Optimization for the Bacterial Cellulose Production of Acetobacter xylinum KJ1 by Factorial Design (Factorial design에 의한 Acetobacter xylinum KJ1의 Bacterial cellulose 생산조건의 최적화)

  • Acetobacter xylinum KJ1 efficiently producing bacterial cellulose(BC) In shaking culture was isolated from a rotten grape. The strain was used to investigate optimum operating conditions for increasing BC production and factorial design model was employed for the optimization. The results of experiments were statistically analyzed by SAS program. Reciprocal effects of each factors(carbon source concentration, shaking speeds(rpm), oxygen pressure, and CSL concentration) and culture condition of BC production were examined by getting regression equation of the dependent variable. Comparisons between experimental results and predicted results about BC concentration were done in total 24 experiments by combination of each factors using SAS program, and the correlation coefficients of BC concentration and BC yield were 0.91 and 0.81, respectively. The agitated cultures were peformed in various operation conditions of factors which affected considerably to BC production in jar fermentor. The results showed that BC concentration was 11.67 g/L in 80 hours cultivation under the condition of carbon source concentration : shaking speeds(rpm) : oxygen pressure : CSL concentration : 4% : 460 rpm : 0.28 : 6%. On the other hand BC yield was 0.42 g/g in 80 hours cultivation under the condition of carbon source concentration : shaking speeds(rpm) : oxygen pressure : CSL concentration : 4% : 564 rpm : 0.21 : 2%. The BC production could be enhanced up to more than 2.4 times by factorial design. The result of a verifying experiment under the optimal conditions determined by the factorial design to the BC production showed that the model was appropriate by obtaining BC concentration of 11.47 g/L in the optimum condition.

  • This study presents the Mid-Lateglacial vegetation history of southwestern Korea. From the result of AMS dating and pollen analysis, four local pollen assemblage zones(BS) were recognized. The four local pollen assemblage zones are BS 1. 12,222cal BC$\sim$9,160cal BC: cool-temperate northern/altimontane mixed coniferous & deciduous broad-leaved forest, BS 2. 9,160cal BC$\sim$ 4,210cal BC: cool-temperate central/montane deciduous broad-leaved forest, BS 3. 4,210cal BC$\sim$125cal BC: cool-Temperate southern/submontane mixed coniferous & deciduous broad-leaved forest. BS 4. BC125cal ${\sim}$present: warm-temperate/cool-temperate southern/submontane coniferous forest. Pine forest expanded since 4,210cal BC and full-scale rice cultivation might started since 2,120cal BC. A radical expansion of Pinus densiflora forest after postglacial stage might be caused by human impacts including full-scale agriculture.

Production and Structural Analysis of Cellulose by Acetobacter sp. V6 Using Static Culture (정치배양을 이용하여 Acetobacter sp. V6의 셀룰로오스 생산 최적화 및 구조 분석)

  • The optimal medium compositions for the production of bacterial cellulose (BC) by a Acetobacter sp. V6, which was isolated from the traditionally fermented vinegar in Korea, were investigated in static cultures. The optimum medium compositions for BC production were 3% glucose, 3% soytone, 0.8% $K_2HPO_4$, and 0.4% ethanol, respectively. Adding $NaH_2PO_4$ or $KH_2PO_4$ had not shown the increase in BC production. Under the optimum medium compositions, the highest BC production was 44.67 g/$m^2$ in 8 days and the thickness of BC pellicle was about 1 cm. Structural properties of BC produced in the optimal medium were studied using Fourier-transform infrared spectroscopy and X-ray diffractometer. BC from the optimal medium was found to be of cellulose type I, the same as typical native cellulose. No difference in the compositions between bacterial and plant celluloses, but BC showed unique micro-network structure and high crystallinity (82%).

Implementation of High Stable Phase-Locked Oscillator for X-Band Satellite Communication (X-Band 위성통신을 위한 고안정 위상 동기 발진기 구현)

  • In this paper, X-band satellite communication oscillator of double phase locked is implemented by constructing a couple of phased-locked loop, and then we have analyzed the phase noise of designed PLL-DRO. The designed phase-locked oscillator is consist of series feedback DRO, frequency divider, phase detector, loop filter and programmable PLL-IC. By dividing oscillation frequency of 12.6 GHz into two frequencies, it exhibits output power of 15.32 dBm at 6.3 GHz. Phase noises of implemented oscillator are -81 dBc/Hz@100Hz, -100.86 dBc/Hz@1 kHz, -111.12 dBc/Hz@10 kHz, -116 dBc/Hz@100 kHz and -140.49 dBc/Hz@1 MHz respectively. These indicate excellent stable operation of oscillator and very good phase noise characteristics.

