Regulation of heavy metal and Growth Adaptation of Meliotus suaveolens Seedlings Treated with Pb (Pb처리에 따른 전동싸리 유식물의 생장적응과 증금속의 조절)

  • Effects of lead(Pb) and calcium(Ca) on growth responses, accumulation of heavy metals and nitrogen fixation activities of Melilotus suaveolens seedlings were quantitatively analyzed during growing period. Pb contents of the root treated with 30, 100 ppm Pb and 100 ppm Pb added 100 ppm Ca were 54.1, 90.9 and 26.1 folds higher than that of the control, respectively, at pH 4.2 in 28th days, and heavy metal content of plant increased with increasing of pH and Pb concentration. The melilot plant was classified as a Pb accumulator by higher accumulation of Pb in shoot than that of root. Pb treatments resulted in inhibiton of height and chlorophyll contet, and Ca treatment increased height and chlorophyll content insignificantly at pH 4.2 in 28 days. The plant biomass reduced 49, 60 and 54% at pH 4.2 and 47,53 and 50% at pH 6.5, respectively, by 30, 100 ppm Pb and 100 ppm Pb added 100 ppm Ca treatment. Specific nitrogen fixation of nodules reduced 68.4% and 46.6% by 100 ppm Pb treatment and 3.7% and 24.9% by 100 ppm Pb added 100 ppm Ca at pH 4.2 and pH 6.5, respectively, so Ca inhibited significantly Pb activity and toxicity in acdic pH. Nodule formation were reduced to 33, 33 and 50% at pH 4.2 and 50, 33 and 38% at pH 6.5 by 30, 100 ppm Pb and 100 ppm Pb added 100 ppm Ca, respectively.

  • The hydrolysis of metampicillin to ampicillin was investigated using high perofrmance liquid chromatography. We developed the simultaneous determination of metampicillin and ampicilin using a Zorbox CN column and 5% acetonitrile and 8% methanol in 0.02 M phosphate buffer (pH 7.0) as mobile phase. Matampicillin was hdyrolyzed to ampicillin with half life of 41.5 min at physiological pH and temperature. In acdic pH, metampicillin was rapidly hydrolyzed to ampicillin within a chromatogrphic separation.

Reduced Addamycin Cytotoxicity in RIF-1 Multicell Spheroid Due to an Acidic Microenvironment

  • Variations in adriamycin uptake and cytotoxicity were studied in tumor cells that were grown in different growth states and microenvironments. RIF-1 tumor cells were maintained in an RPMI 1640 medium, and grown in either a monolayer or multicell spheroids. For exponentially growing cells, adriamycin cytotoxicity increased with increased dosage up to 2.5 $\mu$g/ml, and this cytotoxicity was reduced when the cells were grown in a plateau phase or in an acidic microenvironment (pH 6.6). This reduced cytotoxicity was correlated with the uptake of the drug. For multicell spheroids, the cytotoxicity of the drug was reduced dramatically, and this reduction was also correlated with a reduced uptake of the drug and an acidic pH inside of the spheroids. When the drug cytotoxicity was evaluated at different locations within the spheroids, the cells in the inner regions were least affected by the drug, suggesting that both an acidic microenvironment and noncycling plateau phase cells are contributing factors in decreasing the efficacy of the drug in an organized tissue, such as multicell spheroids.

  • In this study, the effect of an acidic treatment on granular activated carbon (GAC) and activated carbon fibers (ACF) was investigated for a $Pb^{2+}$ and $Ni^{2+}$ ion adsorption. 1.0 M nitric acid solution was used as the acid solution for the surface treatment. Surface properties of the GAC and ACF were characterized by the pH, elemental analysis and pHpzc (pH of the point of zero charge). Their specific surface area and the pore structure were also evaluated by the nitrogen adsorption data at 77K. As a result, the acidic treatment led to an increase of the oxygen-containing functional groups. Furthermore, the adsorption capacity of the acid-treated GAC and ACF was improved in the order of acidic-ACF > untreated-ACF > acidic-GAC > untreated-GAC, though the decrease in specific surface area induced by a pore blocking of the functional groups was observed.

Recovery of Pd(II), Pt(IV), and Rh(III) Using Polyelectrolytes

  • Two methods, precipitation and ultrafiltration, were applied in order to recover platinum group metals(PGM) by complexing them with water-soluble polyelectrolytes, e.g., polyethyleneimine [PEl], poly(2-vinylpyridine) [2-PVP], poly (4-vinylpyridine) [4-PVP], and poly (styrene sulfonic acid) [PSSA]. In the precipitation method, the PGM-polyelectrolyte complex that was formed by mixing first with polybase, e.g.,4-PVP at pH 1 was precipitated by further mixing with polyacid, e.g., PSSA. However, the recovery of PGM obtained by this method was not quantitative(less than 70%). The "sandwiching" binding between the metal anions and two polyelectrolytes was examined by X-ray photoelectron spectroscopy(XPS). The XPS studies indicated that the PGM atom was bound with the acdic and basic polyelectrolyte via its oxygen and nitrogen atom, respectively. The recovery of PGM using polyelectrolyte was further studied by ultrafiltration methods as follows : The PGM ions, eomplexed at pH 1 with polyelectrolyte, allowed the applicntion of membrane filtration by virtue of the great differences in molecular weights between PGM and other low molecular weight species. By applying this method, Pd and Pt (ca. $10^{-4}M$) were selectively separated almost quantitatively from coexisting metal ions, e.g., $Cu^{2+}$ and $Ni^{2+}$. The EPR spectra and viscosity measurements indicated that these polyelectrlytes were not bound to $Cu^{2+}$ and $Ni^{2+}$ ions at this pH, which provided the basis for selective separation of PGM(Pd, Pt and Rh) from these coexisting ions.
