The gene encoding the HN protein from the CBP-1 strain, a heat stable Newcastle disease virus (NDV) isolated from diseased pheasants in Korea, was characterized by reverse transcriptase- polymerase chain reaction(RT-PCR) and the nucleotide and amino acid sequences were analyzed following cloning of the HN gene. In all of the NDV strains studied, a 1.75 kb size cDNA fragment for the HN gene was generated by RT-PCR and smaller specific band sizes harboring the internal portions of the HN gene were also detected by using four pairs of primers. The RT-PCR was sensitive enough to detect viral transcripts when the virus titer was above 25 hemagglutination units. The amplified 1.75 kb cDNA was cloned into a BamHI site of the pVL1393 Baculo transfer vector. The nucleotide sequences of the 1,758 bp HN gene from the CBP-1 strain were determined by the dye terminator cyclic sequencing method. The gene sequences were compared among the strains of CBP-1, Texas GB, Beaudette C, LaSota, B1 and Ulster. The homology of the CBP-1 HN gene to other HN variants was 97.8% to Texas GB, 98.4% to Beaudette C, 95.4% to LaSota, 95.6% to B1 and 90.2% to Ulster. As the deduced 577 amino acid sequences were compared among the strains, the homology for CBP-1 HN appeared to be 96.7% to Texas GB, 97.9% to Beaudette C, 95.5% to LaSota, 95.5% to B1 and 92.7% to Ulster. It was evident that the amino acid sequences included 5 sites for N-asparagine linked glycosylation and 12 cysteine residues. The three conserved leucine residues within the predicted transmembrane domain of the HN protein are amino acid 30, 37 and 44. The three antigenic sites on the HN protein of NDV are amino acids 347(Glu), 481(Asn) and 495(Glu). These data indicate that the genotype of the CBP-1 strain is more closely associated with the strains of Texas GB and Beaudette C than it is for the LaSota, B1 and Ulster strains.
폐경 전후 여성에서의 갱년기 증상 등은 흔하게 발생되는 문제이며 이에 대한 해결책으로서 오랜 기간 동안 여성 호르몬 대체요법이 사용되어져 왔다. 그러나 최근 여성호르몬 대체 요법에 따른 유방암 및 심혈관 질환 위험 증가로, 자연 여성호르몬 대체요법의 사용 효과에 대한 관심이 높아지고 있다. 이 연구는 자연 생약 추출물이 갱년기 증상에 미치는 영향을 관찰하여 복합 자연 요법의 유용성을 밝히고자 하였다. 연구 시점에서 실험군과 대조군간의 기초적인 신체 특성과 혈중 호르몬 수치, 골대사지표, 그리고 혈중 지질은 차이가 없었다. 성장호르몬 수치로서 IGF-1 혈중농도가 실험군에서 증가하였으나, 통계적 유의성은 없었다. 연구시점에 갱년기증상을 가지고 있었던 대상자중 실험군에서는 57.1%, 대조군에서는 16.7%에서 증상이 개선되었으며 (OR=6.67, 95% C.I.0.49-91.33), 연구 개시점에는 특별한 증상을 느끼지는 않았으나 연구 종료 시점에서 이전에 비해 개선된 효과를 보였다고 대답한 대상자는 실험군에서 58.3%, 대조군에서는 21.7%로서 실험군에서 개선된 비율이 통계학적으로 유의하게 높았다(OR=5.04, 95% C.I. 1.40-18.14). 치료 후 3개월에 시행한 실험군과 대조군 사이의 체질량지수, 혈압, 혈중, E2, FSH, Osteocalcin, Total cholesterol, LDL, HDL, TG 등은 연구개시점과 연구종료 시점 사이에서의 평균변화는 통계학적으로 의미 있는 차이를 보이지 않았으나, 골생성 지표인 혈중 ALKP는 시험제 투여 3개월 이후 실험군에서 통계학적으로 유의한 감소를 보여 1년 후 골밀도의 상승효과가 기대되었다(p<0.01). 혈중 중성지방 (TG)은 시험제 투여 1개월 후 대조군에서는 투여전 대비 26.9$\pm$62 (mg%) 증가하였고, 3개월 후 12.852 (mg%) 증가하였으나, 실험군에서는 1개월후 -8.040 (mg%) 감소하였고, 3개월 후 -4.436 (mg%) 감소하여 통계적으로 유의하게 감소하였다(p<0.01, Student t-test). 시험제 3개월 치료 후 실험군의 체중, 수축기 혈압, 이완기 혈압은 대조군에 비하여 감소하는 비율이 높았으나 통계학적인 유의성은 보이지 않았다(p>0.05). 연구시점에 고혈압 상태였으나 시험 제 투여 3개월 후 정상혈압으로 판정 된 사람의 비율은 실험 군에서 대조군에 비해 더 많았으나 통계학적인 의미는 없었다(p>0.05).
