The in vitro Drug Resistance of Escherichia coli Isolated from Piglets, Calve, Lamb and Goats with Diarrhoea (대장균 설사중에 이환된 소, 돼지, 양에서 분리한 대장균의 약제감수성)

  • 대장균 설사중에 걸린 어린 돼지, 송아지, 어린 양에서 분리한 대장균 126주의 항생제와 화학요법제(15종)에 대한 감수성을 disc diffusion technique로 조사한 성적은 다음과 같다. 1. 어린 돼지에서 분리한 62주의 대장균은 gentamicin(GM)에 100%, colistin(CL)에 96.8%, kanamycin(KM)에 93.5%, neomycin(NM)에는 91.9%가 감수성을 가지고 있었으나 ampicillin(AM), erythromycin(EM), lincomycin (LM), novobiocin (NB), penicillin (PC), streptomycin(SM), tetracycline(TC), sulfaisodimidin(SU)에는 내성을 가지고 있었다. chloramphenicol(CP), sulfamethoxazole-trimethoprim(SXT), cephalosporin(CE)에는 각각 75.8%, 64.5%, 50%가 감수성이 있었다. 2. 송아지에서 분리한 32주의 대장균은 GM에 100%, CL에는 87.5%, SXT에는 66.7%가 감수성이 있었으나 CP와 KM에는 각각 40.6%, SU에는 56.2% NM에는 62.5%, SM에는 87.5%가 내성을 가지고 있었다. EM, LM, NB, PC에는 전혀 감수성이 없었으며 AM, SM, TC에도 고도의 내성을 가지고 있었다. 3. 어린 양에서 분리한 32주의 대장균은 GM과CL에 100%, CP에 96.9%, KM과 NM에 90.6%가 감수성이 있었다. SM과 SU에도 71.9%나 감수성이 있었으나 CM, EM, LM, PC, TC에는 대부분 내성을 가지고 있었다. AM에는 21.9%가 감수성이 있었다. 4. 2종류 이상의 약제에 내성을 가진 대장균의 AM, CE, CP, CL, GM, KM, NM. SM, TC, SU등 10종의 약제에 대한 multiple drug resistance pattern(MDRP)을 조사한 바 돼지 유래 약제내성 대장균의 MDRP는 18가지였으며 이중 가장 빈도가 높은 것은 AM, CE, SM, TC, SU 내성형으로 전체의 24.2%나 되었다. 송아지 유래 약제내성 대장균의 MDRP는 17가지였으며, AM, CE, CP, KM, NM, SM, TC, SU 내성형이 28.1%로 가장 빈도가 높았다. 반면에 어린 양에서 분리한 대장균의 MDRP는 9가지였으며 AM, CE, SU의 3약제 내성형이 40.6%로 가장 많았다.

The Stability of Aspalatone and Aspirin in Buffered Aqueous Solution (완충 수용액중 아스파라톤 및 아스피린의 안정성)

