Biological Activity of Aloe Vera Gel and Skin Extracts (알로에 베라 젤 및 껍질 추출물의 생리활성 평가)

  • In vitro biological activities of Aloe vera gel and skin extracts were evaluated. Total polyphenol contents of Aloe vera skin were measured 41.12 mg/g. DPPH radical scavenging activity of Aloe vera skin-70% EtOH extract, Aloe vera skin-water extract, Aloe vera gel-70% EtOH extract and Aloe vera gel-water extract were 55%, 38%, 11% and 10%, respectively. In addition, 70% EtOH extract and water extract were compared with respect to SOD-like antioxidant activity of Aloe vera-70% EtOH extract has higher activity than Aloe vera water extract. Tyrosinase inhibition rate of Aloe vera gel extract was higher than Aloe vera skin extract. Alcohol dehydrogenase (ADH) and Acetaldehyde dehydrogenase (ALDH) relative percentage activity of Aloe vera gel extract were 126% and 216%, respectively. It was suggested that Aloe vera gel and skin extracts could be used as a functional biomaterial for functional food and cosmetics.

Hypoglycemic Effect and Hepatic Detoxification Activity of Extracts from Crataegus fructus and Morus alba L. in Alcohol-treated Rats

  • To consider potentially new sources which have hypoglycemic effect and accelerating alcohol detoxification, this study was designed to investigate the effect of Crataegus fructus and Morns alba L. in alcohol-treated rats. I compared the body weight, glucose, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alcohol dehydrogenase (ADH), and aldehyde dehydrogenase (ALDH) of rats administered both alcohol and extract of experimental plants to rats treated with alcohol alone. Administration of extracts of C. fructus and M alba, respectively, resulted in a significant reduction in the blood glucose level and the activities of ADH of liver compared to the control rats, and administration of extract of M. alba showed significantly lower on bodyweight gain in the rats than in other treated rats. In contrast, the activities of ALDH of liver were increased. The activities of AST and ALT between the only alcohol-treated rats and the alcohol and experimental plants-treated rats were no significant difference. The results suggest that C. fructus and M alba have a hypoglycemic effect, and reduce liver damage by accelerating acetaldehyde metabolism in alcoholic rats, so the combined effect of C. fructus and M alba may be considered as an alternative remedy for hangovers, alcohol-induced overweight and alcohol-induced diabetes.

Flavor Compounds of Cholesterol-Reduced Cheddar Cheese Slurries

  • This study was carried out to find the difference in flavor compounds between cholesterol-reduced Cheddar cheese slurries and control sample. The cheeses were made by 3 different treatments as followings: 1) Control (no homogenization, no ${\beta}$-CD), 2) Trt A (1,000 psi milk homogenization, 1% ${\beta}$-CD) and 3) Trt B (cream separation following by 10% ${\beta}$-CD, mixed with skim milk at 1,000 psi homogenization). The cholesterol removals of the cheeses were 79.30% (Trt A) and 91.22% (Trt B). The cheese slurries made by the cheeses were aged at $32^{\circ}C$ for 3 wk. The production of short-chain fatty acids (SCFA) was significantly increased with storage time in all treatments. Total amount of SCFA was dramatically increased at 2 wk and maintained thereafter in control group. The amounts of acetone and acetaldehyde were slightly increased in control at 3 wk, however, no difference was found in others. Ethanol production was dramatically increased at 1 wk and decreased thereafter in all treatments. Based on our results, cheese slurries for Trt B showed a highest cholesterol removal rate. Although little difference was found in flavor production, lower amount of SCFA was found in Trts A and B in 2 and 3 wk. It may indicate that a certain amount of SCFA is decreased during ${\beta}$-CD treatment.

The Effect of Saponin Fraction of Panax ginseng C.A. Meyer on the Liver of Ethanol Administered Rat (인삼사포닌 분획이 에탄올을 투여한 쥐의 간세포에 미치는 영향)

  • Preventive effect of ginseng saponin fraction against ethanol intoxication of the liver of rats fed width 12% ethanol instead of water for 6 days was investigated. Control group was dosed 12% ethanol instead of water (free access) for 6 days and test group was dosed 0.1% ginseng saponin fraction in the 12% ethanol instead of water for 6 days. Normal rats was given only water freely. It was observed that the activities of alcohol dehydrogenase (ADH) and Microsomal ethanol oxidizing system (MEOS) of both control and test groups were higher than those of normal group while the activity of aldehyde dehydrogenate (ALDH) of control and test groups were lower than that of normal rats, However, the ALDH activity decrease of test group was much less than that of control groups. Electron micrograph showed that severely swollen and disrupted mitochondria and dilated and vesiculated ER can be seen in control group while dilated or vesiculated ER are very few and swollen or disrupted mitochondria can not be seen in test group. From the above experimental result, it seems that ginseng saponin might stimulate ethanol oxidation and the removal of acetaldehyde resulting in the decrease of ethanol intoxication of the liver.

  • In recent years, plastic waste has been recognized as a worldwide environmental issue. To solve the disposal problem of the plastic waste, alternative treatment such as the use of biodegradable plastic(polybutylene succinate: PBS) is indeed highly in demand due to its merit of PBS buried in soil decomposed into carbon dioxide and water. In the present study, for the production of further functional PBS with TiO$_2$ as photocatalyst, which shows the decomposition of detrimental organic compound and pollutant under ultraviolet irradiation, we attempted to prepare photocatalytic TiO$_2$ coatings on PBS substrate by HVOF and plasma spraying techniques under various conditions using three kinds of agglomerated powders (P200: 200nm, P30: 30nm, P7: 7nm). The microstructures of coatings were characterized with SEM and XRD analysis, and the photocatalytic efficiency of coatings was evaluated through the photo degradation of gaseous acetaldehyde. Therefore, such functional TiO$_2$-Plastic composite material is expected to considerably contribute to the reduction of aggravated environmental problem.