Compensation of Aethalometer Black Carbon Data Observed at a Gwangju Site (광주 도심지역에서 측정한 Aethalometer 검댕입자 자료의 보정)

  • $PM_{2.5}$ black carbon (BC) concentrations were measured to investigate the filter spot loading effect in raw BC data at 5-minute time-based resolution using a single-wavelength aethalometer at a Gwangju site. Also the elemental carbon (EC) concentrations from 24-hr integrated filter-based measurements of $PM_{2.5}$ particles were determined to compare with the loading compensated BC values. Close examination of the time-series BC data showed clearly the "gaps" when the filter tape advances, suggesting the correction of raw BC data. Therefore, we calculated the average BC concentration in each range of attenuation (ATN) to decide if there was (or was not) an effect on the aethalometer data according to the loading of the filter spot. A consistent decrease of average BC concentration was found with increasing ATN values for every month, suggesting there was a consistent "spot loading effect" in the raw BC data. The loading compensated BC concentration according to a simple compensation model with loading effect was 1.01~1.15 times greater than the raw BC data. The 24-hr average concentration of EC observed during summer sampling period was about 3% higher than the original 24-hr average BC value and 2% lower than the loading compensated BC concentration.

Analysis of Nutritional Composition and Antioxidant Activity of Black Chestnut by Aging (숙성에 의한 흑밤의 영양성분 분석과 항산화 활성)

  • Black chestnut (BC) was obtained through aging of fresh chestnut (FC) at 80℃ for 15 days. Proximate and mineral compositions along with colors of FC and BC were evaluated. With aging, moisture contents decreased by 50%, whereas sugar contents, carbohydrate contents, and calories increased. Contents of minerals (Fe, P, Ca, Na, Mg, K) were significantly higher in FC than in BC, showing an order of Mg < Ca < P < K in both FC and BC. Using a Hunter color system, it was found that lightness (L), redness (a), and yellowness (b) values of FC were higher than those of BC. Antioxidant and cytotoxic activities of hot water and ethanol (50, 80, 100%) extracts prepared from FC and BC were evaluated. Extraction yields were lower with FC than with BC. Among water and ethanol extracts, water extract showed the highest DPPH radical scavenging activity for both FC and BC. IC50 values for ABTS+ radical scavenging activities increased after aging. Cytotoxicities of FC and BC extracts were similar to each other. They were different against various cell lines (3T3, HeLa, and Sarcoma-180). These results suggest that BC could be used as a new processed food using chestnut.

Design of Ku-Band Phase Locked Harmonic Oscillator (Ku-Band용 위상 고정 고조파 발진기 설계)

  • In this paper, the phase locked harmonic oscillator(PLHO) using the analog PLL(Phase Locked Loop) is designed and implemented for a wireless LAN system. The harmonic oscillator is consisted of a ring resonator, a varactor diode and a PLL circuit. Because the fundamental fiequency of 8.5 GHz is used as the feedback signal for the PLL and the 2nd harmonic of 17.0 GHz is used as the output, a analog frequency divider for the phase comparison in the PLL system can be omitted. For the simple PLL circuit, the SPD(Sampling Phase Detector) as a phase comparator is used. The output power of the phase locked harmonic oscillator is 2.23 dBm at 17 GHz. The fundamental and 3rd harmonic suppressions are -31.5 dBc and -29.0 dBc, respectively. The measured phase noise characteristics are -87.6 dBc/Hz and -95.4 dBc/Hz at the of offset frequency of 1 kHz and 10 kHz from the carrier, respectively.

The Phase Noise prediction and the third PLL systems on 1/f Noise Modeling of Frequency Synthesizer (주파수합성기의 Phase Noise 예측 및 3차 PLL 시스템에서의 1/f Noise Modeling)

