Effects of Ginseng Saponin on Serum Alanine Aminotransferase Activity in Trained Rats (인삼 사포닌이 훈련된 흰 쥐의 혈청 Alanine Aminotransferase 활성에 미치는 영향)

  • The effects of ginseng saponin on the activity of serum alanine aminoiransferase (ALT) in trained rats were examined. The trained group was given a chronic swimming bout (approx. 90 min/day) for 50 days, and ginseng group was given an oral administration of ginseng saponin (150mg/kg/day) for 2 weeks. Ginseng treated-trained group was given an oral administration of ginseng saponin for 2 weeks prior to the termination of a swimming bout. In this experiment, male rats of Sprague-Dawley strain (250 $\pm$ 20 g) were used. The activities of serum ALT in trained and in ginseng groups increased 72.89% (P < 0.01) and 57.14% (P < 0.01) than in control groups, respectively. Also, the activities of serum ALT increased 69.66% (P <0.01) in saline treated-trained group, and 79.31% (P < 0.01) in ginseng treated-trained group than in control groups which were given saline solution and kept sedentary. The effect of ginseng saponin, as revealed by comparing the ginseng treated-trained group with the saline treated-trained group, was not significant. The present study suggests that training and ginseng saponin significantly increased the activity of serum ALT in rats, but that, in ginseng treated-trained group, ginseng saponin did not raise any further the increased activity of serum ALT by training.

Protective Effects of Chalcone Derivatives for Acute Liver Injury in Mice

  • The hepatoprotective effects of chalcone derivatives were evaluated in D-galactosamine/lipopolysaccharide (D-GaIN/LPS)-induced fulminant hepatic failure in mouse. Thirteen chalcone derivatives were synthesized for study and their hepatoprotective effects were evaluated by assessing aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels in serum. Chalcone preparations were injected into mice at 12 hand 1 h before intraperitoneal injection of D-GaIN/LPS. After abdominal administration, changes in AST and ALT between the control and treated groups were observed. Ten of the synthesized chalcone derivatives exhibited inhibitory effects on D-GaIN/LPS-induced levels of AST and ALT in mice. Compounds 2, 3, 8, 9, and 12 markedly reduced serum AST and ALT at 8 h, inhibited hepatocyte necrosis and showed significant hepatoprotective activities. The activity of compound 3 was compared with the bifendate (DDB) through oral administration. Compound 3 showed much higher inhibitory effects than bifendate for decreasing AST and ALT activity. The results indicate that compound 3 has strong hepatoprotective activity through suppression of tumor necrosis factor­alpha (TNF-alpha) preduction, reduction of the histological change in the liver, and attenuated of hepatocyte apoptosis confirmed by DNA fragmentation assay.

Changes in Liver and Serum Guanase Activity following Administration of Carbon Tetrachloride in Rats (四鹽化炭素에 依한 肝損傷時 Guanase 活性 變動)

  • This paper was intented to charily the cause of an increase of serum guanase activity in rats following injection of $CCI_4$. The cause of increasing serum guanase was focused on the change of guanase activities in both serum and liver, and additionally, these results were compared with the previously known alanine aminotransferase (ALT). Concomitantly the microscopic investigation on the histologic changes, and the determination of lipid peroxides of liver were combined in this experiment for a correlation to observe that the activity of guanase would be effected by the various degree of hepatic injury induced by $CCI_4$. The serum levels of guanase were increased about 2 fold in the fatty change stage (3-12 days), 5.2 fold representing the peak value in necrosis stage (21days), 4.5 fold in early cirrhosis stage (48 days), and 2 fold in severe cirrhosis stage (92 days). These changes of serum guanase activity showed similar patterns to those of ALT activity and lipid peroxides in liver cell. The changes of liver guanase activities showed an increase, whereas ALT activities in liver were markedly decreased. It is likely that the increase of serum guanase activity is based on the excess leaking of guanase into blood by the result of accelerated enzyme synthesis in liver cell of $CCI_4$ intoxicated rats. In addition, the possibility could not be ruled out, however, that the increase of serum guanase activity would be caused by the alteration of membrane permeability.

