Effect of Acanthopanax senticosus Extracts on Alcohol Degradation and Anti-Inflammatory Activity in Mice (가시오가피 추출물의 알코올 분해 및 항염증 효과)

  • This study was conducted to investigate the effects of Acanthopanax senticosus extracts(ASE) on alcohol administered mice. The administration of Acanthopanax senticosus extracts(60 mg/kg) had beneficial actions toward alcohol degradation in acute alcohol treated mice. In the acute alcohol degradation experiment, serum alcohol concentration were lower 3 and 6 hours after taking ethanol(5 g/kg) in ASE treated mice. The oral administration of ASE showed decreased gastric mucous membrane damage produced in ethanol treated mice. In addition, intraperitoneal(i.p.) administration of ASE showed antiinflammatory effects in inhibition tests of vascular permeability produced by acetic acid. ASE also reduced concentrations of nitric oxide(NO), tumor necrosis alpha(TNF)-${\alpha}$ and interleukin(IL)-6 in macrophages that were activated by LPS. These results demonstrate that Acanthopanax senticosus extracts possesses the potential to stimulate alcohol degradation and inhibit inflammatory effects in mice.

The Effects of Long-term Administration of Panax Ginseng and Acanthopanax on Egg Production of Hens (인삼 또는 오가피 장기투여가 닭의 산란에 미치는 영향)

  • The effects of Panax ginseng and Acanthopanax on egg production of hens are studied. The hens are fed with three dose levels of ethnaol extract of Panax ginseng and Acanthopanax for 24 months. The weight of each egg and number of egg production per month were compared between control group and treated group. The average weight of egg was not influenced by treatment of Panax ginseng and Acanthopanax. The number of egg production in control group was markedly decreased during cold season (from November to March). Panax ginseng and Acanthopanax markedly improve the egg production during cold season compared with control group.

Studies for Component Analysis and Antioxidant Effect, Antimicrobial Activity in Acanthopanax senticosus HARMS (가시오갈피 (Acanthopanax senticosus) 열매의 성분 분석과 항산화 효과 및 항균활성)

  • In the present studγ, we investigated the component analγsis, measurements of antioxidant activities and anti-microbial activities from fruit of Acanthopanax senticosus HARMS in order to detect the biological activities and develop novel functional resources. Different solvents extraction ratios were different according to different solvent extraction of Acanthopanax senticosus HARMS. Composition analysis we highly composed as 72.33%. The minerals of different organs were highly composed of potasium as 5951.3 mg/100g. The monosaccharldes are composed of the arabinose, xylose, mannose, galactose, glucose. Antioxidamt activity was measured in water extracts: $57.3\;{mu}/ml$, ethanol extracts $630.1\;{\mu}g/ml$, methanol extracts: $248.5\;{\mu}g/ml$, 71% ethanol extracts: $198.97\;{\mu}g/ml$, 75% methanol exracts: $96.77\;{\mu}g/ml$, chloroform extracts. $1194.83\;{\mu}g/ml$ at $IC_{50}$ value. The antimicrobial activities were observed in extracts from Acanthopanax senticosus HARMS against am negative bacteria and gram positive bacteria.

A Study on Effect of Acanthopanax sessiliflorum Seeman Water Extract on the Growth of Lactic Acid Bacteria (오갈피(Acanthopanax sessiliflorum Seeman) 추출액이 유산균의 생육에 미치는 영향)

  • This study examined the effect of medicinal herbs on the growth of 3 strains of lactic starter cultures in a MRS broth by adding a 0, 1, 3, 5, and 10% water extract. The pH, titratable acidity and OD of lactic acid bacteria were investigated in order to obtain fundamental knowledge for the development a new product, box thorn yogurt. In the case of adding the Acanthopanax sessiliflorum Seeman water extract, Streptococcus thermophilus was largely activated by the addition of the 5% extract. The growth of Lactobacillus acidophilus was activated by the extracts but it was depressed by the addition of the 3% Acanthopanax sessiliflorum Seeman water extract. Compared with the control, the growth of Bifidobacterium longum was largely activated by the addition of the 10% extract but it was depressed by the addition of the 5% Acanthopanax sessiliflorum Seeman water extract.

Analysis of Flavonoid Contents in the Fruits of Acanthopanax Species using HPLC

  • Analysis of flavonoid contents in the fruits of Acanthopanax species (A. chiisanensis, A. divaricatus, A. koreanum, A. senticosus, and A. sessiliflorus) was conducted by high performance liquid chromatography. A Discovery$^{(R)}$ C18 ($4.6{\times}250$ mm, 5 ${\mu}m$) column was used with a gradient mobile phase of water and acetonitrile (90 : 10 to 60 : 40 for 60 min) and UV detection was conducted at 350 nm. The contents of rutin, hyperin, quercetin, afzelin, and kaempferol were 0.063~0.540, 0.494~7.480, 0.584~0.704, 0.388~0.567, 0.190~0.471 mg/g, respectively, in the fruits of Acanthopanax species. Total content of flavonoids in the fruits of Acanthopanax species was highest in those of A. chiisanensis. Furthermore, hyperin was the most abundant compound in the fruits of Acanthopanax species. Consequently, our results demonstrate that the fruits of Acanthopanax species containing flavonoids have promising potential as a new income source of agriculture and industry in medicinal natural products, health supplements, and beverages.

