Activities of Phospholipase $A_2$ and Cyclooxygenase, and Syntheses of Thromboxane and Prostacyclin in Streptozotocin Induced Diabetic Rats (Streptozotocin 유도 당뇨쥐에서의 Phospholipase $A_2$, Cyclooxygenase 활성과 Thromboxane 및 Prostacyclin합성)

  • The relation between lipid peroxidation and thrombotic reaction were investigated in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 100$\pm$10gm were randomly assigned to normal and STZ-induced diabetic group(DM). Diabetes was experimentally induced by intravenous injection of 55mg/kg of body weight of STZ in citrate buffer(pH 4.3) after 4 weeks feeding of basal diet. Animals were sacrificed at the 6th day of diabetic states. Body weight gains were lower in diabetic group after STZ injection. Serum levels of thiobarbituric acid reacting substances(TBARS) that were markedly increased in DM group compared with of normal group. TBARS levels of HDL and LDL were similar patterns to total TBARA of serum. Activities of platelet phospholipase A2(PLA2) were higher in diabetic group than those of normal group. Activities of platelet cyclooxygenase were 106% in DM group than normal group. Platelet thromboxane A2(TXA2) formation was increased in DM group than normal group. Production of aortic prostacyclin(PGI2) was lower in diabetic group than that of normal group. PGI2/TXA2 ratios were decreased by 55% in DM groups than those of normal group. The present results indicate that STZ-induced diabetic rats are more sensitive to oxidative stess which leads to acceleration of lipid peroxidation and platelet aggregability. In conclusion, accelerating effect of lipid peroxidation and thrombogenesis in diabetic state is regareded to be resulted from enhancement of PLA2 activity and arachidonic acid metabolism, inhibition of antiaggrgating agent and aortic PGI2 formation.

  • Frost damage of a storage tank for chemical admixtures caused by a low temperature in winter and quality deterioration of chemical admixtures have been often reported. However, there have been few regulations and researches related to the frost damage of chemical admixtures and facilities. Therefore, this paper is intend to investigate not only, the freezing points of chemical admixtures such as AE admixtures and water-reducing AE admixtures, etc., but also the quality variation of concrete used with chemical admixtures before freezing and after freeze-thaw cycles. According to the results of experiments, most chemical admixtures except anti-freezing agent and accelerating water-reducing AE admixtures should be kept from being frozen in low winter temperatures. However, full agitation of frozen chemical admixtures dose not cause the problems of concrete related to the quality of chemical admixtures.

However On domestic mine conditions that couldn't having every time after mine tunnel excavation, it couldn't ensure the field batch plant which is a fixed in an issue of economies and site security, constructing it by supply gets from shotcrete materials combined in neighborhood ready mixed concrete manufacturing plants. carried shotcrete in ready mixed concrete manufacturing plants as migration length and time are falling off in quality. But, it is difficult for construction quality control By quality control arbitrariness absence at on-scene. In the present study, carry out Laboratory Test by kinds and percentages of accelerating agent for evalution of Mechanical Property and Durability of Shotcrete.

  • Recently, viral infectious diseases and new diseases are not limited to one region, but are spreading worldwide, causing serious economic and social damage. In addition, the development cycle of new diseases is shortening, and the rate of spread is accelerating. In order to prevent the spread of disease, passive forms of response after a disease outbreak, such as personal and regional quarantine and border closure, are prioritized. This type of response has many shortcomings as a fundamental response to preventing the spread of disease. Therefore, this study proposes a disease prevention monitoring system including new disease occurrence information. In this study, disease information and user information are collected through the establishment of the IoMT environment. Information collection using an agent collects and classifies data registered in the disease information server. In the IoMT environment, user data is collected, and whether the user is infected with a disease is evaluated and provided to the user. Through this study, individual disease symptom information can be provided and active countermeasures against the spread of disease can be provided.

