Glucose Deprivation and Immunostimulation Induced Death in Rat Primary Astrocytes is Mediated by Their Synergistic Effect on the Decrease in Cellular ATP Level

  • In this study we investigated whether ATP loss was involved in the potentiated death of immunostimulated rat primary astrocytes in glucose-deprived condition. Rat primary astrocytes immunostimulated with LPS plus IFN-${\gamma}$ for 48 h underwent death upon glucose deprivation, which dependent on the production of peroxynitrite. Intracellular ATP level synergistically decreased by glucose deprivation in immunostimulated astrocytes but not in control cells, and the loss of ATP occurred well ahead of the LDH release. The synergistic cell death and ATP loss by immunostimulation and glucose deprivation were inhibited by iNOS inhibitor (L-NAME and L-NNA) or peroxynitrite decomposition catalyst (also a superoxide anion scavenger), Mn(III)tetrakis(N-methyl-4'-pyridyl)porphyrin (MnTMPyP). Exogenous addition of peroxynitrite generator, SIN-l timedependently induced ATP loss and cell death in the glucose-deprived astrocytes. Depletion of intracellular glutathione (GSH) and dis겨ption of mitochondrial transmembrane potential (MTP) were also observed under same conditions. Supply cellular ATP by the addition of exogenous adenosine or ATP during glucose deprivation inhibited ATP depletion, GSH depletion, MTP disruption and cell death in SIN-l treated or immunostimulated astrocytes. This study showed that perturbation in the regulation of intracellular ATP level in immunostimulated astrocytes might make them more vulnerable to energy challenging stimuli.

활성화된 사람 중성 백혈구에서 ATP와 Adenosine 처리에 따른 생화학적 반응의 변경 (Alteration of Biochemical Responses in Activated Human Neutrophils by ATP and Adenosine)

  • 안정 상태 및 활성화된 중성 백혈구에서 ATP는 superoxide 라디칼 생성을 자극하였으나 adenosine은 약간 억제하였다. ATP에 대한 활성화된 중성 백혈구의 반응이 안정상태의 중성 백혈구에서보다 크게 나타났다. 칼슘이 제거된 반응액에서 superoxide 라디칼 생성에 대한 adenosine의 억제효과가 관찰되었으나 ATP는 영향을 주지않았다. Superoxide 라디칼 생성에 대한 ATP의 자극 효과는 adenosine에 의하여 용량에 따라 억제되었다. ATP와 adenosine은 NADPH oxidase 활성도에 영향을 주지 않았다. ATP 또는 adenosine에 의한 superoxide 라디칼 생성의 변경은 다른 triphosphate nucleotide나 nucleoside에 의한 것보다 현저하였다. 활성화된 중성 백혈구에서 ATP와 ADP는 칼슘이온의 흡수를 더 자극하였고 세포질 유리 칼슘농도를 증가시켰으나, adenosine은 칼슘이온의 이동을 억제하였다. APT에 의한 세포질 유리 칼슘이온 농도의 증가는 verapamil에 의하여 효과적으로, tetrodotoxin에 의하여 약간 억제되었다. ATP에 노출된 활성화된 중성 백혈구에서의 superoxide 라디칼 생성에 대한 verapamil 과 tetrodotoxin의 억제 효과는 ATP의 영향이 없는 활성화된 중성 백혈구에서보다 크게 나타났다. Tetraethylammonium chloride는 superoxide 생성에 뚜렷한 영향을 미치지 못했다. CCCP, 2,4-dinitrophenol, diphenylhydantoin과 procaine은 활성화된 중성 백혈구에서 superoxide 라디칼의 생성을 억제하였다. 이들 가운데 CCCP만이 ATP의 자극 효과를 억제하였다. ATP는 활성화된 중성 백혈구에서의 sulfhydryl기의 손실을 더 자극하였으나 adenosin의 영향은 관찰되지 않았다. 이상의 결과로부터 중성 백혈구의 기능적 반응은 부분적으로 purine에 의하여 조절될 것으로 시사되었다. ATP와 adenosine은 칼슘 흡수와 그리고 아마도 세포막 인산화 반응 및 용해성 sulfhydryl기의 산화에 대한 영향을 통하여 활성화된 중성 백혈구의 반응을 더 변경 시킬 수 있을 것으로 추정된다.

