Effects of Dokhwalgisaengtang-gami Water Extract on Osteoclast Differentiation and Osteoblast Function in RANKL-induced RAW 264.7 Cell (독활기생탕가미방(獨活寄生湯加味方)이 파골세포 분화 억제와 조골세포 활성에 미치는 영향)

  • Objectives: This study was performed to evaluate the effect of Dokhwalgisaengtang-gami water extract(DGG) on osteoporosis. Methods: The osteoclastogenesis and gene expression were determined in RANKL-stimulated RAW 264.7 cell. And osteoblastogenesis was also determined in rat calvarial cell. Results: The results were summarized as followes. 1. DGG decreased the number of TRAP positive cell in RANKL-stimulated RAW 264.7 cell. 2. DGG inhibited TRAP activity in RANKL-stimulated RAW 264.7 cell. 3. DGG decreased the expression of NAFTc1, MITF in RANKL-stimulated RAW 264.7 cell. 4. DGG increased the expression of iNOS, COX-2, IL-6 in RANKL-stimulated RAW 264.7 cell. 5. DGG decreased the expression of cathepsin K, MMP-9, TRAP in RANKL-stimulated RAW 264.7 cell. 6. DGG increased cell proliferation of rat calvarial cell. 7. DGG increased ALP activity in rat calvarial cell 8. DGG increased bone matrix protein, collagen synthesis and nodule formation in rat calvarial cell. Conclusions: It is concluded that DGG might decrease the bone resorption resulted from decrease of osteoclast differentiation and it's related gene expression. And DGG might increase the bone formation resulted from increase of osteoblast function.

Effects of Mung Bean (Phaseolus aureus L.) Supplementation on BUN and Hepatic Functional Enzyme Activities in Streptozotocin -induced Diabetic Rats (녹두(Phaseolus aureus L.) 급여가 당뇨성 흰쥐의 BUN 및 간 기능 효소 활성에 미치는 영향)

  • The purpose of this study was to investigate the improvement effect of mung bean (Phaseolus aureus L.) on the hepatic functional enzyme and catalase activity of streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley (SD) male rats were divided into four groups (n=6), and fed experimental diets containing mung bean meal [basal diet+5% mung bean (BM), basal diet+STZ+5% mung bean (SM)], and control (Basal Diet, BD), BS groups (basal diet+STZ). Serum concentrations of Blood Urea Nitrogen (BUN) and creatinine were significantly decreased (p<0.05) by 5% mung bean supplementation diet. The activities of aspartate transaminase (AST), alanine transaminase (ALT), akaline phosphatase (ALP), lactate dehydrogenase (LDH), amylase and lipase were decreased in the BD, BM and SM group than BS group. The catalase (CAT) activity was significantly increased (p<0.05) in mung bean supplementation diet (BM, SM group) than diabetic group (BS). In vivo experiments with diabetic rats showed that ingestion of mung bean supplementation diet were effective in BUN concentration, and hepatic functional enzyme activities.

Effect of Garlic on Enzyme Activities of Rats Fed Lard and Alcohol (마늘성분이 Lard와 Alcohol을 섭취한 흰쥐 혈청중의 효소활성에 미치는 영향)

  • The present study has been undertaken to investigate the effects of garlic added to food on the activities of several enzymes in serum of rats fed lard and alcohol. Thirty-five males of Sprague-Dawley strains weighed about 130g were divided into 7 groups, each group receiving a different diet for 10 weeks; i.e. basal diet plus 15% lard, basal diet plus 5% alcohol, basal diet plus 0.5% garlic, basal diet plus 15% lard and 0.5% garlic. Determinations were carried out in the net weight gain, food efficiency ratio, weight of organs, and AST, ALT, lactate dehydrogenase, alkaline phosphatase activities in serum of rats. The results obtained were as follows; Rats given feed containing lard and alcohol showed significant decrease in net weight gain, but garlic caused an increased in food efficiency ratio. Lard supplementation caused an increase in the weight of liver, kidney, spleen, but another groups did not, AST, ALT, ALP, LDH of serum were significantly increased in lard and alcohol containing group but garlic feeding decreased enzyme activities compared to lard and alcohol containing group. The above results suggest that garlic would prevent the metabolic disease of liver by improving hyperlipemia caused by high fat diet.

