Morphology and histochemistry of gill of the abalone, Haliotis discus hannai were described using light microscopy and scanning electron microscopy (SEM). The abalone has bipectinate gill of protobranch. The cilia and microvilli were well-developed on the free surface of the gill filaments. The epithelial layer of gill filament was simple and composed columnar epithelia and mucous cells. Result of AB-PAS (pH 2.5 and 1.0) and AF-AB (pH 2.5) reaction showed that mucous cells contained mainly acidic carboxylated mucosubstances.
To clarify characteristics of yolk protein of abalone, yolk protein was purified from the ovarian egg extracts of mature female Haliotis discus hannai by a gel chromatography of sepharose CL-4B. From the results of immuno-electrophoresis and Ouchterlony's diffusion test to male and female sera and ovarian egg extracts using antibodies raised against mature female and male sera and male sera and ovarian egg extrascts, it was identified that the mature female serum had female specific serum protein and its antigenecity shared with ovarian egg extracts. A single type of yolk protein was purified from ovarian egg extracts, and it was composed of two subunits. Their molecular weights were estimated to be approximately 166 KDa and 113 KDa by SDSPAGE. The antiserum against yolk proteins cross-reacted with a mature female specific serum protein and extracts of hepatopancreas of vitellogeing females, but did not reacted with extracts of hepatopancreas of mature male.
Basal and heat shock-induced mRNA expression patterns of major heat shock protein (HSP) genes, including those encoding heat shock protein (HSP) 90, HSP70, HSP70-12A, heat shock inducible protein 70 (HSIP70), heat shock binding protein 1 (HSPBP1), HSP60, and HSP40 were examined in the gill and hepatopancreas of 1-year-old diploid and triploid abalone Haliotis discus hannai juveniles. Under non-stimulated conditions at 19℃, triploid abalones displayed, in general, higher mRNA levels of various HSPs (HSP70, HSIP70, HSPBP1, HSP70-12A, and HSP60 in the gill and HSIP70, HSPBP1, and HSP60 in the hepatopancreas) than did communally cultured diploids. Conversely, only the hepatopancreatic expression of HSP70-12A was higher in diploids than in triploids. However, the fold changes in gene expression in response to an acute thermal challenge (elevation from 19 to 30℃) were generally greater in diploids than in triploids, such that the difference in basal expression was diminished, weakened, or even reversed after heat shock treatment. However, unlike other HSP genes, the basal expression of HSP60 (higher in 3N) was more pronounced after heat shock treatment. Collectively, the results of this study suggest that triploid abalones have different capacities for not only basal expression but also the heat-induced expression of HSPs in an HSP member-dependent manner.

The effect of broodstock age on the spawning performance of cultured abalone, Haliotis discus hannai, was assessed for five age groups (3, 4, 5, 6, 7 years old). Spawning performance of the five age groups was assessed in terms of the percentage of spawning abalone, eggs per spawning female, fertilization rates, hatch rates and attachment rates per spawning. The percentage of spawning abalone increased along with broodstock age, reached maximum in female groups of 5- and 6-year age (60%), and in male group of 6-year age (100%), subsequently decreased in 7-year age group. The number of eggs produced per spawning also increased with broodstock age, and the maximum was found in 6-year age. Although the number of spawned eggs for the oldest abalone declined, it still spawned much more eggs than 3, 4, and 5 years old females. The fertilization and hatch rates were obviously larger for 4-6 years old group than the younger and older abalones. The maxima were found in 5-year age group (96.0% and 84.5%), and the minimum were appeared in 7-year age group (79.3% and 58.2%). The attachment rate increased with broodstock age, reached maximum in 6-year age group (33.6%), although the increment gradually declined. The attachment rate for 7-year age group rapidly decreased (16.6%). These results suggested that broodstock age affected the spawning performance of H. discus hannai, which peaked between 5 and 6 years old, and broodstock should be bred during this period for hatchery production.

The purpose of this study was to provide basic information on sexual maturity and reproductive biology for the management of biological resources in abalone Haliotis discus hannai. The nucleus of the oogonium occupied about 42% of the cytoplasm, and had a distinctive basophilic chromatin. The cytoplasm of previtellogenic oocytes was homogeneous and the size of nuclear pores increased. Fine granular and vacuolar yolk granules were observed in the cytoplasm of the initial vitellogenic oocyte. In this stage, the egg stalk and jelly membrane began to develop. The nucleus of the active vitellogenic oocyte was located near the animal pole. Yolk granules were strongly acidophilic. Lampbrush chromosomes were observed in the nucleus and rough endoplasmic reticulum. Annulate lamellae developed in the cytoplasm. The shape of the ripe oocyte was rounded polygonal. The size of ripe oocytes was 202.9±21.40×142.1±18.82 ㎛ and the thickness of the jelly membrane was 10.1±1.52 ㎛. These results show that yolk accumulation in H. discus hannai is based on two methods: exogenous accumulation, through the egg stalk, and endogenous accumulation, through intracellular organelles. Management of biological resources will be necessary when oocytes predominate after the active vitellogenic stage.