  • AM, SM 및 ASA는 수용액중에서 겉보기 1차반응에 따라 분해되었으며 보존온도가 높을수록 분해가 촉진되는 온도 의존성을 나타내었다. AM의 분해경로는 pH 1.22 및 pH 7.0 이상에서는 AM$\longrightarrow$ SM $\longrightarrow$ SA의 경로로 주로 분해되었으며 pH 2.01 - 6.08의 범위에서는 AM $\longrightarrow$ASA$\longrightarrow$SA의 경로로 분해되는 양상을 보였다. 또 pH가 분해에 미치는 영향을 pH-rate profile로 나타낸 결과 AM, SM 및 ASA의 최대안정 pH는 각각 4.0, 3.0, 2.0 부근이 있고 이 조건에서의 분해 반감기는 114, 168, 113 hr로 나타났다. 전체적으로 보면 pH 2.0 이하에서는 ASA가 AM 보다 약간 안정한 편이나 pH 2.0-8.0 사이에서는 AM의 분해속도가 ASA보다 현저히 낮았다. 또 AM은 pH 7.0 이상에서, SM은 pH 6.0 이상에서, ASA는 9.0 이상에서 특수염기촉매반응에 따라 분해가 이루어지는 것을 알 수 있었다. 이온강도($\mu$)의 영향으로는 pH 7.0에서 이온강도가 0.115에서 1.0으로 증가할수록 $\mu$$^{1}$2/에 대해 AM의 분해속도정수가 직선적으로 완만하게 감소되었다. 또 완충수용액 중 AM의 가수분해 억제효과를 검토하기 위해 시클로덱스트린류를 첨가하였을 때, $\beta$-시클로덱스트린과 히드록시프로필기-$\beta$-시클로덱스트린은 AM의 분해를 각각 1.6배 및 1.1배 촉진시켜 촉매적으로 작용하였으며 디메칠-$\beta$-시클로덱스트린은 약 3.2배 분해속도를 억제시켜 안정화제로 작용하였다.Zn^{2+}$, soybean trypsin inhibtor에 의해 25~50% 정도, serine proteinase inhibitor인 phenylmethylsulfonyl floride에 의해 80%정도 활성이 억제되는 특성이 있음을 규명하였다.면역환성 (immunoreactivity)이 나타났고 pyramidal cell layer (PCL)와 glia에 SOD-1이 강하게 염색되었다. APT 병용 투여로 상당수의 경련이 일어나지 않은 흰쥐는 해마의 DG에 FRA가 경미하게 염색되었고, PCL에 SOD-1도 경미하게 나타났으나, 경련이 나타난 쥐에서는 KA만을 투여한 흰쥐와 구별되지 않았다. 이상의 APT의 항산화 효과는 KA로 인한 뇌세포 변성 개선에 중요한 인자로 작용할 것으로 사료되나, 보다 명확한 APT의 기전을 검색하고 직접 임상에 응응하기 위하여는 보다 다양한 실험 조건이 보완되어야 찰 것으로 생각된다. 항우울약들의 항혈소판작용은 PKC-기질인 41-43 kD와 20 kD의 인산화를 억제함에 기인되는 것으로 사료된다.다. 것으로 사료된다.다.바와 같이 MCl에서 작은 Dv 값을 갖는데, 이것은 CdCl$_{4}$$^{2-}$ 착이온을 형성하거나 ZnCl$_{4}$$^{2-}$ , ZnCl$_{3}$$^{-}$같은 이온과 MgCl$^{+}$, MgCl$_{2}$같은 이온종을 형성하기 때문인것 같다. 한편 어떠한 용리액에서던지 NH$_{4}$$^{+}$의 경우 Dv값이 제일 작았다. 바. 본 연구의 목적중의 하나인 인체유해 중금속이온인 Hg(II), Cd(II)등이 NaCl같은 염화물이 함유된 시료용액에 공해이온으로 존재할 경우 흡착에 의한 제거가 가능하다. 한편 이같

Permeation and Enzymatic Degradation of Aspalatone in Gastrointestinal Tract of Rabbit (아스팔라톤의 토끼 위장관 점막 투과 및 효소적 분해)

  • To evaluate the site-specific permeation of aspalatone (acetylsalicylic acid maltol ester, AM) through gastrointestinal tract, the enzymatic degradation and permeation studies were carried out using gastric, duodenal and jejunal mucosae of rabbits. It was found that $15.2{\pm}11.4%$, $11.6{\pm}5.2$ and $0.8{\pm}0.6%$ of the donor dose of AM, salicylmaltol (SM) and aspirin (ASA) permeated through the upper gastric mucosa after 8 hr of permeation, respectively. After 8 hr of AM permeation, SM and ASA were measured to be $15.0{\pm}1.7$ and $2.6{\pm}0.8%$ of the dose in the donor solutions, respectively, and salicylic acid (SA) was not detected even after 6 hr, suggesting a very low gastric damage. For the gastric mucosa, the increase of donor dose from 100 to $1,000\;{\mu}g/ml$ increased the permeation flux dose-dependently (r=0.9905). For the duodenal and jejunal mucosae, however, AM was fully degraded into SM and SA due to the esterase activities within 30 min. AM and ASA were not detected in the receptor solution. This result indicates that AM is not a prodrug of ASA. Addition of potassium fluoride (0.5%) into the donor solution delayed the degradation of AM, but did not allow the permeation through duodenal mucosa even by the inhibition of esterase activity. The addition of $dimethyl-{\beta}-cyclodextrin$ and $2-hydroxypropyl-{\beta}-cyclodextrin$ (5%) into the donor solutions also did not show favorable effects on the permeation of AM through various mucosae. In comparison of permeation rates of AM and ASA through the upper gastric mucosa, the flux of ASA was 4.2 times faster than AM based on the molar concentration. ASA also was fully degraded in the donor solutions faced with duodenal and jejunal mucosae within 2 hr, and was not detected in the receptor solution, suggesting a slower metabolism compared with AM.

