Staphylococcusaureus protein A as a means of assessing sperm penetrability in cervical mucus in vitro

  • Objective: The effectiveness of Staphylococcus protein A (SPA) in improving the penetration ability of sperm and reducing antisperm antibody (ASA) titers in immunologically infertile males was evaluated. Methods: Seminal fluid samples were obtained from 15 infertile men, and ASA titers were assessed with the latex agglutination test. Identification of immunoglobulin (Ig) classes and characterization of the antigens involved in the immune response were performed using indirect immunofluorescence. Local ASAs typically present as a mixture of IgG and IgA classes. The capillary tube penetration method was used to assess the capability of spermatozoa to penetrate the cervical mucus (CM). Results: ASAs associated with the neck region of sperm showed a significantly lower migration distance in the CM of infertile females than ASAs associated with the head or tail segments. ASA-positive seminal fluid exhibited significant increases in the mean migration distance (2.6 ± 1.4 cm vs. 1.54 ± 1.1 cm, respectively; p< 0.001) and sperm concentration (174 ± 121.0 × 103/mL vs. 101 ± 93.7 × 103/mL, respectively; p= 0.033) after treatment with SPA compared to pre-treated samples. A significant reduction (p< 0.01) in the recorded ASA titer was detected. Conclusion: These results indicate that SPA can be used as a sorting regimen for insemination programs. However, further studies are warranted to assess its influence on pregnancy rate.

VARIABLE STARS IN THE REGION OF AN OPEN CLUSTER M50 (산개성단 M50 영역의 변광성)

  • From the short-period variability survey (SPVS), we obtained time-series BV CCD images in the region of an open cluster M50 (NGC 2323) for 10 nights from 2008 February 9 to 27. The observation was performed using a small refracting telescope (${\phi}$=155mm, f=1050mm) in Bohyunsan Optical Astronomy Observatory (BOAO). M50 is included in the observing field of ASAS (The All Sky Automated Survey). In the region of M50, we found 8 new variable stars including two ${\delta}$ Scuti stars with short-periods and very small amplitudes. There are known nine variable stars by the catalogues of ASAS and GCVS in the region. We confirmed the variability of the known variable stars for six and non-variability for three.

Effects of Different Physical Frequency on Food-Dependent Exercise Induced Allergy Anaphylaxis (FDEIA) and Related Mechanisms (운동빈도의 차이가 식이유도 운동알레르기 질환과 관련기전에 미치는 영향)

  • Food allergies have become a serious health concern in the past two decades, especially in developed countries. Foods associated with allergies include vegetables, some fruits, shellfish, wheat, egg, chicken, and nuts. To describe the specific fundamentals, etiological factors, and clinical manifestations, we analyzed the different physical frequency on spleen index in sensitized and regular exercise-trained mice. We also conducted a proliferation assay of lymphocytes to OVA, ROS, ASAS, and we determined the cytokine levels. Female BALB/c mice were bred in the animal laboratory of the P and D university under controlled conditions ($22{\pm}2^{\circ}C$, RH 45-55%, and a 12-hour photoperiod). The animals were 6 weeks old at the start of the study and were fed a standard commercial chow diet from 09:00 to 15:00 for the 8-week study period. All animals had access to distilled deionized water ad libitum. They were divided into four groups: a control group (S; control sensitized, n=25), a low-frequency training group (F2, n=25), a mid-frequency training group (F3, n=25), and a high-frequency training group (F5, n=25) following the treatment of exercise time per week. The results were as follows: The mice spleen index showed the highest grade in the F5 group compared with the other groups; this level showed in an exercise frequency-dependent manner. In the proliferation assay of OVA, the F5 group showed the highest grade compared with the other groups; this level was also showed in an exercise frequency-dependent manner. Peritoneal ROS and ASAS showed a statistically significant increase in the F5 group and decreased in the F2 group compared with the S group. However, there were no significant differences in the F3 group. The highest level of IL-4 was found in the F5 group compared with the other groups. However, the highest level of INF-${\gamma}$ was in the F2 group. The results suggest that FDEIA is positively correlated with the frequency of exercise due to the direct effect of physical exercise on peritoneal ROS and the cytokine profile. Further research is needed on the specific mechanism underlying the combined effects of exercise intensity and frequency on physical-induced allergy anaphylaxis.

