울산지역 노인의 아연 영양상태 및 아연 보충의 효과 (The Zinc Nutriture and Effect of Zinc Supplementation on the Elderly Living in Ulsan)

  • This study was intended to examine the zinc status and effect of zinc supplementation on the zinc nutritional status of the elderly living in the Ulsan area. The zinc intake of 207 subjects(male 97, female 110) was measured by a 24-hour dietary recall and food frequency method. Biochemical analysis were conducted from blood and urine samples to evaluate the changes of zinc nutriture with zinc supplementation. The average dietary zinc intake of subjects was $7.7\pm{2.8mg}$ for male and $7.5\pm{2.6mg}$ for female, which were 51.3% and 62.3% of Korean RDA respectively. The first source of zinc was cereal and grain(36%), and the second was eggs and milk group(27%). After 8 weeks of zinc supplementation, the serum zinc content was significantly increased(p<0.01), although the serum copper content was not significantly decrease. Serum HDL- cholesterol level was not significantly decreased with zinc supplementation. Serum alkaline phosphatase(ALP) activity and urinary zinc excretion were significantly increased(p<0.05). The urinary Zn/Cr was not significantly increased. It is suggested from the results that the daily zinc supplementation can be effective to improve zinc nutriture.

Benzo(a)pyrene에 의해 유도된 간기능장해에 미치는 자근 추출액의 영향 (The Effect of Lithospermi Radix on Benzo(a)pyrene-Induced Hepatotoxicity)

  • 민간에서 주로 사용되는 약용식물들 중 간독성의 발현 저해 및 간기능 보호 효과가 우수한 약물을 찾아 임상적인 약효를 밝혀보고자 하는 실험과정으로 탄소화합물의 불완전 연소 및 열분해에 의해 생성되는 간장해 물질인 benzo(a)pyrene으로 유도한 rats의 간독성 발현에 미치는 자근 수침액의 예방 및 치료 효과를 실험한 결과 자근 수침액의 투여는 B(a)P 투여로 현저하게 증가된 혈청 및 간장의 AST, ALT, LDH, ALP 활성을 유의성 있게 감소시켰고 B(a)P 투여로 증가한 혈청 total cholesterol 및 phospholipid 함량 역시 감소시킬 수 있었으나 그 효과는 현저하지 않았다. 즉, 자근 수침액의 투여는 B(a)P에 의한 간독성 발현을 유의성 있게 감소시켰으며 그 효과는 전처리가 후처리에 비해 우수하였다.

시호추출물이 간장기능 회복에 미치는 영향 (Effect of Siho (Bupleurum falcatum L.) Extracts on Liver Function in $CCl_4$ Intoxicated rats)

  • 시호내에 존재하는 간장기능증진에 효과적인 기능성 물질의 소재를 탐색하기 위한 기초연구의 일차실험으로 $CCl_4$에 의해 간장을 손상시킨 흰쥐에게 MeOH 시호추출물을 수준을 달리 하여 급여한 후 간 손상 치유 및 기능회복에 미치는 영향을 처리군 간에 비교, 검토했다. 그 결과, 혈중 AFP및 Total protein량은 시호추출물처리에 의해 감소했다 그러나 혈중 Albumin의 농도는 일정한 경향을 보여주지 않았다. 혈중 AST와 ALT의 활성치는 시호처 리 군에서 하락하였다. 혈중 ALP와 LDH의 활성치는 시호첨가군에서 감소했다 그러나 ${\gamma}$-G7의 활성치는 처리군간에 일정한 경향을 나타내지 않았다. 혈중 Total cholesterol과 Triglyceride량은 전 처리 군에서 유의한 차이를 보여 주지 않았다.

난소적출로 유발된 랫트 갱년기 장애에 대한 가감귀비온담탕의 생리활성 효과 평가 (Anti-climacterium Effects of Gagamguibiondam-tang in Ovariectomized Rats)

