Chemopreventive Effects of Garlic Extracts on Rat Colonic Aberrant Crypt Foci Induced by 1,2-Dimethylhydrazine (1,2-Dimethylhydrazine에 의해 유발된 Colonic Aberrant Crypt Foci에 대한 마늘추출물의 암예방효과)

  • Garlic (Allium sativum L.) with the food supplement material and medicine was used traditionally in Asia and Europe. Epidemiological studies revealed that the intake of garlic reduced incidences of various cancer including digestive system. The present study was designed to investigate the effect of garlic ethanol extract on the development of colonic aberrant crypt foci (ACF) induced by 1,2-dimethylhydrazine (DMH) in male F344 rats. Five-week-old rats were given four times for two weeks to subcutaneous injections by DMH (30 mg/kg body weight) to induce ACF. The animals were divided into groups that fed diet containing garlic ethanol extract at five different doses (0.1, 0.2, 0.5, 2, 5%), respectively, animals were evaluated for the total number of ACF and total aberrant crypts (AC) per colon detected from methylene blue-stained rat colon. ACF were formed in animals in DMH-treated group. The feeding suppressed potently the appearance ACF in the colon of rats. Especially, fed diet containing garlic ethanol extract at intermediate dose (0.5%) significantly reduced the number of ACF and AC per colon (p < 0.05). Garlic ethanol extract inhibited DMH-induced overexpression of Activator Protein-1 (AP-1) and ${\beta}-catenin$ genes related to cell proliferation that also upregulated the expression of p21Waf1/Cip1 mRNA, a cell cycle-regulating gene. These results suggested that garlic ethanol extract may inhibit ACF formation, ${\beta}-catenin$ gene as the early preneoplastic marker of malignant potential in the process of colon carcinogenesis.

Changing Pattern of Birth Weight and Relationship of Birth Weight with Maternal Age and Parity (출생시 체중변화의 양상과 산모의 연령 및 출산순위와의 관계)

  • The objectives of this study were to examine the changes of birth weight and relationship of birth weight with maternal age and parity. The study population included 13,634 single live births out of 14,346 births delivered at a general hospital in Busan between January 1, 1977 and December 31, 1956. Data were obtained from the delivery record. In 1979 the mean birth weight of male was 3,074 gm and that of female was 2,985 gm. In 1986 birth weight of male was increased to 3,266 gm and that of female to 3,210 gm. Low birth weight ($\leq$2,500 gm) incidence rate was 7.2% in 1977 and it was increased gradually to 10.4% in 1980 but decreased thereafter to 6.5% in 1986. Incidence rate of overweight infant (4,001 gm $\leq$) was 3.9% for 10 years and it ranged from 2.9% to 4.6% but no particular changing pattern was observed. The percentage of mothers who had history of induced abortion was decreased from 51.6% in 1979 to 45.1% in 1986. Also, stillbirth rate was decreased from 2.6% in 1977 to 1.5% in 1986. The proportion of the first and second births was increased from 55.4% in 1977 to 96.0% in 1986 and the proportion of mothers of 25-34 years increased from 72.1% in 1977 to 84.7% in 1986. The incidence rates of low birth weight and over weight infant are lower in the first and second births of 25-34 years old mothers than other parities and age groups. Increased mean birth weight and decreased low birht weight incidence rate indicate that the health status of newborn infants has been improved and substantial portion of these changes can be attributed to increase in family planning practice rate and delayed marriage. There is no evidence, however, for increasing incidence rate of overweight infant.

Transfer Rate of Cross Contamination of Listeria monocytogenes between Pork Meat and Workers' Hands during Pork Meat Processing (포장돈육 가공공정 중 돈육과 작업자 손과의 Listeria monocytogenes의 교차오염 전이율)

  • This study was performed to determine the transfer rates of each foodborne pathogen from pork meat packaging during the processing. We analyzed the transfer rate of Listeria monocytogenes from contaminated pork meat to worker's hands (wearing polyethylene gloves, PEG; cotton gloves, CG; and bare hands), cutting boards and knives, and vice versa. Transfer rate of CG 100.00% was higher than that of bare hands 2.513% and PEG 1.511%. In particular, when wearing Co, the transfer rate from the CG to bare hands with CG was 0.08%. Also, the range of transfer rates from contaminated pork meat to cutting board and knife was 0.352-3.791%. In contrast, transfer rates from the workers' hands (with PEG/CG and bare hands) to cutting board, knife, and pork meat ranged from 0.001 to 0.141%. There was a lower transfer rate from workers' hands than from pork meat. These findings indicate that use of PEG could effectively reduce or prevent the cross-contamination compared to CG and provide important information concerning the consecutive transfer of L. monocytogenes during food processing.

  • In this study, we assessed changes occurring in the characteristics of Aralia continentalis Kitagawa (AcK) Kimchi prepared using different CaCl$_2$ treatments and seasonings by documenting the changes occurring in over the course of preparation and preservation. No visible changes were detected in the degree of total vitamin C although the levels of reducing sugar in the AcK-preserved products decreased over the storage period; the samples overall evidenced relatively high retention rates. Among the qualities of the AcK-preserved products, soluble dietary fiber generally increased over the storage period but the levels of insoluble dietary fiber decreased, and total dietary fiber increased to some degree or was maintained at a constant level. The total soluble solid content evidenced an increasing tendency over the storage period.