Study on the Anti-Cancer Effect of Acanthopanax Sessiliflorus (오가피(五加皮)의 항암효과에 관한 연구)

  • Objectives: Hepatoma is a very serious disease in Korea and vvorldwiclc. Hepatitis B vims (HBV) has proved the most significant cause of hepatoma. We canied out this study to investigate the effect of Acanthopanax sessilifloms on inhibiting cell proliferation and DNA synthesis in HepG2.2.15 cell line and on inhibiting phosphorilation of oncogene (MAP kinase) in NIT/3T3-HBx ceIl. Methods: To investigate the anti-cancer effect of Acanthopanax sessiliflorus, we did the CellTiter 96 Aqueous Non-radioactive Cell Proliferation assay (Promega); MTS/PMS assay, [$^3H$]-thymicline incorporation assay, and we measured the gene expression through westem blotting. Results: Acanthopanax sessiliflorus showed an inhibiting effect on the increase of HepG2.2.15 in the NTS/PMS assay. It also showed an inhibiting effect on DNA synthesis of HepG2.2.15 in the [$^3H$]-thymidine incorporation assay. Acanthopanax sessiliflorus showed an inhibiting effect of phosphorilation of MAP kinase in HBV - X genes. too. Conclusions: The results suggested that this herb had an anti cancer effect. We may discover an effective anti-cancer herb medicine through further studies on this herb medicine.

Acanthopanax senticosus Extract Prepared from Cultured Cells Improves Lipid Parameters in Rats Fed with a High Fat Diet

  • Acanthopanax senticosus was grown by a novel, proprietary method, of culturing isolated cells in a bioreactor. An extract from the cells was evaluated for its effect on lipid metabolism in rats fed a high fat diet. Male Sprague-Dawley rats (n=6) were fed either an AIN-76 diet (control, NDCon), control diet plus Acanthopanax senticosus extract (ND+Ex), a modified AIN-76 diet supplemented with 20% beef tallow (high fat, HFCon), or a high fat diet plus Acanthopanax senticosus extract (HF+Ex), for 5weeks. Body weight gain was significantly higher in the HFCon group than the NDCon group. Feed consumption was significantly lower, but energy intake higher, in the groups fed high fat diets compared with the groups fed control diets. Serum HDL-cholesterol concentrations were significantly increased but serum LDL-cholesterol concentrations were decreased in the groups fed the Acanthopanax senticosus extract. Abdominal fat accumulation and serum leptin levels were significantly higher in the HFCon group than the other groups. Carnitine palmitoyltransferase-I (CPT-I) mRNA levels were increased in the groups fed Acanthopanx senticosus extract. These results suggest that supplementation of cell cultured Acanthopanax senticosus extract regulates CPT-I mRNA levels in liver and has an effect on the normalization of lipids in rats fed a high fat diet.

Effects of Acanthopanax senticosus Extract on Macrophage Activity and the Growth of Lactic Starter Culture during Fermentation (가시오가피 추출물이 마크로파지 활성과 우유의 발효 중 젖산균 성장에 미치는 영향)

  • We examined the effect of Acanthopanax senticosus extract on macrophage activity and its effect on the growth of lactic starter culture when added to fermented milk. Greater amounts of Acanthopanax senticosus extract correlated with higher macrophage activity (p<0.05), in particular at $1,000\;{\mu}g/mL$. Water extract of Acanthopanax senticosus resulted in higher levels of nitric oxide (NO) than 70% ethanol extract at $1,000\;{\mu}g/mL$. In contrast, 70% ethanol extract resulted in higher $interleukin-1{\alpha}\;(IL-1{\alpha})$ and Tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$ levels than water extract. The growth of lactic starter culture was inhibited by increasing amounts of Acanthopanax senticosus water extract, resulting in a lower decrease in pH. A stirred type or drink type fermentation process seemed more suitable than a set type for the proper production of Acanthopanax senticosus extract added to fermented milk.

Antioxidative Activity of Water Extract of Different Parts of Acanthopanax divaricatus var, albeofructus (흰털오가피 부위별 물추출물의 항산화활성)

  • Acanthopanax species have traditionally been used as a tonic, a sedative as well as in the treatment of rheumatism, hypertension and diabetes. In the present study, oxidative stress was induced in Vero cells by incubating the cells with glucose and the cell viability was measured by MTT assay. The concentration of glucose which 50% of cell viability was 125 mM $(IC_{50})$ and the cell viability was increased to $87.6{\pm}8.8%$ by treatment of the extracts of Acanthopanax divaricatus var. albeofructus. The antioxidative activity of water extract of different parts of the Acanthopanax plant was investigated by DPPH (1,1-diphenyl-2-picrylhydrazyl) assay, xylenol orange assay, TBARS (thiobarbituric acid reactive substances) assay and enzyme (superoxide anion and catalase) assay. Each extract (leaves, root, stem and fruits) of the plant showed free radical and $H_2O_2$ scavenging activity. The extract also inhibited lipid peroxidation and recovered enzyme (superoxide anion dismutase and catalase) activity in Vero cells treated with glucose.
