Reduced graphene oxide field-effect transistor for biomolecule detection and study of sensing mechanism

  • Graphene, two dimensional sheet of sp2-hybridized carbon, has attracted an enormous amount of interest due to excellent electrical, chemical and mechanical properties for the application of transparent conducting films, clean energy devices, field-effect transistors, optoelectronic devices and chemical sensors. Especially, graphene is promising candidate to detect the gas molecules and biomolecules due to the large specific surface area and signal-to-noise ratios. Despite of importance to the disease diagnosis, there are a few reports to demonstrate the graphene- and rGO-FET for biological sensors and the sensing mechanism are not fully understood. Here we describe scalable and facile fabrication of rGO-FET with the capability of label-free, ultrasensitive electrical detection of a cancer biomarker, prostate specific antigen/${\alpha}1$-antichymotrypsin (PSA-ACT) complex, in which the ultrathin rGO sensing channel was simply formed by a uniform self-assembly of two-dimensional rGO nanosheets on aminated pattern generated by inkjet printing. Sensing characteristics of rGO-FET immunosensor showed the highly precise, reliable, and linear shift in the Dirac point with the analyte concentration of PSA-ACT complex and extremely low detection limit as low as 1 fg/ml. We further analyzed the charge doping mechanism, which is the change in the charge carrier in the rGO channel varying by the concentration of biomolecules. Amenability of solution-based scalable fabrication and extremely high performance may enable rGO-FET device as a versatile multiplexed diagnostic biosensor for disease biomarkers.

피부 섬유아세포에서 광나무 추출물의 항 노화 효과 (Anti-Aging Effect of Ligustrum japonicum Extract in the Human Fibroblast Cells)

  • 본 연구에서는 천연 추출물을 이용하여 화장품 원료를 개발하고자 다양한 천연물을 대상으로 조사하였고 그중 광나무 추출물을 선별하여 주름 개선 원료로 사용하고자 하였다. 고성능액체크로마토그래피를 수행하여 주름 개선 효과로 잘 알려진 oleanolic acid 31.06 %와 ursolic acid 8.92 %를 확인하였다. 제조된 광나무 추출물은 피부 섬유아세포에서 procollagen type I과 hyaluronan synthase-3의 발현을 증가시키고 matrix metalloproteinase (MMP)-1, MMP-2, cyclooxygenase-2의 발현을 감소시킬 뿐 아니라 elastase의 활성을 억제하여 주름 개선 기능을 갖는 것을 확인하였다. 따라서 광나무 추출물은 주름개선 화장품 소재로 개발될 수 있는 잠재력이 있다.

Change in the Gastro-Intestinal Tract by Overexpressed Activin Beta A

  • Originally, activins were identified as stimulators of FSH release in reproduction. Other activities, including secondary axis formation in development, have since been revealed. Here, we investigated the influence of activin ${\beta}_A$ on the body, including the gastro-intestinal (GI) tract. Initially, the activin ${\beta}_A$ protein was detected in the serum proportional to the amount of pCMV-rAct plasmid injected. The induced level of activin ${\beta}_A$ in muscle was higher in female than male mice. Subsequent results revealed that stomach and intestine were severely damaged in pCMV-rAct-injected mice. At the cellular level, loss of parietal cells was observed, resulting in increased pH within the stomach. This phenomenon was more severe in male than female mice. Consistent with damage of the stomach and intestine, activin ${\beta}_A$ often led to necrosis in the tip of the tail or foot, and loss of body weight was observed in pCMV-rAct-injected male but not female mice. Finally, in pCMV-rAct-injected mice, circulating activin ${\beta}_A$ led to death at supraphysiological doses, and this was dependent on the strain of mice used. Taken together, these results indicate that activin ${\beta}_A$ has an important role outside of reproduction and development, specifically in digestion. These data also indicate that activin ${\beta}_A$ must be controlled within a narrow range because of latent lethal activity. In addition, our approach can be used effectively for functional analysis of secreted proteins.