Effect of Fruiting Body of Cordyceps Militaris on Growth, Lipid and Protein Metabolism, and Enzyme Activities in Male Rats (동충하초 자실체가 흰쥐가 성장률, 지질과 단백질 대사 및 효소활성에 미치는 영향)

  • The effects of fruiting body of Cordyceps militaris on the growth, the food intake, the food efficiency ratio, the lipid metabolism, the serum protein level ad enzyme activity in male rats were studied. Sprague-Dawley rats were fed four types of diets for five weeks, respectiely: a control diet, a control diet supplemented with 2%, 3% or 4% fruiting body of Cordyceps militaris(CF) powder. In rats fed 2% or 3% CF diets the body weight gain, the food intake, the concentratons of hepatic triglyceride and serum LDL-cholesterol, the atherogenic index, and the total lipid, total cholesterol, triglyceride and phospholipid in serum were similar to those in rats fed the control diet. Whereas, in 4% CF diet these were significantly decrased. But the all CF diets feeding could not decrease the food efficiency ratio, the weights of liver, pancreas, spleen, kidney and heart, and the concentration of serum HDL-cholesterol. Also, it was shown that the concentrations of glucose, total protein, albumin, urea and creatinine, and the GOT, GPT, LDH, ${\gamma}$-GTP and ALP activities were the same levels in serum of rats of fed all the experimental diets.

Effects of Chitosan Oligosaccharide Supplementation on Blood Glucose, Lipid Components and Enzyme Activities in Hyperglycemic Rats (키토산 올리고당이 당뇨성 흰쥐의 혈당과 혈중 지질성분 및 효소활성에 미치는 영향)

  • The principal objective of this study was to assess the effects of chitosan oligosaccharide supplementation on the improvement of blood glucose, lipid components and enzyme activities in the serum of streptozotocin(STZ, 55 mg/kg B.W., I.P. injection)-induced hyperglycemic rats fed on experimental diets for 5 weeks. The concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, triglycerides(TG) and phospholipids(PL) in serum were remarkably higher in the hyperglycemic group(group BSW) and STZ(I.P.)+chitosan oligosaccharide supplementation group(group ECW) than those in the control group(group BW, basal diet+water). However the concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, tree cholesterol, cholesteryl ester ratio, TG and PL in serum were lower in the ECW group than in the BSW group, whereas the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration in the ECW group were higher than in the BSW group. The activities of alkaline phosphatase(ALP) and aminotransferase(AST, ALT) in serum were lower in the ECW group than in the hyperglycemic BSW group. The results shown above suggested that chitosan oligosaccharide supplementation effectively improves blood glucose, lipid composition and enzyme activities in the sera of STZ-induced hyperglycemic rats.

Effect of Fed Chlorella on the Change of Lipid Components and Enzyme Activity in Serum of Rat by Lead Exposure (납에 노출된 흰쥐에서 Chlorella 섭취가 혈청내 지방성분 및 효소활성에 미치는 영향)

  • This study was performed to investigate the influence of Chlorella which effected the change of Pb contents, enzyme activity and lipid compounds on the rats fed the beverage involved Pb and the different contented Chlorella added-diets for 18weeks. The rat's weight of 200ppm Pb group was decreased 6.04% and the cause of that was Pb intake. But the rat's weight of Chlorella added-diets +200ppm Pb group was increased 4.02% (p<0.01). When feeding the different(0%, 2%, 5%, 10% ) Chlorella added-diets with the Pb contented beverage to the rats, we could know that the Pb contents accumulated on tissue were decreased to 20.70(0%), 12.88(2%), 14.83 (5%) and 19.56(10%), compared with the quantity of Pb taken in. Total-cholesterol, triglyceride, glucose content and AST, ALT, ALP, LDH enzyme activity in serum were the highest on 200ppm Pb group and those were decreased by the order of different(2%∼5% >10%) Chlorella contents + 200ppmPb group. A significance was recognized on the level of 1%. Therefore, when rats were exposed to Pb, it was thought that the amount of Chlorella intake was adequate on content 2∼5% for reducing the lead toxicity.

  • The present study was undertaken to compare the pharmacological activities of crude Lithospermi radix reported with the clinical uses in the oriental medicine. Crude Lithospermi radix uesd for the treatment of burn, eczema, blister, diuretic, scarlet fever and septicemia as antipyretic, antidotic and antiphlogistics. Therefore we tested the effects of Lithospermi radix water extract on the liver-protective activities in the rats. The results obtained from enzyme assay, measurement of serum and liver alanine. aspartate aminotransferase(ALT, AST) and lipid composition indicated that Lithospermi radix water extract showed significant liver-protective activities against benzo(a)pyrene-induced hepatotoxicity.

