Decreased Expression of TRPV4 Channels in HEI-OC1 Cells Induced by High Glucose Is Associated with Hearing Impairment

  • Purpose: Previous reports have shown that hyperglycemia-induced inhibition of transient receptor potential vanilloid sub type 4 (TRPV4), a transient receptor potential ion channel, affects the severity of hearing impairment (HI). In this study, we explored the role of TRPV4 in HI using HEI-OC1 cells exposed to high glucose (HG). Materials and Methods: HEI-OC1 cells were cultured in a HG environment (25 mM D-glucose) for 48 hours, and qRT-PCR and Western blotting were used to analyze the expression of TRPV4 at the mRNA and protein level. TRPV4 agonist (GSK1016790A) or antagonist (HC-067047) in cultured HEI-OC1 cells was used to obtain abnormal TRPV4 expression. Functional TRPV4 activity was assessed in cultured HEI-OC1 cells using the MTT assay and a cell death detection ELISA. Results: TRPV4 agonists exerted protective effects against HG-induced HI, as evidenced by increased MTT levels and inhibition of apoptosis in HEI-OC1 cells. TRPV4 overexpression significantly increased protein levels of phosphorylated p38 mitogen-activated protein kinase (p-p38 MAPK), while TRPV4 antagonists had the opposite effect. Our results indicated that TRPV4 is a hyperglycemia-related factor that can inhibit cell proliferation and promote cell apoptosis by activating the MAPK signaling pathway in HEI-OC1 cells. Conclusion: Our results show that the overexpression of TRPV4 can attenuate cell death in HEI-OC1 cells exposed to HG.

Transient Abnormalities on Magnetic Resonance Imaging after Absence Seizures (결신발작 이후 확인된 일시적인 자기공명영상 이상 1예)

  • Magnetic resonance imaging (MRI) is recommended for patients with epileptic seizures to rule out an underlying focal lesion. However, abnormalities in idiopathic generalized epilepsy, including childhood absence epilepsy, cannot usually be identified using brain imaging modalities such as MRI. Peri-ictal MRI abnormalities have been most commonly reported secondary to status epilepticus and are rarely observed in patients with focal seizures and generalized tonic-clonic seizures. Transient peri-ictal MRI abnormalities in absence epilepsy are extremely rare. A five-year-old girl presented with a three-day history of absence seizures that persisted despite continued treatment with sodium valproate. Electroencephalography showed bursts of generalized 3-Hz spike-and-wave discharges, during and after hyperventilation. Abnormal cortex thickening in the left perisylvian region was detected on T2-weighted brain MRI, and cortical dysplasia or a tumor was suspected. The patient started treatment with lamotrigine and was seizure-free after one month. The abnormal MRI lesion was completely resolved at the two-month follow-up. We report on a patient with childhood absence epilepsy and reversible brain MRI abnormalities in the perisylvian region. To our knowledge, this is the first report of transient MRI abnormalities after absence seizures. Transient peri-ictal MRI abnormalities should be considered for differential diagnosis in patients with absence seizures and a focal abnormality on brain MRI.

Comparison of Abnormal Grain Growth Behavior of Lead-Free (Na,K)NbO3-M(Cu,Nb)O3, (M = Ca, Sr, Ba) Piezoelectric Ceramics (비납계 (Na,K)NbO3-M(Cu,Nb)O3, (M = Ca, Sr, Ba) 압전 세라믹의 비정상 결정 성장 거동 비교)

