ADP에 의한 생쥐 난자의 외향전류 증가 효과 (Internal Perfusion of ADP in Mouse Oocytes Increases Outward $K^+$ Currents)

  • To find the mechanism underlying the ADP-induced increase in the outward current in ovulated mouse oocytes, we examined changes in voltage-dependent currents using the whole cell voltage clamp technique and the internal perfusion technique. Eggs were collected from the oviduct of superovulated mice with PMSG and hCG. Membrane potential was held at -60 mV (or -80 mV in the case of recording $Ca^{2+}$ currents) and step depolarizations or hyperpolarizations were applied for 300 ms. By step depolarizations, outward currents comprising steady-state and time-dependent components were elicited. They were generated in response to the positive potential more than 20 mV with severe outward rectification and were blocked by external TEA, a specific $K^{+}$ channel blocker, suggesting that they be carried via $K^{+}$ channels. Internally-perused 5 mM ADP gradually increased outward $K^{+}$ currents (IK) 1 min after perfusion of ADP and reached slowly to maximum (150~170%) 5 min later over the positive potential range, implying that ADP might not be acted directly to the $K^{+}$ channels. IK were decreased by 5 mM ATP without affecting the steady-state component of outward current. In contrast to the effect of ADP and ATP on IK, both effect of ATP and ADP on inward $Ca^{2+}$ currents (ICa) could not be detected due to the continuous decrease in current amplitudes with time-lapse ("run-down" phenomena). To check if there is a G protein-involved regulation in the ionic current of mouse oocytes, 1 mM GTP was applied to the cytoplasmic side, and the outward current and inward currents were recorded. ICa was promptly increased in the presence of GTP whereas IK was not changed. from these results, it is concluded that the ATP-dependent regulation is likely linked in the ADP-induced increase in the outward $K^{+}$ current, and G protein-involved cellular signalling might affect ion channels carrying $Ca^{2+}$ and $K^{+}$ in mouse oocytes.

  • 두 종류의 ADP-riboyltransferase(Yac-1과Yac-2)가 최근 mouse의 임파구로부터 cloning되어 그 특성이 규명되어졌다. Yac-1효소는 ADP-ribosyltransferase활성을 보여주나, 대조적으로, Yac-2는 상당한 양의 NAD glycohydrolase 활성도 소유하고` 있으며 이는 NDA를 우선적으로 가수분해 할 수 있다는 사실을 반영한다. Yac-2는 보존된 두 glutamate에 인접한 위치인 219번에 또 다른 glutamate를 소유하고 있다. 효소 활성에 대한 Glu-219의 효과를 알아보기 위해 두 단계의 재조합 중합효소 연쇄 반응 방법에 의해 Glu-219가 glutamate (E219Q)혹은 alanine(E219A)으로 치환되었다. Gln혹은 Ala으로의 치환결과, ADP-ribosyltransferase활성은 56% (E219Q)혹은 66% *E219A)로 감소하였다. Yac-2단백질의 NAD glycohydrolase 활성은 돌연변이에 의해 영향을 받지 않았다. 이러한 결과는 Yac-2효소의 Glu-219가 ADP-ribosyltransferase 활성에 중요한 역할을 하나, NDA glycohydrolase활성에는 관여하지 않음을 시사한다.

  • 6년생 고려인삼근(Panax ginseng C.A. Meyer) 중의 Sucrose synthase, UDP-glucose pyrophosphorylase 및 ADP-glucose pyrophosphorylase의 활성을 생육 시기별로 조사한 결과, Sucrose synthase 와 ADP-glucose pyrophosphorylase는 뿌리저장활성 지표로서 adaptive enzyme의 특성을 나타내는 반면, UDP-glucose pyrophosphorylase는 maintenance enzyme으로서 존재하였다. 평균기온이 24。C 이상일 때 전분합성이 저하되고 중심부의 산소소비량이 급격히 증가되었다.