In abalones, interspecific hybridization has been suggested as a possible means to increase production and desired traits for the industry. In Korea, Haliotis gigantea is considered a species with a larger size and higher temperature tolerance than H. discus hannai. However, H. discus hannai is considered the most valuable and popular fishery resource due to its better acceptance and higher market prices. Thus, viable interspecific hybrids have been produced by artificial inseminating H. gigantea eggs with H. discus hannai sperm. However, the reciprocal hybrid cross was not successful. In this study, the hybridity and the growth and thermal tolerance performance of the interspecific hybrids were examined. A combination of various assays revealed maximum growth occurrence at 21℃ and the higher growth rate in the hybrids than that of H. discus hannai parent. In addition, the growth and survival at high-temperature (28℃) of the hybrids was equivalent to that of the highly tolerant H. gigantea parent, suggesting new possibilities to overcome the mass mortality in H. discus hannai during high temperature periods of summer season in Korea. Furthermore, the induced interspecific hybrid status was confirmed by the presence of species-specific bands for each parental species of the random amplified polymorphic DNA (RAPD) profiles using universal rice primer (URP), which could be used as speciesspecific markers to distinguish the hybrids and their parental species.
The abalone Haliotis discus hannai, is one of the economically important species in the fisheries industry. Abalone intestines are one of the by-products of its processing. To investigate its bioactive potential, abalone intestine was digested using an in vitro gastrointestinal (GI) digestion system containing pepsin, trypsin, and ${\alpha}$-chymotrypsin. The abalone intestine G1 digests (AIGIDs) produced by the GI digestion system were fractionated into AIGID I (> 100 kDa), AIGID II (10-100 kDa), and AIGID III (1-10 kDa) using an ultrafiltration membrane system. Of the three digests, AIGID II and AIGID III exhibited inhibitory effects against matrix metalloproteinase-2 and -9 (MMP-2, MMP-9) in HT1080 human fibrosarcoma cells. Both fractions potently inhibited gelatine digestion by MMP-2 and MMP-9 treated with phorbol 12-myristate 13-acetate (PMA) and migration of HT1080 cells in dose dependently. Furthermore, AIGID II and III attenuated expression of p65, a component of nuclear transcription factor kappa B. These results indicate that of the abalone intestine digests inhibit MMP-2 and MMP-9. Thus, the AIGIDs or their active components may have preventive and therapeutic potential for diseases associated with MMP-2 and MMP-9 activation in fibrosarcoma cells.
To find out suitable indoor rearing tank shape and stocking density of the abalone Haliotis discus hannai, young abalones were reared in the square or ellipse shape tanks with various stocking densities and were fed dried sea mustard, Undaria Pinnatifida HARVEY for three years. There were no significant differences between the square rearing tank and the ellipse one in growth rate and survival rate. With regard to vertical design of rearing tanks with three floors, the significant differences were not found among culture floors. However, the growth and survival rates of young abalones from the running water system were significantly higher than those of the abalones from the circulating system (P< 0.05). For the stocking density experiment with 1,000, 1,500 and 2,000 individuals of 20 mm young abalone, there was no significant differences in growth and survival rates among these groups. However, in that of 200-400 individuals of 40 mm abalone and 100-300 individuals of 50 mm abalone, the lower stocking density of young abalone showed the higher growth and survival rates. The lower stocking density and the smaller size of young abalones showed the higher daily feeding rate and feed efficiency.

Settlement of larvae, growth, and survival of juvenile abalone (Haliotis discus hannai) were examined after feding of 5 species of benthic diatoms (Cocconeis suctellum, Navicula sp Nitzschia longissima Bacillavia paxillifera, Licmophora flabellata). All of the benthic diatoms were isolated from natural populations on plastic plates for the abalone. The settlement of larvae, shell growth, and survival rate of juvenile abalone varied by diatoms species. The highest settlement rate of larvae was $43.1\%$ with the Cocconeis suctellum diet. Daliy growth rate reached a maximum at 64.1 ${\mu}m/day$ with the Navicula sp. diet and a minimum at 22.4 ${\mu}m/day$ with the Licmophora flabellata diet. Survival rate of the juvenile abalone was highest at $62.0\%$ with the Navicula sp., Cocconeis suctellum , and Nitzschia longissima diets. Survival rate of the Juvenile abalone was significantly higher than the control group with mixed diatom population diets (P<0.05). Therefore, diatom . species composition in diets can be a controling factor for the settlement, growth, and survival rates of Juvenile abalone.
A feeding experiment was conducted using juvenile abalone (Haliotis discus hannai) to investigate the optimum level of supplemental vitamin and/or mineral premixes in a formulated diet for practical feed formulation. Three replicate groups of abalone averaging 100 mg were fed the diets containing different levels of vitamin premix ($0\%,\;0.5\%,\;1.0\%$ and $2\%$) and /or mineral premix($0\%,\;2\%,\;4\%$ and $6\%$) for 4 months. Survival rates of abalone showed no distinctive trend by the levels of dietary vitamin premixes. Weight gain, shell growth and soft body weight of abalone were not significantly affected by the different dietary vitamin and/or mineral premixes (P>0,05). Moisture, protein and lipid contents of soft body were influenced by experimental diets (P<0.05) but these differences also had no definite trends by levels of vitamin and/or mineral premixes. These data indicate that supplemental vitamin or mineral premix is not needed in this formulated diet, therefore price of diet for juvenile abalone could be reduced.