PEP-1-FK506BP inhibits alkali burn-induced corneal inflammation on the rat model of corneal alkali injury

  • FK506 binding protein 12 (FK506BP) is a small peptide with a single FK506BP domain that is involved in suppression of immune response and reactive oxygen species. FK506BP has emerged as a potential drug target for several inflammatory diseases. Here, we examined the protective effects of directly applied cell permeable FK506BP (PEP-1-FK506BP) on corneal alkali burn injury (CAI). In the cornea, there was a significant decrease in the number of cells expressing pro-inflammation, apoptotic, and angiogenic factors such as TNF-α, COX-2, and VEGF. Both corneal opacity and corneal neovascularization (CNV) were significantly decreased in the PEP-1-FK506BP treated group. Our results showed that PEP-1-FK506BP can significantly inhibit alkali burn-induced corneal inflammation in rats, possibly by accelerating corneal wound healing and by reducing the production of angiogenic factors and inflammatory cytokines. These results suggest that PEP-1-FK506BP may be a potential therapeutic agent for CAI.

  • In Vivo and In vitro antispasmodic effects of Jun-Si-Baek-Chul-San, a Traditional Korean Polyherbal Medicineconsisted of 7 types of herbs were observed in the present study. To clarify the effects of Jun-Si-Baek-Chul-San, on accelerating small intestinal movement induced by the stimulation of cholinergic neurotransmission, we evaluated the effects of Jun-Si-Baek-Chul-San on In vivo carbachol (an acetylcholinergic agent)-accelerated mice small intestinal transit and on In vitro contractions induced by low-frequency electrostimulation, KCl, histamine or acetylcholine using isolated guinea pig ileum. To induce the acceleration of mice small intestinal transit, Carbachol 1 mg/kg was once subcutaneously dosed 15min before last administration of the test drugs. In the present study, Jun-Si-Baek-Chul-San 500, 250 and 125 mg/kg or domperidone 20 mg/kg were orally pretreated on the carbachol-accelerated mice small intestinal transit once a day for 7 days and the small intestinal transit rateof activated charcoal powder were monitored. In vitro assays, Jun-Si-Baek-Chul-San1, 0.1, 0.01 and 0.001 mg/ml or domperidone $2{\times}10^{-5}M$ were treated 10min before ileal contraction was induced by filed stimulation, acetylcholine, KCl and histamine, and the % changes of contractions were observed compared to the treatment of inducer alone. In spontaneous contraction, the % changes of contractions were observed compared to treatment of vehicle alone at 10min after Jun-Si-Baek-Chul-San or domperidone treatment. The efficacy of Jun-Si-Baek-Chul-San was compared to those of domperidone. High concentration, 1 mg/ml of Jun-Si-Baek-Chul-San was found to decrease the spontaneous contraction of the isolated guinea-pig ileum. In addition, Jun-Si-Baek-Chul-San decrease contractions induced by electrostimulation, acetylcholine, histamine and KCl in the isolated guinea-pig ileum. In addition, Jun-Si-Baek-Chul-San effectively inhibited the accelerated small intestinal movement induced by carbachol stimulation of cholinergic neurotransmission in In vivo. Based on the results, although the exact molecular or action mechanism and which herbs or compound in Jun-Si-Baek-Chul-San are responsible for actions, it was concluded that Jun-Si-Baek-Chul-San normalization in the accelerated intestinal motility might be interfere with a variety of muscarinic, adrenergic and histaminic receptor activities or with the mobilization of calcium ions required for smooth muscle contraction non-specificly. Therefore, it is expected that Jun-Si-Baek-Chul-San will be promising as a prescription of clinical treatment of digestive tract disorders such as accelerated the motility of intestine, diarrhea or intestinal painful contractions.

