METASTASIS OF ADENOID CYSTIC CARCINOMA OF THE MINOR SALIVARY GLAND TO THE SCALP : A CASE REPORT (소타액선 선양낭성암종의 두피 전이 : 증례 보고)

  • Adenoid cystic carcinoma(ACC) is the second most common malignant neoplasm in the salivary gland. In spite of ACC shows slowly growing nature, it is sometimes highly detrimental that it readily invades adjacent tissues and metastasize to distant organs at the early stage of disease. Hence, treatment outcome may be misfortunate due to wide regional infiltration, pathognomonic perineural spread and the tendency of hematogenous metastasis. We present a unusal case of ACC of the scalp in which the patient initially presented with bumpy mass of the scalp who had been diagnosed as the primarily developed ACC of the right hard palate that extended to infratemporal fossa, nasal cavity, and paranasal sinuses and had been treated by total excision and post-operative chemotherapy and radiation therapy for 10 years ago. Although this lesion occured at the scalp, which is a frequent site of primary dermatologic ACC, its histomorphology was the same with that of previous tumor. Complete clinical examination showed no recurrence sign at the primary site and suggested the scalp as a sole treatment failure site. Accordingly, it would be reasonable to consider the present case as either a scalp metastasis or a second primary lesion of salivary gland ACC.

Hybrid 'Sinta' Papaya Exhibits Unique ACC Synthase 1 cDNA Isoforms

  • Five ripening-related ACC synthase cDNA isoforms were cloned from 80% ripe papaya cv. 'Sinta' by reverse transcription-PCR using gene-specific primers. Clone 2 had the longest transcript and contained all common exons and three alternative exons. Clones 3 and 4 contained common exons and one alternative exon each, while clone 1, the most common transcript, contained only the common exons. Clone 5 could be due to cloning artifacts and might not be a unique cDNA fragment. Thus, there are only four isoforms of ACC synthase mRNA. Southern blot analysis indicates that all five clones came from only one gene existing as a single copy in the 'Sinta' papaya genome. Multiple sequence alignment indicates that the four isoforms arise from a single gene, possibly through alternative splicing mechanisms. All the putative alternative exons were present at the 5'-end of the gene comprising the N-terminal region of the protein. 'Sinta' ACC synthase cDNAs were of the capacs 1 type and are most closely related to a 1.4 kb capacs 1-type DNA(AJ277160) from Eksotika papaya. No capacs 2-type cDNAs were cloned from 'Sinta' by RT-PCR. This is the first report of possible alternative splicing mechanism in ripening-related ACC synthase genes in hybrid papaya, possibly to modulate or fine-tune gene expression relevant to fruit ripening.

COMPARATIVE IMMUNOHISTOCHEMICAL ASSAYS FOR THE EXPRESSION OF ANGIOGENIC FACTORS IN TUMORS OF HUMAN SALIVARY GLANDS (타액선 종양에서 혈관형성 인자의 발현에 관한 면역조직화학적 비교 연구)

  • Hallmarks of clinical behaviors of adenoid cystic carcinoma(ACC) of salivary glands are the delayed onset of vascular metastasis and poor responses to classical chemotherapeutic agents. Poor prognoses from salivary ACC are caused by lung metastases that are resistant to conventional therapy. Therefore, cellular and molecular characteristics that influence the dissemination of metastatic cells are important for the design of more effective treatment of salivary ACC. Tumor angiogenesis has been known to be essential for the distant metastasis of malignant cells. So, we determined expressions of angiogenic proteins in benign (pleomorphic adenoma) and malignant (ACC, mucoepidermoid carcinoma) tumors of salivary glands and compared each other and to those in oral squamous cell carcinoma. Using surgical specimens, we performed immunohistochemical assays with anti-vascular endothelial growth factor (VEGF), VEGF receptor-2 (VEGFR-2), phosphorylated VEGFR-2 (pVEGFR-2), matrix metalloproteinase (MMP)-9, and interleukin (IL)-8 antibodies. Most angiogenic factors were overexpressed in malignant salivary tumors than in pleomorphic adenoma which is benign nature. Moreover, ACC demonstrated more expression of VEGFR-2 than that of squamous cell carcinoma which used as control. Conclusively, these data show those angiogenic factors produced by salivary gland tumors may affect the propagation and metastasis of malignant cells of salivary tumors, and could be used as biomarkers for the malignant transformation of salivary gland tumors. Prospectively, although further studies will be needed, these biomarkers related to angiogenesis can be molecular targets for the therapy of salivary ACC, which has propensity for delayed vascular metastasis.