Evaluation of short-term cardiac function by tissue Doppler imaging in pre and postoperative period of congenital heart disease (조직 도플러 영상을 이용한 선천성 심장병 수술 전후의 단기 심기능 평가)

  • Purpose : The objective of this study was to assess ventricular function by tissue Doppler imaging (TDI) in children with congenital heart disease (CHD) who have been undergoing open heart surgery (OHS) using cardiopulmonary bypass. We tried to compare the parameters of tissue Doppler imaging before and after OHS in patients with congenital heart disease. Methods : This study was conducted on 32 patients with CHD after OHS from January 2005 to December 2005 at Kyungpook National University hospital. Patients who underwent 2-D echocardiography before and after their OHS. All patients were divided into three groups, left ventricular volume overloading group (group 1), and right ventricular volume overloading group (group 2), and right ventricular pressure overloading group (group 3). The TDIs were examined before and 1 to 3 months after OHS. Peak early diastolic (E), and peak late diastolic (A) velocity of transmitral flow were measured by pulsed wave Doppler examination. Peak systolic (Sm), peak early diastolic (Em), and peak late diastolic (Am) velocity in apical 4-chamber and 2-chamber views were measured by TDI. The author calculated E/Em ratio. Results : The patients were 14 boys and 18 girls and the average age of patients was 2 years and 3 months. The congenital heart diseases which have to get OHS were ventricular septal defect (13 cases), atrial septal defect (7), atrioventricular septal defect (3), isolated pulmonary stenosis (2) and tetralogy of Fallot (7). There were significant decrease of Sm, Em, Am measured on tricuspid annulus and E/Em measured on mitral annulus in apical 4 chamber view (P<0.05). Conclusion : This study showed significant decrease of Sm, Em, Am measured on tricuspid annulus and E/Em measured on mitral annulus in apical 4 chamber view after OHS. These changes might be due to the effects of cardiopulmonary bypass in OHS and/or hemodynamic changes after correction of congenital heart disease. To clarify these changes, further study on more patients is needed.

Epidemiological properties of salmonellosis of poultry farms in Kyongbuk province at recent year (최근 경북지역의 양계농장에서 발생한 salmonellosis의 역학적 특성)

  • The present study was conducted to investigate the epidemiological properties of salmonellosis of poultry farms in Kyongbuk province during the relied from November 1998 to November 1999 And antibiotic susceptibility and biochemical characteristics of 120 Salmonella cultures isolated from chicken samples were also investigated. The results obtained through this study were summarized as follows, 1. Among 667,200 chickens of 31 flocks in 17 layer farms and 9 broiler farms, 61,350 chickens of 31 flocks were died with salmonellosis. The death rate of 25 farms varied from 0.1% to 75.0%, and the average death rate was 9.2%. 2. According to etiological agents, fowl typhoid was shown the most predominant outbreak among the salmonellosis during a year, which accounted for 8R.0%(22/25) of the total case 3. The serotypes of 120 Salmonella isolates were identified as 7 strains(5.8%) of S pullorum, 10 strains(8.4%) of S typhimurium and 103 strains(85.8%) of S gallinarum. 4. Most outbreak of fowl typhoid were prevalent on the layer chicken farms(77.1%), and the summer season(45.5%) also appeared the most hazardous season during the year. 5. It seemed that the Hyline breed(70.6%) was the most susceptible among the layer chicken breeds, and followed by Isabrown(23.5%), Tetra(5.9%) in order. 6. In layers, 76.4% of fowl typhoid occurred commonly from 14 to 40 weeks including the early laying peroid, but in broiler farms, all cases was outbreak within first second weeks. 7 All the strains of S pullorum were resistant to lincomycin(Lm), penicillin(Pm), and steptomycin(Sm), but sensitive to amikacin(Ak), ampicillin(Am), cephalothin(Ce), ciprofloxacin (CiP), chloramphenicol(Cm), colistin(Co), enrofloxacin(Enr), furazolidone(Fu), gentamicin(Gm), kanamycin(Km), neomycin(Nm), polymyxin(Po), and teracycline(Tc). All the strains of S typhimurium were resistant to Lm(100%), Pm(100%), Po(90%), and Sm(90%), but were sensitive to Ak, Am, Ce, CiP, Cm, Co, Enr, Fu, Gm, Km, Nm, and Tc. 8. Minimum inhibitory concentration(MIC) of 103 strains of S gallinarum were also evaluated and their patterns were much more variable than others. All the strains of S gallinarum were sensitive to Ak, Am, Ce, Cip, Cm, Enr, Fu, Km, and Nm, but resistant to Lm(100%), and Sm(100%), 99(96.1%) to Co, 83(80.6%) to Pm, and 83(80.6%) to Po, 55(53.4%) to Gm, and 33(32.0%) to Tc. 9. The multiple drug resistance patterns of 120 Salmonella strains were CoLmPmPo Sm pattern(34.2%), CoGmLmPmPoSmTc(20.8%), CoGmLmPmPoSm(13.3%), CoGmLmSm(7.5%), LmPmSm(7.5%), LmPm(6.7%), CoLmSmTc(3.3%),, CoGmLmSmTc(1.7%), GmLmSmTc(1.7%), CoGmLmPoSm(0.1%), LmPmPo(0.1%), CoLm Sm(0.1%), and LmSm(0.1%), in order.

