Possibility whether lead ingestion can cause selective toxicity to central serotonergic nervous system in rats was tested. Three groups of wistar rats; 1)Control, 2) Low dose and 3) High dose groups, were prepared. In prenatally lead-exposed rats, until parturition from dams, rat pups were intoxicated via placenta of mother rats having received drinking water containing either 0%(control ), 0.05%(low dose) or 0.2%(high dose) of lead acetate respectively, In postnatally lead-exposed rats, right after parturition from dams rat pups received drinking water containing either 0% (control), 0.05%(low dose) or 0.2%(high dose) of lead acetate. At 2, 4, 6 and 8 weeks of age, tryptophan hydroxylase (TPH) activity and Na$\^$+//K$\^$+/-ATPase activity were measured in 4 areas of rat brain; Telencephalon, Diencephalon, Midbrain and Pons/Medulla. TPH activities were assayed by modified method of Beevers et al. (1983) using L-(5-$^3$H)-tryptophan as substrate. TPH activity was determined as a criterion of lead poisoning to central serotonergic nervous system and Na$\^$+//K$\^$+/-ATPase activity as a criterion of non specific lead poisoning to any kinds of tissues. Selective toxicity of lead poisoning to central serotonergic nervous system was evaluated by the changes of TPH activities without concomitant changes of Na$\^$+//K$\^$+/-ATPase activities. In prenatally lead-exposed rats. this selectivity was found in Telencephalon (2 weeks of age), Diencephalon/Midbrain (2 weeks of age), Midbrain (4 and 6 weeks of age), Pons/Medulla (2, 4 and 6 weeks of age) In rats exposed to low dose of lead and Pons/Medulla (2 weeks of age) to high dose of lead. In postnatal Iy lead-exposed rats, this selectivity was found in Telencephalon (8 weeks of age), Diencephalon(8 weeks of age), Pons/Medulla (6 and 8 weeks of age) in rats exposed to low dose of lead and Pons/Medulla (8 weeks of age) to high dose of lead. These results suggest that lead poisoning may exhibit selective toxicity to central serotonergic nervous system.
Effects of sonication on water-solubilization of myofibril from breast muscle of spent hen effects investigated in this study. To evaluate effect of salt concentration and pH, salt concentration was varied with range from 0.1 to 0.8 M, and pH was varied with range from 6.0 to 8.0. Solubility, SDS-PAGE, viscosity and ATPase activity of sonicated myofibril were measured. Solubility of myofibrillar protein containing 0.1 M NaCl at pH 8.0 after sonication was above 90%. Main components of soluble protein by SDS-PAGE were myosin heavy chain and actin. That is, it indicated breaking of myofibril structure by sonication. Also, viscosity of soluble protein increased, but Ca- and Mg-ATPase activities decreased by increasing sonication time. From these results, we concluded that most of myofibrillar proteins were denatured by sonication.

A study was conducted to investigate cryoprotectant effect of commercially produced oligosaccharides (IMO: isomalto-oligosaccharides, FO: fructo-oligosaccharides and GO: galacto-oligosaccharides) on beef protein and to compare their effectiveness to sucrose or a mixture of sucrose and sorbitol on freezing. The optimal addition level of cryoprotectants was determined by measuring $Ca^{2+}$-ATPase activity of sample treated with different concentration (0 to 12%) after freeze-thaw cycle. Since the stabilization effect was not dramatically increased above 8% sugar concentrations, the 8% was determined as an usage level. During frozen storage (at -18$^{\circ}C$ for 12 week), commercially produced oligosaccharides showed lower cryoprotection ability than sucrose but higher than sucrose+sorbitol as measured by protein solubilities and $Ca^{2+}$-ATPase activities.
