  • Two glycosides, assumed to be one of the potential active principles, is isolate from the root bark of Acanthopanax koreanu Nakai, identified to be acanthoside D, $C_{34}H_{46}O_{18}$, mp $242^{\circ}C$, and syringo side, $C_{17}H_{24}O_{9}$ mp $192^{\circ}C$. Acanthoside D has been found to have s-GPT, s-GOT lowering effect, BSP-retention rate and survival rate in the toxic state through the bio-pharmacological experiments.

  • Acanthopanax koreanum Nakai (Araliaceae) is a indigenous medicinal plant growing throughout Jeju-Do in Korea. The plant extract is used in rheumatism, tonic, paralysis and sedative agent. From the roots of A. koreanum, lignan compounds, a diterpenoid, and a polyacetylene compound were isolated and their structures were elucidated by using IR, PMR, CMR and MS spectral data.

섬오가피 추출물의 항암관련 사이토카인 분비활성 (Effects of Acanthopanax koreanum Extracts on Anticancer Related Cytokine Secretions)

  • Stems and roots of Acanthopanax koreanum Nakai were extracted with water and treated on immune cells in order to determine their immunomodulatory activites. Various Th-1 type cytokines were measured using ELISA including interleukin (IL)-2, IL-12, interferon-gamma (IFN-$gamma$), and tumor necrosis factor-alpha (TNF-$\alpha$) secreted by dendritic cells, T-cells, intestinal epithelial cells, natural killer cells, and macrophages. As a result, there was a significant increase in IL-12 and IFN-$\gamma$, secretion, but there was no change in the secretion of TNF-$alpha$. Additionally T-cells slightly increased the secretion of IL-2, but there was a significant increase of IL-2 in intestinal epithelial cells. Therefore, our results suggest that A. koreanum Nakai may act as an immunomodulator by stimulating the cell-mediated immunity which can help the immune system defend against infections or cancer cells.

Modulation of IL-12 and IFN-γ Secretions by Eleutheroside E, Tortoside A, and Syringaresinol from Acanthopanax koreanum Nakai

  • Acanthopanax koreanum Nakai (Araliaceae) is a medicinal plant indigenous to Korea. The root and stem barks of Acanthopanax species have been used as a tonic and sedative as well as in the treatment of rheumatism and diabetes. In our study, three lignans, eleutheroside E (EE), tortoside A (TA), and syringaresinol (SY), were isolated from the stem and root of A. koreanum in an effort to study the immunomodulating effect. We treated natural killer cells and dendritic cells with lignans (EE, TA, or SY), and analyzed their cytokine (IL-12 and IFN-${\gamma}$) secretion. EE, TA, or SY markedly enhanced IL-12 secretion in mouse lymphoid (DC1) and myeloid type (DC2.4) dendritic cells after 48 hr of treatment. There were no significant differences in the cytokine stimulatory effects between EE, TA, or SY. Moreover, treatment of EE, TA, or SY significantly induced IFN-${\gamma}$ secretion by human NK cells (NK92MI) confirmed by ELISA assay. This study suggests that lignans from A. koreanum modulate cytokines, and that such modulation may provide the mechanism of action for many of their therapeutic effects.

  • A new lupane triterpenoid saponin, 3, 11-dihydroxy-lupan-20-en-28-oic acid 28-o-$\alpha$-L-rhamnopyranosyl-(1$\rightarrow$4)-$\beta$-D-glucopyranosyl-(1$\rightarrow$6)-B-D-glucopyranosyl ester, have been isolated from the leaves of Acanthopanax koreanum Nakai (Araliaceae) together with one known flavonol glycoside, rutin. The structure were elucidated on the basis of spectral and chemical evidence.