Relationship between Elevated Serum Alanine Aminotransferase Concentration and Metabolic Syndrome in Korean Adults (한국 성인에서 혈청 Alanine Aminotransferase 농도의 상승과 대사증후군과의 관계)

  • Metabolic syndrome has been strongly associated with elevated alanine aminotransferase (ALT), a surrogate of nonalcoholic fatty liver disease. We investigated the relationship between metabolic syndrome and elevated ALT in the general Korean population. The study sample was comprised of 4,781 Korean adults who had participated in the 2005 Korean National Health and Nutrition Examination Survey. Metabolic syndrome was defined by National Cholesterol Education Program for Adult Treatment Panel III. Elevated ALT was defined as an enzyme activity > 40 IU/L for men, and > 31 IU/L for women. ALT was measured by enzymatic methods. Among participants, 425 (8.9%) subjects displayed elevated ALT. The odds ratios (ORs) for elevated ALT increased in subjects with obesity or one of components of metabolic syndrome such as abdominal obesity, high blood pressure, high fasting glucose, high triglyceride, and low HDL cholesterol after adjusting for age and sex. The unadjusted OR for elevated ALT increased according to the number of components of metabolic syndrome (OR = 1.5, 95% CI: 0.96-2.32 for 1 component; OR = 3.0, 95% CI: 1.98-4.61 for 2 components; OR = 6.3, 95% CI: 4.29-9.35 for ${\geq}3$ components; p for trend < 0.0001). This trend did not differ after adjustments for putative risk factors including age, sex, BMI, smoking status, and alcohol intake. Metabolic syndrome is implicated as a strong risk factor of elevated ALT in Korean adults.

Effects of sosihotangganogyong on the activity of serum alanine aminotransferase (ALT) and hepatic lipid peroxide in mice with $CCl_4$ solution (소시호탕가녹용(小柴胡湯加鹿茸)이 마우스의 혈청 ALT와 간조직(肝組織) 과산화지질(過酸化脂質)에 미치는 영향(影響))

  • Effects of sosihotangganogyong on the activity of serum transferase and hepatic lipid peroxide in mice with $CCl_4$ solution were determined. The activity of ALT showed a high value in the mice with $CCl_4$, however in the mice with sosihotang and sosihotangganogyong, these values showed a tendency to rapid recovery compared with those of the mice with $CCl_4$ only and the activity of ALT in the group of sosihotang and sosihotangganogyong showed a low values compared with $CCl_4$ only group on the 21 days after treatment. On the 21 days after treatment, the activities of glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT) and lactic dehydrogenase (LDH) in the mice with sosihotang and sosihotangganogyog were similar to those of control group, however these values of $CCl_4$ only group showed a high values compared with those of other groups. The value of hepatic lipid peroxide in the mice with sosihotang and sosihotangganogyong showed a tendency to rapid decrease and recovery compared with those of $CCl_4$ only group and on the 21 days after treatment, this value showed a similar to those of control group. The activity of serum transferase and the value of hepatic lipid peroxide in the mice with sosihotngganogyong showed a tendency to decrease compared with those of sosihotang group, however these values showed a no significantly deference. Results from this study indicated that the sosihotagganogyong can effectively improve the recovery of liver function in mice with $CCl_4$.

  • This experimental study was designed to investigate the effects of Cibotii Rhizoma on the muscle atrophy induced by hindlimb suspension in rats. The measurement has been performed on the activity of CK, aldolase, LDH, AST, ALT and quantity of creatine in serum of hindlimb suspension rats. The results were as ; 1. Cibotii Rhizoma significantly inhibited the increase of the activity of CK in serum. 2. Cibotii Rhizoma significantly inhibited the increase of the quantity of creatine in serum. 3. Cibotii Rhizoma significantly inhibited the increase of the activity of aldolase in serum. 4. Cibotii Rhizoma significantly inhibited the increase of the activity of LDH in serum. 5. Cibotii Rhizoma significantly inhibited the increase of the activity of AST in serum. 6. Cibotii Rhizoma significantly inhibited the increase of the activity of ALT in serum.

Influences of Sodium Fluoride Contents on Hepatic Functional Enzyme Activities in Rats (Sodium Fluoride 함량이 흰쥐의 간 기능 효소활성에 미치는 영향)

  • The purpose of this study was to probe the influences of NaF oral administration on a dose-effect relationship between fluoride levels of serum enzyme activity such as alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) in rats fed experimental diets for 5 weeks. All groups increased the activity of serum ALP, AST, ALT, and LDH levels with increasing NaF. In addition the fluoride levels of serum and organ tissues (liver, brain, heart, lung, kidney) in oral NaF groups (NF3~NF50) were significantly increased by adding sodium fluoride in comparison with normal diet group (ND) (p<0.05). These results, a high concentration of sodium fluoride was determined that the toxicity to various organ tissues.

