Studies on the Errors, in the Microdetermination of Carbon and Hydrogen, due to the Positions of Sample. (탄수원소분석에 있어서 검체충전위치에 기인하는 오차에 대한 연구)

  • To reduce the errors, in the microdetermination of carbon and hydrogen, by the electrical automatic combustion apparatus, measured the errors due to the positions of sample in the carbon and hydrogen combustion tube. The samples used are glucose, acetanilide, phenacetin and diazoaminobenzene. Experimented positions of samples were 3cm, 5cm, 7cm, and 9cm from the front edge of the long stationary burner. In this experiment, it was found that the most suitable position of sample in the carbon and hydrogen combustion tube is 8cm from the long stationary burner.

  • Radio iodination of various aromatic derivatives (aniline, toluene, iodobenzene, acetanilide, benzene, benzoic acid) were achieved at low temperature by a chloroamine-T procedures in presence of polar solvent(dioxane). Organic base (piperidine) was used as the catalyst. Iodine replacement reaction had occured on the aromatic or alicyclic ring by this reaction, and the kind and ratio of iodinated products were proved to be different from those of usual iodide reaction in organic solvent at low temperature. The reactivity of various aromatic or alicyclic compounds towards the present iodination system was evaluated and the scope and limitation of the present procedures in the preparation of radio pharmaceuticals were discussed.

Bioconversion of Aniline to Acetaminophen and Overproduction of Acetaminophen by Streptomyces spp.

  • In order to obtain acetaminophen, a popular analgesic-antipyretic, though microbial p-hydroxylation and N-acetylation of aniline, various Streptomyces strains were screened. Aniline N-acetylation activity was rather ubiquitous but-hydroxylation activity was selective. Microbial conversion pathway of aniline to acetaminophen was considered to be through N-acetylation and p-hydroxylation or vice versa. However, depending on species used, o-hydroxylation and its degradation activity (S. fradiae) and acetaminophen degradation activity (S. coelicolar) were also detected. Among the screened Streptomyces strains, S fradiae NRRL 2702 showed the highest acetanilide p-hydroxylation activity (203% conversion rate). Furthermore, in S. fradiae carbon source and its concentration, phosphate ion concentration and pH of growth medium were found to play the crucial roles in p-hydroxylation activity. Through the proper combination of factors mentioned above, the ten times more activity (26-30% conversion rate) was attained.

Investigation of Fungicide Response of Streptomyces spp. Isolated from Rhizosphere in Zoysiagrass (한국 잔디 근권에서 분리된 Streptomyces spp.의 살균제에 대한 반응조사)

  • Streptomyces spp. isolated from turfgrass rhizosphere and tested for their response to large-patch control fungicides. The tested fungicides were actually used in golf course or turfgrass cultivation to prevent large-patch disease. Tolerance to 3 triazole group of the strains was the highest to the PR fungicide, and following the SR fungicide, whereas the isolated strains were no tolerance to HR fungicide. Tolerances to three kind of Strobilurin group were similar for the all of the tested Streptomyces spp.. Growth and sporulation of the all strain was normal in CB and AP fungicide treatments. However no spore formulated in double concentration. Strains, tolerance to acetanilide fungicides, appeared that KT fungicide tolerance was higher than MK fungicide. The selected strains showed strong tolerance against AT fungicide but have no tolerance to ATR fungicides. In conclusion, the bacterial strains showed tolerance against 1 carbamate, 1 organophosphate and 1 cyanopyrrole group, while have no tolerance against two mixture formulations (1 Quinone + Strobilurin and 1 Imidazole + Triazole).

Effects of Alachlor and Endosulfan on the Survival and Malformation of Bombina orientalis Embryos (제초제 Alachlor과 살충제 Endosulfan이 무당개구리 (Bombina orientalis) 배아의 생존 및 기형유발에 미치는 영향)

  • The chloroacetanilide herbicide alachlor (2-chloro-2', 6'-diethyl-N-(methoxymethyl)-acetanilide) and organochlorine insecticide endosulfan (6, 7, 8, 9, 10, 10-hexachloro-l, 5, 5a, 6, 9, 9a-hexahydro-6, 9-methano-2, 3, 4-benzodioxathiepin-3-oxide)are the highly toxic agricultural chemicals. Bombina orientalis is one of the most common amphibians in the world and comprises a large proportion of their total number. B. orientalis spawns in the farming regions at Spring when the massive application of agricultural chemicals occurs. These chemicals in farmland may threaten the reproduction of this frog. Therefore, we examined the embryotoxic and survival effects of alachlor and endosulfan at various concentrations in B. orientalis embryos. The survival rates of embryos at 312h post fertilization treated with alachlor and endosulfan were decreased with concentration dependent manner. Also, developmental malformations appeared by alachlor and endosulfan in B. orientalis embryos. The malformations showed in order of frequency with bent trunk, tail dysplasia, bent tail, thick-set body and ventral blister in alachlor treated embryos. The exposure of endosulfan produced 7 types of severe external malformations with tail dysplasia, pectoral blister, bent trunk, bent tail, cephalic dysplasia, ventral blister, and thick-set body. Following exposure to alachlor and endosulfan the types of malformations were diverse, suggesting these chemicals target multiple events in embryonic and larval development in this species. These results suggested that alachlor and endosulfan were detrimental for survival and development of B. orientalis embryos.