한·중 피동 표현 대조 연구 - 한국어 행위주 표지와 중국어 피동 표지 대비 중심으로 - (A Contrastive Study on Korean and Chinese Passive Expression: Centered on Korean Act Subject Marks and Chinese Passive Marks)

  • 본고는 피동 표현에서의 한국어 행위주 표지 '-에게(한테)', '-에, -로'와 중국어 피동 표지 '피(被)[$b{\grave{e}}i$]/양(?)[$r{\grave{a}}ng$]/규(叫)[$ji{\grave{a}}o$]/급(?)[$g{\check{e}}i$]'를 연구 대상으로 삼아 그 분포양상 및 특징을 살피고 이들 형태를 비교 대조를 통하여 그 대응 관계를 면밀히 고찰해 보고자 하였다. 대조 분석 시 두 언어의 유형적 특징, 피동 표현에서의 행위주, 피동주에 대한 선택 제약, 그리고 '받다'류 피동 표현에서 행위주(피동) 표지의 사용 제약과 같은 3가지 측면에 중점을 두었다. 본 대조 분석을 통해 확인한바, 한 중 피동 표현에서 한국어 행위주 표지 '-에게(한테)', '-에, -로'와 중국어 피동 표지 '피(被)/양(?)/규(叫)/급(?)'는 각각 행위주와 결합하여 부사어 역할을 하고 있는 공통점에도 불구하고 용법에 있어 차이점을 보였다. 먼저 두 언어 유형적 특징에 따라 피동 표현에서 행위주와의 결합 방식이 각각 달리 나타남을 확인하였다. 그리고 한국어 행위주 표지는 오로지 조사 역할을 하여 '행위주 유정성 유무'에 대한 제약만 받는 반면, 중국어 피동 표지 '피(被)/양(?)/규(叫)/급(?)'는 각각 문법화 정도에 따라 행위주 유무뿐만 아니라 피동주 유정성 유무 그리고 문장에 나타나는 어휘의 의미에 따른 제약 등을 보인다. 특히 한국어 '받다'류 피동 표현에서 한국어의 행위주 표지 '에게(한테), -에, -로'는 그대로 사용되지만 대응하는 중국어에서는 피동 표지 사용에 여러 제약이 있음을 확인하였다.

실제 이미지에서 현저성과 맥락 정보의 영향을 고려한 시각 탐색 모델 (Visual Search Model based on Saliency and Scene-Context in Real-World Images)

  • According to much research on cognitive science, the impact of the scene-context on human visual search in real-world images could be as important as the saliency. Therefore, this study proposed a method of Adaptive Control of Thought-Rational (ACT-R) modeling of visual search in real-world images, based on saliency and scene-context. The modeling method was developed by using the utility system of ACT-R to describe influences of saliency and scene-context in real-world images. Then, the validation of the model was performed, by comparing the data of the model and eye-tracking data from experiments in simple task in which subjects search some targets in indoor bedroom images. Results show that model data was quite well fit with eye-tracking data. In conclusion, the method of modeling human visual search proposed in this study should be used, in order to provide an accurate model of human performance in visual search tasks in real-world images.

바나나 잎 추출물의 주름개선 효과 (Antiaging Effects of Musa sapientum L. (Banana) Leaf Extract)

  • To examine the possibility of using Musa sapientum L. (Banana) leaf extract as a cosmetic raw material, banana leaves grown in Jeju Island were extracted with 70% ethanol. Polysaccharides present in banana leaf extract were discarded by precipitation with cold ethanol. Polysaccharide-discarded banana leaf extract promoted procollagen and COL1A1 gene expression, but inhibited matrix metalloproteinase (MMP)-1 and MMP-2 gene expression in human skin fibroblasts when examined by real-time reverse-transcription polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA). The active compound in banana leaves was identified by fractionation with various solvents. The chloroform fraction showed the highest anti-wrinkle efficacy and the active compound of chloroform fraction was identified as corosolic acid by NMR, FT-IR, EA, and HPLC-MS. In addition, banana leaf extract showed anti-oxidative efficacy with an IC50 value of 67.91 ppm, as determined by DPPH free radical scavenging assay. Finally, the anti-wrinkle efficacy of banana leaf extract-containing cream was confirmed by clinical tests. Based on these results, banana leaves could have an application as a cosmetic raw material with anti-wrinkle efficacy.