ATP meter를 이용한 초음파 탐촉자의 오염도 분류 (Ultrasound Probe Contamination Classification using ATP Meter)

  • In this study, the contamination level was measured using an ATP meter using adenosine triphosphate bioluminescent material to find effective infection control to compensate for the disadvantages of the microbial culture method used for hygiene control of ultrasound probe. The convex probes were selected from six ultrasound probe in the hospital, and the samples were taken in real time before and after cleaning to check the contamination of the probe. In order to classify the pollution degree using the APT meter was classified by category. A total of 78 samples were collected from the ultrasound probe. When the pollution levels before and after cleaning were classified by category, 76.6% of the samples were classified into category 3·4 before cleaning, but they decreased to 23.3% after cleaning. 13.3% before cleaning was in category 1, but increased to 43.3% after cleaning. By classifying the pollution level, it was confirmed that the pollution level was significantly reduced by category. Until now, there was no suitable criterion for determining the contamination level by using ATP meter in medical machines where sample area is small and reused. In this study, criteria for each category were set to measure the contamination level of ATP meter suitable for small sample area such as ultrasound probe, so that contamination level could be determined in real time at the site. Therefore, it is considered that hygiene management for ultrasound probe can be more actively performed.

한국형 틸팅열차 ATP장치 예비위험원분석에 관한 연구 (A Study on the PHA of ATP Equipment for the Korean Tilting Train)

  • In this paper, we derive the hazard which is a criterion of safety assurance by PHA for the ATP equipped in Korea Tilting Train. The ATP onboard equipment in Korea Tilting Train was introduced as ETCS Level l(which is ETCS specification). Also, the ATP trackside equipment was introduced as a ETCS SRS(System Requirement Specification) 2.2.2 and it has a target for commercial operation in 2009 late. In IEC 62278(EN50126), it recommend of PHA to derive a hazard of target system in concept establishment step and estimate if it can control as a tolerable level and then derive safety case to control the hazard by lifecycle. Therefore, we indicate the hazard using criterion of safety assurance by PHA using FRS (Functional Requirement Specification) 5.0. which is a upper criterion than SRS. The hazard of ATP onboard equipment derived in Korea Tilting Train will be able to use as ATP functional hazards of unified onboard equipment in G7 which is a multiple unit and preliminary hazards of ATP onboard equipment in NEL drived by Korail.

정상돈육과 모의 열등돈육의 사후 초기 ATP 및 관련 뉴클레오타이드 함량 변화 (Changes in Early Postmortem Contents of ATP and Other Nucleotides in Normal and Poor Quality-simulated Pork)

  • 도체의 반을 전기로 자극하여 만든 모의 열등육과 전기처리하지 않은 대조군 정상 돈육을 실험육으로 준비하고 ATP 및 관련 뉴클레오타이드의 사후 초기 함량 변화를 추적하였다. 정상육의 경우 사후 2시간 이내에 ATP 함량은 5.00에서 2.04 ${\mu}mole$/g으로 약 60% 감소하였고 전기자극 열등육은 단지 전기자극 만으로 ATP가 25% 감소하였으며 자극 후 2시간 이내에 ATP의 잔존 함량은 거의 고갈되어 초기함량의 5% 정도로 감소하였다. IMP의 함량은 정상육의 경우 사후 2시간 이내에 0.49에서 3.17 ${\mu}mole$/g으로 급증하였고 같은 시간 동안 전기자극한 모의 열등육은 6.64 ${\mu}mole$/g까지 증가하여 ATP보다도 높은 함량을 갖는 가장 주된 뉴클레오타이드로 등장하였다. 정상육의 ADP 함량은 사후 2시간 이내에 1.45에서 0.67 ${\mu}mole$/g으로 감소하였고 전기자극은 이 감소폭을 더욱 증가시켰다. AMP의 함량은 4개의 뉴클레오타이드 함량 중 가장 낮았으며 사후 2시간 이내에 함량이 0.16에서 0.31 ${\mu}mole$/g으로 증가하였고 전기자극은이 증가 추세를 더욱 가속화하였다. 종합적으로, 전기자극한모의 열등육의 사후 초기 ATP 감소 속도는 매우 빨라서 그 함량이 정상육보다 훨씬 낮은 수준을 보였고 다른 뉴클레오타이드들의 증감 범위와 속도도 정상육에 비해 훨씬 높았음을 확인하였다.