An Effect of Methanethiol Treatment on the Liver Function of Rats Previously Fed Low or High Protein Diet (단백식이 조건을 달리하여 성장한 흰쥐에 Methanethiol 투여가 간기능에 미치는 영향)

  • To evaluate an effect of dietary protein on the intoxication of methanethiol in rats, the methanethiol was intraperitoneally injected to the rats fed a low or high protein diet and then the liver weight per body weight and seurm levels of alanine aminotransferase (ALT) activities were determined to investigate the differences in liver damage between the animal groups fed low protein diet and that fed high protein diet. On the other hand, the hepatic glutathione content and its conjugating enzyme, glutathione S-transferase (GST) activity were determined to clarify the cause of difference in liver function between the two groups. The increasing rate of liver weigh/body wt., serum levels of ALT to its control group were higher in methanethiol-treated rats fed low protein diet than those fed high protein diet. The hepatic content of glutathione and GST activity were higher in rats fed high protein diet than those fed low protein diet and the decreasing rate of hepatic glu-tathione content to its control group was higher in rats fed low protein diet than those fed high protein diet. Furthermore, the hepatic GST activity in methanethiol-treated rats was higher in rats fed high protein diet than those fed low protein diet. In case of control group, the GST activity was also higher in rats fed high protein diet than those fed low protein diet.

Effect of Cyclohexane Treatment on Serum Level of Glutathione S-Transferase Activity in Liver Damaged Rats ($CCl_4$ 에 의한 간손상 모델 실험동물에 있어서 cyclohexane 투여가 혈청 glutathione S-transferase 활성에 미치는 영향)

  • To evaluate the effect of cyclohexane(CH) treatment on the serum levels of glutathion S-transferase(GST) activity in liver damaged animals, damaged liver was induced with pretreatment of 50% $CCl_4$ dissolved in olive oil (0.1 m1/100g body weight) intraperitoneally 17 times every other day. To $CCl_4$-treated rats, CH (1.56 g/kg body weight, i.p) was injected once and then the animals were sacrificed at 4 hours after injection of CH. The $CCl_4$-treated animals were identified as severe liver damage on the basis of liver functional findings, 1,e, increased serum levels of alanine aminotransferase(ALT), alkaline phosphate(ALP) and xanthine oxidase(XO) activities. On the other hand, $CCl_4$-treated animals injected with CH once($CCl_4$-pretreated animals) showed more decreased serum levels of ALT and XO, and more increased those of ALP rather than $CCl_4$-treated animals. In case of comparing the GST with ALT activity in liver, both $CCl_4$-treated and pretreated animals showed similar changing pattern of enzyme actvity. Especially $CCl_4$-pretreated animals showed significantly increased serum level of GST actvity compared with the $CCl_4$-treated those, whereas those of ALT showed reversed tendency. In aspects of GST enzyme kinetics, $CCl_4$-pretreated animals showed higher Vmax of liver GST enzyme than $CCl_4$-treated animals. In conclusion, injection of CH to the liver damaged rats led to enhanced liver damage and more increased activity of serum GST which may be chiefly caused by the enzyme induction.

Changes of Biochemical Composition and Activity of Enzyme in the Serum Exposed to Carbon Tetrachloride in Mice (사염화탄소를 투여한 MICE 혈청내 효소 활성 및 생화학적 조성의 변화)

  • Carbon tetrachloride ($CCl_4$) has been used to treat a variety of parasitic infection in both large and small animals, including Fasciola hepatica. Recently, we can easily contest with carbon tetrachloride in air, food, water, rain and Industry area etc. and it is using in order to induce liver injury in laboratory. In this study. we investigated activitis of LDH, ALP, AST and ALT and amount of cholesterol, triacylglycerol, glucose and BUN in mice serum exposed to $CCl_4.$ The mice divided Into a Naive control(A), corn oil control(B) and experimental group(C, D). Naive control group(A) was given feed and water only. Corn oil control group(B) was given corn oil 1ml /100g of body weight(B.W). Experimental group(C) was given carbon tetrachloride 1ml /100g B.W. ($CCl_4$: corn oil=1:20(V /V) ), Experimental group(D) was given carbon teterachloride 1ml /100g B.W. ($CCl_4$: corn oil=1:10(V /V) ). The results obtained were summarized as follows : 1) The body weight was declined after the l0th day In mice exposed to carbon tetra-chloride. 2) The total protein level in serum was significantly in mice exposed to carbon tetra-chloride($P{\leq}0.05$). The albumin and A /G ratio was decresed significantly in mice exposed to carbon tetrachloride($P{\leq}0.01$) 3) All of the activity of LDH, AST, ALT and ALP in mice serum exposed to carbon tetra chloride inclosed significantly activity of LDH ($P{\leq}0.05$), inclosed significantly activity of AST($P{\leq}0.05, \;p {\leq}0.01$), inclosed significantly activityof ALT 3nd ALP($P {\leq}0.05,\;p {\leq}0.01$). 4) The amount of cholesterol and triacylglycerol, lipid metabolite products in serum was inclosed in case of cholesterol but did not change in case of triacylglycerol.