Study of Inorganic Photocatalyst Media for Reused Wastewater (폐수 재이용을 위한 무기계 광촉매 담체 연구)

  • This study focused on effective decomposition methods for low concentrated organic compounds from the reuse of industrial wastewater, and confirmed the possibility through photocatalyst media. Photocatalyst immobilized media was developed to carry on this experiment which confirmed the removal possibility of low concentrated organic compounds. Considering the stability and efficiency of photocatalyst immobilization, inorganic support, hollow bead, and $TiO_2$ nano powder were used. As a result of the removal experiment, the removal efficiencies of acetonitrile, ethanol, IPA(Isopropyl alcohol), methanol were above 75% after 15 minutes while those of acetone, acetaldehyde, urea were 10%, 45%, 20%, respectively after 60 minutes. If further studies were made to increase the surface area of the photocatalyst immobilized media, the efficiency of the removal of low concentrated organic compounds can be improved and this solution can also be used in an actual treatment process.

  • Human mitochondrial aldehyde dehydrogenase(ALDH2) is mainly responsible for the oxidation of acetaldehyde generated during alcohol oxidation in vivo. To investigate the role of ALDH2 in alcohol metabolism, it was needed to get solubilized enzyme. The cDNA of ALDH2 is isolated from cDNA library and ligated to several expression vectors for E. coli. At almost expression system to be constructed, the broad expression band of ALDH2 was detected. But, the large part of the expressed protein consisted as inclusion body, the yield of solubilized enzyme was not more tan 5% of the total expressed amount. Recombinant ALDH2 was verified from the several expression systems.

  • This research is related to the process of refining the raw material, industrial 2-propanone to the 2-propanone of the electronic grade. With this view, the high purity of 2-propanone was obtained through the complex preprocessing(physical adsorption method), distillation process and membrane-filtration of distillate. Impurities were identified by GC and UV, and then we assayed the water content in 2-propanone passing adsorption step made of activated carbon and Zeolite 4A. Furthermore, the distillation was performed with the packed column distillation apparatus to eliminate impurities such as acetaldehyde. Particulates were removed by reduced-pressure filtration through $0.5{\mu}m$ membrane filter and the number of the particulates was measured by particulate counter to confirm the removal of impure particles.

Elucidation of the Regulation of Ethanol Catabolic Genes and ptsG Using a glxR and Adenylate Cyclase Gene (cyaB) Deletion Mutants of Corynebacterium glutamicum ATCC 13032

  • The cyclic AMP receptor protein (CRP) homolog, GlxR, controls the expression of several genes involved in the regulation of diverse physiological processes in Corynebacterium glutamicum. In silico analysis has revealed the presence of glxR binding sites upstream of genes ptsG, adhA, and ald, encoding glucose-specific phosphotransferase system protein, alcohol dehydrogenase (ADH), and acetaldehyde dehydrogenase (ALDH), respectively. However, the involvement of the GlxR-cAMP complex on the expression of these genes has been explored only in vitro. In this study, the expressions of ptsG, adhA, and ald were analyzed in detail using an adenylate cyclase gene (cyaB) deletion mutant and glxR deletion mutant. The specific activities of ADH and ALDH were increased in both the mutants in glucose and glucose plus ethanol media, in contrast to the wild type. In accordance, the promoter activities of adhA and ald were derepressed in the cyaB mutant, indicating that glxR acts as a repressor of adhA. Similarly, both the mutants exhibited derepression of ptsG regardless of the carbon source. These results confirm the involvement of GlxR on the expression of important carbon metabolic genes; adhA, ald, and ptsG.

Quality Properties of Gangha-ju Liquor According to the Preparation Method (제조방법에 따른 강하주의 품질 특성)

  • This studies were performed to develop a Korean traditional folk liquor namely Gangha-ju has been prepared at Bosung district in Korea, and manufacturing conditions and anti-oxidation activity and anti-microbial activity of Gangha-ju were investigated. Ethyl-alcohol 20% and 30% Gangha-ju were brewed with glutinous rice wine, distilled liquor and 6 herbs of ginger, cinnamon, etc. Chemical and physical properties of 30% Gangha-ju were acidity 0.22, pH 4.31, amino acidity 3.26, transmittance 59 and conductivity $911\;{\mu}s/m$, and 20% Gangha-ju were 0.43, 4.20, 6.26, 62 and $924\;{\mu}s/m$. Volatile flavor compounds of ethyl alcohol, acetic acid, butanol, n-amyl alcohol, iso-pentyl alcohol, ethyl acetate, butyl acetate, acetaldehyde and furfural were detected, and main aroma compounds of Gangha-ju were isopentyl alcohol and ethyl acetate. Anti-oxidation activity by DPPH method was evaluated 31.32%, and nitrite scavenging effect was 31.79%. Anti-microbial activity against several microorganisms was pronounced strong activity over a wide range of test organisms, and Leuconostoc mesenteroids and Salmonella Ttyphimurium, Staphylococcus epidermidis were found to be more sensitive to Gangha-ju than Escherichia coli and Aspergillus flavus.