  • 목 적: 이 연구에서는 한의학에서 다양한 부인과 질환에 사용되어온 전통 복합 처방인 귀비탕과 가미온담탕을 합방, 가감한 가감귀비온담탕 열수 추출물의 갱년기 장애 개선 효과를 심혈관 장애, 비만, 고지혈증, 골다공증, 장기 지방축적 및 신경정신 장애를 포함한 다양한 사람의 갱년기 장애를 나타내는 것으로 알려진 난소적출(Ovariectomized, OVX) 랫트 모델을 이용하여 평가하였다. 방 법: 난소적출수술 28일 후부터 가감귀비온담탕 추출물(수율 : 22.03%)을 각각 100, 50 및 25 mg/ml의 농도로 멸균 증류수에 용해시키고, 체중 kg 당 5 ml의 용량(500, 250 및 125 mg/kg)으로 매일 1회씩 84일(12주 : 3개월)간 경구 투여한 다음, 항비만효과, 항자궁위축효과 및 골다공증 억제 효과의 3가지 생리활성 효과로 구분하여 평가하였다. 항비만 효과 및 항자궁위축효과를 평가하기 위해, 체중 및 증체량, 혈청중 에스트라디올 함량, 복부 축적 지방 및 자궁의 중량의 변화와 복부 축적 지방의 두께 및 평균 지방세포 직경, 자궁 전체, 상피 및 점막의 두께와 점막내 자궁샘이 차지하는 비율의 변화를 각각 평가하였다. 또한 골다공증 개선효과, 즉 골 보호효과를 평가하기 위해, 대퇴골, 경골 및 요추골의 습, 건조 및 탄화 중량, 골밀도, 골강도, 혈중 osteocalcin 및 bone specific alkaline phosphatase(bALP) 함량, 골량 및 구조와 골흡수에 대한 조직병리학적 변화를 각각 측정하였다. 실험군(5개군; 군당 8마리의 랫트 사용) 거짓수술 대조군(거짓수술 후, 증류수 투여 대조군) 난소적출 대조군(난소적출 수술 후, 증류수 투여 대조군) GGOT500(난소적출 수술 후, 가감귀비온담탕 추출물 500 mg/kg 투여 고용량 실험군) GGOT250(난소적출 수술 후, 가감귀비온담탕 추출물 250 mg/kg 투여 중간용량 실험군) GGOT125(난소적출 수술 후, 가감귀비온담탕 추출물 125 mg/kg 투여 저용량 실험군) 결 과: 난소적출 대조군에서는 거짓수술 대조군에 비해 현저한 체중 및 증체량, 사료 및 물 섭취량, 축적 복부 지방 중량, 혈청 중 osteocalcin 함량의 증가가 자궁, 대퇴골, 경골 및 L5 중량과 혈중 bALP 및 에스트라디올 함량의 감소와 함께 인정되었으며, 현저한 복벽 축적 지방 두께의 증가 및 자궁의 위축, 대퇴골, 경골 및 L4의 골량 및 구조의 감소 소견이 골 흡수 지표(Ocn 및 OS/BS)의 현저한 증가와 함께 조직병리학적 및 조직형태계측학적으로 인정되었다. 즉, 전형적인 에스트로겐 결핍성 갱년기 장애가 난소적출에 의해 유발되었다. 한편 이러한 난소적출에 의한 에스트로겐 결핍성 폐경기 관련 갱년기 장애 소견이, 가감귀비온담탕 추출물 500, 250 및 125 mg/kg의 84일에 걸친 연속 경구 투여에 의해 투여 용량 의존적으로 억제되었다. 결 론: 이상의 결과에서, 가감귀비온담탕 500, 250 및 125 mg/kg의 경구투여는 난소적출 랫트에서 에스트로겐 결핍성 폐경기 관련 갱년기 장애 개선 효과를 투여 용량 의존적으로 나타내었다. 따라서 가감귀비온담탕은 효과적인 갱년기 장애 개선제로서 개발 가능성이 높을 것으로 기대되며, 특히 에스트로겐 결핍성 비만 및 골다공증의 개선에 유효할 것으로 판단된다. 한편 가감귀비온담탕은 총 18종의 약제로 구성되어 있고, 각각 수많은 생리활성 물질을 함유하고 있어, 이후 생리활성을 나타내는 화학성분의 검색과 더불어 다양한 방면으로 기전적인 연구가 체계적으로 수행해야 할 것으로 판단된다.