10%의 $\alpha-(1\rightarrow3)$가지 결합을 갖고 $\alpha-(1\rightarrow6)$으로 연결된 댁스트란을 합성하는 텍스트란수크라제를 coding하는 유전자 (dsCB)를 Leuconostoc mesenteroides B-742CB로부터 분리하여 염기서열과 아미노산 서열을 결정하였다. dsCB가 포항된 6 6.lkb띄 DNA fragment는 4,536 bp의 염기로 구성된 하나의 open reading 잔ame(ORF)를 가지고 있었다. 추정된 아미노산 서열은 ORF의 698벤째 nucleotide 위치에 있는 start codon (ATG)으로부터 5,223번째 위치에 있는 slop codon(TAA)까지 였다. 구조 유전자의 아마노산은 1,5087H로 구성되고 분자량의 계산값은 168.6 kDa었고 non-denatured SDS-PAGE를 이용하여 활성 band툴 분석한 결과 170 kDa이었다. pDSCB를 까지고 있는 재조합 E. coli는 2 % sucrose배치에서 세포외로 댁스트란수크라제를 생산하였으며, soluble과 insoluble 텍스트 란을 생산하였다. 텍스트란수크라체의 효소 촉매작용에 관여 하는 것으로 얄려져있는 conserved region의 아미노산 중 Asp-492를 Asparagine으로 바꾸고자 point mutation을 시도하였고, 결과로 얻어친 D492N은 돌연변이 이전의 균에 비하여 활성이 1.6배 감소함을 확인하였다.
우렁쉥이의 육에 존재하는 carotenoprotein을 추출하여 이온교환수지 및 겔크로마토그래피를 이용하여 정제하였으며, 정제된 carotenoprotein의 분자량, pH 안정성, 열안정성, 변성제의 영향, 아미노산 조성 및 지방산 조성을 분석하였다. Carotenoprotein의 정제도는 5배, 수율은 2.9%였으며, 1 mg당 $0.15\;{\mu}g$ carotenoid가 함유되어 있었다. Carotenoprotein의 최대 흡수파장은 UV/Vis 스펙트럼에서 463 nm 및 439 nm였으며, 분자량은 64.4 kDa으로 단일 polypeptide로 이루어져 있었다. Carotenoprotein은 pH 7.0부근의 중성영역에서 안정하였고, 열안정성은$65^{\circ}C$에서 2시간 이내에 90%이상 유지하였으며, SDS 변성제에 의해 5 nm 단파장 영역으로 이동하였다. Carotenoprotein의 아미노산 조성은 Gly, hsn, Gln 및 Ser의 함량이 전체 아미노산의 35.3%로 전체적으로 산성의 성질을 보였으며, 구성지방산 조성의 함량은 monoene산이 45.2%로 가장 많았고, 다음으로 포화지방산 및 polyene산이 각각 38.1% 및 11.7%였다.
중쇄 acyl-CoA 탈수소효소 결핍증은 미토콘드리아에 존재하는 효소 중 하나인 MCAD의 부족으로 인하여 적절한 지방산 산화가 이루어지지 못하는 대사질환으로 지방산 산화와 관련된 대사 질환 중 가장 흔한 형태이다. 다양한 임상증상으로 저혈당, 발달지연, 발작, 돌연사 등이 나타날 수 있다. 저자들은 신생아 선별검사상 C6, C8, C10:1 acylcarnitine, C8/C2 ratio 혹은 C8/C10 ratio의 증가를 보이는 무증상의 신생아에서 유전자 분석검사를 통해 MCAD 결핍증을 진단하였다. 생후 10개월 경, 고열을 동반한 전신 강직성간대경련 발생하였으나 혈액검사 상 저혈당은 관찰되지 않았고 발열 호전된 후 추가적인 경련은 없었다. 이후 생후 25개월까지 추적관찰 하였을 때 경련을 포함한 증상 없었고, 정상적인 성장과 발달을 보였다. 무증상의 신생아에서 신생아 선별검사를 통해 우연히 MCAD 결핍증으로 진단된 후 1회의 열성경련 발생하였으나 대사성 위기없이 정상적인 성장 및 발달을 보이고 있는 환아가 있어 이에 증례를 보고하는 바이다.
