Effect of the Ethanol Extract from the Aleurone Layer of Anthocyanin-Pigmented Rice on Blood Glucose and Lipid Metabolism in Streptozotocin Induced Diabetic Rats

  • The present study was designed to investigate the hypoglycemic activity and lipid metabolism of ethanol (EtOH) extract from the aleurone layer of anthocyanin-pigmented (AP) rice in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 210~240 g were divided into 4 groups, normal, diabetic control, and two experimental groups, and diabetes in rats was induced by injection of STZ (45 mg/kg, body weight) into tail vein. The EtOH extract of the powdered aleurone layer of AP rice was administered orally in diabetic rats for 14 days. In order to find the hypoglycemic effects in the animal model, the body weight, plasma glucose levels, cholesterol, HDL-cholesterol, triglyceride (TG), free fatty acid (FFA), aspartate aminotransferase (AST) and alanine amino- transferase activities (ALT) were determined. Oral administration of 1.0 81kg on the EtOH extract for 14 days resulted in a significant reduction in blood glucose, ALT, TG and FFA. However, in the case of 2.0 g/kg, the hypo-glycemic effects were not considerable. This results suggest that the EtOH extract might induce hypoglycemic effects in STZ-induced diabetic rats due to some photochemical components in the aleurone layer of AP rice.

The Effect of Kamihaengche-tang Plus Yukmijihwang-tangon Oxidant and Hg-induced Rabbit's Liver Cell Injury (가미행체탕 합 육미지황탕이 Oxidant 및 Hg에 의한 가토 간세포손상에 미치는 영향)

  • Objectives : This study was carried out to determine whether Kamihaengche-tang plus Yulanijihwang-tang (KCYH) exerts a protective effect against oxidant-induced liver cell injury. Methods : Cell injury was estimated by measuring lactate dehydrogenase (LDH) and alanine aminotransferase (ALT) release, and lipid peroxidation was estimated by measuring malondialdehyde, a product of lipid peroxidation in rabbit liver slices. Results : Oxidants (tBHP and $H_2O_2$) increased dose-dependently LDH release which was significantly prevented by 1% KCYH. The protective effect of KCYH against oxidant-induced cell injury was dose-dependent in the range of 0.05-1 % concentrations. Similarly, KCYH inhibited oxidant-induced lipid peroxidation in a dose-dependent manner. When liver tissues were exposed to Hg (0.5 mM), ALT activity in the medium and lipid peroxidation in tissues were markedly increased. These changes were prevented by 1% KCYH, KCYH restored toxicant-induced inhibition of cellular GSH content. KCYH increased the activities of catalase and glutathion peroxidase in oxidant-treated tissues. Conclusions : These results indicate that KCYH exerts a protective effect against oxidant-induced liver cell injury, and this effect is attributed to prevention of lipid peroxidation. These effects may be due to an increase in concentration of endogenous antioxidants.

YH439, a Hepatoprotective Agent, Suppresses Cytokines and Nitric Oxide Production in LPS-primed Rats Administered with $CCL_4$ ($CCI_4$와 Lipopolysaccharide로 유도한 흰쥐 간 독성에 대한 YH439의 방어작용 : cytokines 및 nitric oxide 생성의 억제)

  • The aim of the present investigation was to examine whether YH439, a hepatoprotective agent, exerts protective effect against hepatotoxicity and reduces the production of cytokines and NO in lipopolysaccharide (LPS)-primed rats with carbon tetrachloride ($CCl_4$). Administration of LPS following a single dose of CCl4 injection resulted in remarkable elevations of the serum $TNF{\alpha},{\;}IL-l{\beta$ and IL-6 level. The serum NO level was moderately elevated and severe liver damage was evidenced by increases in serum alanine aminotransferase (ALT) and sorbitol dehydrogenase (SDH) activities. YH439 decreased the levels of TNF, $IL-l{\beta}$, IL-6, ALT, SDH as well as NO in the serum elevated by CCl4+LPS in a dose-dependent manner. Inducible nitric oxide synthase (iNOS) level was decreased in the liver of rats treated with YH439. The increased iNOS activity induced by LPS and $interferon-{\gamma}$ was significantly decreased in RAW 264.7 cells by YH439 treatment. YH439 increased the GSH level decreased by $CCl_4+LPS$ and suppressed the ratio of GSSG/GSH. The reduction of hepatotoxicity by YH439 may associated with the decrease in the production of cytokines as well as suppression of iNOS protein in conjunction with an increase in the GSH level.