Regeneration Process in Gap of Quercus mongolica Forest (신갈나무림의 Gap 내 재생과정)

  • The pattern of gap regeneration and vegetational changes were carried out in gaps with different ages and in an intact forest in a Quercus mongolica (mongolian oak) stand located at Munsubong of Mt. Worak. In the early stage of gap formation, Lespedeza maximowiczii, Fraxinus rhynchophylla, Tripterygium regelii, Quercus mongolica, and Stephanadra incisa were dominants in shrub layers. The numbers of shrubs with smaller diameters at ground surface were abundant in the early stage of gap formation. On the other hand, as gap age increased, the number of individuals of Quercus mongolica and Fraxinus rhynchophylla forming tall tree layer decreased. However the diameter at ground surface of Quercus mongolica and Fraxinus rhynchophylla increased. There were few young Quercus mongolica, but those over 130 years old appeared in mature Quercus mongolica stands. The last regeneration episode ended about 130 years ago and the new one started 40 years ago in this study site. Considering the presence of Quercus mongolica below 40 years old in gap and the absence of Quercus mongolica from 40 to 130 years old in closed stand, it is concluded that Quercus mongilica forest is maintained by discontinued regeneration.

Effect of Seed Position on Seed Size, Contents of Ginsenosides, Free Sugars and Fatty Acids in Panax ginseng (종자의 착생위치가 인삼종자의 크기, 사포닌, 유리당 및 지방산함량에 미치는 영향)

  • This study was carried out to know the effect of seed position on the size, contents of ginsenosides, free sugars, and fatty acids in ginseng seeds. Seed positions were classified by the three portions as center, middle and border in a seed cluster. Seed weight at center was light remarkably in comparison with those of seeds of at border and middle. The weight of embryo plus endosperm was in same tendency as seed weight. Percentage of single-seeded berry was smaller than that of the double-seeded, and the triple-seeded was rare. The percentage of the single-seeded increased from the border to the center. Size of the single -seeded seed was smaller than that of the double- seeded. Rate of dehiscence did not differ among different seed positions. The major ginsenosides in seed were Re, Rb$_1$, and Rb$_2$. The contents of Rb$_2$ and total saponin were highest in border, least in center, but reversed in Re and Rd. Major free sugars in seed were sucrose and glucose. The sucrose content was gradually decreased according to the seed position from border to center. Major fatty acids in the seed were oleic and linoleic acid. Contents of palmitic and linolenic acid were different according to the seed position.

Inhibitory Effect of YBR on Hepatic Fibrogenesis (YBR의 간섬유화(肝纖維化)억제 효과(效果)에 관한 연구(硏究))

  • Objective : This study was performed to investigate the anti-fibrogenic effect and changes of inflammation-related genes by YBR I and YBR II (YBR I: Arteisiae Capillaris Herba, Atractylodis Rhizoma Alba, Hoelen/ YBR II: YBR I +Sanguisorbae Radix, Biotae Cacumen, Cirsii Japonici Herba) on HSC(hepatic stellate cells)-T6 and TAA-induced rat liver tissue. Materials and Methods : HSC-T6 were treated with various concentrations of distilled-water extract YBR I and YBR II extract for 24, 48 and 72 hours. After the treatment, cell viability, proliferation, procollagen levels and IL-6 levels were measured by using MTT Assay, BrdU Assay, Procollagen Type 1 C-peptide EIA kit, and Murine IL-6 ELISA Development kit. Rat liver fibrosis was induced by intraperitoneal TAA injection of 150mg/kg 3 times a week for 6 weeks. After the treatment, body weight, liver & spleen weights, liver function test, complete blood cell count and change of portal pressure were studied. In addition, gene expressions of ASMA, IL-6, MMP-2, TIMP-1 and TIMP-2, all of which are known to be associated with liver fibrosis, were analyzed by using Real-Time PCR. After YBR I and YBR IItreatment, percentages of collagen in TAA-induced rat liver tissue were measured. Results : The viability and proliferation of the HSC-T6 decreased as the concentration increased. The production of procollagen decreased as the concentration increased. The production of IL-6 was little influenced by YBR I and YBR II. There was no difference in rat body weight between the TAA-only group and the YBR groups. Compared with rat liver weight of TAA-only group, that of the YBR groups increased. In the YBR I group, the serum level of AST elevated by TAA injection significantly decreased and in the YBR I and II group, the serum level of ALP and ALT elevated by TAA injection decreased. In the YBR I group, white blood cell count elevated by TAA injection decreased but platelets increased. In the YBR I group, the portal pressure elevated by TAA injection significantly decreased. Decreases in the gene expression of ASMA and MMP-2 were observed in the YBR I group. The gene expression of IL-6 was little influenced by YBR I and YBR II -treated groups. In the histological finding, TAA injections caused severe fibrosis, but YBR I and YBR II treatment significantly reduced the amounts of hepatic collagens. Conclusions : These results suggest that YBR I and II have inhibitory effects on the hepatic fibrogenesis.

