Yolk platelets, one of the main food stores in the embryonic development, are composed of proteins. However, little is known about the identity of proteins utilized at certain stages of embryogenesis. In this study, we followed the fates of embryonic storage proteins by using an anti-polyubiquitin monoclonal antibody (mAB) as a probe. The mAb recognized the major storage proteins of Drosophila, Xenopus and chicken eggs. In the Drosophila embryo, the mAb-reactive 45-kDa protein was not used until stage 11 but was used up at stage 16 when the embryo completed segmentation. In the Xenopus embryo, the mAb-reactive 111 kDa protein was mostly utilized between stages 42 and 45 implying that the protein might be an energy source used just prior to feeding on food. By N-terminal sequencing the storage protein of Xenopus embryo was identified as a lipovitellin 1. This study confirms that storage proteins are used almost simultaneously at certain stages of embryogenesis and that vitellogenin 1 is the last storage protein in Xenopus embryogenesis.
This study was conducted to examine how a low-head dam removal (partial removal) could affect benthic macroinvertebrate communities in a stream. Benthic macroinvertebrates and substrates were seasonally sampled before and after dam removal (March 2006-April 2007). Benthic macroinvertebrates and substrates were quantitatively sampled from immediately upstream (upper: pool) and downstream (lower: riffle) sites, the location of the dam itself (middle), and immediately above the impoundment (control: riffle). After the removal, species richness and density of benthic macroinvertebrates as well as the EPT group (Ephemeroptera, Plecoptera and Trichoptera) increased to higher levels than those before the removal, while functional feeding groups and habitat orientation groups changed more heterogeneously at the upper site. At the lower site, species richness and density decreased somewhat immediately after dam removal, which was associated with an increase of silt and sand, but recovered after monsoon floods which helped to enhance substrate diversity at the upper site. Decreased dominance index and increased diversity index in both the upper and lower sites are evidence of positive effects from the dam removal. In conclusion, we suggest that even a partial removal of a dam, resulting in increased substrate diversity in the upper site, could sufficiently help rehabilitate lost ecological integrity of streams without major habitat changes.
The present study was conducted to assess the dietary effect of T-2 toxin on the antioxidant systems of the liver in growing quail and to comparatively evaluate the protective properties of two different mycotoxin-adsorbent additives, Mycosorb and zeolite, in preventing inhibition of the antioxidant system. Four groups of 4 day old quail were formed with 20 birds in each group. The birds were maintained on the floor for the course of the study. The three treatment diets consisted of the basal diet with T-2 toxin added in the form of Fusarium sporotrichioides culture (8.1 mg/kg feed), T-2 toxin (8.1 mg/kg) plus zeolite (30 g/kg feed), and T-2 toxin (8.1 mg/kg) plus Mycosorb (1 g/kg feed). After 30 days of feeding (34 days old) all birds were sacrificed and liver samples for biochemical analyses were collected from five quail in each of the four groups. Antioxidant concentrations were evaluated by HPLC-based methods. Inclusion of T-2 toxin in the quail diet was associated with a significant (p<0.05) decrease in concentrations of all forms of antioxidants studied, including ${\alpha}$- and ${\gamma}$-tocopherols, ascorbic acid, retinol and retinyl esters. At the same time, liver susceptibility to lipid peroxidation significantly (p<0.05) increased. Inclusion of zeolite in the quail diet at the level of 3% was ineffective in preventing antioxidant depletion in the liver by mycotoxicosis. In contrast, Mycosorb in the diet at a 0.1% level was able to significantly inhibit liver antioxidant depletion and as a result decreased lipid peroxidation in the liver. Concentrations of all forms of antioxidants studied were significantly higher in the livers of the quails fed the basal and T-2 toxin/Mycosorb combination in comparison to birds fed the basal with T-2 toxin alone.
Steers (n = 335) of known genetic background from four fundamentally different growth types were subjected to two production systems to study differences in cooking loss (CL), tenderness, and chemical composition. Growth types were animals with genetic potential for large mature weight-late maturing (LL), intermediate mature weight-late maturing (IL), intermediate mature weight -early maturing (IE), and small mature weight-early maturing (SE). Each year, in a nine-year study, calves of each growth type were weaned and five steers of each growth type were developed on pasture or feedlot and harvested at approximately 20 and 14 mo of age, respectively. Data collected were CL and Warner-Bratzler shear force (WBS) for the Longissimus dorsi (LM), Psoas major (PS), and Quadriceps femoris (QF) muscles. Chemical composition was also determined from the right fore- and hindquarter. Data were analyzed using least squares analysis of variance for unequal subclass numbers. The beef growth $type{\times}production$ system interaction was significant for CL and WBS of the LM and ash in the lean trim of the forequarter. Growth types of LL and IL had greater (p<0.05) mean percentage CL in the PS and QF muscles than did IE and SE steers. Growth type LL had the highest (p<0.05) mean for both moisture and protein in the fore- and hindquarters; while SE had the lowest numerical mean value for moisture and protein in the fore- and hindquarters. Shear force of the PS did not differ (p>0.05) among steers of the four growth types. Increasing challenges to the cattle feeding industry may dictate that pasture development play a larger role in future production regimes. Producers should strive to match genetic growth type with available resources in order to remain viable and continue producing a quality product.