적혈구를 이용한 Daunorubicin의 배송시스템 (Delivery System of Daunorubicin by Red Blood Cells)

  • Drug delivery system by the use of red blood cells was established to sustain the release of drugs in the circulatory system by the intravenous injection. The entrapment method by the preswelling technique was re-examined and evaluated for searching the new entrapping conditions without hemolysis. The addition of 4 volume of $0.6{\times}\;hank's$ balanced salt solution (HBSS) into 1 volume of 50% red blood cells suspension did not induce the hemolysis and change the hematocrit level in this experimental condition (within 15 min). Most of daunorubicin could be entrapped into red blood cells within 15 min. While the intracellular adenosine triphosphate (ATP) level followed by the entrapment was reduced to 86% of normal ATP level, the membrane fluidity and the shape factor of red blood cells were not altered. The release rate of daunorubicin from red blood cells was affected by the hemolysis under this condition. To maintain the intracellular ATP in red blood cells, the new reaction buffer was made With the addition of ATP and sodium pyruvate during the entrapment procedure because the hemolysis during the release test would reflect the loss of intracellular ATP that might result in the decrease of the viability in vivo. The addition of ATP raised the intracellular ATP level, which protect the hemolysis during the release test.

  • 본 논문에서는 철도신호시스템분야에서 중요한 신뢰성과 안전성 수준을 국제규격에 부합할 수 있도록 하기 위한 접근방법을 모색하며, 국철 구간의 차상신호시스템 (ERTMS/ETCS Level 1)도입에 따라 현실적이고 접근 가능한 구체적인 활동 사항들을 프로젝트 수행과 연관하여 기술한다. 이에 따라 시스템 전반에 걸친 RAMS 및 안전성 평가를 통한 인증활동에 대한 부분을 제시하며, 국내 철도 신호시스템 적용을 위한 차상신호 시스템(ERTMS/ETCS Level 1)에 있어 차상장치(On-Board)를 중심으로 품질 및 안전 무결성(SIL) 수준 확보에 따른 안전성 확보방안의 구체적인 위험도분석 및 평가 활동을 제시 하고자 한다

적혈구내 비용혈 약물봉입과 약물방출 (Nonhemolytic entrapping method into red blood cells and its release pattern)

  • Drug delivery by red cells was established to maintain the release of drugs in the blood. The entrapment method by amphotericin B was re-examined and evaluated for obtaining the suitable entrapping conditions without hemolysis. The amphotericin B treatment below $10{\mu}g/ml$ induced the non-hemolysis to entrap daunorubicin into red cells within 10min. Under these conditions intracellular ATP level was decreased as $18\%$. Membrane fluidity and the shape factor of red cells were maintained. To maintain intracellular ATP, ATP and sodium pyruvate were added during the entrapment procedure because hemolysis during the release test would reflect the loss of intracellular ATP that would be postulate the decrease of the viability invivo. Consequently, the addition of ATP in the reaction solution can raise the intracellular level of ATP.

