Effect of Planting Density on the Growth and Yield in Staking Cultivation of Bitter Gourd (Momordica charantia L.) under Non-heated Greenhouse (여주 무가온 하우스내 입체재배시 재식밀도가 생육 및 수량에 미치는 영향)

  • This experiment was conducted to determined the optimum planting density for the production of high quality bitter gourd (Momordica charantia L.) adapted in spring cultivation with the unheated greenhouse condition. 'Erave' variety was planted at three different planting densities (235, 305, $380plants{\cdot}10a^{-1}$) on March 26. The training method was six lateral vines with pinching the main one. The light intensity was lower in the higher planting density than the lower one. Net photosynthetic rates of the bitter gourd leaves in the higher density were significantly lower (41 to 71%) than the lower one. There was no difference in the fruit characteristics among treatments. But the root weight was heavier in the lower planting density ($235plants{\cdot}10a^{-1}$) as 113.1g than 96.0g of the higher planting density ($380plants{\cdot}10a^{-1}$). The number of the harvested fruit also higher in the lower planting density ($235plants{\cdot}10a^{-1}$) with 60.7 than 39.9 of the higher planting density ($380plants{\cdot}10a^{-1}$). The average fruit weight was the highest in the plot of $305plants{\cdot}10a^{-1}$ as 338.7g and lowest in the lower planting density ($235plants{\cdot}10a^{-1}$) as 285.2g. The total yield of $305plants{\cdot}10a^{-1}$ density was $5,359kg{\cdot}10a^{-1}$, which was higher than $4,068kg{\cdot}10a^{-1}$ of the lower planting density ($235plants{\cdot}10a^{-1}$). Marketable yield was increased by 24% in the planting density of $305plants{\cdot}10a^{-1}$($4,767kg{\cdot}10a^{-1}$) as compared to the lower density in $235plants{\cdot}10a^{-1}$($3,629kg{\cdot}10a^{-1}$) and increased by 13% in the planting density as $380plants{\cdot}10a^{-1}$($4,137kg{\cdot}10a^{-1}$). Therefore, the planting density of bitter gourd was desirable in $305plants{\cdot}10a^{-1}$ density for the higher yield and quality in the protected cultivation.

The Relationship between the Polymorphism of CYP1A1, GSTM1, GSTT1, GSTP1 and p53 Overexpression in Head and Neck Squamous Cell Carcinoma (두경부 편평세포암종에서 CYP1A1, GSTM1, GSTT1, GSTP1 유전자 다형성 및 p53 과발현)

  • Background and Objectives: Individual genetic susceptibilities to chemical carcinogens have been recognized as a major important host factors in human cancers. The cytochrome P450 family (CYPs) and glutathione S-transferase(GST) have been reported to be associated with risks to the smoking-related human cancers. Inactivation of tumor suppressor genes like p53 playa key role in tumor progression. The purpose of this study is to demonstrate an association between p53 overexpression and the prevalence of the genetic polymorphisms of CYP1A1 and GSTs in Korean head and neck squamous cell carcinoma (HNSCC). Materials and Methods: The polymorphisms of CYPIA1 and GSTs were analyzed by PCR and PCR-RFLP in 98 Korean head and neck squamous cell carcinoma patients. The expression of p53 was analyzed by immunohistochemistry with anti-p53 Ab (DO7). Results: Overexpression of p53 detected in 45.9% of HNSCC. The odds ratio for p53 overexpression in GSTM1(-), GSTT1(-), GSTP1(val/val) and CYP1A1(val/val) were 1.53, 1.83, 1.17 and 1.47, respectively. Among the combined genotypes, the odds ratio of the CYP1A1 val/val, GSTM1 (-), CYP1A1 val/val, GSTT1(-), and CYP1A1 val/val, GSTT1(-) were 2.0, 2.34 and 4.68, respectively. Conclusion: Based on our results, it might be suggested that p53 overexpression is slightly increased in GSTM1(-), GSTT1(-), GSTP1 val/val, CYP1A1 val/val genotypes. The further study is needed to evaluate the relationship and mechanism between the p53 overexpression and the specific CYP1A1 and GSTs genotypes.

