A Study on Protein Separation with AOT-Isooctane Reverse Micellar System (AOT-Isooctane 역마이셀계를 이용한 단백질 분리에 관한 연구)

  • For the selective separation of proteins, the solubilization and desolubilization of proteins in sodium-di-2-ethylhexyl sulfosuccinate (AOT)-isooctane reverse micellar system were investigated. Protein solubilization increased with increasing the concentration of AOT to 200 mM and then decreased above that concentration. Protein was solubilized into reverse micelles in the pH range below the isoelectric Point of each protein, pH 4-10 for lysozyme and pH 5-6 for trypsin and ${\alpha}-chymotrypsin$, Lysozyme, trypsin and ${\alpha}-chymotrypsin$ were efficiently extracted in the precence of KCl and NaCl while larger molecular weight proteins such as pepsin and BSA had high solubilization with $CaCl_2$. At higher ionic strength all proteins exhibited murk less tendency to solubilize and the increase of ionic strength resulted in the decrease of micelle size. Lysozyme was successfully back transfered at pH 12.2 and 1.0M KCl; trypsin at pH 12.6 and 0.5M KCl; and ${\alpha}-chymotrypsin$ at pH 6.7 and 0.5M KCl. In a test group separation experiments, complete separation of lysozyme from BSA could be obtained.

Optical Properties of Aerosol at Gongju Estimated by Ground-based Measurements Using Sky-radiometer (스카이라디오미터(Sky-radiometer)로 관측된 공주지역 에어로솔의 광학적 특성)

  • We investigate the optical properties of aerosols over Gongju by an indirect method using the pound measurement, Sky-radiometer. The analysis period is from January to December, 2004. Skyrad. pack.3 is used to estimate the optical properties, such as the aerosol optical thickness (AOT), single scattering albedo (SSA), ${\AA}ngstron$ exponent $({\alpha})$ and size distribution, of aerosols from the ground measured radiance data. And qualify control is applied to minimize the cloud-contaminated data and improve the quality of analysis results. The 12-month average of AOT, ${\alpha}$, and SSA are 0.46, 1.14, and 0.91, respectively. The average volume spectra of aerosols shows a bi-modal distribution, the first peak at fine mode and the second peak at coarse mode. AOT and coarse particles clearly increases while SSA decreases during the Asian dust events. The optical properties of aerosols at Gongju vary with?seasons, but those are not influenced by the wind direction.

The Effect of Action Observation Training on Sit to Walk with Chronic Stroke Patients

  • Purpose: This study was performed to examine the effect of action observation training on sit to-walk times in chronic stroke patients. Methods: Twelve stroke patients were randomly allocated to two groups, as follows: an action observation training (AOT) group and a scenery picture observation (SPO) group. The AOT group was engaged in video observation for 2 minutes 30 seconds with 12 ADL functional activities related to the sit-to-walk task: physical training was carried out in two repetitions lasting 2 minutes 30 seconds. The landscape imagery control group observed landscape picture for 2 minutes 30 seconds and then carried out the same physical training. The intervention involved a total of 12 sessions over 4 weeks, and each session lasted 30 minutes. The pre and post-tests measured the timed up and go test (TUG), Sit to stand test (STS), and Dynamic gait index (DGI). The Wilcoxon signed ranks test was used to compare pre-test and post-test result, and the Mann-Whitney U test was employed for comparison between groups. Results: The TUG time was significantly different between the AOT group and the SPO group. According to the findings, the experimental group's TUG was significantly decreased (p<0.05). In comparison of the two groups, there was no significant in STS or DGI between the AOT group and SPO group. Conclusion: This study showed that the intervention can be used to decrease TUG time. Our research suggests that action observation training has a positive effect on sit-to-walk times in patients with chronic stroke.