백서 간손상에 의한 구기자의 유리자유기 소거능에 관한 연구 (The Study of Free Radical Scavenging Effect of Lycii Fructus by Liver Injury of Rats)

  • In the present study, we investigated the protective effect of the Lycii Fructus water extracts (LFE) against CCl4-induced hepatotoxicity and the mechanism underlying these protective effects in the rats. The pretreatment of LFE has shown to possess a significant protective effect by lowering the serum alanine and aspartate aminoteansferase (AST and ALT) and alkaline phosphatase (ALP). This hepatoprotective action was confirmed by histological observation, In addition, the pretreatment of LFE prevented the elevation of hepatic malondialdehyde (MDA) formation and the depletion of reduced glutathione (GSH) content and catalase activity in the liver of CC1₄-injected rats. The LFE also displayed hydroxide radical scavenging activity in a dose-dependent manner (IC50 = 83.6 μg/ml), as assayed by electron spin resonance (ESR) spin-trapping technique. Moreover, the expression of cytochrome P450 2E1 (CYP2E1) mRNA, as measured by reverse transcriptase-polymerase chain reaction (RT-PCR), was significantly decreased in the liver of LFE-pretreated rats when compared with that in the liver of control group. Based on these results, it was suggested that the hepatoprotective effects of the LFE may be related to antioxidant effects and regulation of CYP2E1 gene expression.

건지황(乾地黃) 추출물이 Rat fetus 두개골로부터 분리한 조골세포에 미치는 영향 (The Effect of Dried Roots of Rehmannia glutinosa Extract on Osteoblast in Rat Fetus Calvarial Cells)

  • Objectives: Osteoporosis is characterized by bone loss and morbidity with osteoporotic fracture. In this study, the author aimed to evaluate the effect of dried roots of Rehmannia glutinosa extract (RGE) on osteoblast proliferation in murine calvarial cells. Methods: The osteoblast separated from murine calvariae was cultivated for 6 days and evaluated the cell function. After the addition of RGE on the culture medium, we determined the effect of RGE on the cell viability, cell proliferation, protein synthesis, alkaline phosphatase activity, collagen synthesis and calcified nodule formation of the cultivated osteoblast. Results: The results were summarized as follows. 1. RGE did not change the survival rate of rat calvarial osteoblast. 2. RGE increased the proliferation of rat calvarial osteoblast. 3. RGE increased ALP activity of rat calvarial osteoblast., 4. RGE slightly affected protein synthesis of rat calvarial osteoblast. 5. RGE increased collagen synthesis of rat calvarial osteoblast. 6. RGE slightly affected calcified nodule formation of rat calvarial osteoblast. Conclusions: From these results, it is concluded that RG might improve the osteoporosis resulted from augmentation of osteoblast proliferation.

Acacia ferruginea Inhibits Tumor Progression by Regulating Inflammatory Mediators-(TNF-α, iNOS, COX-2, IL-1β, IL-6, IFN-γ, IL-2, GM-CSF) and Pro-Angiogenic Growth Factor-VEGF

  • The aim of the present investigation was to evaluate the effect of A ferruginea extract on Dalton's lymphoma ascites (DLA) induced tumours in BALB/c mice. Experimental animals received A ferruginea extract (10 mg/kg.b.wt) intraperitoneally for 14 consecutive days after DLA tumor challenge. Treatment with extract significantly increased the life span, total white blood cell (WBC) count and haemoglobin (Hb) content and decreased the level of serum aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), gamma glutamyl transferase (${\gamma}$-GT) and nitric oxide (NO) in DLA bearing ascites tumor models. In addition, administration of extract significantly decreased the tumour volume and body weight in a DLA bearing solid tumor model. The levels of pro-inflammatory cytokines such as tumor necrosis factor-alpha (TNF-${\alpha}$), interleukin-1 beta (IL-$1{\beta}$), interleukin-6 (IL-6) and granulocyte monocyte-colony stimulating factor (GM-CSF), as well as pro-angiogenic growth factors such as vascular endothelial growth factor (VEGF) and inducible nitric oxide synthase (iNOS) were elevated in solid tumour controls, but significantly reduced by A ferruginea administration. On the other hand, the extract stimulated the production of interleukin-2 (IL-2) and interferon-gamma (IFN-${\gamma}$) in animals with DLA induced solid tumours. Increase in $CD4^+$ T-cell population suggested strong immunostimulant activity for this extract. GC/MS and LC/MS analysis showed quinone, quinoline, imidazolidine, pyrrolidine, cyclopentenone, thiazole, pyrazole, catechin and coumarin derivatives as major compounds present in the A ferruginea methanolic extract. Thus, the outcome of the present study suggests that A ferruginea extract has immunomodulatory and tumor inhibitory activities and has the potential to be developed as a natural anticancer agent.

