Acute and Subacute Effect of Lead acetate on Enzyme Activities and Ultrastructure in Mouse Diencephalone (초산납이 생쥐 간뇌의 미세구조 및 Catecholamine 대사에 미치는 영향)

  • The present experiment was performed to investigate the acute and subacute effect of lead acetate on ultrastructural and biochemical changes in mouse diencephalon. In acute case, mouse were peritoneally injected with lead acetate at a dose of 0.26 mmole/kg body weight, and after treatment, mouse were sacrificed at time intervals of 12, 24, 48, and 96 hours. In subacute case, mouse were injected at doses of 0.07 mmoie/kg B. W. and 0.13 mmole/kg B.W. once at two days, and after treatment, mouse wee sacrificed at 1 week, 2 weeks, and 3 weeks. It was observed that after acute treatment, changes composed of increased monoamine oxidase activity, $Na^{+}-K^{+}$ ATPase activity, decreased $Mg^{2+}$-APTase activity, wrinkled myelin, swollen Golgi apparatus and more dense synaptic vesicle in nerve terminal. After subacute treatment, decreased monoamine oxidase activity, increased $Mg^{2+}$-ATPase, $Na^{+}-K^{+}$ ATPase, lose of myelin, uneven mitochondrial distribution, synaptic vesicular density and edema, but at a higher dose the effect was more severe. Therefore, lead acetate caused abnormal change of diencephalon, and at a subacute, it appears metal accumulative toxicity.

Four-Week Repeated Dose Safety Test of Rhus-II Orally Administrated to Rats (랫드에서 옻나무 추출액(Rhus-II)의 4주 반복 투여 안정성 평가)

  • The purpose of this study was to examine the four week repeated toxicity in Sprague-Dawley rats orally administrated with Rhus-II (water fraction of Rhus Veniciflua). In acute toxicity test, three groups (40 rats of both sex) were administrated different dosages of Rhus-II, 2 g/kg (high dosage group), 1 g/kg, 0.5 g/kg and one group (10 rats of both sex) were received by orally only saline according to the Regulation on Korea Food and Drug Administration, respectively. There was no difference in body weight change, feed intake and water consumption among different dose groups. There was no alteration in relative organ weight by the administration of Rhus-II. No death of abnormal clinical signs was observed during the experimental period. Between the groups orally administered Rhus-II and the control group, there was no statistical significance in urinalysis, hematological test or serum biochemical values. There were no gross findings at final sacrifice. There was no evidence of histopathological alteration mediated by four week treatment with Rhus-II. These results suggest that no observable effect level(NOEL) of the test orally administration was considered to be more than 2g/kg in rats under the conditions employed in this study.

  • Osteochondroma is a common benign tumor of the axial skeleton, especially the distal metaphysis of the femur and the proximal metaphysis of the tibia, however, is occurred rarely on the facial skeleton. Development of the tumor is most frequently seen in the second or third decades of life. Typically, it arises directly from the cortex of the underlying bone, without any intervening zone of abnormal osseous tissue, and is covered by a cap composed of cartilage undergoing calcification. In case of osteochondroma of the mandibular condyle, its clinical features are occulusal change, facial asymmetry, headache, pain and click on temporomandibular joint, mouth opening limitation, and jaw deviation to involved site. This is a case report of a 13-year old woman who had mouth opening limitation and severe temporomandibular joint pain. We obtained successful results with surgical removal of the osteochondroma on the mandibular condyle.

Renin Response to Intravenous Furosemide in Hypertension of Chronic Renal Failure (만성신부전(慢性腎不全)의 고혈압(高血壓)에서 Furosemide 정주(靜注)에 대(對)한 Renin 반응(反應))

  • It has been suggested that plasma renin activity (PRA) and its response to volume depletion may be abnormal in that it shows little or exaggerated change in patients with chronic renal failure and hypertension. Intravenous furosemide stimulation test was performed in 46 control subjects and 51 patients with chronic renal failure and/or malignant hypertension in order to evaluate PRA response. In contrast to the consistent increase in PRA in control subjects (from $2.5{\pm}1.95\;to\;4.5{\pm}2.51ng/ml/hr$), no consistent increase was observed in patients with chronic renal failure, especially in those who showed favorable response to antihypertensive therapy (from $2.5{\pm}2.21\;to\;2.9{\pm}2.46ng/ml/hr$). But poor responder to antihypertensive treatment showed considerably higher PRA before and after furosemide stimulation (from $4.9{\pm}1.96\;to\;6.4{\pm}1.71ng/ml/hr$) than the responder group did. Moreover, this group seemed to retain the ability to increase PRA in response to intravenous furosemide stimulation. Thus it became apparent that responder group was unable to increase PRA normally in response to furosemide as well as volume depletion, while poor responder seemed to retain that ability. Thus intravenous furosemode may serve as a convenient way to differenfiate those who might be benefited by conservative antihypertensive measures from those who would require more drastic measures such as bilateral nephrectomy for their optimal blood pressure control.

