miR-380-3p promotes β-casein expression by targeting αS1-casein in goat mammary epithelial cells

  • Objective: αS1-Casein is more closely associated with milk allergic reaction than other milk protein components. microRNA (miRNA) is a class of small non-coding RNAs that modulate multiple biological progresses by the target gene. However, the post-transcriptional regulation of αS1-casein expression by miRNA in ruminants remains unclear. This study aims to explore the regulatory roles of miR-380-3p on αS1-casein synthesis in goat mammary epithelial cells (GMEC). Methods: αS1-Casein gene and miR-380-3p expression was measured in dairy goat mammary gland by quantitative real-time polymerase chain reaction (qRT-PCR). miR-380-3p overexpression and knockdown were performed by miR-380-3p mimic or inhibitor in GMEC. The effect of miR-380-3p on αS1-casein synthesis was detected by qRT-PCR, western blot, luciferase and chromatin immunoprecipitation assays in GMEC. Results: Compared with middle-lactation period, αS1-casein gene expression is increased, while miR-380-3p expression is decreased during peak-lactation of dairy goats. miR-380-3p reduces αS1-casein abundance by targeting the 3'-untranslated region (3'UTR) of αS1-casein mRNA in GMEC. miR-380-3p enhances β-casein expression and signal transducer and activator of transcription 5a (STAT5a) activity. Moreover, miR-380-3p promotes β-casein abundance through target gene αS1-casein, and activates β-casein transcription by enhancing the binding of STAT5 to β-casein gene promoter region. Conclusion: miR-380-3p decreases αS1-casein expression and increases β-casein expression by targeting αS1-casein in GMEC, which supplies a novel strategy for reducing milk allergic potential and building up milk quality in ruminants.

LVDC 배전을 위한 출력 380V DC-DC 컨버터 설계에 관한 연구 (A study on the Design of Output 380V DC-DC Converter for LVDC Distribution)

  • 본 연구에서는 LVDC 배전용 출력 380V DC-DC 컨버터를 3가지 유형으로 설계하였고, 시뮬레이션을 통해 3가지 유형의 DC-DC 컨버터의 전압과 전류 특성을 비교 분석하였다. 전력용 MOSFET와 2개의 전류억제용 IGBT를 병렬구조로 적용하여 컨버터를 구성한 경우, 출력 전압이 DC 380V로 안정화 된 시간이 9ms로 비교적 짧았으며, 출력 측 전류 변화 폭도 44.8~50.2A로 IGBT를 적용하지 않았을 경우(68~83A) 보다 훨씬 변화 폭도 작고 전류억제 효과도 더 뛰어남을 알 수 있었다. 이러한 결과는 제안한 LVDC 배전용 DC-DC 컨버터가 스마트 그리드 구축에 적용 가능성이 있음을 시사한다.

THE APPLICATION OF THE TOMS AEROSOLS RETRIEVAL ALGORITHM TO GLI MEASUREMENTS

  • We have applied the TOMS aerosols retrieval algorithm to GLI measurements. TOMS has utilized the aerosol index, which is a measure of the change in spectral contrast due to the wavelength-dependent effects of aerosols. We have retrieved the GLI aerosol index, which is made by the pair of 380/400nm, 380/412nm, 380/460nm, and 412/460nm. We have found that the biomass burning aerosols represent the absorbing aerosols. In addition, the pair of 380/460nm has shown the best signal for detecting aerosols in Principal Component Analysis(PCA) and comparison of aerosol optical thickness from AERONET data. The theoretical aerosol index is also shown the best signal in the pair of 380/460nm.

  • In this paper, we established a prediction model of fuel consumption at the aircraft's taxi operation. To look for countermeasures to reduce fuel consumption and carbon emissions, Airbus A380's actual ground taxi data was used. As a result, the number of stops or turnings during the taxi operation was not related to fuel consumption. It was confirmed that the amount of fuel consumption in the taxi operation was the taxi time and the thrust change. It can be confirmed that ground control optimization, which is the result of close cooperation between the control organization and the airline, is absolutely necessary to reduce taxi time and minimize the occurrence of thrust change events.

  • In the manufacturing process of metal matrix composites parts, thixoforging is one of the most effective forming processes. The major purpose of the current study is to provide the proper conditions such as the die shape, the forging velocity, the forging time, the forging pressure and reinforcement injection velocity and pressure on various defects in thixoforged cylinder liner, filling tests were performed by MAGMA S/W. In order to evaluate the effectiveness of the calculated conditions which is given by computer aided engineering, A357, A380 and SiC$_{p}$/A380 cylind~5$mu extrm{m}$r liner were fabricated under the calculated conditions. SiC$_{p}$/A380 composite billets were fabricated by both the mechanical stirring and electrical magnetic stirring process. Incase fo SiC$_{p}$/A380 composite cylinder liner, reinforcement distribution and effect of reinforcement(SiC$_{p}$) content(10~20 vol. %)and size(5.5~14${\mu}{\textrm}{m}$) on the mechanical properties were investigatedstigated.

자궁내피임장치 카파티 380A에 관한 임상적 고찰 (Clinical Expenience with Copper T 380A Intrauterine Contraceptive Device)

  • Copper T 380A intrauterine devices were inserted to nine thousand and nine hundred twenty women by 51 KAVS-member physicians since January 1, 1993 through December 31, 1993. This study was undertaken for the clinical analysis on a total of 9,920 women wearing Copper T 380A to grasp the effectivness as well as the expulsion rate due to a various side effects. The following features were the results of the study: 1. During the twelve months period, from January 1993 to December 1993, the largest number of IUD insertion per one physician is 740 cases and the smallest number 60 cases, the mean 195 cases. 2. After insertion of Copper T 380A, a total of 117 cases were removed the IUD;81 cases(0.8%) were due to extensive bleeding and 36 cases(O.36%) were due to pain. The pregnancy failure rate represented comparatively lower rate, 0.08% (8 cases) and the expulsion rate was 0.06%(6 cases). 3. The removal rate of Copper T 380A was different by physicians. The highest removal rate for bleeding was 3.13% and 13 physicians(25.5%) did not experience any bleeding cases. 4. For the removal rate for pain, 30 physicians(58.8%) did not experience any removal of IUD due to pain while 21 physicians(41.2%) removed IUD due to pain: 17 physiciansC33.4 %) had 0.1-1.5% of removal rate and 4 physiciansC7.0%) had 1.6-2.1% of removal rate. 5. A total of 43 physicians(84.3 %) did not experience any pregnancy failure case while 8 (15.7%) physicians was experienced pregnancy failure rate. Four physicians(7.8%) had 0.1-0.5% of pregnancy failure rate and 4 physicians had 0.6-1.0% of pregnancy failure rate. 6. Expulsion rate showed comparatively lower, 0.06% in this study. A total of 45 physicians (88.2%) did not experience any expulsion cases while 6 physicians(11.7%) had expulsion rate with the highest expulsion rate of 2.0%.