식물엽의 ATP함량에 미치는 Pb과정 및 P결핍의 영향 (Effects of Pb-Surplus and P-Deficiency on ATP Content in Plant Leaves)

  • Kidneybean(Phaseolus vulgaris L.) and buckwheat(Fagopyrum esculentum M nch) seedlings grown with Hoagland solution to a height of 7 to 10 cm in the earthen pot containing sand were used for experimental plants. One group of the experimental plants was irrigated with Hoagland solution composed of various Pb-concentratons of 0 to 1000ppm containing 30ppm, and the other group was irrigated with Hoagland solution composed of various P-concentrations of 0 to 1,000ppm containing 30ppm Pb for a month during June and July in 1979. By Pb-surplus over 100ppm, the margins of buckwheat were curled down and turned into yellow-brown chlorosis, but the leaves of kidneybean were become dark-green at an early stage, and then developed chlorosis, finally shedded earlier than the control. Pb-toxicity was similar to the symptoms of P-deficiency. The ratio, 3/1 of chlorophyll a/b at an early stage, was unaffected by Pb-surplus or P-deficiency, but at the later stage it was altered. ATP content with Pb-surplus of 100ppm was decreased by 69% in kidneybean leaves and by 38% in buckwheat leaves, and it with P-deficiency was decreased by 75% in kidneybean leaves and by 43% in buckwheat leaves. In the assay of ATP content of the leaves at intervals of three hours for one day of July in the year, the rhythms of ATP level were unaffected by light or temperature although the amplitude of the level was modified. The rhythms of Pb-surplus and P-deficiency plants were observed at the lower range of ATP content than that of control.

ATP and Purinergic Receptor Agonists Stimulate the Mitogen-Activated Protein Kinase Pathway and DNA Synthesis in Mouse Mammary Epithelial Cells

  • The effects of adenosine 5'-triphosphate (ATP) and ATP analogs, P/sub 2y/ purinoceptor agonists, on growth of normal mouse mammary epithelial cells (NMuMG) were examined. Cells were plated onto 24 well plates in DMEM supplemented with 10 % fetal calf serum. After serum starvation for 24 hours, ATP, P/sub 2y/ purinoceptor agonists (AdoPP[NH]P, ATP-α-S, ATP-γ-S, β, γ-me-ATP and 2me-S-ATP), P/sub 2u/ purinoceptor agonist (UTP) and P/sub 2y/ purinoceptor antagonists (Reactive Blue 2, more selective to P/sub 2y/ receptor than PPADS; PPADS) were added. DNA synthesis was estimated as incorporation of 3H-thymidine into DNA (1 hour pulse with 1 μ Ci/ml, 18~19 hours after treatment). ATP, Adopp[NH]P, ATP-α-S or ATP-γ-S, significantly increased DNA synthesis at 1, 10 and 100 μM concentrations with dose-dependency (P<0.05), and the maximum responses of ATP and ATP analogs were shown at 100 μM concentration (P<0.05). The potency order of DNA synthesis was ATP≥ATP- γ -S>Adopp [NH]P>ATP-α-S. β, γ -me-ATP, 2me-S-ATP and UTP did not increase DNA synthesis. In autoradiographic analysis of percentage of S-phase cells, similar results were observed to those of DNA synthesis. Addition of 1, 10 or 100 μM Reactive Blue 2 or PPADS significantly decreased ATP (100 μM)-induced DNA synthesis, however, PPADS was less effective than Reactive Blue 2. In Elvax 40P implant experiment, ATP directly stimulated mammary endbud growth in situ suggesting the physiological regulator of ATP in mammary growth. ATP 100 μM rapidly increased MAPK activity, reaching a maximum at 5 min and then gradually decreasing to the base level in 30 min. ATP analogs, Adopp[NH]P and ATP-γ-S also increased MAPK activity, however, β, γ-me-ATP and 2me-S-ATP did not. The inhibitor of the upstream MAPK kinase (MEK), PD 98059 (25 μM), effectively reduced ATP (100 μM) or EGF(10 ng/ml, as positive control)-induced MAPK activity and DNA synthesis (P<0.05). These results indicate that ATP-induced DNA synthesis was prevented from the direct inhibition of MAPK kinase pathway. Overall results support the hypothesis that the stimulatory effects of normal mouse mammary epithelial growth by addition of ATP or ATP analogs are mediated through mammary tissue specific P/sub 2y/ purinoceptor subtype, and MAPK activation is necessary for the ATP-induced cell growth.