  • NKN [(Na,K)NbO3] is a candidate lead-free piezoelectric material to replace PZT [Pb(Zr,Ti)O3]. A single crystal has excellent piezoelectric-properties and its properties are dependent of the crystal orientation direction. However, it is hard to fabricate a single crystal with stoichiometrically stable composition due to volatilization of sodium during the growth process. To solve this problem, a solid solution composition is designed (Na,K)NbO3-Ba(Cu,Nb)O3 and solid state grain growth is studied for a sizable single crystal. Ceramic powders of (Na,K)NbO3-M(Cu,Nb)O3 (M = Ca, Sr, Ba) are synthesized and grain growth behavior is investigated for different temperatures and times. Average normal grain sizes of individual specimens, which are heat-treated at 1,125 ℃ for 10 h, are 6.9, 2.8, and 1.6 ㎛ for M = Ca, Sr, and Ba, respectively. Depending on M, the distortion of NKN structure can be altered. XRD results show that (NKN-CaCuN: shrunken orthorhombic; NKN-SrCuN: orthorhombic; NKN-BaCuN: cubic). For the sample heat-treated at 1,125 ℃ for 10 h, the maximum grain sizes of individual specimens are measured as 40, 5, and 4,000 ㎛ for M = Ca, Sr, and Ba, respectively. This abnormal grain size is related to the partial melting temperature (NKN-CaCuN: 960 ℃; NKN-SrCuN: 971 ℃; NKN-BaCuN: 945 ℃).

Transient Prolonged Stunning by Dipyridamole Stress Proved by Post-stress(1 hour) and 24 hour Tc-99m-MIBI Gated SPECT (반복 게이트 심근 Tc-99m-MIBI SPECT로 확인한 디피리다몰 부하에 의한 일과성 심근기절현상)

  • We performed 1st day Tc-99m-sestamibi gated SPECT with dipyridamole/rest T1-201 SPECT and 2nd day 24 hour delay T1-201 SPECT/rest Tc-99m-sestamibi gated SPECT in 27 patients with coronary artery disease(24) or having chest pain(3). Stress and rest Tc-99m-sestamibi gated SPECT was acquired at 60min post-injection. A 4-point scoring system(0 to 3 for normal to absent tracer uptake) for 17 segments was used. Wall motion was scored on another 4 point scale(0 to 3 for normal to dyskinesia) in the 1st day post-stress gated and the 2nd day rest gated SPECT. Post-stress gated SPECT showed wall motion abnormality in 94 segments(20%). Fifty-five segments among these 94 showed the same wall motion between post-stress and rest gated SPECT: i.e. 1-1 23 segments, 2-2: 29 segments, 3-3: 3 segments. Remaining 39 segments(41.5%) showed different wall motion between post- stress and rest Tc-99m-sestamibi gated SPECT. Twenty one segments with wall motion abnormality had normal perfusion(rest : 15 segments, 24 hour delay: 6 segments) at either rest or 24 hour delay. Fifteen among these 21 segments showed persistent post-stress and the 2nd day rest wall motion abnormality(persistent stunning). However, in 6 segments with pro-longed (1 hour after stress) stunning, abnormal wall motion did improve in the 2nd day rest Tc-99m-sestamibi gated SPECT(transient prolonged stunning). These 6 segments had normal perfusion at rest(n=4) or at 24 hour delay(n=2). Post stress wall motions showed significantly higher scores in persistent stunning than in prolonged transient stunning(P value<0.05). It was concluded that we could find stunned myocardium with gated Tc-99m-sestamibi SPECT at either post-stress or rest and that some myocardial walls of post-stress 1 hour gated SPECT did not show truly rest wall motion. So, we should be cautious if we use post-stress Tc-99m-sestamibi wall motion to assess rest wall motion.

Regulation of ANKRD9 expression by lipid metabolic perturbations

  • Fatty acid oxidation (FAO) defects cause abnormal lipid accumulation in various tissues, which provides an opportunity to uncover novel genes that are involved in lipid metabolism. During a gene expression study in the riboflavin deficient induced FAO disorder in the chicken, we discovered the dramatic increase in mRNA levels of an uncharacterized gene, ANKRD9. No functions have been ascribed to ANKRD9 and its orthologs, although their sequences are well conserved among vertebrates. To provide insight into the function of ANKRD9, the expression of ANKRD9 mRNA in lipidperturbed paradigms was examined. The hepatic mRNA level of ANKRD9 was repressed by thyroid hormone ($T_3$) and fasting, elevated by re-feeding upon fasting. However, ANKRD9 mRNA level is reduced in response to apoptosis. Transient transfection assay with green fluorescent protein tagged- ANKRD9 showed that this protein is localized within the cytoplasm. These findings point to the possibility that ANKRD9 is involved in intracellular lipid accumulation.

