APEX-1 Regulates Cell Proliferation through GDNF/GFRα1 Signaling (APEX-1은 GDNF/GFRα1 시그널을 통해 세포증식을 조절한다)

  • Human apurinic/apyrimidinic endonuclease (APEX-1) is a multifunctional protein that is capable of repairing abasic sites and single-strand breaks in damaged DNA. In addition, it serves as a redox-modifying factor for a number of transcription factors. Identifying the transcriptional targets of APEX-1 is essential for understanding how it affects various cellular outcomes. Expression array analysis was used to identify glial cell-derived neurotropic factor receptor ${\alpha}1$ ($GFR{\alpha}1$), which is an encoding receptor for the glial cell-derived neurotropic factor (GDNF) family, the expression of which is induced by APEX-1. A target of GDNF/$GFR{\alpha}$ signaling, c-Src (Tyr418) was strongly phosphorylated by GNDF in the APEX-1 expressing cells. Moreover, GDNF initiated cell proliferation, measured by counting the number of cells, in the APEX-1 expressing cells. Importantly, the down-regulation of APEX-1 by siRNA caused a marked reduction in the $GFR{\alpha}1$ expression level, and it reduced the ability of GDNF to phosphorylate c-Src (Tyr418) and stimulate cell proliferation. These results demonstrate an association between APEX-1 and GDNF/$GFR{\alpha}$ signaling and suggest a potential molecular mechanism for the involvement of APEX-1 in cell survival and proliferation.

Evaluating the Performance of APEX-Paddy Model using the Monitoring Data of Paddy Fields in Iksan, South Korea (국내 논필지 모니터링 자료를 이용한 APEX-Paddy 모델 적용성 평가)

  • The APEX model has been developed for assessing agricultural management efforts and their effects on soil and water at the field scale as well as more complex multi-subarea landscapes, whole farms, and watersheds. Recently, a key component of APEX application, named APEX-Paddy, has been modified for simulating water quality by considering paddy rice management practices. In this study, the performance of the APEX-Paddy model was evaluated using field data at Iksan experimental paddy sites in Korea. The discharge and pollutant load data during 2013 and 2014 were used to both manually and automatically calibrate the model. The APEX auto-calibration tool (APEX-CUTE 4.1) was used for model calibration and sensitivity analysis. Results indicate that APEX-Paddy reasonably performs in predicting runoff discharge rate and nitrogen yield. However, sediment and phosphorus yield is not correctly predicted due to the limitation of model schemes. With APEX-Paddy, the performance in reproducing the discharge and nitrogen yield is found to be a satisfactory level after manual calibration. The manually calibrated model performed better than the automatically calibrated model in nearly all comparisons. For runoff, manual calibration reduced PBIAS while R2 and NSE values of the automatically calibrated model were the same as the manual calibration. For T-N, NSE and PBIAS were reduced when using manual calibration, whereas R2 value was the same as manual calibration. The limitation of the APEX-Paddy model for predicting sediment, as well as the phosphorous yield, was discussed in this study.

Effects of Apex Removal on the Growth and Yield of Artichoke by Planting Times (정식시기별 생장점 제거가 아티초크(Cynara scolymus L.)의 생육 및 수량에 미치는 영향)

  • Apex removal is a common practice in artichoke cultivation to harvest heads of lateral shoots. This experiment was carried out to investigate the effect of apex removal by different planting times on the growth and yield of Artichoke (Cynara scolymus L.) in open field. Two treatments (apex removal and no apex removal) at three different planting times ($1^{st}$: Sep. 27, 2011, $2^{nd}$: March 29, 2012, and $3^{rd}$: Sep. 21, 2012) were tested using 'Green Globe' variety. There was no difference in the head characteristics and the number of harvested head between the treatments. The head weight was heavier in no apex removal of 242.7 g than the apex removal of 170.8 g. The yield also increased in no apex removal by 25% (1,249 kg/10a) compared to the apex removal of 997 kg/10a at the first planting time. At the second planting time, there was no difference in the head weights between the treatments. But the number of the harvested head was higher in no apex removal with 10.8 than 8.2 of the apex removal. The yield of no apex removal was 2,660 kg/10a, which was higher than 1,848 kg/10a of apex removal. At the third planting time, the head weight increased in no apex removal with 253.5 g compared to the apex removal with 218.7 g. The yield of no apex removal was 1,405 kg/10a, which was higher than 1,148 kg/10a of the apex removal. All the tests at 3 different planting times showed that the artichoke yields were higher in no apex removal than in apex removal. Therefore, it is desirable to cultivate artichoke without removing the apex for the higher yield and labor saving in open-field cultivation in Jeju island.