  • In this paper, we designed 2303.15MHz frequency synthesizer for the purpose of the phase noise prediction. For the modeling of phase noise generated in the designed system through introducing the noise-modeling method suggested by Lascari we analyzed a variation of phase noise as according as that of offset frequency. Especially, for the third-order system of the PLL among some kinds of phase noise generated from VCO we analyzed the aspect of 1/f-noise appearing troubles in the low frequency band. Since it is difficult to analyze mathematically 1/f-noise in the third-order system of the PLL, introducing the concept of pseudo-damping factor has made an ease of the access of the 1/f-noise variance. we showed a numerical formula of 1/f-noise variance in the third-order system of the PLL which is compared with that of 1/f-noise variance in the second-order system of the PLL. As a result, In case of txco we found the reduce rapidly along the offset frequency after passed through that phase-noise was -160dBc/Hz before passed through a loop at 10kHz offset frequency and -162.6705dBc/kHz after passed through the loop, -180dBc/Hz at 100kHz offset frequency and -560dBc/kHz after passed through the loop. We can notice that the variance of third-order system more occurs (or the variance of second-order system in connection with noise bandwidth and variance factor of second-order and third-order system.

Inheritance Study of Male Sterile Transformants Containing Pollen-specific Promoter and Diphtheria Toxin A Gene (수술특이프로모터와 디프테리아 독성 유전자에 의한 웅성불임 형질전환체의 후대 유전분석)

  • The objectives of this study were to investigate the genetic and phenotypic features of male sterile transformants by pollen-specific expression of diphtheria toxin gene and to find out inheritance patterns of transgene to the next generation. When backcrossed (BC) progenies were tested for expression of kanamycin resistance ($Km^R$), 9 lines out of 13 lines, except 4 lines ($BC_{1}5-13,\;BC_{1}5-23,\;BC_{1}5-28,\;BC_{1}5-32$), showed the ratio of $Km^R$ to kanamycin sensitive ($Km^S$), from 1:30 to all $Km^S$. As a result, they were much lower than Mendelian segregation of a dominant gene. To determine whether male sterility is a heritable and stable trait, 5 male sterile plants ($BC_{1}5-13,\;BC_{1}5-14,\;BC_{1}5-23,\;BC_{1}5-32,\;BC_{1}5-33$ lines) which had different transgene copy numbers were backcrossed as female parents with pollens from wild type. To confirm the existence of the DTx-A gene in the genome of the progenies, PCR was conducted using specific primers of the DTx-A coding region. A PCR band of 428 bp was obtained from each generation, which is the predicted size of the DTx-A gene fragment. Trangenes were inherited to the next $BC_4T_0$ progenies and showed male sterility, however, based on the copy numbers of DTx-A gene male sterile plants did not show predicted ratio. When male sterile plants were backcrossed with fertile plants, fruit capsule sizes and seed settings were relatively reduced from those of selfing wild type plants. The fruit sizes and seed settings were reduced in proportion to the increase in the copy number of DTx-A gene.

TREATMENT OF ECHOLALIA IN CHILDREN WITH AUTISM (자폐아동의 반향어 치료)

  • The purpose of this study was to investigate the possibility of providing familiar tasks as a treatment option to decrease echolalia. Two comparisons were made:One was to compare ‘conversation condition’ and ‘task performance condition.’ and the other was to compare ‘task performance alone condition’ and ‘task performance along with contingency of reinforcement condition.’ Two echolalic children aged 12 and 13 years participated in the experiment and A-B-A-B-BC-B-BC design was used, in which A was conversation only, B was task performance, and C was task performance along with contingency of reinforcement. In the A condition, the therapist asked easy and short questions to the child;in the B condition the child was given familiar tasks with short instruction, and in BC condition, each child was reinforced for his performance on given tasks, in which immediate echolalia was controlled through his hands being held down for 5 seconds. Delayed echolalia was recorded without any intervention being given. Each child was put into each of the 7 treatment conditions. With a 15 minutes session, each child went through 5 to 6 sessions per day for 2 weeks. The mean echolalia(immediate) rates across the 7 treatment conditions were:For child 1, A(99%)-B(65%)-A(95%)-B(10%)-BC(7%)-B(6%)- BC(7%) and for child 2, A(67%)-B(62%)-A(63%)-B(35%)-BC(8%)-B(4%)-BC(0%). As to the generalization of the treatment effect of immediate echolalia to the untreated delayed echolalia, there was shown a drastic reduction of delayed echolalia in child 2:A(35%)-B(57%)-A(56%)-B(40%)-BC(8%)-B(5%)-BC(9%). Child l’s delayed echolalia was negligible(mean=3%) pre-and post treatments. In conclusion, the results of this study clearly show that providing a task performance setting with familiar tasks can certainly be helpful for minimizing echolalic response, and along with the use of the contingency of reinforcement technique it can further not only correct echolalic behavior to a negligible degree but also help the echolalic child generalize its treatment effect to the child’ overall language improvement.