Difference in Lung Functions according to Genetic Polymorphism of Tobacco Substance Metabolizing Enzymes of Korean Smokers (한국인 흡연자들의 담배 물질 대사 효소의 유전자 다형성에 따른 폐기능 차이)

  • This study aimed to determine whether there was a difference in lung functions of smokers according to the presence of carcinogenic genetic-metabolizing enzymes by comparing the results of lung functions and the presence of genetic metabolizing enzymes that metabolize tobacco substances. To achieve this, 31 smokers without no illness and no psychiatric history were selected (28 males and 3 females); they were aged 20 to 27 years and were physically and mentally healthy students attending K University. Their lung functions were measured, and gene polymorphisms of cytochrome P-450 1A1 (CYP1A1) related to metabolic activation of tobacco components and gene polymorphism of tumor protein 53 (TP53) related to lung cancer were analyzed. As a result, the mean values of lung function of TT and Arg / Arg without genetic mutations were the highest, and ANOVA analysis of CYP1A1 and lung functions showed that the P-value of FVC was 0.049, which was different between groups. In other words, there is no high mutation in Cytochrome P-450 1A1 (CYP1A1) gene, which is associated with the metabolic activation of tobacco components. In other words, In the absence of the mutant Cytochrome P-450 1A1 (CYP1A1) gene, which is associated with the metabolic activation of tobacco components, the value of FVC was high.

Vaccinium uliginosum L. Improves Amyloid β Protein-Induced Learning and Memory Impairment in Alzheimer's Disease in Mice

  • The present study investigated the effects of Vaccinium uliginosum L. (bilberry) on the learning and memory impairments induced by amyloid-${\beta}$ protein ($A{\beta}P$) 1-42. ICR Swiss mice were divided into 4 groups: the control ($A{\beta}40$-1A), control with 5% bilberry group ($A{\beta}40$-1B), amyloid ${\beta}$ protein 1-42 treated group ($A{\beta}1$-42A), and $A{\beta}1$-42 with 5% bilberry group ($A{\beta}1$-42B). The control was treated with amyloid ${\beta}$-protein 40-1 for placebo effect, and Alzheimer's disease (AD) group was treated with amyloid ${\beta}$-protein 1-42. Amyloid ${\beta}$-protein 1-42 was intracerebroventricular (ICV) micro injected into the hippocampus in 35% acetonitrile and 0.1% trifluoroacetic acid. Although bilberry added groups tended to decrease the finding time of hidden platform, no statistical significance was found. On the other hand, escape latencies of $A{\beta}P$ injected mice were extended compared to that of $A{\beta}40$-1. In the Probe test, bilberry added $A{\beta}1$-42B group showed a significant (P<0.05) increase of probe crossing frequency compared to $A{\beta}1$-42A. Administration of amyloid protein ($A{\beta}1$-42) decreased working memory compared to $A{\beta}40$-1 control group. In passive avoidance test, bilberry significantly (P<0.05) increased the time of staying in the lighted area compared to AD control. The results suggest that bilberry may help to improve memory and learning capability in chemically induced Alzheimer's disease in experimental animal models.

Ca2+ Regulators affect the Gravitropism and Ethylene Production Induced by Malformin A1 in Maize Root (옥수수 뿌리에서 칼슘 이온 조절제가 malformin A1에 의해 유도된 굴중성과 에틸렌 생합성에 미치는 영향)