  • One of the most important factors for successful endodontic therapy is an accurate length determination of physiological root apex. Some methods suggested for the measurement of root canal length, include digital-tactile sense and roentgenographic technique with measuring wire, scale and grid. But these methods do not derermine an accurate working length to physiological root apex. Recently electronic measuring devices are used to locate the physiological root apex in root canal length determination and these devices are accepted as an effective apparatus. The 89 patients (116 teeth, 144 canals) among the out-patients of Yonsei University Dental Infirmary, who had had an endodontic treatment in the Department of Operative Dentistry, were measured by the Root-Canal Meter$^{(R)}$ as an electronic device, and radiographs to determine the distribution and location of physiological root apex, then the following results were made: (1) Range of ${\pm}$1mm from the radiographic root apex were present in 88.88% (128 canals) of the subjects. (2) Physiological root apex and radiographic root apex were coincided in 31.94% (46 canals) of the subjects. IN VITRO EVALUATION OF THE CONSISTENCY OF TWO ELECTRONIC APEX LOCATORS (2종 전자근관장측정기의 일관성에 관한 in vitro 연구)

  • The purpose of this study was to evaluate the consistency of two electronic apex locators in vitro model. Materials consisted of fifty two extracted premolars and two electronic apex locators; Root ZX (J. Morita, Osaka, Japan) and E-Magic Finder Deluxe (S-Denti. Cheonan, Korea). After access preparation, the teeth were embedded in a saline-mixed alginate model. Canal lengths of each tooth were measured at "0.5" and "Apex" mark of the apex locators, respectively so that each tooth had two measurements from 0.5 and Apex points. The file was fixed at final measurement using a glass ionomer cement. The apical 4 mm from the apex was exposed to measure the distance from the file tip to the major apical foramen of each tooth. Average distances and standard deviations were used to evaluate the consistency. Results showed that all measurements of both Root ZX and E-Magic Finder located the major foramen the range of ${\pm}0.5\;mm$ level. Both apex locators showed better consistency at Apex mark than at 0.5 mark. The average distance of file tip-major foramen was - 0.18 mm at 0.5 mark and - 0.07 mm at Apex mark in Root ZX, - 0.25 mm at 0.5 mark and - 0.02 mm at Apex mark in E-Magic Finder. Standard deviation was 0.21 at 0.5 mark and 0.12 at Apex mark in Root ZX, 0.12 at 0.5 mark and 0.09 at Apex mark in E-Magic Finder.

EVALUATION OF THE ACCURACY OF THE ELECTRONIC APEX LOCATOR USING THE POLARIZING MICROSCOPE (편광현미경을 이용한 전자적근관장측정기의 정확성에 관한 연구)

  • The purpose of this study was to evaluate the accuracy of an electronic apex locator, the Neosono - M, in determining the location of the cementodentinal junction. A total of 26 teeth with 46 canals were evaluated. The apex locator was used to locate the file and the teeth extracted. The specimen were prepared with highspeed burs and sandpaper discs to a thickness of $250{\sim}300{\mu}m$. Distances were measured from the cementodentinal junction with the use of polarizing microscope. Measurements made by the apex locator were also compared with those by the X - ray. The results were as follows. 1. The apex locator and X - ray were to measure a mean value of 0.17, 0.45mm coronal to the CDJ respectively. There was no significant difference between the accuracy of the apex locator and that of X - ray. 2. There was no significant difference between the accuracy of apex locator in vital and in nonvital teeth, and between in narrow and in wide canals. 3. The files in nonvital and narrow group were apical to the CDJ significantly than those of vital and narrow, vital and wide, and non vital and wide groups.

