Effect of Cold Brine Immersion Time on Sliced Raw Fish Texture (생선회의 육질향상에 미치는 저온 브라인 침지시간의 영향)

  • The purpose of the present study was to investigate physicochemical changes in olive flounder and korean rockfish muscle subjected to the cold brine, and to find the optimal immersion condition. Effects of various immersion conditions in a sliced raw fish quality were investigated in the rigor index, breaking strength, ATP related compounds, and lactate accumulation in the muscle. The onset of rigor-mortis was accelerated by the increase of immersion time. Also, the time reaching full rigor was shortened remarkably too. However, the rigor index of samples immersed in the cold brine decreased more than that of samples killed instantly. Rigor index was the highest in samples killed instantly, followed by 2.5 min, 5min, 7.5 min, 10 min and 15 min in order. Breaking strength of all samples immersed in the cold brine decreased significantly after reaching the maximum values. The content of ATP related compounds in olive flounder, korean rockfish were $12.0{\mu}mole/g$ and $8.3{\mu}mole/g$, respectively. However, ATP decreased in the samples immersed in the cold brine. The content of ATP was lower, at the optimal condition and then it was increased. Also, lactates increased, at the optimal condition and then it decreased, in contrast with the ATP. The results suggested that the optimal immersion condition of olive flounder and korean rockfish was at -${12.5}^{\circ}C$ for 5min.

Development of sea water cooling apparatus for fish hold storage (어창용 해수 냉각장치 개발에 관한 연구)

  • The objective of this study was to manufacture the sea water cooling apparatus for fish hold storage to extend the freshness of fish. Using the sea water cooling apparatus manufactured, it makes observations on the physicochemical and sensual properties of mackerel such as ATP and its related compounds, K-value, VBN, TMA, TBA, salt-concentration and skin color through storage. The results of this investigation showed that the values of physicochemical properties such as ATP and its related compounds, K-value, VBN, TMA, TBA increased gradually during storage in sea water cooling apparatus manufactured and the changes of skin color was inhibited also. From these results, it could be suggested that developed sea water cooling apparatus is effective in shelf-life extension of fishes.

Comparison of Extractive Nitrogenous Constituents in the Three Species of Raw Bloody Clams, Scaphrca broughtonii, S. subcrenata, and Tegillarca granosa Extracts (피조개, 새고막 및 고막의 함질소 엑스성분 조성 비교)

  • Extracts of raw bloody clams, broughton's ribbed ark (Scaphrca broughtonii), subcrenated ark (S. subcrenata), and granulated ark (Tegillarca granosa), were analyzed and compared foe extractive nitrogen, free amino acids, combined amino acids, ATP and related compounds, quaternary ammonium bases, and guanidino compounds. The contents of extractive nitrogen in broughton's ribbed ark, subcrenated ark, and granulated ark were 479, 506, and 432 mg/100 g, respectively. Twenty-eight or twenty-nine types of free amino acids were detected in all three extracts, among which taurine, ${\beta}-alanine$, glutamic acid, and alanine were the major ones. The composition of the major extractive components such as free amino acids, combined amino acids, ATP and related compounds, TMAO, TMA, and creatine in the extracts were similar among the extracts, but their contents were different.

Extractive Nitrogenous constituents of Echiuroid Urechis unichinctus (개불의 함질소 엑스성분)

  • In order to investigate the composition and the actual status of extractive nitrogenous compounds in the fresh 'Gae-bul' (echiuroid), a kind of echiurida (Urechis unicinctus), the extract was analyzed separately into extractive nitrogen, free amino acids (FAAs), oligopeptides, nucleotides and related compounds, quaternary ammonium bases, and guanidino compounds, using specimens collected at fish market in April 1988. The extractive nitrogen of echiuroid was $601{\sim}610mg/100g$. Thirty-two kinds of FAAs were found, and the total of them in it was $2,437{\sim}2,609\;mg$. Glycine, alanine, taurine, and serine were the major FAAs in the echiuroid extracts. The large amount of glycine $(1,075{\sim}1,171mg)$ was noted in the extract. The sum of ATP and its related compounds was $3.04{\sim}3.12\;{\mu}mol/g$, and predominant compound was the AMP. Besides, CTP, GTP, UTP and their related compounds were also detected, and the total amount of them was $1.92{\sim}3.74\;{\mu}mol/g$. The lower homarine, trigonelline, TMAO, TMA, and creatine were detected in the extracts. The extractive nitrogenous constituents of medium size and large size echiuroid were almost the same level each other. The total nitrogens of the compounds analyzed for each samples accounted for more than 90% of the extractive nitrogen in this study.