We have determined and analyzed the full-length cDNA sequence of a coxsackievirus B3 (CVB3) Korean isolate (CVB3-Korea/97) which has been known as a general human pathogen. The whole genome contains 7,400 nucleotides and has a single large open reading frame with 6,555 nucleotides that encodes a potential polyprotein precursor of 2,185 amino acids. The genome also contains a 5' non-coding region (NCR) of 741 bases and a 3' NCR of 104 bases followed by poly(A) tail. Sequence homologies of nucleotides and deduced amino acids between the CVB3-Korea/97 strain and the prototype (Nancy strain) were 81.7% and 91.5%, respectively. The genes encoding the functional proteins including viral protease and RNA dependent RNA polymerase showed higher homology than those encoding the structural proteins. We have further analyzed the sequences of 5' NCR, VP1 and VP2 of CVB3-Korea/97, which are known as cardiovirulent determining factors at the nucleotide and amino acid levels. Although the CVB 3-Korea/97 strain was isolated from an aseptic meningitis patient without cardiomyopathy, its 234th nucleotide and 165th amino acid were uracil and Asn as same as those of other cardiovirulent strains one. However, the 155th amino acid of VP1, which closely associated with cardiovirulence, was replaced with $Arg^{155}$ by single nucleotide substitution from $A^{2916}$ to $T^{2916}$. Moreover, additional amino acid substitutions were observed in the flanking region of $Asp^{155}$. Taken together, amino acid(s) substitution in VP1 may playa critical role in determining cardiovirulence of the CVB3-Korea/97 strain rather than individual nucleotide replacements in the 5' NCR and/or an amino acid substitution in VP2.
We report the expression, purification, and characterization of L-asparaginase (AnsA) from Rhizobium etli. The enzyme was purified to homogeneity in a single-step procedure involving affinity chromatography, and the kinetic parameters $K_m$, $V_{max}$, and $k_{cat}$ for L-asparagine were determined. The enzymatic activity in the presence of a number of substrates and metal ions was investigated. The molecular mass of the enzyme was 47 kDa by SDS-PAGE. The enzyme showed a maximal activity at $50^{\circ}C$, but the optimal temperature of activity was $37^{\circ}C$. It also showed maximal and optimal activities at pH 9.0. The values of $K_m$, $V_{max}$, $k_{cat}$, and $k_{cat}/K_m$ were $8.9{\pm}0.967{\times}10^{-3}$ M, $128{\pm}2.8$ U/mg protein, $106{\pm}2s^{-1}$, and $1.2{\pm}0.105{\times}10^4M^{-1}s^{-1}$, respectively. The L-asparaginase activity was reduced in the presence of $Mn^{2+}$, $Zn^{2+}$, $Ca^{2+}$, and $Mg^{2+}$ metal ions for about 52% to 31%. In addition, we found that $NH_4{^+}$, L-Asp, D-Asn, and ${\beta}$-aspartyl-hydroxamate in the reaction buffer reduced the activity of the enzyme, whereas L-Gln did not modify its enzymatic activity. This is the first report on the expression and characterization of the L-asparaginase (AnsA) from R. etli. Phylogenetic analysis of asparaginases reveals an increasing group of known sequences of the Rhizobial-type asparaginase II family.

Equine chorionic gonadotropin (eCG) is a unique molecule that elicits the response characteristics of both follicle-stimulating hormone (FSH) and luteinizing hormone (LH) in other species. Previous studies from this laboratory had demonstrated that recombinant eCG (rec-eCG) from Chinese hamster ovary (CHO-K1) cells exhibited both FSH- and LH-like activity in rat granulosa and Leydig cells. In this study, we analyzed receptor internalization through rec-eCGs, wild type eCG ($eCG{\beta}/{\alpha}$) and mutant eCG ($eCG{\beta}/{\alpha}{\Delta}56$) with an N-linked oligosaccharide at $Asn^{56}$ of the ${\alpha}-subunit$. Both the rec-eCGs were obtained from CHO-K1 cells. The agonist activation of receptors was analyzed by measuring stimulation time and concentrations of rec-eCGs. Internalization values in the stably selected rat follicle-stimulating hormone receptor (rFSHR) and rat luteinizing/chorionic gonadotropin receptor (rLH/CGR) were highest at 50 min after stimulation with 10 ng of $rec-eCG{\beta}/{\alpha}$. The dose-dependent response was highest when 10 ng of $rec-eCG{\beta}/{\alpha}$ was used. The deglycosylated $eCG{\beta}/{\alpha}{\Delta}56$ mutant did not enhance the agonist-stimulated internalization. We concluded that the state of activation of rFSHR and rLH/CGR could be modulated through agonist-stimulated internalization. Our results suggested that the eLH/CGRs are mostly internalized within 60 min by agonist-stimulation by rec-eCG. We also suggested that the lack of responsiveness of the deglycosylated $eCG{\beta}/{\alpha}{\Delta}56$ was likely because the site of glycosylation played a pivotal role in agonist-stimulated internalization in cells expressing rFSHR and rLH/CGR.