Drug Resistance of Bacterial Flora Isolated from Kunsan Bay (군산 내만에서 분리된 세균총에 대한 약제 내성 조사)

  • One hundred and twenty three strains of bacterial flora collected from Kunsan bay and examined for drug resistance to 9 antibiotics. The isolated and examined bacteria were Vibrio spp.(44 strains), Pseudomonas spp.(42 strains), Aeromonas spp.(26 strains), Moraxella spp.(9 strains), Enterobacteria spp.(6 strains), Bordetella spp.(3 strains), Alkaligenesis spp.(3 strains), Staphylococcus spp.(3 strains), and Flavobacterium spp.(2 strains). The drugs used were Ampicillin(AM), Penicillin-G(PM), Rifampicin(RF), Streptomycin(SM), Oxolinic acid (OA), Nalidixic acid(NA), Oxytetracycline(OT), Amikacin(AK), and Enorfloxacin(EF). Forty two strains were found to be sensitive to all drugs. The remaining strains showed resistance to various combinations of drugs. Among the resistant strains were mostly restricted to AM(54 strains/43.9%), PM(47 strains/38.2%), RF(35 strains/28.4%), SM(9 strains), OA(5 strains/ 4.06%), and NA(1 strains/0.8%), in combination at high degree showing 15 different drug resistant patterns. The most frequently showed resistant patterns were AM-PM-RF(16 strains/13.4%), AM-PM(8 strains/6.5%), and PM-RF(7 strains/5.6%). These results suggested that Kunsan bay were contaminated with various strains of highly resistant strains to drugs(AM, PM and RF). These results suggest that high levels of various antibiotics have already been introduced to Kunsan bay. Furthermore it seems that chemotherapy of fish disease has become extremely difficult because of the acquirement of multi-drug resistance to wide range of antibiotics.

Genetic properties of R plasmids in Salmonella isolates of swine and bovine origin in Korea I.   • This paper dealt with the distribution of Salmonella (S) infection on 4 herds in Kyungju and Taegu during the period from May to October 1986. Isolated Salmonella were examined for serotypes, antimicrobial drug resistance and detection of R plasmid. The results obtained were summarised as followings: 1. Of total 4.622 samples from 4 herds, 67 Salmonella were isolated from 51 samples(1.1%), and their serovar strains were S typhimurium 6, S derby 5, S infantis 4, S bareilly 4, S dublin 3, S anatum 2, S montevideo 2 and untypable 41. 2. The isolation rate of Salmonella was higher in summer and autumn. 3. Of the 67 strains examined, 45 (67.2%) were resistant to one or more antibiotics, such as ampicillin (Am), cephalothin (Ce), chloramphenicol (Cm), rifampicin (Rf), sulfadimethoxine (Su), and tetracycline (Tc), and higher resistant to Sm (40.2%), Ce (31.3%), Am (23.9%). 4. Of the 45 resistant Salmonella strains, 44 (97.8%) harbored conjugative R plasmids and the transfer frequency of Sm (100%), Ce (95.2%), Tc (91.0%) and Su (80.0%) resistance was much higher than that of the other drug resistance. 5. The most common resistant patterns were Sm, Ce, AmCeCmSmSuTc, and AmCe. 6. In 4 herds, the incidience of drug resistance was 57.7%~100% and transfer frequency of conjugative R plasmid was 96.1%~100%.