  • This research was conducted to develop and analyze a wireless monitoring system for judging if sick or dead layers (SDL) exist in multi-tier layers battery (MLB) by machine vision, and to evaluate the performance between a wired monitoring system and it. This study used the AP (Access Point), the RS-285 to RS-232 converter, RS-232 to Ethernet converter, PICBASIC board and upgraded lump image processing method to change wired monitoring system into wireless monitoring system. The system was tested at a pilot farm and farm layer house. Results showed that monitoring judgement success rate at a pilot farm on normal cage (without SDL) was 82.3% and that on abnormal cage (with SDL) was 87.5%, respectively. And communication performance test results showed at farm layer house was $700{\sim}900$ kbps while equipments operated. There were dropped slightly than performance of wired monitoring system, however, the quantity was too small to make a significant difference of performance of the controling system developed for wireless communication.

  • Recently, intentional arsons for exploitation and wildland fire caused by abnormal change of weather are increasing as well as the damage scale due to such fire in the world. The number of such big wildland fire is also increasing in Korea these days. Fire prevention activity can be said as more important than fire putting-out activity after a fire occurrence for the most effective way to cope with wildland fire, and the research on wildland fire prevention system is what we need to do urgently, In this study I examined and analyzed the experiments and data about the factors influencing wildland fire and stated the dangerousness of each factor for all of the 6 factors and set up the general dangerousness rating applying each factor's contribution to the dangerousness.

Acute Toxicity and Antigenicity of Guamerin (Guamerin의 단회투여독성 및 항원성 평가)

  • This study was carriet out to evaluate the acute intravenous toxicity and antigenicity of Guamerin, newly developed by Mogam Biotechnology Research Institute (MBRI). In acute intravenous toxicity test, ICR mice were administered intravenously with single dose of 1,000mg/kg, and body weights and clinical signs were observed for 14 days. No dead animal, clinical signs, body weight change and abnormal autopsy findings were found in control and Gumerin treated group. Therefore, the 50% lethoal dose (LD50) of Guamerin for ICR mice was more than 1,000mg/kg on intravenous route for male and female. And the antigenic potential of Guamerin was examined by active systemic anaphylaxis(ASA) and passive cutaneous anaphylaxis(PCA) tests. In the ASA test, low and high doses (10 and 100ug/animal, respectiwely) of Guamerin were administed subcutaneously to guinea pigs for 9 times 3 weeks. All experimental groups showed negative responses whereas the positive control group given ovalbumin plus Freunds complete adjuvant (FCA) showed severe anaphylactic responses. PCA test using rats with mice anti-serum against Guamerin, low and high doses(10 and 100 ug/animal, respectively) of Guamerin were administered to mice for 9 times in 3 weeks. The anti-serum against Guamerin was administed intradermally at the back of rats, however, any positive responses were not detected in the experimental groups. These results strongly indicate that Guamerin does not induce IgE production and is not a PCA reaction inducer under these experimental conditions.

A Protective Mechanism in Lungs of Rats Experimentally Infected with Aspergillus fumigatus

  • Aspergillus fumigatus is associated with invasive disease aspergillosis in immunocompromised individuals. The major aim of this study was to investigate the biochemical and immunological responses of male Wistar rats against A. fumigatus experimentally-induced pulmonary fungal infection. Nostril experimental exposure of male Wistar rats to a high dose of A. fumigatus freeze-dried preparation for only 24 hr resulted in a significant increase in levels of catalase, nitric oxide and lipid peroxide in lung homogenates, compared to those of the control animals. However, the oxidative status of the lungs of rats challenged with killed fungus did not change significantly, except for the stimulation in the level of lipid peroxide. IgG level was significantly elevated only in rats that received two low doses of fungus, compared to unexposed animals (p < 0.005). Examining the lung of rats exposed to A. fumigatus revealed no abnormal changes, except for pus in bronchial lumen spaces and per bronchial inflammation. Histologically, large numbers of granuloma cells were evident in the lungs of challenged rats, while no granuloma formation was evident in the lungs of rats exposed to killed fungus.

Dose-Incidence Relationships on the Prenatal Effects of Gamma-Radiation

  • The objective of this investigation was to evaluate of dose-incidence relationships on the prenatal effects of gamma-radiation. Pregnant ICR mice were exposed on day 11.5 after conception, coincident with the most sensitive stage for the induction of major congenital malformations, with 0.5-4.0 Gy of gamma-radiations. The animals were sacrificed on day 18 of gestation and the fetuses were examined for mortality, growth retardation, change in head size and any other morphological abnormalities. With increasing radiation dose, incidence of small head, growth retarded fetus, cleft palate, dilatation of cerebral ventricle and abnormalities of the extremities in live fetuses rose. The threshold doses of radiation that induced cleft palate and dilatation of cerebral ventricle, and abnormal extremities were between 1.0 and 2.0 Gy, and between 0.5 and 1.0 Gy, respectively.

  • We propose a novel monitoring system for diagnosing crack faults of the wind turbine using image information. The proposed method classifies a normal state and a abnormal state for the blade parts of the wind turbine. Specifically, the images are input to the proposed system in various states of wind turbine rotation. according to the blade condition. Then, the video of rotating blades on the wind turbine is divided into several image frames. Motion vectors are estimated using the previous and current images using the motion estimation, and the change of the motion vectors is analyzed according to the blade state. Finally, we determine the final blade state using the Support Vector Machine (SVM) classifier. In SVM, features are constructed using the area information of the blades and the motion vector values. The experimental results showed that the proposed method had high classification performance and its $F_1$ score was 0.9790.