글로벌 R&D센터에 대한 현장조사연구: 갭접근방법에 의한 글로벌 IT산업정책중심 (A Field Study on Global R&D Centers in Korea: Focusing on Global IT Industry Policy with Gap Approach)

  • 우리나라 IT산업성장에서 혁신잠재력과 혁신수행능력이 중요한 역할을 했다. 글로벌 R&D센터는 IT산업의 혁신의 중요한 요소이다. IT산업에서 혁신환경에 대한 글로벌 R&D센터구조를 파악하기 위하여 R&D센터구성원과 정책입안자 각 50명씩 현장설문을 실시하였다. 본 실증분석결과를 보면 단기적으로 R&D정책은 연구영역과 시장역역의 인식차이를 해소하는 촉진자로서 역할이 중요하며, 중기적으로 R&D정책은 개발과 시장의 영격의 인식차이를 좁히는 조정자로 그리고 장기적으로는 기술능력과 시장의 확산 자로서 역할이 중요함을 보여준다. IT 정책입안자는 R&D정책에서 글로벌 R&D센터를 혁신체계의 요소로 활용하는 것이 요구된다.

광나무의 주성분, Ursolic acid와 Oleanolic acid의 항노화 효능 (Excellent Anti-aging Effects of Ursolic acid and Oleanolic acid Present in Ligustrum lucidum)

  • 광나무(L. lucidum)는 ursolic acid와 oleanolic acid를 다량 포함하고 있다. 본 연구에서는 광나무 열매, 줄기, 잎 세 부위 추출물의 항주름 효능을 평가하였다. 광나무 추출물은 human skin fibroblasts에서 독성이 없을 뿐만 아니라 MMP-1과 MMP-2의 발현을 감소시키고 COL1A1의 발현을 증가시켰다. 이들 추출물은 모두 농도 의존적으로 COL1A1의 발현을 증가시켰으며 MMP-1과 MMP-2의 발현을 감소시켰다. 광나무 세 부위 추출물 가운데, 열매 부위에 가장 많은 양의 ursolic acid 와 oleanolic acid가 함유되어 있었으며 가장 강한 COL1A1 upregulating 효과와 MMP-1과 MMP-2 downregulating 효과를 나타냈다. 이처럼 항주름 효능을 보이는 광나무 열매 추출물은 기능성 화장품 소재로 개발될 수 있는 가능성이 있다.

Revision of Biotechnology Support Act for Accelerating the Bioeconomy

  • The Biotechnology Support Act has provided a legal basis for promoting the biotechnology field as the highest legal authority in the biosciences since its enactment in 1983, and has contributed to enhancing Korea's biotechnology research and development (R&D) capabilities and to revitalizing bioscience ecosystems by establishing policies for supporting biotechnology, expanding the government research and development, and promoting industrial applications. The revised bill of the Biotechnology Support Act is the law that reflects changes in the technological and social environment and that provides a legal basis for practical support of government policies such as total periodic research support, commercialization empowerment, and creation of an innovative research environment, and it will come into effect in November, 2020 after a six-month grace period. The main contents of the revised bill are 1) increasing status as a general norm in the biotechnology field, 2) establishing data evidence-based policies, 3) inducing biotechnology innovation, and 4) promoting regulatory science, especially research and development. This revision has been reorganized in a timely manner in accordance with the current technological advancements, changes in society and environment, and both quantitative and qualitative growth of the domestic bioscience ecosystems and its competitiveness are expected by systematic supports and promotions of the government during the whole period in terms of research and development (R&D) as well as business and industrialization.