The selective toxicity of lead was tested in central catecholaminergic nervous system of postnatally lead exposed rats. Three groups of animals were prepared; 1) rats exposed to low dose of lead (0.05%PbAc); 2) rats exposed to high dose of lead(0.2%PbAc); 3) age-matched normal control rats. At2, 4, 6 and 8 weeks of age brain and body weight gain, and lead concentrations in brain tissues were measured. At the same ages tyrosine hydroxylase and Na-K ATPase activities were measured in the 4 brain areas of each animal. Body weight gain was decreased after 6 weeks of age in rats exposed to high dose of lead. Concentrations of lead in whole brain tissues were increased from 0.37 to 0.83 (ng/mg wet tissue) in these animals. in lead exposed rats, tyrosine hydroxylase activities were higher but Na-K ATPase activities were lower than those of age-matched control animals. Brain areas where tyrosine hydroxylase activities were detected without concomitant changes of Na-K ATPase activities were pons-medulla (2 weeks of age) and telencephalon (6 weeks of age) in rats exposed to low dose of lead, and those in rats exposed to high dose of lead were midbrain (4 and 6 weeks of age). These data indicate that catecholaminergic nervous system in the brain areas described above could selectively be affected by lead.
Aging effect of vacuum-packaged beef loins was investigated at $0\pm1^{\circ}C$ for 8 weeks. Hardness, chewiness, and shear force value of beef loins decreased steadily for the first 5 weeks of chilling period, but thereafter they did not changed significantly. Myofibrillar protein extractability, Mg-ATPase activity, and myofibrill fragmentation index of beef loins were increased for the first 6 weeks, and did not change for the rest period The Hunter's value in $L^{\ast},\;a^{\ast}\;and\;b^{\ast}$ of beef loin showed that no significant changes had occurred over the whole storage time. The palatability of beef loin was improved progressively for the first 5 weeks, but it became inferior thereafter. It can be concluded that the storage time to obtain optimum aging effect for the vacuum-packaged beef loin would be 5 weeks at $0^{\circ}C$ at the longest.

To treat peptic ulcer diseases, many potent proton pump inhibitors have been developed for suppressing the gastric acid secretion in clinical patients. However, most of these agents have common irreversible mechanisms against H$\^$+/, K$\^$+/-ATPase which might be the cause of hypergastrinemia and ECL cell hyperplasia. Therefore, the development of new reversible inhibitors is prompted. In this study, we investigated the inhibitory mechanism of a newly synthesized proton pump inhibitor, YJA20379-8 using lyophilized hog gastric microsomes. YJA20379-8 inhibited K$\^$+/-stimulated H$\^$+/K$\^$+/-ATPase activity uncompetitively with respect to K$\^$+/, and in the other hand, showed competitive inhibitory pattern with ATP, respectively. From these data, we suggest that YJA20379-8 may be a proton pump inhibitor with a new inhibitory mechanism.
The RecA protein of Deinococcus radiodurans is essential for the extreme radiation resistance of this organism. The central steps involved in recombinational DNA repair require DNA-dependent ATP hydrolysis by recA protein. Key feature of RecA protein-mediated activities is the interactions with ssDNA and dsDNA. The ssDNA is the site where RecA protein filament formation nucleates and where initiation of DNA strand exchange takes place. The effect of sequence heterogeneity of ssDNA was examined in this experiment. The rate of homopolymeric synthetic ssDNA-dependent ATP hydrolysis was constant or nearly so over a broader range of pHs. For poly(dT)-dependent ATP or dATP hydrolysis, rates were generally faster, with a broader optimum between pH 7.0 and 8.0. Activities of RecA protein were affected by the ionic environment. The ATPase activity was shown to have different sensitivity to anionic species. The presence of glutamate seemed to slimulate the hydrolytic activity. Dr RecA protein was shown to require $Mg^{2+}$ ion greater than 2 mM for binding to etheno ssDNA and the binding stoichiometry of 3 nucleotide for RecA protein monomer.