  • In an effort to elucidate degradation mechanisms of an acetanilide herbicide, Butachlor, by soil microorganisms, a common soil fungus, Chaetomium globosum which is known to be powerful was selected and incubated in a Butachlor-contained medium. The results obtained from the resulting metabolites are as follows: (1) Dechlorination from Butachlor occurred very easily, remaining almost constant after 180 hrs. of incubation. (2) More than 10 metabolites were isolated and characterized, of which the metabolites, m/e 205, 177, 223, 182, and 206 were the main products. (3) In this paper, the structures and pathways of formation of metabolites, m/e 206, 182, 223, 225, and 189 were tentatively proposed.

Microbial Transformation of Aniline to Acetaminophen

  • In order to obtain acetaminophen, a popular analgesic-antipyretic, through microbial p-hydroxylation and N-acetylation of aniline, various fungi and bacteria were secreened. Among them, Streptomyces species were chosen for strain improvement by the use of interspecific protoplast fusion technique. Two interspecific fused strains were developed between S. rimosus (N-cetylation function) and S. aureofaciens (p-hydroxylation function) and also between S. lividans and S. globisporus. For efficient protoplast fusion and cell wall regeneration, various conditions were examined. In a typical experiment of mixed S rimosus ($pro^- \;his^-$) and S. aureofaciens ($ilv^-$) protoplasts with 40% (w/v) polythylene glycol 3350 (PEG) for 3 min gave $8.3\times10^{-7}$ of fusion frequency. Treatment of mixed S. lividans (pant-) and S. globisporus (leu-) protoplasts with 50% (w/v) PEG for 3 min at $30^\circ{C}$ gave $1.2\times10^{-6}$ of frequency. Among the fused strains, up to 40-50% increase in p-hydroxylation power was observed. To investigate the possibility of plasmid involvement in p-hydroxylation power was observed. To investigate the possibility of plasmid involvement in p-hydroxylation of acetanilide, plasmid curing was attempted. We found that cells treated with acriflavine (at the frequency of 100%) and cells regenerated from protoplsts of S. auroefaciens (2% frequency) lost their p-hydroxylation function.

  • The effects of alachlor [2-chloro-2', 6' diethyl-N-(methoxymethyl) acetanilide] treatment on nucleic acid, amino acid and protein synthesis were studied. The amide herbicide alachlor blocks the biosynthesis of the amino acids isoleucine, valine and aromatic amino acid in oat root tips. Nucleic acid was inhibited, but was not proportional to reduction in protein synthesis. $1{\times}10^{-4}M$ of alachlor treatment of oat roots inhibited 36% DNA synthesis, but DNA synthesis was not inhibited at $1{\times}10^{-5}M$. RNA synthesis was inhibited by $1{\times}10^{-5}M$ and $1{\times}10^{-4}M$ of alachlor 16 and 27%, respectively, while inhibition of protein synthesis did occur at same concentrations. Inhibition of protein synthesis also did not occur at concentration below $1{\times}10^{-4}M$ alachlor. It suggest that inhibition of protein sythesis caused significantly by alachlor($1{\times}l0^{-3}M$) result from secondary action.

  • Effect of Butachlor(2-chloro-2, 6-diethyl N-(buthoxymethyl) acetanilide), Nitrofen(2,4-dichloro-4-nitrodiphenyl ether), Benthiocarb+Simetryne(s-(4-chlorobenzyl)-N, N-diethylthiocarbamate $7\%$+2-methylthio-4, 6-bis(ethylamino)-s-triazine $1.5\%)$, Propanil (3,4-dichloropropionanilide), and Perfluidone {1. 1. 1-trifluoro-N, N-(2-methyl-4-(phenylsulfonyl) Pheny1) methanesulfon amide} on urea hydrolysis and subsequent nitrification was investigated in a flooded soil incubated at $24\pm1^{\circ}C$ for 9 weeks. 1. Butachlor and Perfluidone at the rate of 1,440 and 1,200g, ai/10a, respectively, slightly inhibited the early stage of urea decomposition, and caused a slight decrease in the production of ammomium, which, however, was recovered readily. 2. Propanil at the rate of 2,800g, ai/10a imhibited the first stage of nitrification, and brought about a slight increase in the ammonium conentration and a decrease in the concentration of nitrite and nitrate. This inhibitive effect was a little more evident at higher concentration of applied nitrogen. The other herbicides caused no inhibition of urea decomposition and subsequent nitrification even at the highest rate of application. 3. pH and Eh of the soil were not significantly affected by the herbicides tested.

Effects of Butachlor on the Growth of PurpleNnon-sulfur Bacteria (홍색 비유황광합성 세균에 미치는 제초제 Butachlor의 영향)

  • The effects of a herbicide butachlor[2-chloro-2', 6'-diethyl-N-(butoxymethyl) acetanilide] on the growth of the purple non-sulfur bacteria were investigated. The butachlor inhibited the growth of all species tested by 18-51%, except Rhodospirillum rubrum at concentrations of M, which would be field capacity. The photosynthetic growth rate of the species in the presence of butachlor was influenced by the nitrogen source. Cultures supplied with (NH&S04 showed a somewhat higher growth rate than those fixing dinitrogen, but they were more susceptible to butachlor (26-51%). On the contrary, butachlor enhanced the growth rate of Rhodospirillum rubrum in nitrogen gas conditions. When the culture was performed in medium with butachlor as the carbon source, the cells of fixing dinitrogen showed a higher exhaustion of butachlor than those supplemented with (N&)2S04, which exhaustion was examined by a decrease of the major absorbance at 213 nm and 260 nm. The exhaustion of butachlor as the carbon source had relation with the growth of the cells. The alkalization of culture supplemented with nitrogen gas was found in the cells treated with butachlor or untreated. The butachlor affected the carotenoid region but bacteriochlorophyll remained unaffected.