  • Introduction: Epidermal growth factor receptor (EGFR) is expressed in human epithelial tumors including salivary cancers, and known to be correlated with tumor progression and poor clinical courses in some epithelial tumors. In this study, we determined the therapeutic effect of the anti-EGFR monoclonal antibody Erbitux (C225, cetuximab) in combination with the DNA topoisomerase I inhibitor irinotecan (CPT-11) on human salivary adenoid cystic carcinoma (SACC) cells growing in nude mice. Materials and Methods: At first, immunocytochemical analysis for the expression of EGFR and phosphorylated EGFR (pEGFR) on a human salivary ACC cell line (ACC3). To determine the in vivo effects of Erbitux and CPT-11, nude mice with orthotopic parotid tumors were randomized to receive intraperitoneal Erbitux (1 mg) two times per week, intraperitoneal Irinotecan (50 mg/kg) once per week, Erbitux plus CPT-11, or placebo. (control) Tumor volume and weight were measured. And mechanisms of in vivo activity of Erbitux and/or CPT-11 were determined by immunohistochemical/ immunofluorescent analyses. Results: Immunocytochemical staining of ACC3 demonstrated that EGFR was expressed and phosphorylated. CPT-11 inhibited ACC tumor growth in nude mice. Tumors of mice treated with CPT-11 and CPT-11 plus Erbitux exhibited increased tumor cell apoptosis and decreased microvessel density, which correlated with a decrease in the tumor volume in nude mice. But, CPT-11 seems not to be synergistic with Erbitux in our ACC3 model system. Conclusion: These results suggest that anti-EGFR monoclonal antibody and the DNA topoisomerase I inhibitor will be effective in the treatment of recurred or metastatic lesions of salivary ACC.

ON ENDOMORPHISM RING OF H-INVARIANT MODULES

  • The relationships between submodules of a module and ideals of the endomorphism ring of a module had been studied in [1]. For a submodule L of a moudle M, the set $I^L$ of all endomorphisms whose images are contained in L is a left ideal of the endomorphism ring End (M) and for a submodule N of M, the set $I_N$ of all endomorphisms whose kernels contain N is a right ideal of End (M). In this paper, author defines an H-invariant module and proves that every submodule of an H-invariant module is the image and kernel of unique endomorphisms. Every ideal $I^L(I_N)$ of the endomorphism ring End(M) when M is H-invariant is a left (respectively, right) principal ideal of End(M). From the above results, if a module M is H-invariant then each left, right, or both sided ideal I of End(M) is an intersection of a left, right, or both sided principal ideal and I itself appropriately. If M is an H-invariant module then the ACC on the set of all left ideals of type $I^L$ implies the ACC on M. Also if the set of all right ideals of type $I^L$ has DCC, then H-invariant module M satisfies ACC. If the set of all left ideals of type $I^L$ satisfies DCC, then H-invariant module M satisfies DCC. If the set of all right ideals of type $I_N$ satisfies ACC then H-invariant module M satisfies DCC. Therefore for an H-invariant module M, if the endomorphism ring End(M) is left Noetherian, then M satisfies ACC. And if End(M) is right Noetherian then M satisfies DCC. For an H-invariant module M, if End(M) is left Artinian then M satisfies DCC. Also if End(M) is right Artinian then M satisfies ACC.

Effect of Benzyladenine on the IAA-Induced Ethylene Production in the Primary Roots of Maize (옥수수 일차뿌리에서 benzyladenine이 IAA에 의해 유도된 에틸렌 생성에 미치는 영향)

  • This study was conducted to examine the effect of cytokinin ($N^6$-benzyladenine; BA) and/or an IAA on ethylene production of maize (Zea mays) primary roots. When the two hormones were applied exogenously, both hormones synergistically increased ethylene production, which was greater than the sum of the level of each hormone's effect. For example, the ethylene production was stimulated between about 87% and 170% of the control by $10^{-4}\;M$ BA with $10^{-4}\;M$ IAA for 8 hours respectively, whereas the ethylene production was increased by about 480% of the control when the two hormones were treated simultaneously. Such a synergistic effect was also found in changes in the activity and gene expression level of ACC synthase. However, in the case of ACC oxidase did not show any observable effects. Based on our results, it is possible to conclude that BA and IAA stimulated the ethylene production synergistically by affecting the ACC synthase in maize roots.