Apolipoprotein A5 3'-UTR variants and cardiometabolic traits in Koreans: results from the Korean genome and epidemiology study and the Korea National Health and Nutrition Examination Survey

  • BACKGROUND/OBJECTIVES: This study aimed to test the association between APOA5 3'-UTR variants (rs662799) and cardiometabolic traits in Koreans. SUBJECTS/METHODS: For this study, epidemiological data, Apolipoprotein A5 (APOA5) genotype information, and lymphoblastoid cell line (LCL) biospecimens from a subset of the Ansung-Ansan cohort within the Korean Genome and Epidemiology study (KoGES-ASAS; n = 7,704) as well as epidemiological data along with genomic DNA biospecimens of participants from a subset of the Korea National Health and Nutrition Examination Survey (KNHANES 2011-12; n = 2,235) were obtained. APOA5 mRNA expression was also measured. RESULTS: APOA5 rs662799 genotype distributions in both the KoGES-ASAS and KNHANES groups were 50.6% for TT, 41.3% for TC, and 8.1% for CC, which are similar to those in previous reports. In both groups, minor C allele carriers, particularly subjects with CC homozygosity, had lower high-density lipoprotein (HDL) cholesterol and higher triglyceride levels than TT homozygotes. Linear regression analysis showed that the minor C allele significantly contributed to reduction of circulating HDL cholesterol levels [${\beta}=-2.048$, P < 0.001; ${\beta}=-2.199$, P < 0.001] as well as elevation of circulating triglyceride levels [${\beta}=0.053$, P < 0.001; ${\beta}=0.066$, P < 0.001] in both the KoGES-ASAS and KNHANES groups. In addition, higher expression levels of APOA5 in LCLs of 64 healthy individuals were negatively associated with body mass index (r = -0.277, P = 0.027) and circulating triglyceride level (r = -0.340, P = 0.006) but not significantly correlated with circulating HDL cholesterol level. On the other hand, we observed no significant difference in the mRNA level of APOA5 according to APOA5 rs662799 polymorphisms. CONCLUSIONS: The C allele of APOA5 rs662799 was found to be significantly associated with cardiometabolic traits in a large Korean population from the KoGES-ASAS and KNHANES. The effect of this genotype may be associated with post-transcriptional regulation, which deserves further experimental confirmation.

The Effects of Isotypes and Regional Distribution of Antisperm Antibodies on Semen Parameters and Fertilizing Ability (항정자항체가 정액성상 및 수정능력에 미치는 영향)

  • To investigate the influences on semen parameters and fertilizing capacity of immunoglobulin (Ig) isotypes and regional distribution of antisperm antibody (ASA) on the human sperm surface. Sixty-seven ASA-positive patients were compared with 96 ASA-negative donors. ASAs in semen showed significant negative effects on both semen parameters and fertilizing capacity; in those with ASAs in the sperm head and/or tail, the reductions were significant. In the head as well as the tail, there was close correlation between fertilizing capacity and both IgG and IgA. Both semen parameters and fertilizing capacity are significantly affected by the presence of ASA in semen. In particular, antibodies IgG to sperm head and/or tail, and antibodies IgA to sperm tail appeared to have a highly detrimental effect on fertilizing capacity.

