Bacillus subtilis A405 균주가 생성하는 내열성 항균 peptide의 특성 검정 (Characterization of a heat-resistant antimicrobial peptide secreted by Bacillus subtilis A405)

  • 전국에서 분리한 세균으로부터 항균력이 있는 길항세균들을 먼저 분리한 다음 그중 배양배지 속으로 강력한 항균력을 나타내는 peptide를 분비하는 A405균주를 선발, Biolog System 및 rDNA 염기서열을 이용하여 Bacillus subtilis로 동정하고 이 균주가 생성하는 항균 peptide의 항균력을 조사한 바 Botrytis cinerea, Cercospora sp., Fusarium oxysporum, Penicillium digitatum, Celletotrichum gloeosporioides, Rhizoctonia solani, Pythium ultimum, Pyricularia oryzae등의 식물병원균과 Escherichia coli, Pseudomonas spp. 등의 세균 및 효모인 Candida albicans의 성장도 억제하는 강력한 항균력을 보였다. 또한 이 균주가 생산하는 항균 peptide는 알콜, 아세톤과 같은 유기용매에 내성을 가지고 있었으며 proteinase K와 phenol 처리시에는 항균력이 감소되거나 소실되는 특성을 가지고 있었다. 이 항균 peptide를 SDS-PAGE, Native-PAGE 및 Tris-Tricine 전기영동으로 분자량이 3kDa인 작은 peptide임을 확인하였고 aspartic acid, glycine, serine, glutamine, valine, leucine, isoleucine, proline, tyrocine 등의 9가지 아미노산으로 구성되어 있음을 Amino Acid Analyzer로 확인하였으며 냉동건조한 항균 peptide 50 ug/ml을 체리토마토에 처리하여 과일의 선도유지를 조사한 결과 대조구에 비하여 월등한 선도유지 효과가 있음을 입증하였다.

Stemphylium lycopersici에 의한 고려엉겅퀴 점무늬병의 발생 (Occurrence of Leaf Spot Caused by Stemphylium lycopersici on Cirsium setidens in Korea)

  • 2015년 8월경 강원도 영월군의 고려엉겅퀴 재배 농가에서 잎에 소형 점무늬 증상이 발생하였다. 초기에는 고려엉겅퀴 잎에 회갈색 내지 갈색의 작은 점이 나타나며, 병이 진전되면 부정형의 진한 갈색의 병반으로 커지면서 크게 확대되고 결국 잎 전체가 진한 갈색 내지 흑색으로 변하였다. 병든 잎에서 Stemphylium균이 분리되었으며, 분생포자 크기와 분생포자경의 길이 등을 고려한 균학적 특성에 의해 Stemphylium lycopersici 혹은 Stemphylium solani, Stemphylium xanthosomatis와 유사한 것으로 나타났다. Ribosomal internal transcribed spacer (ITS) 영역, elongation factor 1 alpha (EF-$1{\alpha}$), glyceraldehydes-3-phosphate dehydrogenase (gpd), vacuolar membrane ATPase catalytic subunit A gene (vmaA)와 vacuolar biogenesis gene (vpsA) 사이의 noncoding 영역을 대상으로 한 다자위 염기서열 분석에 의해 분리균은 모두 S. lycopersici로 확인되었다. 고려엉겅퀴 잎을 대상으로 상처구와 무상처구로 나누어 분리균의 포자 현탁액을 접종하여 병원성을 확인한 결과, 접종 3일 후부터 상처의 유무에 관계없이 분리균을 접종한 부위에서 병반이 관찰되었다. 따라서 본 결과를 토대로 본 병을 S. lycopersici에 의한 고려엉겅퀴 점무늬병으로 명명하며, 고려엉겅퀴 점무늬병의 발생을 국내 최초로 보고한다.

In vitro control of plant pathogens by using anti-fungal hydroxy fatty acid obtained from microbial bioconversion of linolenic acid

  • Bioconverted linolenic acid (bLNA) obtained from linolenic acid by Pseudomonas aeruginosa PR3, showed anti-fungal activity against plant pathogens such as B. cinerea, F. oxysporum, F. solani, P. capsici and C. capsici. The oil sample also showed anti-fungal activity with MIC values, ranging from >250 to >1,000 ${\mu}g/ml$. Varied concentrations of bLNA had a great potential effect on spore germination of different fungi.

식물병원성 곰팡이에 대한 몇 가지 식물정유 및 주성분의 성장억제 효과 (Antifungal Activity of Some Essential Oils and Their Major Constituents on 3 Plant Pathogenic Fungi)

  • 11 plant essential oils are screened in vitro for their antifungal activities against Botrytis cinerea, Fusarium oxysporum and Rhizoctonia solani, which are causative agents of serious plant diseases. The radial growth of the test fungi were reduced in response to the oils. Among them, the essential oil from the bark of Cinnamomum zeylanicum inhibited 3 tested fungi growth, strongly, followed by those of oregano and thyme. The major constituents of the three essential oils, cinnaldehyde, carvacrol and thymol were tested for their effects on the fungi. From the results obtained, cinnamaldehyde, the major constituents of C. zeylanicum bark esential oil, has potential to be developed as a biopesticide for controlling phytopathogenic fungi causing serious damages on the important crops cultivated in Korea.