  • A detailed procedure was described for the isolation of cratine kinase (ATP-Creatine phosphotransferase, E. C. 2. 7. 3. 2.) from the muscle of the snake Bungarus fasciatus. The original isolation procedure of Kuby et al. for the rabbit muscle enzyme has been modified and extended to include a chromatographic step. The properties of the enzyme have been investigated and kinetic constants for the reverse reactions determined as the followings: 1) A molecular weight of the enzyme was determined by gel filteration on Sephadex G-100 and by electrophoresis on SDS-polyacrylamide was 86,000. 2) Two reactive sulphydryl groups were detected with dithiobis nitrobenzoic acid (DTNB). 3) The nucleotide substrate specificity in the reverse reaction was determined as ADP*2'-dADP>GDP>XDP>UDP with magnesium as the activating metal ion. 4) The order of the metal specificity in the reverse reaction Mg>Mn>$Ca{\sim}Co$ was determined with ADP as substrate. 5) A detailed kinetic analysis was carried out in the reverse direction with $MaADP^-$ as the nucleotide substrate. Initial velocity and product inhibition studies($MaADP^{2-}$ competitive with respect to MgADP- and noncompetitive with respect to $N-phosphorycreatine^{2-}$ ; Creatine competitive with respect to $N-phosphorycreatine^{2-}$ and noncompetitive with respect to Ma $ADP^-)$ indicated that the reaction obeyed a sequential mechanism of the rapid equilibrium random type.

Oxidative Phosphorylation계에 의한 Phosphocreatine의 생성 기작 (Evidences for Functionally Direct Coupling between Mitochondrial Phosphocreatine Formation and Oxidative Phosphorylation)

  • ADP 농도를 증가시킴에 따라 미토콘드리아에 의한 Phosphocreatine의 생성속도는 포화 경향을 보이면서 증진되었다. 이 조건에서 Phosphocreatne 생성 속도를 측정하여 가해준 ADP에 대한 Km값을 구해본 결과 0. 0185 mM 임을 알았고, 이 Km값은 미토콘드리아의 Oxidative Phosphoryla-tion게와 연계되지 않은 (추출한) 미토콘드리아 Creatine Kinase의 ATP에 대한 Km 값인 0. 21mM보다 훨씬 낮음을 알 수 있었다. 이 외에도 ADP 존재하에 Oxidative Phosphorylation에 의한 ATP 생성에 대한 Creatine Kinase의 활성의 영향을 살펴본 결과 이 조건에서 Phosphocreatine은 반응시간에 정비례하게 생성 되었으나, 생성된 ATP는 반응시간에 무관함을 보였다. 또한 Oxidative Phosphorylation에 의해 미토콘드리아 외부 Respiration 용액 내에 이 ATP가 축적되는 속도도 미토콘드리아 Creatine Kinase의 Phosphocreatine 생성과 무관함을 알수 있었다. 이상의 결과들은 Mitochondrial Creatine Kinase가 Oxidative Phosphorylation계와 기능적으로 밀접하게 연계되어 ATP가 아닌 Phophocreatine이 에너지 전달 물질로 직접 이용될 가능성을 시사해준다.

  • Further scaling the semiconductor devices down to low dozens of nanometer needs the extremely shallow depth in junction and the intentional counter-doping in the silicon gate. Conventional ion beam ion implantation has some disadvantages and limitations for the future applications. In order to solve them, therefore, plasma source ion implantation technique has been considered as a promising new method for the high throughputs at low energy and the fabrication of the ultra-shallow junctions. In this paper, we study about the effects of DC bias and base pressure as a process parameter. The diluted mixture gas (5% $PH_3/H_2$) was used as a precursor source and chamber is used for vacuum pressure conditions. After ion doping into the Si wafer(100), the samples were annealed via rapid thermal annealing, of which annealed temperature ranges above the $950^{\circ}C$. The junction depth, calculated at dose level of $1{\times}10^{18}/cm^3$, was measured by secondary ion mass spectroscopy(SIMS) and sheet resistance by contact and non-contact mode. Surface morphology of samples was analyzed by scanning electron microscopy. As a result, we could accomplish the process conditions better than in advance.