  • Treatment of malformin A1 is known to increase ethylene production 130% at 4 hr and 56% at 8 hr after treatment in maize root compared to untreated plants. The ethylene production by malformin A1 was maximum level at 4 hr and slowly decreased up to 8 hr. Calcium ion regulators such as A23187 (calcium ionophore) and verapamil (calcium channel blocker) stimulated ethylene production. Treatment of both calcium ion regulators increased about 30% of ethylene production at 4 hr, and 20% at 8 hr. Both calcium ion regulators did not stimulate malformin A1-induced ethylene production at 4 hr as malformin A1 itself did. However, the treatment of calcium ion regulators with malformin A1 maintains the ethylene production for 8 hr. These results suggested that the proper concentration of calcium might need to confer the effect of malformin A1 on the ethylene production. Malformin A1 suppressed the gravitropic curvature of maize root about 58% at 4 hr and 42% at 8 hr compared to control plant. Verapamil inhibited the gravitropic curvature about 54% at 4 hr and 23% at 8 hr compared to control, respectively. But A23187 could not. In addition, verapamil showed more inhibition in malformin A1-induced gravitropic curvature than A23187 in malformin A1 induced. These data suggested that calcium ion regulators affect the malformin A1-induced ethylene production and gravitropic curvature, and give the evidence that calcium ion play an important role in gravitropic curvature in maize root.

The 30 isolates of Xanthomonas oryzae collected from various places in Korea were classified into 2 strains in A' and B, by the 4 kinds of Japanese bacteriophages as follow; A' strain; Susceptible to $OP_1$ and $OP_2$ but not to $OP_1h$ and $OP_1h_2$. B strain; Susceptible to $OP_1h$, $OP_1h_2$ and $OP_2$ but not to $OP_1$. Of these bacterial strains, A' strain, a little different to A strain in Japan, was susceptible to $OP_1h_2$, while A' strain was resistant to $OP_1h_2$, 2. A' strain was widely and commonly distributed throughout this country, In geographical distribution, it was generally known that A' strain was distributed in south of Kyunggi-Do, whereas B strain was distributed north of Suwon area. 3. No specific tendency was found in the distribution of these strains in same variety.

mTOR Signal Transduction Pathways Contribute to TN-C FNIII A1 Overexpression by Mechanical Stress in Osteosarcoma Cells

  • Osteosarcoma is the most common primary malignant bone tumor with a very poor prognosis. Treating osteosarcoma remains a challenge due to its high transitivity. Tenascin-C, with large molecular weight variants including different combinations of its alternative spliced FNIII repeats, is specifically over expressed in tumor tissues. This study examined the expression of Tenascin-C FNIIIA1 in osteosarcoma tissues, and estimated the effect of mechanical stimulation on A1 expression in MG-63 cells. Through immunohistochemical analysis, we found that the A1 protein was expressed at a higher level in osteosarcoma tissues than in adjacent normal tissues. By cell migration assay, we observed that there was a significant correlation between A1 expression and MG-63 cell migration. The relation is that Tenascin-C FNIIIA1 can promote MG-63 cell migration. According to our further study into the effect of mechanical stimulation on A1 expression in MG-63 cells, the mRNA and protein levels of A1 were significantly up-regulated under mechanical stress with the mTOR molecule proving indispensable. Meanwhile, 4E-BP1 and S6K1 (downstream molecule of mTOR) are necessary for A1 normal expression in MG-63 cells whether or not mechanical stress has been encountered. We found that Tenascin-C FNIIIA1 is over-expressed in osteosar-coma tissues and can promote MG-63 cell migration. Furthermore, mechanical stress can facilitate MG-63 cell migration though facilitating A1 overexpression with the necessary molecules (mTOR, 4E-BP1 and S6K1). In con-clusion, high expression of A1 may promote the meta-stasis of osteosarcoma by facilitating MG-63 cell migration. Tenascin-C FNIIIA1 could be used as an indicator in metastatic osteosarcoma patients.