Clinical Course of Aplasia Cutis Congenita (선천성 피부 무형성증의 임상 경과)

  • Purpose: Aplasia cutis congenita (ACC) is a rare condition characterized by a localized absence of skin, and it can be associated with other congenital anomalies. This study was done to evaluate clinical course and outcome of ACC in neonates. Methods: Based on the medical records, we retrospectively reviewed 8 neonates diagnosed with ACC at Cheil General Hospital and Women's Health Care Center, Kwandong university College of Medicine from January 2004 to December 2010. We classified ACC by Frieden's classification and analyzed the patient's demographic data, clinical course and outcome. Results: Among 8 patients with ACC, 5 patients were classified to group 1; scalp ACC without anomalies and 3 patients to group 7; ACC localized to extremities without blistering. Defect size was from 0.3 cm to 1.5 cm, limited in the superficial skin or subcutaneous tissue without associated anomalies. Defects were healed under conservative treatment with mild scar formation within four months. Conclusion: ACC could be diagnosed easily through physical examination in neonates. All cases showed good clinical outcome without surgical treatment. However because of small numbers and small sized defects of cases, further study including lesions of large size is needed.

Effects of Medicinal Plant Water Extracts on Expression of Anti-diabetic Enzymes mRNA (약용식물 물 추출물이 항당뇨 효소의 유전자 발현에 미치는 영향)

  • This study evaluated the anti-diabetic effects of medicinal plant water extracts on expression of hepatic glucokinase (GCK), pyruvate dehydrogenase (PDH) and acetyl-CoA carboxylase (ACC) mRNA. GCK, PDH and ACC mRNA expression levels were measured by RT-PCR. The medicinal plants used in our study were Cordyceps militaris (CM), Perilla sikokiana (PS), Salvia miltiorrhiza Bunge (SMB), Panax notoginseng (PN) and Angelica utilis Makino (AUM). We found that GCK mRNA expression was increased to about 181% at the 250 ppm of CM water extract. Furthermore, we also found that CM and AUM water extracts stimulated PDH mRNA expression level related to glucose metabolism, however, PS, SMB and PN did not stimulate PDH mRNA expression as expected. Expression of ACC mRNA was also significantly higher in both CM and AUM water extracts. Overall, the results of our study suggest that CM and AUM water extracts stimulate expression of hepatic GCK, PDH and ACC mRNA.

Ethylene Biosynthesis of an Alkalophilic Bacillus sp. Alk-7 (알카리성 Bacillus sp. Alk-7에 의한 Ethylene 생합성과 그 경로)

  • AH alkalophilic Bacillus SP. AIk-7, isolated from soil, produced ethylene. The characteristics of this microorganism is the ability to grow well under the alkaline condition, at pH 10.3. This strain is similar to Bacillus alkalophilus in terms of morphological, physiological and biological characteristics. In observation of relationship of cell growth and ethylene production according to incubation times, the ethylene synthesis mostly occur from the late exponential phase to the death phase of growth. The purpose of this paper is to study the effects of various substrates on the biosynthesis of ethylene in the intact cell and the cell-free system by the Bacillus sp. AIk-7. In both intact cell and cell-free extract, optimum conditions for ethylene production was achieved at pH 10.3 and 3$0^{\circ}C$. Ethylene was effectively produced from L-Met and 1-aminocyclopropane-1-carboxylic acid (ACC). In this case, ACC as the substrate on ethylene production were two fold higher than L-met at each concentration of substrates. On the other hand, the cell-free ethylene-forming system was used as a tool for the elucidation of the biochemical reaction involved in the formation of ethylene by Bacillus sp. AIk-7. Ethylene production in the cell-free system required the presence of manganese and cobalt ion to be stimulated a little. The result obtained in this work suggests that L-met and ACC may be a precursor more directly related to bacterial ethylene production than any other substrates tested.

The Patterns of Deviation in Urban Music Festival: Focusing on the ACC World Music Festival (도시음악축제에서 나타나는 일탈성 양상 연구 - ACC월드뮤직페스티벌을 중심으로 -)

  • The purpose of this study is to examine the patterns of deviation in urban music festival through the theories and a case study. A deviation help contemporary people in the routine of daily life to give a lot of energy. Such a deviation is strongly experienced from the festival. So it is important to study the patterns of deviation in urban festival. For this purpose, the relation between deviation and festival is examined first. And then the patterns of deviation in festival are drawn from the preceding research and case studies. The patterns of deviation in festival are identified as spatio-temporal deviation, active deviation, and situational deviation. Spatio-temporal deviation is divided into non-dailiness, space separation, and space appropriation. Active deviation is divided into make-over and expression. Situational deviation is divided into overturning and new meeting. This patterns of deviation applies to case study of ACC world music festival held in Gwangju metropolitan city with a content analysis methodology. The research finding is that Spatio-temporal deviation is most evident in ACC world music festival, on the other hand active deviation and situational deviation are weak or no evident. It is expected that various deviant elements presented in this study will be used strategically in festival planning to help strengthen the festivity of modern urban festivals.