Evaluation of the Mutagenic Properties of Two Lignans from Acanthopanax koreanum Nakai

  • Acanthopanax koreanum Nakai, a well known traditional herb grown in Jeju Island, South of Korea, has been used as a tonic and sedative agent, as well as in the treatment of diabetes and immune diseases. Mutagenicity of two lignans, syringaresinol and tortoside A isolated from A. koreanum, was assessed using Salmonella/microsome (Ames) test. Tester strains used were Salmonella typhimurium TA98, TA100, TA1535, and Escherichia coli WP2uvrA. The mutagenic activity was determined both in the absence or presence of S9 mixture. As a result, tortoside A did not cause any increase in the number of $his^+$ revertants in S. typhimurium and E. coli WP2uvrA strains in the presence or absence of S9 mix, compared to the controls. Similarly, low concentrations of syringaresinol (750 and 1,500 ${\mu}g$/plate) did not show any mutagenic properties in all bacterial strains, in the presence or absence of S9 mixture. However, in the high concentration of syringaresinol (3,000 ${\mu}g$/plate), the number of revertants were increased in TA1535 strains, in the absence of S9 metabolic activation. Therefore, in vivo experiments such as comet assay are needed to further determine the genotoxic/carciogenic potential of syringaresinol isolated from A. koreanum.

  • This study was conducted to identify the effect of shading and pinching on growth and acanthoside-D content of Acanthopanax divaricatus var. albeofructus and A. koreanum Nakai. Different pinching heights showed no significant differences in terms of plant growth and acanthoside-D content but higher values showed that pinching A. divaricatus at 60 cm and A. koreanum at 30 cm favored good growth and higher fresh weight in the shoots. The content of acanthoside-D was not significantly affected by pinching heights. Also, no significant difference in acanthoside-D content was found between the lower and upper part of plant in the first year. However it was much higher in the lower part than the upper part in the second year, which indicated that the content of acanthoside-D was comparatively high in the lower part where lignification is much advanced. Shading showed benefits in terms of growth of A. divaricatus while only 50%-shading was favorable for A. koreanum to achieve superior growth. Overall, results indicated that shading had favorably affected the growth of the 2 Acanthopanax species while no-shading is better if we opt to achieve higher acathoside D content.

The Glycosides of Araliaceaus Drugs and their Biological Activities

  • Among the Araliaceae plants indigenous to Korea, those whose medicinal usage are comparatively high have been selected in this serial studies. Chiisanoside and acanthoside D were isolated and identified from the Acanthopanax chinensis leaves and root bark. Chiisanoside and acanthoside D have been found to have the lowering S-GPT, S-GOT value and BSP-retention rate and survival rate, anti-histaminic effect in the toxic state through the bio-pharmacological experiments. ${\alpha}-hederin$, hederagenin pentaglycoside were isolated both stem bark of Kalopanax pictum Nakai var. magnificum and Kalopanax pictum Nakai var. Max. respectively. Syringoside, acanthoside D were also isolated from the root bark of Acanthopanax koreanum. The biological activity of ginsenoside $Rb_1$, $Rg_1$, Re were examined. Ginsenoside $Rb_1$, $Rg_1$, Re promotes the antileaking effect in X-ray (Co 60) irradiated toxic state.

오가피잎의 생약학적 연구 (Pharmacognostical Studies on the 'O Ga Pi Ip')

  • 'O Ga Pi Ip' has been used in Korea to cure rheumatism, neuralgia, edema, dematopathy, and impotence. The botanical origin of the crude drug has not been confirmed pharmacognostically. To clarify the botanical origin of 'O Ga Pi Ip', the morphological and anatomical characteristics of the leaves of Acanthopanax species growing in Korea and Japan, A. chi-isanensis Nakai, A. Koreanum Nakai, A. rufinerve Nakai, A. senticosus (Rupr.et Maxim). Harms, A senticosus (Rupr.et Maxim.) for inermis Harms, A. seoulense Nakai, A sessiliflorus (Rupr. et Maxim.) Seem, A. divaricatus Seem, A. sieboldianum Makino were compared. As a result, it was determined that 'O Ga Pi Ip' was the leaf of A. sessiliflorus (Rupr. et Maxim.) Seem, A chiisanensis Nakai, and A. koreanum $Nakai_I$.