Study on Physical and Chemical Properties of CaO-Al2O3 System Melting Compound (CaO-Al2O3계 용융화합물의 물리·화학적 특성에 관한 연구)

  • This study is aimed to identify the method to use the CaO-$Al_2O_3$ system of rapidly cooled steel making slag (RCSS) as the environment-friendly inorganic accelerating agent by analyzing its physical and chemical properties. The fraction of rapidly cooled steel making slag is distinguished from its fibrous, and the contents of CaO and $Fe_2O_3$ are inversely proportional across different fractions. In addition, as the content of CaO decreased and the content of $Fe_2O_3$ increased, the loss ignition tended to become negative (-) and the density increased. The pore distribution by mercury intrusion porosimetry is very low as compared to the slowly cooled steel-making slag, which indicates that the internal defect and the microspore rate are remarkably lowered by the rapid cooling. To analyze the major minerals the rapidly cooled steel-making slag, XRD, f-CaO quantification and SEM-EDAX analysis have been performed. The results shows that f-CaO does not exist, and the components are mainly consisted of $C_{12}A_7$ and reactive ${\beta}-C_2S$.

Hitological Changes on the Wound Healing Process of Alkali Burned Mouse Cornea (알칼리 화상을 입은 마우스 각막에서 상처 치유과정 중 관찰된 조직학적 변화)

  • Purpose: To better understand the corneal regeneration after alkali burn regarding the initial clinical progression and the therapy, we investigated the changes of the multi factors following chemical injury in cornea. Methods: This study was performed to observation on the healing process of alkali burned cornea in aspect of immunohistochemistry by immunofluorescence or H-E staining and TUNEL assay. Results: The results showed that although a healing process occurred after alkali burn, apoptosis of epithelial, stromal and endothelial cells in the cornea was continuously observed. Neovascularization and expression of ${\alpha}$-smooth muscle actin (${\alpha}$-SMA) from limbus and from injured cornea, respectively, were observed after 3 days of alkali burn. Formation of collagen III in corneal stroma and increased expression of chondroitin sulfate are coincident with expression of ${\alpha}$-SMA and transforming growth factor-${\beta}$ (TGF-${\beta}$). Conclusions: These data suggest that medical treatment within 3 days of alkali burn will be effective to inhibit neovascularization and formation of collagen III and chondroitin sulfate. This study extends our immumohistochemical understanding of healing process in alkali burned cornea, and the results get in this study will be cornerstones in the development of therapeutic agent for accelerating renewal of chemical damaged cornea.

Neuroprotective effect of Puerariae Radix extract on focal cerebral ischemia in mice (갈근 에탄올추출물의 국소뇌허혈 모델에 대한 뇌신경보호 효과)

  • Objectives : The purpose of this study was to evaluate the neuroprotective effect of Pueraria lobata extract on focal cerebral ischemia in mice. Methods : Focal cerebral ischemia was induced by occlusion of the right middle cerebral artery using the intraluminal filament model. ICR male mice underwent 90 minutes of middle cerebral artery occlusion (MCAo) followed by 24 hours of reperfusion. Mice were administered Pueraria lobata extract orally at the dose of 300mg/kg just prior to reperfusion. Rotarod test and balance beam test were practiced to assess sensory-motor function 23 hours after MCAo. In rotarod test, the latency to fall on the accelerating rotarod was recorded for 5 min. In balance beam test, the score was graded according to number of slips and latency to cross. The infarct volume was measured 24 hours after MCAo using 2% 2,3,5-triphenyltetrazolium chloride (TTC) staining. Results : Pueraria lobata extract treated group showed significant reduction in infarct volume by 27.3% compared to control group (p<0.05). In rotatod test, it also showed significant extension of latency time compared to control group ($67.82{\pm}15.08$ vs. $5.62{\pm}1.06$, p<0.001). In contrast to performance in rotarod test, that in balance beam test did not improve with Pueraria lobata extract treatment. Conclusions : We conclude that Pueraria lobata extract has a significant neuroprotective effect and reduces damage of sensory-motor function in MCAo model. These findings suggest that Pueraria lobata could be a potent neuroprotective agent.