A feeding trial was carried out over 238 days to determine the effect of compensatory growth in crossbred calves having 166 kg body weight. Fifteen crossbred calves were divided into two groups of five calves (G1 group) and ten calves (G2 group) as per randomized block design. Growth study was conducted on the feeding of wheat straw based diet containing 60 and 30 percent concentrate supplying equal amount of protein in group G1 and G2 respectively for 119 days (phase - I). At the end of phase-I, calves of G2 group were subdivided in to two groups (G3 and G4). One sub group (G4) received 60% concentrate in their diet (during 120 to 238 days of experiment) while other subgroup G3 received 30% concentrate in their diet (phase-II). The calves of G1 group continued to receive the same diet as during phase-I experiment. Mean DM intake was significantly higher in calves fed high level of concentrate (in G1 and G4 groups), which resulted in significantly higher digestibility of all nutrients except NDF. Nitrogen balance was positive in all the groups and showed significant differences in phase-II (higher nitrogen retention in G4 group than G1 group). ME intake was significantly affected by the level of dietary concentrate, being higher in high concentrate fed group (G1 and G4 than G2 and G3 group). Higher daily body weight gain in the calves of G4 group during phase-II than in G1 and G3 groups was due to compensatory growth on shifting animals from low concentrate to high concentrate based ration. Average daily body weight gain was higher in phase-I than in the phase-II. Protein and energy intake per unit body weight gain were significantly lower in calves fed high concentrate diet.
The purpose of this study is to increase the part recovery rate (to more than 70%) during the casting of a ductile cast iron turbo charger housing using a heater around the riser. Before creating a casting mold, various runner and riser systems were designed and analyzed with a casting simulation analysis tool. The design variables were the heater temperature, top insulation, riser location, riser diameter and the riser shape. During the feeding from the riser to the part, the reverse model was better than the forward model. When heating the riser (above $600^{\circ}C$), solidification of the riser was delayed and the feeding effect was suitable compared to that without heating. At a higher heating temperature, less solidification shrinkage and porosity were noted inside the part. On the basis of a casting simulation, eight molds were fabricated and casting experiments were conducted. According to the experimental conditions, external and internal defects were analyzed and mechanical properties were tested. The ultimate tensile strength and elongation outcome were correspondingly more than 540MPa and 5% after a heat treatment. In addition, a maximum part recovery rate of 86% was achieved in this study.

Objective: We aimed to evaluate the color and oxidative stability of beef gluteus medius (GM) from cattle raised in organic and non-organic production systems. Methods: The GM samples (n = 10) were obtained from organic (ORG; n = 5) or nonorganic (NORG; n = 5) beef samples, sliced into 2.54-cm steaks, packaged in aerobic conditions, and stored for nine days at 4℃. ORG and NORG steaks were compared regarding myoglobin concentration, pH, instrumental color, delta E (ΔE), metmyoglobin reducing activity (MRA), and lipid oxidation on days 0, 5, and 9. Results: Feeding system did not influence (p>0.05) the myoglobin concentration. ORG steaks exhibited greater (p<0.05) meat pH, yellowness, and MRA, whereas NORG steaks exhibited greater (p<0.05) redness, chroma, R630/580, delta E, and lipid oxidation. ORG and NORG steaks exhibited similar (p>0.05) lightness and hue angle. During storage, ORG and NORG exhibited an increase in muscle pH, hue angle, and lipid oxidation; and a decrease (p<0.05) in redness, yellowness, chroma, and color stability (R630/580). Both samples exhibited a stable (p>0.05) pattern for lightness and MRA. Conclusion: Therefore, the production system can affect beef color and lipid stability during storage.