Effects of Garlic Shoot Extract on Lipid Metabolism in Hyperlipidemic Rats Fed a High-fat Diet (마늘종 추출물이 고지방식이로 유도된 고지혈증 흰쥐의 지질개선에 미치는 영향)

  • This study was designed to investigate the effect of garlic shoot extract administration on serum and liver tissue lipid levels in hyperlipidemic rats. The rats were fed a 45% high-fat diet to induce hyperlipidemia. They were then administered garlic shoot extract (50%) and ethanol extract (75%) at 200 mg/kg B.W./day (GSA-1, GSB-1) and 400 mg/kg B.W./day (GSA-2, GSB-2) for 5 weeks. The total lipid and triglyceride contents in serum were lowest in the GSB-2 group, and the total cholesterol content was significantly lower in the GSA-2 and GSB-2 groups than in the control group. The high density lipoprotein (HDL)-cholesterol content of the GSB-2 group was similar to that of the normal group. The low density lipoprotein (LDL) and very low density lipoprotein (VLDL)-cholesterol contents were significantly lower in the GSB-1 and GSB-2 group than in the control group. atherogenic index (AI) and cardiac risk factor (CRF) were lowest in the GSB-2 group (0.58 and 1.57, respectively). Aspartate aminotransferase (AST), alanine aminotransferase (ALT), and alkaline phosphatase (ALP) activities were significantly higher in the control group than in the normal group, and the levels of AST, ALT, and ALP were significantly lower in the GSA-2 and GSB-2 groups than in the control group. The total lipid content in liver tissue was significantly lower in all the experimental groups compared to that of the control group, but it was not significantly different among the experimental groups. The total cholesterol and triglyceride content in liver tissue was lowest in the GSB-2 group. Antioxidant activity in serum and liver tissue was highest in the GSB-2 group (40.16% and 47.41%, respectively).The thiobarbituric acid reactive substance (TBARS) content in serum and liver tissue was lowest in the GSB-2 group, with the significant difference. Our results suggest that garlic shoot extracts may improve lipid metabolism in serum and liver tissue and potentially reduce hyperlipidemia.

Effects of Supplemental Agents Enhancing Calcium Absorption on Bioavailability of Starfish Calcium in Rats (흰쥐에서 불가사리칼슘의 체내이용성에 대한 칼슘흡수증진물질의 첨가 효과)

  • This study was conducted to investigate the bioavailability of starfish calcium with substances enhancing calcium absorption. Three week-old young female rats (Sprague-Barley) were divided into 5 groups according to calcium sources and testing agents; calcium carbonate (C), starfish calcium (S), starfish calcium + casein phosphopeptide (S-CPP), starfish calcium+citrate-malate (S-CM), starfish calcium+isoflavone (S-ISO), and were fed experimental diets containing AIN-93G based Ca (0.35% w/w) diet with CPP, CM and ISO for 6 weeks. Blood, femur, urine and feces samples were collected. There was no significant difference among groups in terms of growth and food intake. Serum Ca concentrations were normal in all 5 groups. Serum P concentrations and ALP activities were not significantly different among groups. Ca absorption and retention were significantly increased both in S-CPP and S-CM groups compared to C group (p<0.05). p absorption was significantly higher in S-CPP group than in other groups. While the amount of soluble Ca of intestinal contents did not differ among groups, the amount of insoluble Ca was significantly lower in S-CPP, S-CM and S-ISO groups than in C and S groups. However, the weight, Ca and P concentrations of femur were not significantly different among groups. These results suggest that the addition of CPP and citrate-malate were more effective for enhancing the bioavailability, intestinal absorption and solubility of starfish calcium.

The effect of different crystallization temperature of the hydroxyapatite coating produced by ion beam-assisted deposition on anodizing-treated titanium disks on human osteosarcoma cells (양극산화처리된 티타늄 표면에 이온빔보조증착방식을 이용한 수산화인회석 코팅시 소결온도의 차이가 조골세포에 미치는 영향)

  • Purpose: The aim of this study was to study the effect of hydroxyapatite (HA) coating crystallinity on the proliferation and differentiation of human osteosarcoma cells. Materials and methods: Surface roughness of the titanium disks increased by anodizing treatment and then HA was coated using ion beam-assisted deposition (IBAD). HA coating was crystallized by heat-treated at different temperature ($100^{\circ}C$, $300^{\circ}C$, $500^{\circ}C$, $800^{\circ}C$). According to the temperature, disks were divided into four groups (HA100, HA300, HA500, HA800). With the temperature, crystallinity of the HA coating was different. Anodized disks were used as control group. The physical properties of the disk surface were evaluated by surface roughness tests, XRD tests and SEM. The effect of the crystallinity of HA coating on HOS cells was studied in proliferation and differentiation. HOS cells were cultured on the disks and evaluated after 1, 3, 5, and 7 days. Growth and differentiation kinetics were subsequently investigated by evaluating cell proliferation and alkaline phosphatase activity. Results: Regardless of the heat-treated temperature, there is no difference on the surface roughness. Crystallinity of the HA was appeared in the groups of HA500, HA800. HOS cells proliferation, ALP activity were higher in HA500 and HA800 group than HA100 and HA300. Conclusion: Within the results of this limited study, heat treatment at $500^{\circ}C$ of HA coating produced by IBAD has shown greater effect on proliferation and differentiation of HOS cells. It is considered that further in vivo study will be necessary.