In this study, we have cloned a novel cDNA encoding for a papain-family cysteine protease from the Uni-ZAP XR cDNA library of the polychaete, Periserrula leucophryna. This gene was expressed in Escherichia coli using the T7 promoter system, and the protease was characterized after partial purification. First, the partial DNA fragment (498 bp) was amplified from the total RNA via RT-PCR using degenerated primers derived from the conserved region of cysteine protease. The full-length cDNA of cysteine protease (PLCP) was prepared via the screening of the Uni-ZAP XR cDNA library using the $^{32}P-labeled$ partial DNA fragment. As a result, the PLCP gene was determined to consist of a 2591 bp nucleotide sequence (CDS: 173-1024 bp) which encodes for a 283-amino acid polypeptide, which is itself composed of an 59-residue signal sequence, a 6-residue propeptide, a 218-residue mature protein, and a long 3'-noncoding region encompassing 1564 bp. The predicted molecular weights of the preproprotein and the mature protein were calculated as 31.8 kDa and 25 kDa, respectively. The results of sequence analysis and alignment revealed a significant degree of sequence similarity with other eukaryotic cysteine proteases, including the conserved catalytic triad of the $Cys^{90},\;His^{226},\;and\;Asn^{250}$ residues which characterize the C1 family of papain-like cysteine protease. The nucleotide and amino acid sequences of the novel gene were deposited into the GenBank database under the accession numbers, AY390282 and AAR27011, respectively. The results of Northern blot analysis revealed the 2.5 kb size of the transcript and ubiquitous expression throughout the entirety of the body, head, gut, and skin, which suggested that the PLCP may be grouped within the cathepsin F-like proteases. The region encoding for the mature form of the protease was then subcloned into the pT7-7 expression vector following PCR amplification using the designed primers, including the initiation and termination codons. The recombinant cysteine proteases were generated in a range of 6.3 % to 12.5 % of the total cell proteins in the E. coli BL21(DE3) strain for 8 transformants. The results of SDS-PAGE and Western blot analysis indicated that a cysteine protease of approximately 25 kDa (mature form) was generated. The optimal pH and temperature of the enzyme were determined to be approximately 9.5 and $35^{\circ}C$, respectively, thereby indicating that the cysteine protease is a member of the alkaline protease group. The evaluation of substrate specificity indicated that the purified protease was more active towards Arg-X or Lys-X and did not efficiently cleave the substrates with non-polar amino acids at the P1 site. The PLCP evidenced fibrinolytic activity on the plasminogen-free fibrin plate test.
최근에 발표된 연구 결과에 의하면 대장균 트립토판 중합효소(tryptophan synthase) $\alpha$ 소단위체에서 33번 잔기의 세린과 112번 잔기의 아스팔산은 이 효소의 구조 형성 과정(folding)에 관여하는 것으로 보여진다. 이들 효소를 대장균내에서 과발현시켰을 때, SL33(잔기 33번의 세린이 류신으로 치환) 효소는 대부분 웅집된 덩어리형태의 효소로 생성되고, DG112(잔기 112번의 아스팔산이 글리신으로 치환) 효소와 DN112(아스랄산이 아스파라진으로 치환) 효소는 야생형 효소와 비슷한 양의 수용성형태로 생성된다고 보고된 바 있다. 본 연구에서는 이 효소의 구조 형성 과정동안 33번 잔기와 112번 잔기가 상호 작용하는 가를 조사하기 위하여 주 자리 잔기 치환 단백질을 만들어 이들의 성질을 단일 잔기 치환 단백질들과 비교하였다. 이들을 대장균내에서 젖당으로 과생산하여 전기영동으로 조사하여 본 결과 SL33 단일 잔기 치환 효소에 비해 SL33/DG112 이중 치환 효소는 현격히 보다 많은 양이 응집된 형태의 불용성 효소로 생성 되며, SL33/DN112효소는 많은 양의 효소들이 가용성 효소로 생성되었다. Densitometer로 정량한 결과 SL33/DN112 치환 소단위체는 SL33 치환 소단위체에 비해 가용성 형태의 양에 대한 응집된 형태의 양의 비가 6시간 젖당처리시 약 5배, 22시간 젖당처리시 약 13배 증가하였으며, SL33/DN112 치환 $\alpha$ 소단위체는 그 비가 1/4-1/3로 감소했다. 이러한 결과는 331번 위치에서 세린이 류신으로 치환되었을 때 나타나는 구조 형성 과정의 변화를 112번 위치에 새로 치환된 글리신 또는 아스파라진이 영향을 미쳐, 구조 형성 솨정 변화가 더욱 더 변화했거나 원상태로 어느 정도 회복되었음을 의미한다. 이 결과는 대장균 트립토판 중합효소 $\alpha$ 소단위체의 N 말단 도메인(N-terminal domain)의 구조 형성 과정시 33번 잔기와 112번 잔기가 상호작용을 하고 있음을 시사해 주고 있다.

② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
