Potential use of dried persimmon (Diospyros kaki) byproducts as feed sources for ruminants

  • The aim of this study was to evaluate the chemical composition, in vitro digestibility, and palatability of dried persimmon byproducts (persimmon peel [PP] and damaged whole persimmons [WP]) ensiled with rice straw in different mixing ratios. PP and WP were ensiled with rice straw at ratios of 3:7 (PP3R7, WP3R7), 5:5 (PP5R5, WP5R5), 7:3 (PP7R3, WP7R3), and 8:2 (PP8R2, WP8R2) for 70 d. WP3R7 had the highest (p < 0.05) crude protein and lactate contents compared to the other combinations. On the other hand, PP3R7 and PP8R2 had lower concentrations of neutral and acid-detergent fibers (p < 0.05) and produced lower amounts of ammonia-N (p < 0.05). The silages were compared to rice straw silage (RS), maize silage (MS), whole-crop rye silage (WCRS), and sorghum-sudangrass silage (SSGS) during an in vitro study. The results showed that PP8R2 and WP7R3 had higher (p < 0.05) dry matter digestibility values than RS, MS, WCRS, and SSGS in a 6 h incubation period. In addition, a palatability test of the silages was conducted on Hanwoo cattle, goats, and deer, using the cafeteria method. The palatability index rate of PP7R3 was the highest (p < 0.05) for the goats and the Hanwoo cattle, whereas PP8R2 had the highest (p < 0.05) rate for the deer and the Hanwoo cattle. In conclusion, dried persimmon byproducts in the form of PP and WPs can be used as ruminant feed when ensiled with RS at ratios of 7:3 and 8:2.

Physico-chemical stability of pesticide formulations under different storage conditions (저장조건별 농약제품의 물리.화학적 안정성)

  • To confirm physico-chemical stability of formulated products, this study was conducted under different storage conditions; room temperature and accelerated temperature of $54^{\circ}C$. The tested pesticide formulations were dichlorvos 50% EC, acephate 50% WP, hymexazol 4% DP, thiram 80% WP and isoprothiolane 12% GR. The selected formulations were stored for 10 weeks and 5 years under the given temperature in maximum and contents of active ingredients were also analyzed by GLC or HPLC after each time of storage. The degradation rates of 5 active ingredients under the two conditions showed a similar trend except acephate. Acephate was rapidly decomposed at $54^{\circ}C$ but slowly decomposed at room temperature, and the degradation rate under the accelerated condition was 2.4 to 5-fold higher than that under the room temperature. Consequently, the stability test on active ingredients in pesticide formulations was able to recommend to be carried out under the accelerated condition except acephate. And the physical properties of all formulations tested under two storage conditions were good.

Endosymbionts of Acanthamoeba Isolated from Domestic Tap Water in Korea

  • In a previous study, we reported our discovery of Acanthamoeba contamination in domestic tap water; in that study, we determined that some Acanthamoeba strains harbor endosymbiotic bacteria, via our molecular characterization by mitochondrial DNA restriction fragment length polymorphism (Mt DNA RFLP). Five (29.4%) among 17 Acanthamoeba isolates contained endosymbionts in their cytoplasm, as demonstrated via orcein staining. In order to estimate their pathogenicity, we conducted a genetic characterization of the endosymbionts in Acanthamoeba isolated from domestic tap water via 16S rDNA sequencing. The endosymbionts of Acanthamoeba sp. KA/WP3 and KA/WP4 evidenced the highest level of similarity, at 97% of the recently published 16S rDNA sequence of the bacterium, Candidatus Amoebophilus asiaticus. The endosymbionts of Acanthamoeba sp. KA/WP8 and KA/WP12 shared a 97% sequence similarity with each other, and were also highly similar to Candidatus Odyssella thessalonicensis, a member of the $\alpha$-proteobacteria. The endosymbiont of Acanthamoeba sp. KA/WP9 exhibits a high degree of similarity (85-95%) with genus Methylophilus, which is not yet known to harbor any endosymbionts. This is the first report, to the best of our knowledge, to show that Methylophilus spp. can live in the cytoplasm of Acanthamoeba.