Favorable Hepatoprotective Effects of Gongjin-dan on the Acute Ethanol-induced Liver Damaged C57BL/6 Mice

  • To observe the potential hepatoprotective effects of Gongjin-dan on the acute ethanol (EtOH)-induced liver damages in C57BL/6 mice with its possible action mechanisms. EtOH-mediated acute hepatic damages were induced by oral administration of EtOH total 3 doses. The changes on the body weight, liver weight, albumin, TG, AST, ALP, ALT, hepatic TG contents, hepatic antioxidant defense system, TNF-α, CYP 2E1 activity and mRNA expressions of hepatic lipogenic genes - SREBP-1c, SCD1, ACC1, FAS, PPARγ and DGAT2 or genes involved in fatty acid oxidation - PPARα, ACO and CPT1 were observed with final liver histopathological inspections after 15 days of continuous administration of silymarin 200 mg/kg, Gongjin-dan (GJD) 400, 200 and 100 mg/kg. The results were compared with silymarin 200 mg/kg treated mice. Marked decreases of body and liver weights, increases of serum AST, ALT, Albumin and TG levels, hepatic TG contents, TNF-α level, CYP 2E1 activity and mRNA expressions of hepatic lipogenic genes or decreases mRNA expressions of genes involved in fatty acid oxidation were observed with histopathological changes related hepatosteatosis increases of immunolabelled hepatocytes, as the results of a binge drinking of EtOH in the present study. Also destroys of hepatic antioxidant defense systems were demonstrated in EtOH control mice as compared with intact vehicle control mice, respectively. The results suggest that oral administration of 400, 200 and 100 mg/kg of GJD favorably protected the liver damages from acute mouse EtOH intoxications.

BMP-2를 함유한 2상 알지네이트 담체를 이용한 골수줄기세포의 골분화 (Osteogenic Differentiation of Bone Marrow Stem Cell using Bi-phase Alginate Scaffold Including BMP-2)

  • Purpose: The object of this study is to develop a novel BMP-2 delivery system for continuous osteogenic differentiation and to induce osteogenesis of stem cells using a bi-phase alginate carrier in vitro. Methods: Alginate nanoparticle loaded BMP-2 was prepared by the reverse emulsification-diffusion technique. Physical properties and release profiles of alginate carriers were measured by Instron and ELISA kit, respectively. Cell viability and alkaline phosphate activity of hBMSCs differentiation was also evaluated by MTS and Metra BAP assays, respectively. Results: Optimal concentration for bi-phase alginate carrier was determined as 2 wt% by evaluating mechanical and biological properties, and differentiation of BMSCs for bone regeneration. The 2% bi-phase alginate carrier had the lowest initial and final release ratio. In addition, the 2% bi-phase alginate carrier had a little higher ALP activity than the homogeneous carrier. An improved controlled release profile was obtained by combining alginate hydrogel with lyophilized particles. Conclusion: Bi-phase alginate carrier has many advantages such as biocompatibility and controlled release capability. It is expected to be effective as a scaffold and carrier in bone tissue engineering.

사염화탄소 처리한 SD(rat)에 대한 진균발효쌍화탕의 간독성 치료효과 연구 (Efficacy of fungus-fermentated Ssangwhatang on liver protection in SD male rats treated with $CCl_4$)

  • In this research, as a method for verifying the efficacy of Ssangwhatang and fungus-fermentated Ssangwhatang, a comparative study on the liver protection effect was conducted using animal experiments by inducing the liver toxicity with the $CC_{l4}$ treatment. Inducing the liver damage resulted in the increase in the serum AST and ALP activity, and one day administration of the test material($CCI_4$: 0.5 ml/kg/day) caused 520 IU/L of the ASP activity leading to 29% enhancement in comparison with the normal group and 93% and 81% reductions in the fungus-fermentated Ssangwhatang-administered groups, BFST1 and BFST2, respectively. ALT is 42 IU/L for the normal group and 99 IU/L for the negative control group leading to 135% enhancement. 15 ml/kg/day and 30 ml/kg/day administrations of fungus-fermented Ssangwhatang(BFST) resulted in 51% and 45% decreases in the ALT concentration, respectively. One day administration of 30 ml/kg Ssangwhatang and fungus-fermentated Ssangwhatang caused the LDH in the plasma to tend to decrease. $CCI_4$(1.0 ml/kg/day) administered at the 0th and 4th days led to the observation of the tendency toward the decrease in AST, ALT, and LDH contents. The results indicate that the function of Ssangwhatang is partly reinforced under the fungus-fermentated Ssangwhatang performed in order to verify the efficacy of Ssangwhatang' s effect on the recovery from fatigues.
