Anastral Spindle 3/Rotatin Stabilizes Sol narae and Promotes Cell Survival in Drosophila melanogaster

  • Apoptosis and compensatory proliferation, two intertwined cellular processes essential for both development and adult homeostasis, are often initiated by the mis-regulation of centrosomal proteins, damaged DNA, and defects in mitosis. Fly Anastral spindle 3 (Ana3) is a member of the pericentriolar matrix proteins and known as a key component of centriolar cohesion and basal body formation. We report here that ana3m19 is a suppressor of lethality induced by the overexpression of Sol narae (Sona), a metalloprotease in a disintegrin and metalloprotease with thrombospondin motif (ADAMTS) family. ana3m19 has a nonsense mutation that truncates the highly conserved carboxyl terminal region containing multiple Armadillo repeats. Lethality induced by Sona overexpression was completely rescued by knockdown of Ana3, and the small and malformed wing and hinge phenotype induced by the knockdown of Ana3 was also normalized by Sona overexpression, establishing a mutually positive genetic interaction between ana3 and sona. p35 inhibited apoptosis and rescued the small wing and hinge phenotype induced by knockdown of ana3. Furthermore, overexpression of Ana3 increased the survival rate of irradiated flies and reduced the number of dying cells, demonstrating that Ana3 actively promotes cell survival. Knockdown of Ana3 decreased the levels of both intra- and extracellular Sona in wing discs, while overexpression of Ana3 in S2 cells dramatically increased the levels of both cytoplasmic and exosomal Sona due to the stabilization of Sona in the lysosomal degradation pathway. We propose that one of the main functions of Ana3 is to stabilize Sona for cell survival and proliferation.

Analysis about Autonomic Nervous Activity and Balance of Workers (근로자들의 자율신경 활성 및 균형도에 대한 분석)

  • Objective : This study was designed to analyze the results of autonomic nervous activity(ANA) and autonomic nervous balance(ANB) by heart rate variability test. The subjects were workers in the automobile manufacturing company. Method : The subjects consisted of 32,049 workers who had answered about questionnaires of a job position, age, smoking, drinking, exercise history and sex. The ANA and ANB were examined by SA3000P. I analyzed the differences of ANA and ANB according to job positions, ages, the state of smoking, drinking, exercise and sex by T-test or ANOVA with SPSS ver. 17.0. Results : Regarding the differences of ANA among job positions, the ANA was lowest in sales positions, highest in production workers. Among ages, the ANA was highest in 50s, lowest in 30s. In smoking, the ANA was higher in non-smoking group. In drinking, the ANA was higher in non-drinking group. In exercise, the ANA was higher in exercise group. In sex, there was no significant difference. The case of ANB, there were no significant differences except for ages. Among ages, the ANB was highest in 20s, the lowest in 40s. Conclusion : According to these results, we should establish the methods of controlling ANA and ANB from the perspective of Korean traditional medicine.

FURTHER PURIFIED GINSENG EXTRACT FRACTION (D-O-ANA) FOR INSULIN RELEASE AND ITS MODE OF ACTION COMPARED WITH THE ISOLATED RESIDUAL COMPONENTS (인삼성분 D-O-ANa이 인슐린 분비에 미치는 영향 및 작용기전에 관한연구)

  • A further purified fraction (D-O-ANa) was obtained from DPG 3-2 fraction of Ginseng Radix by complete removal of saponins, nucleosides, nucleic acid bases, amino acids, and sugars. D-O-ANa - induced insulin release was investigated to compare with that of DPG 3-2 and other isolated components. Among the sub fractions of DPG 3-2, D-O-ANa exhibited the most potent release of insulin with or without high concentrations of glucose, and it particularly enhanced the second phase of glucose-induced insulin release. DGP 3-2 potentiated significantly the glucose-induced insulin release from the isolated islets of diabetic mice at increasing concentrations of extracellular calcium ions (0.16 - 2.5 mM). A definite relationship was found between calcium $(^{45}Ca)$ uptake and insulin release. Ginsenoside $(G)-Rb_1\;and\;G-Rg_1$ did not enhance the glucose-induced insulin release. The effect of ginseng saponins was blocked by glucose (16.7 mM), being distinctly different from the glucose-potentiated effect of DPG 3-2. The insulin release effect of $G-Rg_1$ was unaffected by the presence or absence of extracellular $Ca^{2+}$ and theophylline. Adenosine also increased insulin release from isolated islets, but had no effect on perfused rat pancreas. Arginine stimulated insulin release less evidently than D-O-ANa, though arginineand adenosine-induced glucagon releases were more remarkable. In conclusion, D-O-ANa appears to be a major fraction in insulin release activity of ginseng and its mode of action may be related to $Ca^{2+}$ ion uptake. This physiological mechanism was distinct from that of the abnormal release induced by ginseng saponins.

Properties of Acid Tolerance of the Adipic acid-resistant Mutant of Leuconostoc paramesenteroides (Adipic acid 저항성 변이주 Leuconostoc paramesenteroides의 내산성 특성)

  • Properties of acid tolerance of an adipic acid-resistant mutant, Leuconostoc paramesenteroides (ANaP100) were studied and compared with those of its paired wild type of Leu. paramesenteroides (LPw). The value of protons permeability of LPw after an acid shock at pH 5.0 was 4.3 min, while the value of ANaP100 was 4.8 min at the same pH. The maximal specific activities of ATPase of LPw and ANaP100 were 0.59 unit/mg protein and 0.63 unit/mg protein at pH 6.0, respectively. The release of magnesium ion from the mutant strain was about 27.3% at pH 4 after 2 hrs, while the wild strain was about 52.2% under the same conditions. The contents of $C_{19:0,cyclo}$ and $C_{18:1}$ in a membrane fatty acid of ANaP100 and LPw were higher and lower, respectively, than that of LPw. These results indicated that acid tolerance of ANaP100 was improved in comparison with that of its wild type, LPw.

Characteristics of Acid Tolerance of Adipic Acid-Resistant Mutant Strain, Leuconostoc mesenteroides (Adipic acid-resistant 변이주 Leuconostoc mesenteroides의 내산성 특성)

  • To determine an increased acid tolerance of an adipic acid-resistant mutant Leuconostoc mesenteroides(ANaM100) developed for use as a Kimchi starter, proton permeability of cytoplasm, activities of H+-ATPase, Mg++ release and fatty acid composition of cytoplasmic membranes of strain ANaM100 were studied and compared with those of its wild type (LMw). The value of protons permeability of LMw after an acid shock at pH 5.0 was 5.4 min., while the value of ANaM100 cells was 8.4 min. at the same pH. The pH of maximal specific activ-ities of ATPase originated from the LMw and ANaM100 were 0.87 unit/mg protein at pH 6.0 and 0.92 unit/mg pro-tein at pH 5.5, respectively. The release of magnesium ion from ANaM100 was observed about 12.8% at pH 4 after 2 hours, while the wild strains of LMw released Mg++ about 27.6% under the same conditions. The content of C19:0,cyclo and C18:1 in a membrane fatty acid of ANaM100 was higher and lower, respectively than that of LMw. These results indicated that acid tolerance of adipic acid-resistant strain, ANaM100 was significantly improved in comparison with that of its wild type, LMw. In addition, the strain ANaM100 was adipic resistance based on the result of growth of the strain in comparison with that of strain LMw in a broth containing adipic acid.