SIMULATED AP1000 RESPONSE TO DESIGN BASIS SMALL-BREAK LOCA EVENTS IN APEX-1000 TEST FACILITY

  • As part of the $AP1000^{TM}$ pressurized water reactor design certification program, a series of integral systems tests of the nuclear steam supply system was performed at the APEX-1000 test facility at Oregon State University. These tests provided data necessary to validate Westinghouse safety analysis computer codes for AP1000 applications. In addition, the tests provided the opportunity to investigate the thermal-hydraulic phenomena expected to be important in AP1000 small-break loss of coolant accidents (SBLOCAs). The APEX-1000 facility is a 1/4-scale pressure and 1/4-scale height simulation of the AP1000 nuclear steam supply system and passive safety features. A series of eleven tests was performed in the APEX-1000 facility as part of a U.S. Department of Energy contract. In all, four SBLOCA tests representing a spectrum of break sizes and locations were simulated along with tests to study specific phenomena of interest. The focus of this paper is the SBLOCA tests. The key thermal-hydraulic phenomena simulated in the APEX-1000 tests, and the performance and interactions of the passive safety-related systems that can be investigated through the APEX-1000 facility, are emphasized. The APEX-1000 tests demonstrate that the AP1000 passive safety-related systems successfully combine to provide a continuous removal of core decay heat and the reactor core remains covered with considerable margin for all small-break LOCA events.

  • The purpose of this study is to explain theoretically the mechanism of the frequency-dependent apex locator which can detect the apex of the root in moist canals with blood or other conductible canal irrigants. The frequency-dependent apex locator is based on measuring the ratio of the two impedances of the two alternating currents with different frequencies. We analyzed the changes of the ratio between the two impedances by using differential calculus. Our analysis shows that : 1. When the file is in the moist canal, the ratio between the two impedances is almost constant. 2. As the file approaches the apex of the root, the ratio decreases sharply. By this mechanism, the frequency-dependent apex locator can detect the apex of the-root quite accurately in moist canals.

  • 근래 농촌지역에서의 하천 및 저수지의 수질오염에 관하여 관심이 고조됨에 따라 비오염원에 대한 파악과 대책을 세우기 위해 관련분야에서 많은 연구가 진행 되고 있다. 비점오염원은 주로 강우나 유출에 의해 배출되기 때문에 배출 장소와 경로가 불분명하고 다양하다. 비점오염의 관리를 위한 유역모델로 SWAT (Soil and Water Assessment Tool) 모델을 이용한 연구가 광범위하게 사용되고 있다. 그러나 SWAT 모델은 유역모델로써 농촌지역에 논, 밭에서의 비점오염원 기작을 표현하기에는 공간적 범위의 한계가 있다. 이에 본 연구에서는 예당저수지 유역(465.12 km2)을 대상으로 유역규모의 SWAT 모델과 유역-필드규모에 적용 가능한 SWAT-APEX (Agricultural Policy/Environmental eXtender) 모델의 수질(T-N, T-P) 모의결과를 비교하여 SWAT-APEX 모델의 적용성을 평가하고자 하였다. 모형의 적용을 위한 입력자료로 기상자료와 지형자료를 구축하였으며 기상자료로 예당저수지유역 3개의 강우관측소 자료를 수집하여 구축하였으며, 지형자료로 격자크기 30m의 DEM (Digital Elevation Model)과 농촌진흥청에서 제공하는 1:25,000 정밀토양도와 토지이용도는 환경부로부터 1:25,000 중분류 토지이용도를 이용하였다. 또한 환경부에서 제공하는 월단위 하천수질 자료(기간)를 구축하여 모형의 검증을 실시하였다. 분석과정으로 SWAT 모델에서의 유역차원 수문, 수질 모의를 한 후, APEX 모델을 이용하여 소유역별 논, 밭에 대한 필드단위에 오염물질 모의 후 각각 소유역 출구에서 APEX 모델에 결과를 반영한 SWAT-APEX 모의를 거쳐 최종 유역출구에서의 유출량과 수질항목을 분석하였다. 모의 결과 유출량에 대해 Nash와 Sutcliffe (1970)가 제안한 모델효율성계수 (Model Efficient, ME)는 0.67, 결정계수는 0.69 그리고 수질항목의 결정계수는 각각 0.77, 0.75으로 분석되었다. 또한, SWAT-APEX 모의 결과 수질항목의 결정계수는 각각 0.80, 0.72이었다. 따라서, 본 연구에서 농촌지역의 비점오염원 모의는 필드모의를 반영한 SWAT-APEX 모델 결과가 SWAT 모델만 적용한 결과보다 정확한 비점오염 모의가 이루어졌다고 판단 할 수 있다.
