro-pyrazole-1-yl)-3(2H)-pyridazinone-2-yl]acetic acid were prepared and tested for their in vivo analgesic and anti-inflammatory activity by using p-benzoquinone-induced writhing test and carrageenan induced hind paw edema model, respectively. The analgesic and anti-inflammatory activity of the compounds, 7c, 7d and 7k were found to be equipotent to aspirin (as an analygesic) and indometacin (as an anti-inflammatory drug), respectively. The other amide derivatives generally resulted in lower activity on comparision with reference compounds.

현호색, 창출, 천수근 약욕이 체성내장통에 미치는 영향 (Effects of Herbal Bath on Acetic Acid-induced Somato-visceral Pain in Mice)

  • As an effective non-pharmacological method of pain relief, hydrotherapy was widely used. And bath additive has been used for enhancing the efficacy of hydrotherapy, In the present study, as a bath additive, the analgesic activity of HAC, which composed of Corydalis turtschaninovii, Atractylodes japonica, and Harpagophytum procumbens(HAC), was investigated in the ventrolateral periaqueductal gray (VIPAG), lateral PAG (IPAG), central nuclei of amygdala (CeA), and the paraventricular nucleus of the hypothalamus (PVN) in mice, using writhing test and immunohistochemistry for c-Fos. Male C57BU6 mice weighing $25{\;}{\pm}{\;}2g$ (8 weeks of age) were used for this experiment. The animals were divided into five groups: the control group, the acetic acid treatment group, the acetic acid treatment and 0.01 g/L HAC-immersed group, the acetic acid treatment and 0.1 g/L HAC-immersed group, and the acetic acid treatment and 1.0 g/L HAC-immersed group. To induce somato-visceral pain in the experimental animals, a single intraperitoneal (i.p.) injection of acetic acid was administrated to each animal, and the animals of the control group received injections of equivalent doses of normal saline. The animals of the HAC-immersed groups were immersed the water with HAC powder at the respective doses deep enough to cover the mice body, and those of the control group and the acetic acid treatment group immersed the water without HAC powder at 10 min immediately after the acetic acid injection. Our present study has shown that the HAC reduced the acetic acid-induced abdominal constrictions and the acetic acid-Induced increase of numbers of c-Fos-positive cells in the VIPAG, IPAG, PVN, and CeA. The most potent analgesic effect appeared with the treatment of 1.0 g/L KB-immersed group. Based on our present results, it is very possible that HAC can be a potent therapeutic bath additive for alleviating pain without the fear of addiction to the drugs and side-effects associated with the prescription of multiple analgesic drugs.