Chitinase 3-like-1, a novel regulator of Th1/CTL responses, as a therapeutic target for increasing anti-tumor immunity

  • Chitinase-Like Proteins (CLPs) are an evolutionarily conserved protein which lose their enzymatic activity for degrading chitin macromolecules. Chitinase-3-like-1 (Chi3l1) is a type of CLP that is highly expressed in epithelial cells, macrophages, etc., and is known to have correlations with type 2 inflammation and cancer. Although the increased level of Chi3l1 in the blood was reported in various disease patients, the function of Chi3l1 in adaptive immunity has been totally unknown. Recently, we found that Chi3l1 is expressed in T cells and has a negative regulatory role in T-cell activation and proliferation. A genetic ablation study of Chi3l1 in T cells showed hyperresponsiveness to TcR stimulation, which increased proliferation and Th1 differentiation. A significant increase of $IFN{\gamma}$ signaling in Chi3l1-deficient T cells synergistically increased Th1 and CTL functions against melanoma cells in vitro and in vivo. In addition, targeted knockdown by Chi3l1 siRNA complexed with the cell-penetrating peptide dNP2, which showed decreased pulmonary melanoma metastasis with increased infiltration of Th1 and CTL in the lung. This study first suggests that Chi3l1 is a novel regulator of Th1/CTL responses and could be a target for treating cancer to increase tumor immunity.

  • Reactions of $Ir(ClO)_4(CO)(PPh_3)_2$ with dicyano olefins, cis-NCCH = CH$CH_2$$CH_2$CN (cDC1B), trans-NCCH = CH$CH_2$$CH_2$CN (tDC1B), trans-NC$CH_2$CH = CH$CH_2$CN (tDC2B), and NC$CH_2$$CH_2$$CH_2$$CH_2$CN (DCB) produce binuclear dicationic iridium (I) complexes, $[(CO)(PPh_3)_2Ir-NC-A-CN-Ir(PPh_3)_2(CO)](ClO_4)_2$ (NC-A-CN = cDC1B (1a), tDC1B (1b), tDC2B (1c), DCB (1d)). Complexes 1a-1d react with hydrogen to give binuclear dicationic tetrahydrido iridium (Ⅲ ) complexes, $[(CO)(PPh_3)_2(H)_2Ir-NC-A-CN-Ir(H)_2(PPh_3)_2(CO)](ClO_4)_2$ (NC-A-CN = cDC1B (2a), tDC1B (2b), tDC2B (2c), DCB (2d)). Complexes 2a and 2b catalyze the hydrogenation of cDC1B and tDC1B, respectively to give DCB, while the complex 2c is catalytically active for the isomerization of tDC2B to give cDC1B and tDC1B and the hydrogenation of tDC2B to give DCB at $100^{\circ}C$.

A Study on the Rational Utilization of Resource Catched by Multiple Fisheries (복수의 어업에 의해 어획되는 수산자원의 관리를 위한 연구)

  • This study is attempted to serve the fundamental theory for ‘The reorganization of Korean coastal and adjacent water fishery.’ On the Korean coastal and adjacent water fishery where one species stock is catched by multiple fisheries, traditional analysis is not suitable, as analyzing through adjusting the heterogeneous fishing effort among the fisheries to an unit having same fishing strength. Therefore, this study presents the ‘Multi-Variable Model’, adopting fishing effort from each fishery as independent variable, respectively, in order to analyze the quantitative fluctuation of fishery resource not with fishing strength but with amount of fishing effort, measured by the unit of each fishery. For the sake of simplication, this study assumes that one species is catched by two fishery, premise two assumption. 1) Every fishery has not the selectivity in fishing 2) The promotion of fishing efficiency is accomplished in the same speed. Resource equilibrium equation of each fishery is; $$CPUE_1=\frac{Y_1}{E_1}=a_1+ b_1\cdot E_1+c_1\cdot E_1$$ $$CPUE_1=\frac{Y_1}{E_1}=a_1+ b_1\cdot E_1+c_1\cdot E_1$$ Sustainable yield equation is; $$SY_1=a_1\cdot E_1+\cdot b_1E{_1}{^2}+c_1\cdot E_1\cdot E_1$$ $$SY_1=a_1\cdot E_1+b_1\cdot E_1\cdot E_1+c_1\cdot E{_1}{^2}$$ This study is rudimentary, hereafter, refinemental analysis will be supplemented.
