Characterization of the Interaction of Sulfiredoxin (Srx1) with a Vacoular Protein $\alpha$-Mannosidase (Ams1) in Saccharomyces cerevisiae (설피리독신과 알파-만노시다제 간의 단백질 결합 특성에 관한 고찰)

  • Most redox-active proteins have thiol-bearing cysteine residues that are sensitive to oxidation. Cysteine thiols oxidized to sulfenic acid are generally unstable, either forming a disulfide with a nearby thiol or being further oxidized to a stable sulfinic acid, which have been viewed as an irreversible protein modification. However, recent studies showed that cysteine residues of certain thiol peroxidases (Prxs) undergo reversible oxidation to sulfinic acid and the reduction reaction is catalyzed by sulfiredoxin (Srx1). Specific Cys residues of various other proteins are also oxidized to sulfinic acid ($Cys-So_2H$). Srxl is considered one of the oxidant proteins with a role in signaling through catalytic reduction of oxidative modification like in the reduction of glutathionylation, a post-translational, oxidative modification that occurs on numerous proteins. In this study, the role of sulfiredoxin in cellular processes, was investigated by studying its interaction with other proteins. Through the yeast two-hybrid system (Y2HS) technique, we have found that Ams1 is a potential and novel interacting protein partner of Srxl. $\alpha$-mannosidase (Ams1) is a resident vacuolar hydrolase which aids in recycling macromolecular components of the cell through hydrolysis of terminal, non-reducing $\alpha$-D-mannose residues. It forms an oligomer in the cytoplasm and under nutrient rich condition and is delivered to the vacuole by the Cytoplasm to Vacuole (Cvt) pathway. Aside from the role of Srxl as a catalyst in the reduction of cysteine sulfenic acid groups, it may play a completely new function in the cellular process as indicated by its interaction with Ams1 of the yeast Saccharomyces cerevisiae.

Comparison of Design Rainfalls From the Annual Maximum and the Non-annual Exceedance Series (연최대치계열과 비연초과치계열으로부터 산정한 확률강우량의 비교·분석)

  • The annual maximum series (AMS) is usually used to estimate hydrological quantiles in practice because it is simple to construct and straightforward to probabilistic interpretation. However, it is limited to use the AMS in Korea due to the lack of reliable observed data which leads to the overestimation of design rainfall and/or flood. Using the 40-year observations of rainfall provided by the Korea Meteorological Administration, this study constructed the AMS and non-annual exceedance series (NAES) after identifying the independent storm event, analyzed the correlation between design rainfalls estimated from the AMS and NAES, and proposed a new method of point frequency analysis to estimate design rainfalls from the small number of observations.

Health Impact Assessment for Cadmium Exposure: Comparison of Residents around Abandoned Mines with the General Population (인구집단의 비교를 통한 폐금속광산 지역 주민의 카드뮴 노출수준 및 건강영향평가)

  • Objective: We compared the level of blood cadmium exposure with health impacts by using data from a survey of residents near an abandoned mine and a national health survey. Methods: For this study, we used data from 7,046 individuals who participated in the Health Effects Survey of Abandoned Metal Mines (AMS, 2008-2011) and 6,871 individuals who participated in the Korea National Health and Nutrition Examination Survey IV-VI (KNHANES, 2008-2013). To evaluate the health impacts, the quartiles (S1 to S4) were classified according to blood cadmium concentration, and then the odds ratios of S2 to S4 over S1 for exceeding the reference values of renal function, blood pressure, and bone density were compared. Similarly, the odds ratio of AMS over KNHANES was confirmed. Results: In the AMS, adjusted for general characteristics, the geometric mean of blood cadmium concentration was 1.34 ㎍/L, which was statistically significantly higher than that of the KNHANES 1.22 ㎍/L (p<0.001). In the integrated data of AMS and KNHANES, the estimated odds ratio of S4 over S1 for exceeding the reference value was found to be 1.70 for serum creatinine, 1.71 for hypertension, and a T-score 2.02 for the tarsal bone. They were all statistically significant. Conclusion: Residents around abandoned metal mines had a higher blood cadmium concentration than the general population, and the odds for exceeding the reference values were higher for some health indicators. Continuous biomonitoring should be conducted for vulnerable areas such as around abandoned metal mines, and measures to reduce cadmium exposure and management of chronic diseases are needed.

Comparison of the fit of automatic milking system and test-day records with the use of lactation curves

  • Objective: The aim of the paper was to compare the fit of data derived from daily automatic milking systems (AMS) and monthly test-day records with the use of lactation curves; data was analysed separately for primiparas and multiparas. Methods: The study was carried out on three Polish Holstein-Friesians (PHF) dairy herds. The farms were equipped with an automatic milking system which provided information on milking performance throughout lactation. Once a month cows were also subjected to test-day milkings (method A4). Most studies described in the literature are based on test-day data; therefore, we aimed to compare models based on both test-day and AMS data to determine which mathematical model (Wood or Wilmink) would be the better fit. Results: Results show that lactation curves constructed from data derived from the AMS were better adjusted to the actual milk yield (MY) data regardless of the lactation number and model. Also, we found that the Wilmink model may be a better fit for modelling the lactation curve of PHF cows milked by an AMS as it had the lowest values of Akaike information criterion, Bayesian information criterion, mean square error, the highest coefficient of determination values, and was more accurate in estimating MY than the Wood model. Although both models underestimated peak MY, mean, and total MY, the Wilmink model was closer to the real values. Conclusion: Models of lactation curves may have an economic impact and may be helpful in terms of herd management and decision-making as they assist in forecasting MY at any moment of lactation. Also, data obtained from modelling can help with monitoring milk performance of each cow, diet planning, as well as monitoring the health of the cow.