Salt Effect on Interaction between Acridine Orange and Aerosol OT (Acridine Orange와 Aerosol OT 사이의 상호작용에 관한 염효과의 연구)

  • The changing shape of absorption and fluorescence spectra will increasing S/D in AO-AOT system decreases and then increases again. The aggregation and disaggreation in AO-AOT system occurres. According to increasing temperature, the absorbance increases in pure AO system. But it gradually decreases in premicellar range(S/D = 10, 20, 50 and 80). The disappearance degree of metachromasy according to the concentration of added salt to AO-AOT system is increased. However, the case of $NaNO_3$is reversed. The order of disappearance degree of metachromasy on cationic salts is $Li^+$, $Na^+$ > $Mg^{2+}$ > $Ca^{2+}$, and the order of it on anionic salts is $Cl^-$ > $SO_4{^{2-}}$ > $NO_3{^-}$.

Aerosol Vertical Distribution Measured by LIDARs in Baengnyeongdo, Munsan, and Gunsan during 10~11 May 2010 (백령도, 문산, 군산의 라이다로 측정한 에어로졸 연직분포 -2010년 5월 10~11일 황사를 중심으로-)

  • This study aims to analyze the vertical distribution of Asian dust measured by LIDARs at three weather stations in Baengnyeongdo (BND), Munsan (MS), and Gunsan (GS) during 10~11 May 2010, and thereby investigate their effectiveness. Asian dust passed through from central to south-western part of Korea. Although dust particles were detected over the surface in MS and GS, LIDAR data showed that the Asian dust with non-spherical particles was observed in all of the three regions. It seems that the naked-eye observation could not detect dust over the surface of BND due to the temperature inversion below a height of 0.45 km. During the Asian dust events, the duration time of dust presented 9.5 hr (BND), 19.5 hr (MS), and 24.5 hr (GS), respectively with the longest time in GS, whereas dust altitudes ranged from 0.4 to 1.3 km (BND), 0.1 to 2.8 km and 4.1 to 4.2 km (MS), and 0.2 to 2.0 km (GS), respectively, while showing the highest altitude in MS. Aerosol optical thickness (AOT) retrieved by LIDAR and skyradiometer (SR), located close to the LIDAR sites, was compared. MS (LIDAR) and Seoul (SR) attained the AOT of 0.64 and 0.50, and GS (LIDAR) and Gongju (SR) attained the AOT of 0.38 and 0.54, respectively. As SR-derived angstrom exponents (AE) during the time period determined as Asian dust by LIDAR data were 0.17 in Seoul (near MS) and 0.30 in Gongju (near GS), it can be said that the characteristics of dust particles were appeared. During the study period, depolarization ratio could serve as a useful indicator to determine dust aerosol. But, it still seems essential to conduct further investigation with longer period of data to better describe the discrepancy of AOT between LIDARs and SR.

  • The interaction between pyronine G(PG) and surfactants has been examined by the absorption and fluorescence spectroscopy. The optimum condition for ${\gamma}$-band in PG-AOT system was found when S/D(concentration of surfactant / concentration of PG) was in the vicinity of 80. The disassociation of stacking conformation occured gradually at S/D>80. In PG-cationic and nonionic surfactant systems the absorptivity of monomeric PG increased. With the increment of the concentration of NaCl in PG-AOT system, the absorptivity of ${\gamma}$-band decreased at S/D<10 but, increased remarkably at S/D>20. PG-AOT system with the addition of NaCl showed greater dissociation than that without the addition of NaCl.

The Effect of Action Observation Training with Acoustic Stimulation on Balance and Gait in Stroke Patients