Studies of Exercise-Induced Allergy Anaphylaxis Mechanisms and the Effects of Vitamin C and Catalase Supplementation in Exercise-Induced Allergy Anaphylaxis Models (운동 유발성 알레르기 질환분석 및 비타민 C와 catalase 투여 효과 분석)

  • Exercise-induced anaphylaxis (EIA) is defined as the onset of allergic symptoms during, or immediately after, exercise, the clinical signs being various degrees of urticaria, angioedema, respiratory and gastrointestinal signs, and even anaphylactic shock. Food-dependent exercise-induced anaphylaxis (FDEIA) is a specific variant of exercise-induced anaphylaxis that requires both vigorous physical activity and the ingestion of specific foods within the preceding several hours. To describe the physiopathologic mechanism, etiologic factors, and clinical manifestations, we evaluated the supplementation of vitamin C and catalase on spleen index, proliferation assay, ROS, and ASAS in sensitized and exercise trained mice. The results were as follows: Spleen index showed the highest level in the ST12 group compared to other groups; this level increased in a time dependent manner and in significant amounts. In proliferation assay of Med and OVA, the ST12 group showed the highest level compared to other groups; this level also increased in a time dependent manner. On the other hand, spleen ROS did not show a statistically significant difference, and peritoneal ROS showed the highest level in the ST12 group. ASAS showed the highest level in the ST12 compared to the S; this was also in a time dependent manner and in significant amounts. From the results, we chose the ST9 and ST12 groups to evaluate allergy anaphylaxis with supplementation of Vitamin C and catalase. In both the ST9 and ST12 groups, peritoneal ROS and ASAS were lower in vitamin C treatment group than in the catalase treatment group. This was a statistically significant difference. From the results, allergy anaphylaxis showed a higher level in the long trained group than in the short trained group. Also, treatment with vitamin C was more effective in lowering allergy anaphylaxis than catalase treatment.

Development of Automatic Subjective Assessment System Using Adjectives (형용사를 이용한 자동 주관적 평가 시스템의 개발)

  • The objective of this research is the development of the Automatic Subjective Assessment System (ASAS). The proposed subjective assessment system is designed to evaluate human emotion and sensibility (or "gamsung" in Korean terminology) with subjective responses of volunteers about the experiment of emotion and sensibility. Once volunteers enter their subjective responses about the experiment into the developed system, the proposed system can automatically generate statistical results of human emotion and sensibility using Statistical Package for the Social Sciences (SPSS). Then, the system stores the statistical results in the database which will be open to public through internet. The proposed system will be integrated into the universal" gamsung" assessment system for evaluation of human emotion and sensibility.

Air Surveillance Using Mode-S Multilateration (모드-S 다변측정법을 이용한 항공감시기술 분석 및 전망)

  • Surveillance is an enabler of safety with respect to aircraft separation and as a consequence capacity and efficiency with respect to aircraft operations. The new emerging technology among modern civil aviation surveillance is Multilateration (MLAT) which would affect on the surveillance capacity with both side of surveillance signal and operational properties. Multilateration system is needed to receive the signal which must reach at least 3 ground receivers simultaneously and has the effect that will have on with the ultimate accuracy. In this paper, the principle and the system configuration are reviewed. And its benefit of development is considered with use in situations where it is difficult to locate tranditional radar. This MLAT requires no additional avionics equipment to supply service with more accurate and less expensive. And it is able to enhance performance that meets international standards and extend the investment of air navigation service providers with reducing environmental impact by utilizing a small footprint on existing structures. Finally, it can be added to meet a wide range of coverage requirements and future surveillance needs.

EFFICIENT PERIOD SEARCH FOR TIME SERIES PHOTOMETRY

  • We developed an algorithm to identify and determine periods of variable sources. With its robustness and high speed, it is expected to become an useful tool for surveys with large volume of data. This new scheme consists of an initial coarse. process of finding several candidate periods followed by a secondary process of much finer period search. With this multi-step approach, best candidates among statistically possible periods are produced without human supervision and also without any prior assumption on the nature of the variable star in question. We tested our algorithm with 381 stars taken from the ASAS survey and the result is encouraging. In about $76\%$ cases, our results are nearly identical as their published periods. Our algorithm failed to provide convincing periods for only about $10\%$ cases. For the remaining $14\%$, our results significantly differ from their periods. We show that, in many of these cases, our periods are superior and much closer to the true periods. However, the existence of failures, and also periods sometimes worse than manually controlled results, indicates that this algorithm needs further improvement. Nevertheless, the present experiment shows that this is a positive step toward a fully automated period analysis for future variability surveys.