Aerial Application Tests with Some Low toxicity Insecticides Against the Fall Webworm, Hyphantria cunea Drury (Lepidoptera: Arctiidae) (저독성약제(低毒性藥劑)를 이용(利用)한 미국흰불나방의 항공방제효과(航空防除效果))

  • Aerial application tests with a biological insecticide (Thuricide$^{(R)}$) and two low toxicity insecticides, Diflubenzuron (Dimilin$^{(R)}$) and Triflumuron (Alsystin$^{(R)}$) were carried out against the fall webworm (Hyphantria cunea Drury). For experimental application by helicopter, dilutions of 30x and 45x Thuricide, 180x and 240x Alsystin, and 180x Dimilin were utilized. The solutions were applied at a rate of 30 liters per hectare. The results are as follows: 1) All treatments, Dimilin 25% wp, Thuricide wp, and Alsystin 25% wp, were found to very effec tive against fall webworm larvae. Percentages of mortality averaged between 93.9% and 97%. 2) No significant differences in percentages of larval kill were found between dilutions of Thuricide (30x and 45x) or Alsystin (180x and 240x). 3) Assessment of parasite densities in the treated areas 35 days after treatment indicated that percentages of reduction for all five species found, including Brachymeria lasus(Walker) (=obscurata [Walker]), were lower than for areas treated with the conventional insecticide Diplox$^{(R)}$ (trichlorfon) 25% wp. In particular, the percentage of reduction in Thuricide-treated areas was only 21% as compared with the untreated (control) areas. 4) Aerial applications of Dimilin were very effective and economical for control of the fall webworm by comparison with [aerial] applications of Thuricide.

  • There is well known algorithm is a Gale-Shapley algorithm(GSA) for stable marriage problem. The GSA is performed as each man propose to his most favorite woman(MP), then the woman accepts more than one proposal rejects all but her favorite from among those who have proposed to her. This algorithm always gets a stable set of marriages with man-optimal and woman-pessimal. But the woman proposal and man-accept/reject method(WP) is can be get the distinct result. Also, the optimal stable matching may be fail using MP or WP. This paper suggests always get the optimal stable matching on all occasions in order to overcome the shortcomings of MP and WP. The proposed algorithm perform k-opt, k-women exchange with each other for the result of delete at less preference in each woman from MP result. As a result of applied to various experimental data, this algorithm can be get the optimal stable matching that the MP or WP failed to it.

Assessment of the Effects of Some Herbicides on Mortality of Earthworm (Eisenia fetida) (제초제가 줄지렁이 치사에 미치는 영향평가)

  • The toxic effects of 9 commercially available herbicides on the earthworm, Eisenia fetida (Savigny) were evaluated with recommended dose, 2-fold and 4-fold dose using soil surface spray, immersion and contact filter tests in order to find out whether herbicides actually influence the life of earthworm in the soil or not. In the surface sprayed soil test, the earthworm mortality to nine herbicides were not significantly different from the comparison with the control regardless of level of dose. In the immersion test, the mortality of 4-fold dose was 34% in the napropamide and 64% in the alachlor. In contact filter test, the mortality appeared 80% in alachlor, 37% in napropamide, and 10% in triclopyr at 2-fold dose level and those of 4-fold dose were as follows: napropamide 96%, alachlor 80%, glyphosate 47%, triclopyr 37%, paraquatdichloride 37%; glufosinate ammonium 33%, pyributicarb 10%. As a result of these tests by three methods, the mortality of earthworm in terms of the recommended dose level was no observation about eight herbicides while the napropamide appeared 33% in contact filter test.

