Antimicrobial Effect of Acanthopanax sessiliflorum Fruit Extracts against Selected Oral Bacteria

  • This study aimed to evaluate the antimicrobial effects of Acanthopanax sessiliflorum fruit (ASF; Ogaza) extracts on Streptococcus mutans and Streptococcus sobrinus, which are agents that cause dental caries, and on Streptococcus mitis and Streptococcus salivarius, the microbial flora of the oral cavity. The ASF extracts obtained using 70% ethanol were fractionated in the order of ethyl acetate and n-Butanol, concentrated under reduced pressure, and lyophilized to give powdery solvent extracts. The antimicrobial activity of ASF extracts from each solvent was examined using the disk diffusion method. As a result, only those extracts obtained using an ethyl acetate solvent showed antimicrobial activity. These extracts were selected, and the minimum inhibitory concentration was measured by disk diffusion method at various extract concentrations. Results showed a minimum inhibitory concentration of 32 mg/ml. The viable cell count was measured to confirm the minimum bactericidal concentration. Results showed a minimum bactericidal concentration of 64 mg/ml. In the cytotoxicity test using normal human dermal fibroblast cells, the absorbance value of the test group was similar to that of the control group at 0.64, 1.28, and 6.4 mg/ml. The bacteria and their colonies were examined using a scanning electron microscope. Boundaries between the antimicrobial activity region and non-antimicrobial activity region were observed around the paper disk, which was immersed in the extract with 32 mg/ml concentration. Bacterial colonization was not observed in the area with antimicrobial activity. This finding suggests that ASF extracts can inhibit the growth of some microorganisms in the oral cavity, in addition to the effects of these extracts known to date. In particular, ASF extracts may be used as a preparation for preventing dental caries by adding the extract to the toothpaste or oral mouthwash.

오가피(Acanthopanax sessiliflorum) 열매 추출물의 생리활성 (Biological Activity of Extracts from Acanthopanax sessiliflorum Fruit)

  • 오가피열매 추출물의 다양한 생리활성을 측정하였다. 오가피열매 추출물의 페놀성물질 함량은 열수 추출물과 60% ethanol 추출물에서 각각 21.4, 15.8 mg/g으로 측정되었으며, 항산화효과 측정에서는 200 ${\mu}g/mL$ 농도의 페놀성 화합물이 첨가되었을 때 DPPH에 대한 전자공여능은 열수 추출물에서는 $92.4{\pm}0.1%$, 60% ethanol 추출물에서는 $89.2{\pm}1.1%$로 측정되었다. $ABTS^+$ radical decolorlization에서는 열수 추출물은 $98.3{\pm}1.1%$, 60% ethanol 추출물은 $96.5{\pm}3.5%$로 측정되었고, antioxidant protection factor측정 결과, 열수 추출물은 $2.0{\pm}0.6$ PF, 60% ethanol 추출물은 $1.2{\pm}2.8$ PF로 측정되었다. TBARs 측정에서는 열수 추출물이 $66.3{\pm}0.8%$, 60% ethanol 추출물이 $61.4{\pm}2.3%$의 저해능을 나타내었다. Angiotensin converting enzyme (ACE)과 xanthine oxidase (XOase) 저해능은 200 ${\mu}g/mL$ 농도의 페놀성 화합물이 첨가 되었을 때 열수 추출물에서 ACE가 $59.3{\pm}1.5%$의 저해능을 나타내었고, 에탄올 추출물에서 ACE가 $85.1{\pm}3.2%$, XOase 가 $9.52{\pm}0.8%$의 저해능을 가지는 것으로 확인되었다. 오가피열매 추출물의 tyrosinase 억제효과는 열수 추출물에서 $56.6{\pm}1.8%$, 60% ethanol 추출물에서 $33.7{\pm}2.2%$의 저해능을 나타내었고, elastase의 경우 열수 추출물과 60% ethanol 추출물에서 각각 $53.1{\pm}1.1%$$22.4{\pm}3.1%$의 저해능을 나타내었다. Astringent 효과는 열수 추출물에서 $50.5{\pm}0.9%$, 60% ethanol 추출물에서 $11.5{\pm}4.1%$의 효과를 가지는 것으로 확인되었다.

복합생약 HO-Series의 숙취개선 임상적 유용성 평가 (Evaluation of Clinical Usefulness of Herbal Mixture HO-Series for Improving Hangover)

  • The purpose of this study is to investigate the hangover relieving effect of HO-series. HO-S1 is an herbal mixture, which consists of extracts from Flower of Pueraria lobata Ohwi, Glycyrrhiza glabra Linné, Fruit of Lycium chinense Miller, Poria cocos Wolf, Acanthopanax sessiliflorum Seeman, Scutellaria baicalensis Georgi, Atractylodes lancea De Candlle and Zingiber officinale Roscoe. HO-S2 is a candidate that has been performed to ultra filtration based on HO-S1. HO-S3 is a mixture of amino acids and vitamins based on HO-S2. HO-01 is the final beverage base produced based on HO-S3. The antioxidant activity of HO-series was similar to that of vitamin C or trolox. The production of t-BHP induced reactive oxygen species(ROS) was significantly blocked in the presence of HO-series. In vivo study, AUC of alcohol and acetaldehyde concentrations in HO-S2 and HO-S3 treated groups significantly decreased. Hepatic alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH) activity were significantly higher in HO-S2 and HO-S3 treated groups. And 2E1 activity and glutathione were significantly elevated, while the malondialdehyde level was not significantly in liver tissue. After alcohol exposure, the sensitivity scores of blood alcohol and acetaldehyde concentration and hangover symptoms were significantly decreased in the HO-01 intake group compared with the non-intake group. ALDH activity was significantly increased in the HO-01 intake group. HO-series have antioxidant activity and a protective effect from ROS. HO-S2, HO-S3 and HO-01 are potentially highly beneficial in relieving hangover, as it scavenges reactive free radicals and boosts the endogenous antioxidant system.