Studies on the Performance Variation of a Variable Speed Vapor Compression System under Fault and Its Detection and Diagnosis (가변속 증기압축 냉동시스템에서 고장시의 성능변화와 고장 감지 및 진단에 관한 연구)

  • An experimental study has been peformed to develop a scheme for fault detection and diagnosis(FDD) in a vapor compression refrigeration system. This study is to analyze fault effect on the system performance and to find efficient diagnosis rules for easy determination of abnormal system operation. The refrigeration system was operated with a variable speed compressor to modulate cooling capacity. The FDD system was designed to consider transient load conditions. Four major faults were considered, and each fault was detected over wide operating load range by separating the system response to the load change. Rule-based method was used to diagnose and classify the system faults. From the experimental results, COP degradation due to the faults in a variable speed system is severer than that in a constant speed system. The method developed in this study can be used in the fault detection of refrigeration systems with a variable speed compressor.

Note on the appearance of Freak Waves from in-situ ocean wave data

  • Freak waves in the ocean are recently drawing much attention as a natural disaster to ocean structures and navigating ships as well. Several observation data, among them the Draupner New Year Wave, show the very impressive feature of Freak waves whose wave height is up to three times as high as the significant wave height of surrounding waves, In addition, Freak wave appears as an isolated very high crest in somewhat stationary random waves of same order in their wavelengths. Bearing such characteristics in mind, one notices its extraordinary steepness. This strongly suggests that Freak wave is not long lived but transient nature on the whole. A great number of studies to explain these natures were published from both theoretical and numerical point of view. However it is not sure if they are applicable to actual ocean environment. In this paper, we deal with the results concerning abnormal and/or Freak waves from in-situ ocean wave data and point out several remarks to the problems lain behind the contributions in this context. A physical experiment is described to reinforce the subject discussed from the observation data.

Serious Blood-Brain Barrier Disruption after Coil Embolization of Unruptured Intracranial Aneurysm : Report of Two Cases and Role of Immediate Postembolization CT Scan

  • Abnormal contrast enhancement on brain computed tomography (CT) scan after diagnostic or interventional angiography is not rare, and has known to be induced by temporary blood-brain barrier (BBB) disruption from contrast media. Furthermore, it has been regarded as clinically subtle, but reported to have no symptom or mild transient symptoms. However, we recently experienced two cases of serious BBB disruption during the acute period after coiling of an unruptured intracranial aneurysm. One patient presented with an unruptured paraclinoid internal carotid artery (ICA) aneurysm on the right and the other with an unruptured right supraclinoid ICA aneurysm. Both patients showed similar findings on immediate postembolization CT scan and clinical courses after coiling. Typical radiological, clinical characteristics of BBB disruption were described. In addition, the role of immediate postembolization CT scan are also discussed.

Calculation of Inrush Current of a Transformer using FEM (유한요소법에 의한 변압기의 돌입전류 계산)

  • The inrush current of transformer can flow when the overvoltage caused by surge or external faults is applied. In this paper, an algorithm for the calculation of this inruch current is proposed. The capacitances of windings are precalculated by using 3 dimensional FEM and are appended to circuit of the transformer. And transient characteristics of the transformer are analyzed by axisymmetric FEM which is coupled magnetic field of transformer and circuit of transformer. When a transformer encounters abnormal voltage, using the proposed method, internal magnetic field of transformer, voltages and currents of windings are calculated.

Wavelet Analysis of Swells in the East Sea (동해 너울에 대한 웨이블릿 분석)

  • Swell data observed in the East Sea in February, 2008 were analyzed using wavelet method. The wavelet analyzed results show detailed time series variation of wave group, peak frequency and spectrum. The comparison of time averaged wavelet spectrum with fourier spectrum turn out to be very similar in terms of spectrum shape and peak frequency evolution but the peak frequency wave energy and the significant wave height show discrepancies. Wavelet analysis can detect the change of spectrum in time as well as in frequency and very efficient to study transient and irregular phenomena such as freak waves and abnormal swells in the ocean. More analysis with more wave data are needed for future application.