Comparison of Size-Exclusion Chromatography and Flow Field-Flow Fractionation for Separation of Whey Proteins

  • Whey protein (WP) is a mixture of proteins, and is of high nutritional values. WP has become an important source of functional ingredients in various health-promoting foods. In this study, size-exclusion chromatography (SEC) and asymmetrical flow field-flow fractionation (AsFlFFF) were used for separation and analysis of whey proteins. It was found that a lab-prepared WP from raw milk is mostly of ${\beta}$-lactoglobulin with small amount of higher molecular weight components, while a commercial whey protein isolate (WPI) powder contains relatively larger amount of components other than ${\beta}$-lactoglobulin, including IgG and protein aggregates. Results suggest that AsFlFFF provides higher resolution for the major whey proteins than SEC in their normal operation conditions. AsFlFFF could differentiate the BSA and Albumin, despite a small difference in their molecular weights, and also was able to separate much smaller amount of aggregates from monomers. It is noted that SEC was able to show the presence of low molecular weight components other than the major whey proteins in the WP samples, which AsFlFFF could not show, probably due to the partial loss of those low molecular weight species through the membrane.

Buffering Capacity of Dairy Powders and Their Effect on Yoghurt Quality

  • Preheating conditions (low-, medium-, and high heat-) did not significantly affect the buffering capacity (BC) of skim milk powder (SMP), whereas the level of demineralization significantly affected the BC of whey powders (WP). Heat treatment ($85^{\circ}C$ for 30 min) of both SMP and WP (90% demineralized) mixtures (88:12, 76:24, 64:36 and 52:48; SMP:WP) resulted in a reduced BC, and the extent of this reduction increased with the proportion of WP increased in the samples. High-buffering milk prepared by the addition of phosphate salts (40 mM $NaH_2PO_4$ and 60 mM $Na_2HPO_4$) delayed the rate of pH decline during yoghurt fermentation. The high-buffering yoghurt showed a significantly higher water holding capacity (WHC) than that of control yoghurt (p<0.05), as well as a more uniform and interconnected microstructure with small pore sizes than those of control yoghurt. No significant differences were found between high-buffering and control yoghurt regarding the viable bacterial counts of starter. The manipulation BC can potentially improve the quality characteristics of yoghurts, such as WHC and texture.

  • A Weyl type algebra is defined in the book ([4]). A Weyl type non-associative algebra $\={WP_{m,n,s}}$ and its restricted sub-algebra $\={WP_{m,n,s_{\gamma}}}$ are defined in various papers ([1], [12], [3], [11]). Several authors 0nd all the derivations of an associative (Lie or non-associative) algebra in the papers ([1], [2], [12], [4], [6], [11]). We find all the non-associative algebra derivations of the non-associative algebra $\={WP_{0,2,0_B}$, where $B=\{{\partial}_0,\;{\partial}_1,\;{\partial}_2,\;{\partial}_{12},\;{\partial}^2_1,\;{\partial}^2_2\}$.

The Mechanism of Antimutagenic Effect of Cinnamaldehyde on Chemical Mutagenesis (화학적 변이에 대한 Cinnamaldehyde의 항돌연변이)

  • The antimutagenic mechanism of cinnamaldeyde on mutagenesis induced by 4-nitroquinoline-1-oxide(4-NQO) and N-metyl-N'-nitro-N-nitrosoguanidine (MNNG) was investigated in various DNA repair-deficient strains, E. coli B/r and K-12 series. Cinnamaldehyde did not show any effects not only on the $\beta$-galactosidase activities of GW1060 and GW1103(recA441) which synthesizes $\beta$-galactosidase consitutively at 41$^{\circ}C$ but also on that of GW1107[lexA51 (Def)] in which the SOS response always occur. These results suggest that cinnamaldehyde dose not change the function of RecA which positively controls the SOS response as well as not acting as the repressor like LexA. In addition, no inhibitory effect of cinnamaldehyde was observed on the growth of Trp+ revertant and the delay of viable cell growth was also not found by adding cinnamaldehyde. Despite the decrease in the number of revertants, a significant increase in survival of 4-NQO treated cells was observed in E. coli WP2s(uvrA), ZA159($\Delta$uvrB) and TK603(uvrA). But these effects disappeared in excision-proficient strain WP2(uvrA+) and lexA-deficient strains(CM561 and CM611). The enhancement of survival was not found in WP67(uvrA polA) deficient in polymerase I which ligates the gap between complementary DNA. From the above results, we assume that cinnamaldehyde might show antimutagenic effect by enhancing an error-free recombinational repair system.