Effects of the Guibi Tang(GT) and Guibi Tang Blended Placenta Hominis(GTPH) on Osteoporotic Rats Induced by Ovariectomy (귀비탕(歸脾湯) 및 귀비탕가자하거(歸脾湯加紫河車)가 난소적출(卵巢摘出)로 유발(誘發)된 흰쥐의 골다공증(骨多孔症)에 미치는 영향(影響))

  • Purpose: This study was carried out to investigate the effects of Guibi Tang(GT) and Placenta Hominis blended Guibi Tang(GTPH) on the postmenopausal osteoporosis. Methods: In order to observe the potential effects of GT and GTPH on the osteoporosis induced by ovariectomy, antioxidative effects of GT and GTPH were measured. Antioxidative activity for the samples was investigated by Rancimat method. Animals were observed for 8 weeks, and vehicle and extracts of prescriptions were administered ad libitum. And we measured the variance of body weight, femoral bone weight, serum level of osteocalcin, calcium, alkaline phosphatase(ALP), estradiol, cholesterol, triglyceride, alanine aminotransferase(ALT) and creatinine. Results: As a result, GTPH showed antioxidative effect, but did not in GT. Body weight increased significantly in 6 weeks after ovariectomy, and the weight of GTPH administered group decreased significantly. GT and GTPH decreased the serum level of cholesterol and triglyceride. Administration of GTPH increased the serum level of ALP when compared with control group. Ovariectomy and GT, GTPH administration had no influence on serum level of ALT and creatinine. Conclusion: According to above results. We suggest that GT and GTPH may be used prevention and curing the postmenopausal osteoporosis.

Lipid lowering activity of Ponciri Fructus and Aurantii Fructus Immaturus on hyperlipemia rats induced by Triton WR-1339. (국산과 중국산 지실(枳實)의 Triton WR-1339 유발 고지혈증(高脂血症) 흰쥐에 대한 효능 연구)

  • Objective : In order to elucidate hyperlipidemic effects of the fruits of Poncirus trifoliata and Citrus aurantium, our experimental study was performed on hyperlipidemia rats induced by Triton WR-1339. Method : The hyperlipidemia rats induced by intraperitonial injection of triton WR-1339 were treated with the fruits of P. trifoliata (rare and preparata) and C. aurantium 50 and 200mg/kg. The amount of triglyceride. total cholesterol. HDL-cholesterol, LDL-cholesterol, AST. and ALT levels were measured. Results: The levels of triglyceride. total cholesterol and LDL-cholesterol were significantly reduced in fruits of P. trifoliata (rare and preparata) and C. aurantiu. Simvastatine treated group compared with those of control but the level of HDL-cholesterol was incresed. The level of AST and ALT were not significantly reduced by the treatment to the fruits of P. trifoliata (rare and preparata) and C. aurantium. Conclusion : In the hyperlipidemia rats induced by triton WR-1339. the fruits of P. trifoliata (rare and preparata) and C. aurantium showed a hyperlipidemic effect.

The Effect of Codium fragile (Chlorophyta) Extract on Hepatic Dysfunction and Hyperlipidemia in Rats

  • To examine the effect of Codium fragile on blood cholesterol and lipid metabolism, hyperlipidemia was induced in experimental animal rats through the administration of a hypercholesterolemic diet. Codium fragile powder was then administered to the rats for 5 weeks, after which, blood biochemical changes such as blood cholesterol, Aspartate Aminotransferase (AST: serum SGOT) and Alanine Aminotransferase (ALT: serum SGPT) enzyme activity, etc. were determined. And histological changes in liver cells were examined using an electron microscope. Codium fragile treatment resulted in a significant reduction of the levels of total cholesterol, blood triglyceride and low-density cholesterol (LDL. Chol) compared to the control rats. In contrast the expression levels of high-density cholesterol (HDL. chol.) were increased. The AST value of the Codium fragile administration group was significantly reduced and the blood ALT value of the Codium fragile group showed a significant decrease in comparison to the negative control group. In summary, this study demonstrated the beneficial possibilities of Codium fragile in improving the abnormality of lipid metabolism caused by liver cell damage and hyperlipidemia.