  • PURPOSE: This study examined the effects of action observational training with acoustic stimulation (AOTA) on the balance and gait ability in stroke patients. METHODS: Forty-five chronic stroke patients were divided into three groups. The AOTA group (n = 15) received training via a video that showed a normal gait with the sound of footsteps. The action observation training (AOT) group (n = 15) received AOT without acoustic stimulation. The control group (n = 15) received physical training. Each intervention was applied once per day, three times per week for six weeks. The participants in the AOTA and AOT groups had five minutes of AOT. The participants in the all group had 20 minutes of physical training. All participants were measured using the Berg Balance Scale, the Timed Up and Go Test, the Functional Reaching Test, 10 Meter Walk Test, six Minute Walk Test, and Dynamic Gait Index. The collected data were analyzed using SPSS version 20.0 for Windows. The between- and within-group comparisons were analyzed using the one-way analysis of variance (ANOVA) test and a paired t-test, respectively. For all statistical analyses, the significance level was set to .05. RESULTS: The one-way ANOVA test identified significant differences among the measurement results of the three groups (p < .05). Post hoc analyses indicated the AOTA group to undergo more significant balance and gait changes than the control group (p < .05). CONCLUSION: The gait and balance abilities could be improved effectively for patients with stroke when action observation training and acoustic stimulation were applied simultaneously.

Visibility Estimated from the Multi-wavelength Sunphotometer during the Winter 2011 Intensive Observation Period at Seoul, Korea (2011년 겨울철 서울시 대기 집중 관측 기간 동안 다파장 복사계로 분석된 에어러솔 연직분포와 시정 거리)

  • The aerosol extinction vertical profile and surface visibility have been derived from the Microtops-II sunphotometer observation during the winter 2011 intensive observation period (IOP) at Seoul, Korea. Using models of degradation of aerosol optical thickness (AOT) and aerosol scale height, we have performed extinction-visibility modulation to determine the height dependent aerosol extinction and visibility. It is shown that the aerosol loading is relatively low during IOP (mean $AOT_{550}=0.22{\pm}0.08$, ${\AA}$ngstr$\ddot{o}$m exponent=$1.14{\pm}0.26$). Modeled extinction by use of Microtops II sunphotometer data shows good agreement with measurements by the Multi-wavelenth Polarization Lidar (MPoLAR), and the derived surface visibility are consistent with data from the transmissometer. These results emphasize the use of a vertically resolved extinction from AOT to predict visibility conditions at ground level.

Tropospheric Ozone Patterns in the Metropolitan Seoul Area During 1990~1997 Using Two Ozone Indices of Accumulation over the Threshold Concentrations (한계농도 누적 오존지표로 본 1990~1997년의 수도권 오존농도 변화)

  • In order to assess the chronic impact of tropospheric ozone on vegetation in the Seoul metropolitan area, it is necessary to quantify ozone exposure. Two ozone indices commonly used to relate ozone exposure to injury of vegetation were calculated. SUM06(SUM of hourly concentrations at or above 0.06 ppm) and AOT40(Accumulated exposure Over a Threshold of 40 ppb) which are widely used as ozone indices in the US and Europe were calculated based on hourly ozone concentrations in 5 areas of Seoul and 5 cities of Kyunggido during 1990~1997. Most SUM06 levels were 1~5ppm.hr, however several areas in Northern and Eastern Seoul reached about 5~7 ppm.hr in 1996~1997. AOT40 values were as high as 17~24 ppm.hr. Although measured SUM06 levels would not be expected to significantly impact vegetation, the overall ozone index, as well as annual average, 95th, and 99th percentile have increased continuously over the last 8 years. Often, ozone concentrations are lower in cities where there is a significant NOx concentration, than in outlying rural agricultural areas where NOx scrubbing is not as important. Concentrations greater than 40 ppb, which can cause chronic ozone toxicity to vegetation, were found mostly in the summer and constitutued about 5~15% of total hourly ozone cocentrations.

Stability of O/W Emulsions Prepared from Winsor Systems-1 (Winsor계에서 제조된 O/W 에멀젼의 안정성 연구-1)

  • We have investigated the stability of macroemulsions of AOT prepared from Winsor systems. The emulsion system was prepared from heptane+aqueous NaCl+AOT. As the concentration of NaCl increases, the stability of the emulsion increases, reaches a maximum, and then decreases. At low salt concentration the creaming occurred due to density difference of oil and aqueous phases. We developed a model for the creaming. Behavior of the emulsions containing low salt could be described very well by the model, however, at high salt concentration the electrostatic repulsion decreased due to screening and droplets started to attract each other and the stability of the emulsion decreased.
