Characteristics of Structure and Expression Pattern of ADSF/resistin Gene in Korean Native Cattle

  • Adipocyte-specific secretory factor (ADSF)/resistin, a hormone, is a small cysteine-rich protein secreted from adipose tissue and has been implicated in modulating adipogenesis in humans and rodents. The objective of this study was to clone a gene encoding ADSF/resistin and to characterize its function in Korean Native Cattle (Hanwoo). The coding sequence was 330 base pairs and it encoded a protein of 109 amino acids. An NCBI BLAST-search revealed the cloned cDNA fragment shared significant homology (82%) with the cDNA encoding the human ADSF/resistin. The nucleotide sequence homology of the Hanwoo sequence was 73% and 64% for the rat and mouse, respectively. A 654 bp ADSF/resistin gene promoter was cloned and putative binding sites of transcription factors were identified. Tissue distribution of ADSF mRNA was examined in liver, skeletal muscles (tenderloin, biceps femoris), subcutaneous fat, and perirenal fat by RT-PCR. ADSF mRNAs were detected in fat tissues but not in liver and muscles, suggesting that ADSF/resistin expression may be induced during adipogenesis. Although, the physiological function of ADSF/resistin in the cow remains to be determined, these data indicate ADSF is related to the adipocyte phenotype and may have a possibly regulatory role in adipocyte function.

Analysis of the ADSF/resistin Gene Polymorphism Associated with Carcass Traits in Hanwoo (한우 ADSF/resistin 유전자의 단일 염기 다형과 육질관련형질 상관 분석)

  • Adipocyte-specific secretory factor(ADSF)/resistin, an hormone, is a small cysteine-rich protein secreted from adipose tissue and ADSF/resistin has been implicated in modulating adipogenesis in human and rodents. Although the exact role of ADSF/resistin in bovine has not been identified, it may have directly or indirectly involved in adipocyte differentiation. The objective of this study was to investigate its DNA polymorphism associated with carcass traits in Korean Native Cattle(Hanwoo). To investigate DNA polymorphism in Hanwoo ADSF/resistin gene, blood samples were taken from 295 Hanwoo steers belonging to progeny testing at Hanwoo Improvement Center in Korea. Seven single nucleotide polymorphisms(SNPs) were found in intron regions but not in any other regions including promoter (1.7kb) and 4 exons. The highest frequency among SNPs was C186A(0.16/0.84) following G964A (0.156/0.884). The significant correlation(P<0.05) between the SNPs and economic traits was found on 764Ains associated with marbling but not from any other SNPs determined.  A computer simulation was also conducted to assess the efficiency of marker assisted selection(MAS) versus the conventional breeding scheme.  Results revealed that MAS was more efficient as a breeding tool compared to the conventional. In conclusion, ADSF/Resistin gene is one of candidate genes to evaluate the quality, especially marbling score, in Hanwoo.

  • To correspond with the unpredictable future tactical environment, Ive expanded the application of M&S(Modeling & Simulation) that is more scientific and more economic in a field of weapon system development process. This paper describes experience in development of ADSF(Air Defense Simulation Framework) that supports not only HLA(High Level Architecture) which is an international standard in M&S but also TCP/IP as well as real-time distributed simulation. ADSF has been applied to the M-SAM(Medium Range Surface to Air Missile) System Simulator, and satisfying test results through GPS(Global Positioning System) timer has been acquired. As a result, an ADSF which is able to support HLA and TCP/IP as veil as precise real-time simulation has been successfully made. we were in need or a real-time simulation engine to support Air Defense System Simulators that were consisted of several subsystems.

Molecular Cloning of Adipose Tissue-specific Genes by cDNA Microarray

  • In an attempt to isolate novel molecules that may play a regulatory role in adipocyte differentiation, we devised an experimental strategy to identify adipose tissue-specific genes by modifying cDNA microarray technique. We used genefilter membranes containing approximately 15,000 rat non-redundant EST clones of which 4,000 EST were representative clones of known genes and 11,000 ESTs were uncharacterized clones. A series of hybridization of genefilter membranes with cDNA probes prepared from various rat tissues and nucleic acids sequence analysis allowed us to identify two adipose-tissue specific genes, adipocyte-specific secretory factor (ADSF) and H-rev107. Verification of tissue-specific expression patterns of these two genes by Northern blot analysis showed that ADSF mRNA is exclusive expressed in adipose tissue and the H-rev107 mRNA is predominantly expressed in adipose tissue. Further analysis of gene expression of ADSF and H-rev107 during 3T3-L1 adipocyte differentiation revealed that the ADSF and H-rev107 gene expression patterns are closely associated with the adipocyte differentiation program, indicating their possible role in the regulation of adipose tissue development. Overall, we demonstrated an application of modified cDNA microarray technique in molecular cloning, resulting in identification of two novel adipose tissue-specific genes. This technique will also be used as a useful tool in identifying novel genes expressed in a tissue-specific manner.

An experimental study on characteristics of mixture turbulence and flame scale (미연혼합기의 난류특성과 화염 스케일에 관한 실험적 연구)

  • The high loading combustion is accomplished by making the turbulent intensity strong and the scale small in the premixed combustor. The Da-mkoler number, which is decreased by short turbulent characteristic time or by long chemical reaction time, can make the distributed reaction flame. So we developed a doubled jet burner for high loading combustion. The doubled jet burner was designed to make the scale of the flame small by the effect of impingement and increasing shear stress with doubled jet. We investigated the turbulence characteristics of unburned mixture and visualized several flames with the typical schlieren photography. Then we studied the influence of several factors that related the scale of flame. Consequently, the doubled jet burner can make the eddy very small. And we can obtain the detail information of the flame scale through ADSF(the Average Distance between Successive Fringes) in the micro- schlieren photography. The ADSF is not a exact flame scale, but it has qualitative trend with increasing turbulent intensity. The ADSF is diminished remarkably with increasing turbulent intensity. The reason is that strong turbulent intensity makes the flame zone thick and flamelets numerous. We can confirm this fact by the signal analysis of ion currents.