Quality Evaluation of Red seabream, Pagrus major by Chemoenzymatic Method (효소 화학적 방법에 의한 참돔, Pagrus major의 품질판정)

  • Chemoenzymatic method was used in this study to evaluate the quality of cultured red seabrim. Level of ATP related compounds, ATPase and Adenylate energy charge (AEC) well reflected changes in condition of fish. ATP level in the muscle of exercised cultured red seabream was decreased significantly during cultured period. In contrast, the level of ADP in the muscle was increased during the period. Level of ATP related compounds in the muscle of cultured red seabream without exercise didn't show significant difference compared to the exercised group. AEC value in muscle of cultured red seabream without exercise was 0.89$\pm$0.02, while AEC value of the exercised group was 0.88$\pm$0.04. ATP and AEC levels among different size of fish did not show any significant difference. AEC value in all sample was higher than 0.8, indicating that the condition of cultured fish was healthy. Activity of Mg2$^{+}$ (+Ca2$^{+}$), Mg2$^{+}$ (-Ca2$^{+}$) and Ca2$^{+}$-ATPase during culture period did not change significantly, regardless of exercise. Therefore, the AEC appears to have some validity as fish health indicator.

Changes of Components in Salt-Fermented Blenny, Enedrias nebulosus Sauce during Fermentation (베도라치액젓의 숙성 중 성분변화)

  • To investigate changes of components in salt-fermented blenny, Enedrias nebulosus sauce during fermentation, various chemical properties were examined at 2$\~$3 months intervals during 18 months of fermentation. The degree of hydrolysis increased sharply until 6 months of fermentation and showed the gentle increasement after that. On the other hand, the contents of total and amino nitrogen, total ATP related compounds increased gradually during 18 months of fermentation, The hypoxanthine and uric acid were the most abundant in ATP related compounds, ranging from $80.1\% to $90.5\%. After 18 months of fermentation, sauce was rich in free amino acids such as glutamic acid, alanine, lysine, valine, leucine in that order.

Effects of Nucleic Acid Related Compounds on Cytidine Deaminase Activity Produced by Bacillus subtilis ED 213 (Bacillus subtilis ED 213 Cytidine Deaminase 활성에 미치는 핵산관련물질의 영향)

  • This study was carried out to investigate the effects of nucleic acid related compounds and metal ions on activities of cytidine deaminase from Bacillus subtilis ED 213. The purified cytidine deaminase was weakly inhibited by 1mM GMP, IMP and ATP, but not affected by other nucleic acid related compounds such as CMP and UDP. The apparent Km values for cytidine, deoxycytidine, 5 methylcy tidine, fluorodeoxycytidine, and 5 bromocytidine were calculated to be 6.6$\times$10-4M, 6.0$\times$10-4M, 0.9$\times$10-4M, 0.8$\times$10-4M, and 2.0$\times$10-3M, respectively. The cytidine deaminase was completely inhibited by 1mM Hg2+, and mildly inhibited over 40% by metal ions such as Na+ and Fe2+. However the enzyme activity was activated more than 40% by 1mM Mg2+.