  • This paper is concerned on design of organic flow deposition system and development of the deposition process for pentacene thin film by OFD and on electrical characteristics of pentacene films deposited by it. 19F NMR Investigation of F1-ATPase of Escherichia coli Using Fluorinated Ligands

  • Asymmetry amongst nucleotide binding sites of Escherichia coli $F_1$-ATPase was examined using $^{19}F$ NMR signal from fluorinated analogs of adenine nucleotides bound to nucleotide binding sites. ADP-$CF_2-{PO_3}^{2-}$ showed no inhibitory effect to $F_1$-ATPase. But ADP-CHF-${PO_3}^{2-}$ (racemic mixture) showed competitive inhibition of $F_1$-ATPase with $K_i$ of $60\;{\mu}m$. ADP-CHF-${PO_3}^{2-}$ shows only negligible binding to $EF_1$ in the absence of $Mg^2+$. With the addition of $Mg^2+$ to the medium, the $^{19}F$ resonance of free ADP-CHF-${PO_3}^{2-}$ disappeared and the new broad resonances appeared. Appearance of more than two new asymmetric resonances following the binding of ADP-CHF-${PO_3}^{2-}$ to $EF_1$ may indicate that at least one of the isomers showed split resonances. This may suggest that the region between ${\alpha}$-and ${\beta}$-phosphate of ADP-CHF-${PO_3}^{2-}$ which is bound to catalytic sites is experiencing a different environment at different sites.

미래 식량자원으로써 장수풍뎅이 유충의 영양학적 평가 (In vivo assessment of the nutritional value of Allomyrina dichotoma larva protein as future protein resource)

  • 식용 곤충은 인구증가에 따른 식량 문제를 해결하기 위한 하나의 방법으로 제시되며 최근 곤충의 생리활성 기능과 관련한 연구가 많이 수행되고 있다. 반면 식품으로서 활용 가능한 기초연구는 부족한 실정으로 본 연구에서 장수풍뎅이 유충의 단백질 대체 자원으로서 영양학적 기능을 규명하기 위한 실험을 실시하여 장수풍뎅이 유충 단백질(ADP)의 식이 효율, 단백질 효율, 순 소화율 등을 동물실험을 통해 알아보았다. 4주령 SD-rat 24마리를 이용하여 각 그룹 당 8마리 3그룹으로 실험을 실시하였다. 실험군은 사료 내 전체 kcal 20%가 Casein 단백질로 구성된 사료를 식이 하는 일반 식이군(Con), 전체 kcal 20%가 장수풍뎅이 유충의 단백질로 구성된 사료를 식이 하는 장수풍뎅이 유충 단백질식이군(ADP), 그리고 체내 대사작용에 의해 배설되는 단백질(metabolic fecal nitrogen)을 측정하기 위하여 단백질 무 첨가 식이군(NP, non-protein diet)으로 설정했다. 4주간 식이를 공급하여 장수풍뎅이 유충 단백질의 식이 효율(Food Efficiency Ratio, FER), 단백질 효율(Protein Efficiency Ratio, PER), 순 소화율(true digestibility, TD)을 평가했다. 본 실험 결과 식이 효율은 Con군에서 0.52, 장수풍뎅이 유충 단백질 섭취군에서 0.41로 ADP에서 유의적으로 낮은 수준을 보였다. 단백질 효율은 ADP군에서 2.39, Con군에서 2.63으로 ADP군은 Con군에 비해 낮았으나 유의적인 차이는 없었다. 순 소화율은 Con군에서 91%, ADP군에서 80%로 나타났으며 통계적인 유의적 차이는 없었다. 장수풍뎅이 유충의 순 소화율은 80%로 나타났으며 콩과류의 단백질 순 소화율 64-77%에 비해 높은 수준을 보였다.