For the purpose of investigate the inhibition effects of Injinchunggantang on Hepatitis and C Virus. The inhibition experiments were as follows : 1. The inhibition effects of Hepatitis B surface antigen production was remarkably increased in the pretreated group of Injinchunggantang compared with control group. 2. Amplified PCR products of HBV-DNA in culture media was considerably decreased in the pretreated group of Injinchunggantang compared with control group. 3. Results of ATP quantitation and ATPase inhibition percent was increased pretreated group of Injinchunggantang. Also ATPase activity of HCV helicase inhibition assay was increased with a dose dependent manner. These results may suggest that Injinchunggantang will have the inhibition effects of Hepatitis B and C Virus.
$Na^{+}$ balance was studied in Rana temporaria, which hibenates in fresh water in the winter time. $Na^{+}$ uptake rate, skin $Na^{+}$ loss rate, urinary $Na^{+}$ loss rate and $Na^{+}-K^{+}$ adenosine triphosphatase(ATPase) were measured at two different temperatures $1{\sim}2^{\circ}C\;and\;20{\sim}24^{\circ}C$ respectively. The results obtained were as follows: 1. $Na^{+}$ uptake rates in the frog in an artificial Pond water (APW) were found to be $8.28{\pm}0.73\;and\;2.19{\pm}0.37\;{\mu}Eq/g/day\;at\;20{\sim}24^{\circ}C\;and\;1.0{\sim}2.5^{\circ}$ respectively. 2. $Na^{+}$ loss rate through the frog skin to APW were found to be $4.26{\pm}0.72\;and\;0.93{\pm}0.21\;{\mu}Eq/g/day$ at the same temperatures. 3. Mean rates of urinary $Na^{+}$ loss at $20{\sim}24^{\circ}C\;and\;3{\sim}4^{\circ}C$ were found to be $3.02{\pm}0.73\;and\;0.78{\pm}0.13\;{\mu}Eq/g/day$ respectively. 4. The activities of $Na^{+}-K^{+}$ activated ATPase of frog skin fragments were found to be $258{\pm}39.4\;and\;49.6{\pm}7.1\;{\mu}M\;Pi/g$ protein/hr at $24^{\circ}C\;and\;2^{\circ}C$ respectively. From the above results, it may be concluded that frogs can take up enough $Na^{+}$ through the skin from APW exceeding skin loss Plus urinary loss at $1{\sim}2^{\circ}C$. It is suggested that $Na^{+}$ transport across frog skin is closely related with $Na^+-K^+$ ATPase since $Q_{10}\;of\;Na^{+}$ uptake is much similar to that of the activities of $Na^{+}-K^{+}$ ATPase.
Using a PEG- dextran two phase partition method, plasma and intracellular membrane separated from microsomal membrane of canola (Brassica napus) leaves have been fractionated by centrifugation. $K^{+}$- ATPase specific activity in the plasma membrane (U$_2$ phase) of plants grown at $25^{\circ}C$ and 1$0^{\circ}C$ were 6.6 and 4.6 times, respectively that of the microsomal membrane. Plasma membrane had a lower cytochrome- c- oxidase specific activity than the microsomal membrane or intracellular membrane, while intracellular membrane (L$_2$ phase) had a high cytochrome-c- oxidase but little $K^{+}$- ATPase specific activity. The plasma membrane of canola grown at 1$0^{\circ}C$ had higher 18:3 to 18:2 (linolenic to linoleic acid) ratio (29.2% ) and higher degree of unsaturation than that grown at $25^{\circ}C$ The double bond index of plasma membrane from canola grown at 1$0^{\circ}C$ increased by 8.9% relative to canola grown at $25^{\circ}C$. Similar, intracellular membrane increased by 19.7% at 1$0^{\circ}C$. Canola grown at 1$0^{\circ}C$ was lower in chlorophyll contents (17.3%) than that grown at $25^{\circ}C$. These changes in fatty acid unsaturation were attributable largely to change in Cl8 fatty acid, with major changes occurring in linolenic acid (18 :3) which might have a physiological role of membrane to adaptation on low temperature.ure.