The quality of meat is highly variable in many properties. This variability originates from both animal production and meat processing. At the pre-slaughter stage, animal factors such as breed, sex, age contribute to this variability. Environmental factors include feeding, rearing, transport and conditions just before slaughter (Hildrum et al., 1995). Meat can be presented in a variety of forms, each offering different opportunities for adulteration and contamination. This has imposed great pressure on the food manufacturing industry to guarantee the safety of meat. Tissue and muscle speciation of flesh foods, as well as speciation of animal derived by-products fed to all classes of domestic animals, are now perhaps the most important uncertainty which the food industry must resolve to allay consumer concern. Recently, there is a demand for rapid and low cost methods of direct quality measurements in both food and food ingredients (including high performance liquid chromatography (HPLC), thin layer chromatography (TLC), enzymatic and inmunological tests (e.g. ELISA test) and physical tests) to establish their authenticity and hence guarantee the quality of products manufactured for consumers (Holland et al., 1998). The use of Near Infrared Reflectance Spectroscopy (NIRS) for the rapid, precise and non-destructive analysis of a wide range of organic materials has been comprehensively documented (Osborne et at., 1993). Most of the established methods have involved the development of NIRS calibrations for the quantitative prediction of composition in meat (Ben-Gera and Norris, 1968; Lanza, 1983; Clark and Short, 1994). This was a rational strategy to pursue during the initial stages of its application, given the type of equipment available, the state of development of the emerging discipline of chemometrics and the overwhelming commercial interest in solving such problems (Downey, 1994). One of the advantages of NIRS technology is not only to assess chemical structures through the analysis of the molecular bonds in the near infrared spectrum, but also to build an optical model characteristic of the sample which behaves like the “finger print” of the sample. This opens the possibility of using spectra to determine complex attributes of organic structures, which are related to molecular chromophores, organoleptic scores and sensory characteristics (Hildrum et al., 1994, 1995; Park et al., 1998). In addition, the application of statistical packages like principal component or discriminant analysis provides the possibility to understand the optical properties of the sample and make a classification without the chemical information. The objectives of this present work were: (1) to examine two methods of sample presentation to the instrument (intact and minced) and (2) to explore the use of principal component analysis (PCA) and Soft Independent Modelling of class Analogy (SIMCA) to classify muscles by quality attributes. Seventy-eight (n: 78) beef muscles (m. longissimus dorsi) from Hereford breed of cattle were used. The samples were scanned in a NIRS monochromator instrument (NIR Systems 6500, Silver Spring, MD, USA) in reflectance mode (log 1/R). Both intact and minced presentation to the instrument were explored. Qualitative analysis of optical information through PCA and SIMCA analysis showed differences in muscles resulting from two different feeding systems.

The treatment efficacy of oral administration of praziquantel against the blood-sucking monogenean Microcotyle sebastis under the commercial rockfish-culture conditions was evaluated. The quantity of praziquantel added to the food was calculated so that the amount of praziquantel ingested daily by fish was 200mg or 400mg/kg body weight (bw). The fish were fed a moist-pelleted fish meal supplemented with praziquantel at an rate of 0 (control), 200 or 400 mg of praziquantel per kg bw, respectively, for 3 times at an interval of 24 h. On 1 day to 8 days after the treatment, 10 fish were taken randomly from each net-pen daily (80 fish in each group in total), and the efficacy of each treatment was confirmed. Abundance of worms were significantly reduced in groups fed praziquantel-supplemented diets. The results clearly demonstrated that feeding the praziquantel-supplemented diet was effective for controlling M. sebastis infestation in practical commercial rockfish-culture systems without imposing any handling stress.
Substructure shake table testing is a class of real-time hybrid simulation (RTHS). It combines shake table tests of substructures with real-time computational simulation of the remaining part of the structure to assess dynamic response of the entire structure. Unlike in the conventional hybrid simulation, substructure shake table testing imposes acceleration compatibilities at substructure boundaries. However, acceleration tracking of shake tables is extremely challenging, and it is not possible to produce perfect acceleration tracking without time delay. If responses of the experimental substructure have high correlation with ground accelerations, response errors are inevitably induced by the erroneous input acceleration. Feeding the erroneous responses into the RTHS procedure will deteriorate the simulation results. This study presents a set of techniques to enable reliable substructure shake table testing. The developed techniques include compensation techniques for errors induced by imperfect input acceleration of shake tables, model-based actuator delay compensation with state observer, and force correction to eliminate process and measurement noises. These techniques are experimentally investigated through RTHS using a uni-axial shake table and three-story steel frame structure at the Johns Hopkins University. The simulation results showed that substructure shake table testing with the developed compensation techniques provides an accurate and reliable means to simulate the dynamic responses of the entire structure under earthquake excitations.