  • This study was carried out to determine whether Kamihaengche-tang plus Yukmijihwang-tang (KCYH) exerts the protective effect against oxidant-induced liver cell injury. Cell injurt was estimated by measuring lactate dehydrogenase (LDH) and alanine aminotransferase (ALT) release, and lipid peroxidation was estimated by measuring malondialdehyde, a product of lipid peroxidation in rabbit liver slices. $H_2O_2$increased LDH release which was significantly prevented by 1% KCYHT. The protective effect of KCYH against $H_2O_2$-induced cell injury was dose-dependent in the range of 0.05-1% concentrations. These result indicate that KCYH exerts the protective effect oxidant-induced liver cell injury, and this effect is attributed to prevented to prevention of lipid peroxidation. These dffects may be due to an increase in concentration of endogenous antioxidants.

Effect of Garlic on Enzyme Activities of Rats Fed Lard and Alcohol (마늘성분이 Lard와 Alcohol을 섭취한 흰쥐 혈청중의 효소활성에 미치는 영향)

  • The present study has been undertaken to investigate the effects of garlic added to food on the activities of several enzymes in serum of rats fed lard and alcohol. Thirty-five males of Sprague-Dawley strains weighed about 130g were divided into 7 groups, each group receiving a different diet for 10 weeks; i.e. basal diet plus 15% lard, basal diet plus 5% alcohol, basal diet plus 0.5% garlic, basal diet plus 15% lard and 0.5% garlic. Determinations were carried out in the net weight gain, food efficiency ratio, weight of organs, and AST, ALT, lactate dehydrogenase, alkaline phosphatase activities in serum of rats. The results obtained were as follows; Rats given feed containing lard and alcohol showed significant decrease in net weight gain, but garlic caused an increased in food efficiency ratio. Lard supplementation caused an increase in the weight of liver, kidney, spleen, but another groups did not, AST, ALT, ALP, LDH of serum were significantly increased in lard and alcohol containing group but garlic feeding decreased enzyme activities compared to lard and alcohol containing group. The above results suggest that garlic would prevent the metabolic disease of liver by improving hyperlipemia caused by high fat diet.

Hypoglycemic Effect and Hepatic Detoxification Activity of Extracts from Crataegus fructus and Morus alba L. in Alcohol-treated Rats

  • To consider potentially new sources which have hypoglycemic effect and accelerating alcohol detoxification, this study was designed to investigate the effect of Crataegus fructus and Morns alba L. in alcohol-treated rats. I compared the body weight, glucose, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alcohol dehydrogenase (ADH), and aldehyde dehydrogenase (ALDH) of rats administered both alcohol and extract of experimental plants to rats treated with alcohol alone. Administration of extracts of C. fructus and M alba, respectively, resulted in a significant reduction in the blood glucose level and the activities of ADH of liver compared to the control rats, and administration of extract of M. alba showed significantly lower on bodyweight gain in the rats than in other treated rats. In contrast, the activities of ALDH of liver were increased. The activities of AST and ALT between the only alcohol-treated rats and the alcohol and experimental plants-treated rats were no significant difference. The results suggest that C. fructus and M alba have a hypoglycemic effect, and reduce liver damage by accelerating acetaldehyde metabolism in alcoholic rats, so the combined effect of C. fructus and M alba may be considered as an alternative remedy for hangovers, alcohol-induced overweight and alcohol-induced diabetes.

Effect of Siho (Bupleurum falcatum L.) Extracts on Liver Function in $CCl_4$ Intoxicated rats (시호추출물이 간장기능 회복에 미치는 영향)

  • Effect of Siho (Bupleurum falcatum L.) extracts on liver function were investigated in CCl4 intoxicated rats. Plasma $\alpha$-fetoprotein and total protein concentration showed a tendency to decrease in Siho extracts groups. However plasma albumin concentration showed no significantly different in all treatment groups. Activities of plasma asparate aminotransferase and alanine aminotransferase in Siho extracts groups showed a lower values than control group. Alkan phosphatase and lactate dehydrogenase activities showed a tendency to decrease in Siho extracts groups. However ${\gamma}$-glutamyl transferase activity showed no significantly different in all treatment groups. Concentration of plasma triglyceride and total cholesterol showed no significantly different in all treatment groups.