Changes of the Nucleotides and their Related Compounds according to the Ripening Process of Low Salt Fermented Squid (저염 오징어 젓갈의 숙성에 따른 핵산관련물질의 변화)

  • This study shows the changes of the nucleotides and their related compounds of squid during fermentation for 8 weeks at $10^{\circ}C$ in 5% salt solution. Among nucleic acid related matters, ATP and ADP were vanished not to be detected, AMP existed only at the early stage and then rapidly decreased until the mid-stage of the ripening. Inosine and hypoxanthine were the main components of nucleotides and their related compounds. As the salt concentration was decreased and fermentation temperature raised, pH was significantly increased to the latter stage of the ripening and hence fermentations was enhanced. The titrable acidity was continuously decreased until the latter stage of the ripening. Considering the above result, it is possible to make an estimate that the suitable fermentation conditions of squids are $10^{\circ}C$ of fermentation temperature, 10% of salt concentration and 5 weeks of ripening period.

Comparison of Seasonal and Regional Variation in Extractive Nitrogenous. Constituents of the Raw Anchovy (Engraulis japonica) (생 멸치의 함질소 엑스성분조성 및 지역과 계절에 따른 변동)

  • In order to investigate the composition, seasonal and regional variation of extractive nitrogenous components in the raw anchovy (Engraulis japonica) harvested from June 1991 to June 1992 at Nam-hae and Ki-jang in the southern coast of Korea, the contents of extractive nitrogen (EN), free amino acids (FAA), oligopeptides (OP), ATP and its related compounds (ARC), quaternary ammonium bases, and guanidino compounds were measured. The EN contents of raw anchovy were $444{\~}773\;mg/100\;g$. Thirty-four kinds of FAA were found and their total amount were $1,049{\~}2,079\;mg/100\;g$. Histidine, taurine, alanine, leucine and glutamic acid were the major free amino acids in the anchovy extracts. The amount of ARC was average $5.02\;{\mu}mol/g$, and IMP was the major compound among them, who and creatine contents in the extracts or raw anchovy were average 164 and 229 mg/100 g, respectively. The contents of glycinebetaine,${\brta}-alaninebetaine,\;{\gamma}-burobetaine$, homarine, TMA, and creatinine were extremely small. As for the regional and seasonal disparities, the Nam-hae samples contained more EN, FAA, OP, betaines and creatine than the Ki-jang samples. On the other hand the Ki-jang samples contained more ARC and TMAO. The spring samples contained more EN, FAA, OP, ARC and betaines than the autumn samples. On the other side the autumn samples contained more TAMO and creatine.

Extractive Nitrogenous Constituents and Their Monthly Variation of Fresh Laver Porphyra dentata (잇바디돌김의 함질소 엑스성분조성 및 월별변동)

  • In order to investigate the composition and monthly variation of extractive nitrogenous components in the laver Porphyra dentata cultured at the south coast of Korea, the fresh laver was analyzed separately for the amounts of free amino acids, combined amino acids, ATP and its related compounds, and quaternary ammonium basis in fresh laver were measured. The extractive nitrogen contents of fresh laver extracts were 760~870 mg/100g (dry basis). Twenty-seven to thirty-one kinds of free amino acids were detected in the laver extracts and their total amounts were 2,404~3,966 mg/100g (on dry basis). The laver extracts showed rich in free amino acids such as alanine, taurine, glutamic acid, glutamine and aspartic acid. Sixteen to twenty-three kinds of combined amino acids were detected in the extracts and their total amounts were 1,429~2,692 mg/100g (on dry basis). Proline, glutamic acid, glycine, phosphoserine, serine were the amin combined amino acids in the extracts. The amounts of ATP and related compounds were 73.3~94.4 mg/100 g (2.04~4.43 $\mu$mol/g, on dry basis). Homarine and trigonelline were detected in all specimens but $\beta$-alaninebetaine, ${\gamma}$-butyrobetaine were found in some. Small amounts of trimethylamine were detected in all samples. Free and combined amino acids were occupying almost 90% of extractive nitrogen. Most of free and combined amino acids showed a marked monthly variation with a maximum in January and March, and a minimum in February and April. The fresh laver P. dentata did not differ much from the fresh laver P. yezoensis in qualitative com-position of extractive components, but their contents were generally low level.