Screening for In Vitro Antifungal Activity of Soil Bacteria Against Plant Pathogens

  • Antifungal bacteria for biological control of plant diseases or production of novel antibiotics to plant pathogens were isolated in 1997 from various soils of Ansung, Chunan, Koyang, and Paju in Korea. Sixty-four bacterial strains pre-screened from approximately 1,400 strains were tested on V-8 juice agar against eight plant pathogenic fungi using in vitro bioassay technique for inhibition of mycelial growth. Test pathogens were Alternaria mali, Colletotrichum gloeosporioides, C. orbiculare, Fusarium oxysporum f. sp. cucumerinum, F. oxysporum f. sp. lycopersici, Magnaporthe grisea, Phytophthora capsici, and Rhizoctonia solani. A wide range of antifungal activity of bacterial strains was found against the pathogenic fungi, and strain RC-B77 showed the best antifungal activity. Correlation analysis between inhibition of each fungus and mean inhibition of all eight fungi by 64 bacterial strains revealed that C. gloeosporioides would be best appropriate for detecting bacterial strains producing antibiotics with potential as biocontrol agents for plant pathogens.

Enhancing Effect of Shimizuomyces paradoxus on Seed Germination and Seedling Growth of Canola, Plant Growth of Cucumber, and Harvest of Tomato

  • Shimizuomyces paradoxus showed no inhibitory effect against plant pathogen fungi, such as Fusarium oxysporum f. sp. lycopersici and Alternaria solani. The S. paradoxus culture filtrate showed higher seed germination and seedling growth rates in canola than distilled water and potato-dextrose broth. A conidial suspension of $1.0{\times}10^4/mL$ resulted in the highest growth stimulating effects on total plant length, and fresh and dry weight of shoots and roots in cucumber, when compared to the highest suspension concentration. Total plant length and shoot weight increased with the foliar spray treatment, and root length and root weight increased by simultaneous treatments of soil drenching and foliar spray in cucumber. Lower concentrations of the S. paradoxus conidial suspension increased the harvest of tomato fruit.

  • Twenty seven out of ca. 5,000 actinomycete strains, which were isolated from soil collected throughout the country, showed antibicrobial effects against fungai, Rhizopus stronifer (ATCC 6227a), Rhizoctonia solani (KCCM 11271) and yeast, Candida albicans (ATCC 10231). From these antifungal microorganisms, we further selected seven strains which seemed to produce insecticidal substances with in vivo test, using silkworm, Bombyx mori and beet armyworm, Spodoptera exigua. Morphological and biochemical experiments revealed that three strains out of seven were Streptomyces. Further investigations on the physical and chemical properties of these antifungal and insecticidal substances are now in progress.

점액세균 KR025의 분리 동정 및 생리활성물질의 탐색 (Isolation and Identification of Myxobacteria KR025 and Searching of Their Bioactive Compounds)

  • Fifty isolates of myxobacteria were isolated from soils from several areas in Korea during 1996-1997 and bioactivity against plant pathogenic fungi of these isolates was examined. A myxobacterial isolate KR025 showed good antifungal activities against Pyricularia oryzae, Cryphonectria parasitica, Colletotrichum lagenarium, and C. gloeosporioides but did not against Rhizoctonia solani, Fusarium oxysporum and Pythium ultimum. The bacterium was identified as Myxococcus fulvus based on morphological and physiological characteristics. Antifungal substances were extracted from culture broth and bacterial cell of Myxococcus fulvus KR025 by ethyl acetate. Antifungal substance of Myxothiazole (100 ${\mu}{\textrm}{m}$/ml) produced by Myxococcus fulvus KR 025 controlled 97.0% rice blast, tomato late blight, wheat leaf rust, and barley powdery mildew and showed 45.0 and 82.6% disease control of rice sheath blight and cucumber gray model, respectively.

  • A multifunctional antagonistic bacterium, producing both antifungal antibiotic and chitinase that could be used as biocontrol agents against fungal plant pathogens was isolated from the plant-disease suppressive soil. The isolate was identified as Promicromonospora sp. KH-28 from various morphological, biochemical and physiological tests. The antifungal antibiotic produced by Promicromonospora sp. KH-28 was soluble in n-butanol, methanol, toluene, n-hexane, ethanol but insoluble in H2O, acetone, chloroform, ethylacetate and ethylether. It inhibited the growth of fungal plant pathogens of Fusarium solani, F. oxysporum, Alternaria mali and Phytophthora capsici. The antagonistic Promicromonospora sp. KH-28 produced optimally the antifungal antibiotic when it was cultivated at pH 7, 3$0^{\circ}C$ for 5 days.

Composition of Methanol Extract from Hwangryeon (Coptidis rhizoma) and Antimicrobial Activity against Food Spoilage and Foodborne Disease Microorganisms

  • Hwangryeon (Coptidis rhizoma) was extracted by methanol and its antimicrobial activities against food spoilage and foodborne disease microorganisms were investigated by the paper-disc method. The microorganisms used in this experiment included 5 species of bacteria (Escherichia coli, Pseudomonas aeruginosa, Salmonella typhimurium, Klebsiella pneumoniae, and Staphylococcus aureus) and 3 species of fungi (Fusarium solani, Aspergillus flavus, and Penicillium citreonigrum). The Hwangryeon extract showed antimicrobial effect against P. aeruginosa, S. aureus, S. typhimurium, and K. pneumonia. The minimum inhibitory concentration on S. aureus, S. typhimurium, and K. pneumoniae was 300 mg/mL, but on P. aeruginosa it was 200 mg/mL. In the methanol extracts from Hwangryeon, 190 compounds were separated by GC/MS. The extraction yields of phenols, furans, alcohols, acids and esters, ketones, and miscellaneous compounds were 28.10%, 2.67%, 1.79%, 12.89%, and 2.35%, respectively. The phenolic compounds, generally understood to be an antimicrobial active substance, was measured at 28.10%, a relatively remarkable amount.