Microbiological Studies on Feed Supplements (사료첨가제(飼料添加劑)의 미생물오염(微生物汚染)에 관(關)하여)

  • Eighty one products from 36 kinds of vitamin and mineral feed supplement collected during August, 1984 to February, 1985 were examined for microbiological contamination. In addition, 83 strains of coliform isolated from the samples were tested for the resistance to 8 kinds of antimicrobial drugs and distribution of R plasmid. General bacteria were detected in all of samples tested. Bacterial population was varied from less than 10 per gram of the sample to 1,400,000 per gram and 34 (42%) of 81 samples were contaminated with 100 to 1,000 cells per gram. Coliform isolation, which was more frequent in samples with larger number of general bacteria, was possible in 14 (17.3%) out of 81 samples tested and 6 (33.3%) out of 18 companies were coliform positive in their products. Forty one (49.4%) out of 83 coliform isolates were fecal coliform. The frequency of resistant strains was the highest to sulfadimethoxine (Sa) with 92.8% and followed by streptomycin (Sm, 67.5%), tetracycline (Tc, 50.6%), kanamycin (Km, 26.5%), chloramphenicol (Cm, 18.1%) and ampicillin (Am, 15.7%). No strain was resistant to nalidixic acid (Na) and gentamicin (Gm). The resistance frequency of fecal coliform strains were higher compare to non-fecal coliform strains. There were minimum inhibitory concentration (MIC) of $3,200{\mu}g/m{\ell}$ or higher in 7 strains to Am, 3 to Sm and 3 to Km, and 70 strains had MIC of $1,600{\mu}g/m{\ell}$ of higher to Sa while Tc had MICs from $1.6{\mu}g/m{\ell}$ to $400{\mu}g/m{\ell}$. All strains had MICs of $6.3{\mu}g/m{\ell}$ of lower to Na and $3.1{\mu}g/m{\ell}$ of lower to Gm. Seventy nine (95.2%) of 83 strains were resistant to one or more drugs tested. The most frequent resistance patterns were SaSm (14.5%) and followed by SaSmTc(12%), SaSmTcKm(8.4%) SaTc (8.4%) and SaSmKm (7.2%) ; total 19 different patterns were noted. Thirty two (40.5%) of 79 resistant strains were transferred all of a part of their resistance to Escherichia coli ML 1410. The frequency of transferable resistance was high in Am (100%) and Cm (80%) while low in Tc (38.1%), Sa (18.2%), Sm (17.9%) and Km (4.5%).

Theoretical Studies on the Competitive Sn2 Reactions of O-Imidomethyl Derivatives of Phenols with OH-

  • Nucleophilic substitution reactions of O-imidomethyl derivatives of phenols with OH- were studied theoretically using the semiempirical AM1 and Solvation Model 2.1 (SM2.1) methods in the gas phase and aqueous solution, respectively. In the gas phase, the two reaction paths, in which the imide (1a) or phenol (1b) is functioning as a leaving group, can occur competitively. In contrast, in aqueous solution, path (1b) becomes more favorable than (1a) because the transition states (TS) of path (1b) are more stabilized by solvent. Differences in solvation energies are caused by the structural differences of TS, i.e., the TS via path (1b) is more dissociative than that via path (1a). Therefore we conclude that the solvent effects play an important role in the hydrolysis of O-imidomethyl derivatives of phenols. However, reactivity is dependent on the acidities of both the imide and the phenol fragments since the ρz values vary progressively from 4.2 (Z' = I) to 2.5 (Z' = IV) as the acidities of imide increase. These are in good agreement with the experimental results.

  • This paper dealt with the distribution of Shigella spp. on 5 piggeries in Taegu and Kyungpook during the period from August to October 1987. Isolated Shigella were examined for serogrouping, antimicrobial drug resistance and detection of R plasmid. Genetic properties of R plasmid in Shigella have examined to fertility inhibition (Fi) and gel electrophoresis was performed for the isolation of plasmid DNA. The results obtained were summarised as followings; 1. Of total 2,978 samples from 5 piggeries, 82 strains (2.8%) of Shigella spp. were isolated from 82 samples. The isolated strains were identified as S dysenteriae (60 strains), S flexneri (20 strains) and S sonnei (2 strains). 2. Of the 82 strains examined 67 (95.1%) were resistant to one or more antibiotics, such as ampicillin (Am), chloramphenicol (Cm), kanamycin (Km), nalidixic acid (Na), rifampicin (Rf), streptomycin (Sm), sulfademethoxine (Su), and tetracycline (Tc) and higher resistant to Su (90.2%), Sm (63.4%) and Tc (63.4%). 3. Of the 78 resistant Shigella strains 26 (33.3%) harbored conjugative R plasmids and the transfer frequency of Sm (50.0%), Cm(33.3%) resistance was much higher than that of the other drug resistance. 4.