Anatomical studies on pattern of branches of bile and hepatic ducts in Korean native cattle (한우의 담관 및 간관의 분지에 관한 해부학적 연구)

  • The distribution of the bile and hepatic ducts within the liver of 24 Korean native cattle were observed. Vinylite solution was injected into common bile ducts on 13 cases for cast preparation. The angiography was prepared in 11 cases which were injected 30% barium sulfate solution into the common bile ducts and then radiographed on X-ray apparatus. The results were summarized as follows; 1. The dorsal, intermediate and ventral ducts of left hepatic lobes, ducts of quadrate lobes, and ducts of papillary process were collected into left hepatic ducts. The dorsal and ventral ducts of right hepatic lobe, ducts of caudate process and papillary process were collected into right hepatic ducts. 2. The bile ducts in the left hepatic lobes were collected into dorsal, ventral and intermediate ducts of left hepatic lobes. The intermediate ducts of left hepatic lobes was absent in some cases(5 cases, 20.83%). 3. In some cases, the ventral ducts of right hepatic lobes were collected into right hepatic ducts directly. 4. The ducts of quadate lobes were collected into left hepatic ducts(8 cases, 33.33%), ventral ducts of left hepatic lobes(13 cases, 54.17%), and dorsal duct of left hepatic lobes, intermediate and ventral ducts of left hepatic lobe, common trunk collected into intermediate and ventral duct of left hepatic lobe and left hepatic duct(1 cases, 4.17%) respectively. 5. The ducts of papillary process were collected into left and right hepatic ducts(12 cases, 50%), left hepatic ducts only(11 cases, 45.83%) or right hepatic duct only(1 cases, 4.17%). 6. The average length of right hepatic ducts was 26.87(6-45.8)mm. The average length of left hepatic ducts was 56.68(6-127)mm. The length of gallbladder was 128.60(104-160)mm on the average. The breadth of gallbladder was 67.16(38.4-153.4)mm on the average. But there was a difference in the length and the breadth according to the quantity of bile. 7. The ramified angles of dorsal, intermediate and ventral ducts of left hepatic lobes, ducts of quadrate lobes, ducts of papillary processes and caudate processes, dorsal and ventral ducts of right hepatic lobes and cystic ducts were 130-170, 160-200, 190-250, 240-300, 90-140, 320-350, 30-100, 270-330 and 240-300 degrees respectively.

Investigation of the Characteristic Velocity of Geomorphologic Instantaneous Unit Hydrograph (지형형태학적 순간단위도의 특성속도에 대한 고찰)

  • The GIUH (Geomorphologic Instantaneous Unit Hydrograph) is to be applied to the ungauged or insufficiently gauged basins. For tIris purpose, an accurate estimation of the charactenstlc velocity is one very important part, but any proper method for this has not been developed yet. In case that we have enough rainfall and runoff clata, the estimation of the characteristic velocity may be an easy job, but it is out of the purpose of the GIUH. Remindmg that the purpose of GIUH the characterisbc veloclty should be estimated based on the geomorpholog1c analysis and also be snnple for easy apphcation. In tIris research analysis cmd application of the GruH was given to several sub-basins in Wi-stream river basin, Gono, Donggok and Hyoryung. After deriving the characteristic velocity througn a optimizatlOn process with real data, it is compared w1th several velOCIties der1ved from geOlnoI1Jhoclimatic instantaneous unit hydrograph theory and several other concentration time formulae. The estimated charactenstic velocities using Kerby, Kim, KInematic Wave, and Brasby- Williams formulae found to g1ve the appropriate results. Hmvever, as the Kerby, and the Kinematic Wave require user's decision of the IvIanning's n value, the K1m and the Braby-Williams seem to be more applicable and recommended as characteristic velocity formula.

Physico-Chemical and Sensory Properties of Emulsified Sausages containing Mulberry and Persimmon Leaf Powder (뽕잎과 감잎분말을 첨가한 유화형 소시지의 이화학적 및 관능적 특성)

  • The purpose of this study was to investigate the effects of mulberry and persimmon leaf powder with various contents(0.04%, 0.08%) on physico-chemical and sensory properties of emulsified-sausages. There were no differences(P>0.05) in moisture and crude ash contents(%), but crude fat contents(%) of the sausages containing 0.8% persimmon leaf powder showed the highest, compared to other treatments. pH values of all treatments decreased during storage period. Sausages containing mulberry and persimmon leaf powder had more bright and yellow, but less red color than the control. Hardness values of the sausages containing 0.04% mulberry and persimmon leaf powder were higher than those of a control and other treatments. The addition of mulberry leaf powder had better flavor and overall palatability of sausages than persimmon leaf powder. However, no differences in color, flavor, tenderness and overall palatability were observed on 60 days of storage. These results indicated that the addition of mulberry and persimmon leaf powder for the manufactured of emulsified-sausages partially affected physico-chemical and sensory properties. Further research will be performed why the addition of these powders affect the product quality.