Production of Monoclonal Antibody Against the Plasmalemma of Amoeba and its Application in Determining the Role of Membrane Components (아메바 세포막에 대한 단항체 생산 및 이를 이용한 막 조성 물질의 역할규명)

  • Monoclonal antibodies (MAbs) reacting with the plasmalemma of Amoeba proteus were produced. Specificity of the 3 MAbs was determined by transfer blotting of the SDS polvacryfamide gel. AMS antibody reacted with the mucopolysaccharide bands of the spacer gel, 220 KD and 50 KD proteins of the resolving gel. The maior glycoprotein bands (175 KD, 165 KD) and 50 KD protein of the plasmalemma were recognized by AUG antibody. A third, AMP antibody reacted with the 50 KD protein only. In immunofluorescence microscopy of the enzyme treated cells, the antigens of these MAbs were sensitive to proteases, but not sensitive to neuraminidase. In the assay of cell to substratum attachment after binding with the antibody, AMG and AMP antibodies exerted no effect, but AMS hindered the attachment and cell spreading. Thus the effective components of the plasmalemma in cell to substratum attachment appear to be the mucopolysaccharides and 220 KD protein. The membranes of latex particle infested phagosomes did not show any distinction from the plasmalemma in fluorescence microscopy. Phagosome membranes of amoebae appear to be derived from the plasma membrane without selection in terms of the antigen composition. Amoeba Proteus의 세포막과 반응하는 단세포군 항체를 생산하였다. SDS polyacrylamide gel을 transfer blotting하여 이들 항체의 반응 특이성을 조사해 본 결과 AMS 단항체는 PAS로 염색되는 spacer gel의 mucopolysaccharide 린드, resolving gel의 220 KD 및 50 KD 단백질과 반응하였으며, 세포막의 주요 당단백질인 175 KD 및 165 KD 빈드와 50 KD 단백질은 AMG 단항체에 의해서 인지되었다. 그리고 AMP단항체는 공통인 50 KD 단백질과 특이하게 반응하였다. 효소처리한 아메바의 면역형광칠미경적 조사에서 이들 항체에 대한 항원분자들은 모두 단백질분해효소에 민감하였으며 neuraminidase에 대해서는 변화가 없었다. 이들 항체를 결합시킨 아메바의 용기표면 부착 가능성을 분석한 결과 AMP 및 AMG 단항체는 아무런 영향을 미치지 못하였으며 AMS 단항체는 세포의 용기표면 부착 및 세포의 펴짐을 저해하였다. 따라서 아메바의 용기표면 부착은 mucopolysaccharide 및 220 KD 단백질에 의해서 매게되는 것으로 나타났다. 그리고 latex particle을 담고 있는 식포막은 면역형광형미경적 조사에서 세포막과 차이가 없었다. 따라서 겐포막은 항원 조성에 있어서 비 선택적으로 세포막에서 유도되는 것으로 나타났다.

  • The long-lived radionuclide, $^{10}Be$, is produced by cosmic-ray effects in the atmosphere of the earth as well as its surface and that of other planetary surfaces and atmospheres. Accelerator mass spectrometry (AMS) was developed in late 1970s, which made $^{10}Be$ terrestrial measurements more feasible. Since then, many research applications of $^{10}Be$ for both terrestrial and extraterrestrial applications have been developed, which parallel the wide range of radiocarbon ($^{14}C$) research applications. This paper summarizes production mechanisms of $^{10}Be$ both in the atmosphere and on the surface of terrestrial and extraterrestrial environments and also provides numerous $^{10}Be$ research applications in the fields of geomorphology, oceanography, archaeology, glaciology, cosmochemistry, climatology, and planetary science. We also review some $^{10}Be$ AMS research applications.

Anisotropy of Magnetic Susceptibility (AMS) of Anorthositic Rocks in the Hadong-Sanchong Area (하동-산청지역에 분포하는 회장암질암에 대한 대자율 비등방성 연구)

  • Low-field anisotropy of magnetic susceptibility (AMS) was measured with 247 samples from 17 sites of Pre-Cambrian anorthositic rocks in the Hadong-Sanchong area, southwestern part of the Ryongnam Block. Tectonic stress-direction is defined by the minimum susceptibility (k3) direction, and flow-direction by the maximum susceptibility (k1) direction. Five sites rendered self-consistent NW-SE site-mean tectonic stress-direction. Even though a general fold test for every site was not possible due to the homoclinal nature of the bedding attitudes, a site with various bedding attitudes shows far better clustering of the k3-direction before the bedding-tilt correction. The in-situ NW-SE tectonic stress-direction is consistent over the study area and compatible with petrographic foliation observed in metamorphic rocks in and arround the study area, suggesting a regional compressive force acted after the emplacement of the anorthositic rocks. On the other hand, flow-directions obtained from six sites varies from site to site. Strong-field IRM experiments show predominance of titanomagnetites over a small amount of hematite in some samples.

Holocene Sea Level Reflected from Marine Terrace in Geoje Island and its Influences on Coastal Morphogenesis (거제도 동부해안에서 파악되는 홀로세 고해수준면과 지형발달과정)

  • Coastal terrace was developed at 7.2m height near Shinchon village in Geoje Island. It is located on the east side of southern coast in Korean Peninsula, where sea-level changes caused by ebb and flow of the tide, embayment are relatively low. Due to the breccia layer by mass-movement, dark grayish clayey formation, marine origin's rounded gravel are deposited sequentially in a cross-section of coastal terrace, so it provides a good example which understand Holocene sea level changes to determine the effect on the various sedimentary environments. For the purpose of identifying the morphogenetic process, Grain size, Roundness, XRD, AMS dating analysis was attempted. As a result, after last glacial age, Holocene sea level rise to +5.6m(4,740±100yrs BP). At that time, various geomorphological features are considered to be formed.
