CD-ROM 구동 시 발생소음 분석 및 저감 방안에 관한 연구 (A Study of Acoustic Noise Analysis and Reduction Method for Driving CD-ROM)

  • Optical disk drive device is improved in rotating speed for faster data reading. In the case of CD-ROM, rotating speed is over 10000 rpm in the practical use. As a result of high rotating speed, unexpected effects as like increasing disk fluctuation and acoustic noise are raised by the air friction on the rotating disk and the eccentricity of rotating parts. The overall acoustic noise of running CD-ROM could be classified into two different characterized noise. The first is the structural-borne noise which is generated from vibrating solid body. By the reason, the signal of structural borne noise has very similar to the signal of surface vibrating one. It has dense noise energy at specific frequency region. The other is the air-borne noise which is generated from turbulence or vortex caused by friction between disk and air. The signal of air-borne noise has no dominant peak point at acoustic pressure-frequency domain. The noise energy is widely distributed while comparatively high and large frequency region. The structural-borne noise could be reduced by reducing vibration of structure and in addition it's target reducing frequency is narrow. However the air-borne noise reduction is effectively needed of enclosing method for the noise source located near the disk surface because it is difficult to define target frequency point. In this study, the acoustic noise at driving CD-ROM is classified by the sides of it's character and tried to reduce the overall acoustic noise.

돌돔(Oplegnathus fasciatus) somatolactin cDNA의 분석 (Characterization of Somatolactin cDNA from Rock Bream (Oplegnathus fasciatus))

  • 돌돔 (Oplegnathus fasciatus) SL을 암호화하는 cDNA clone을 뇌하수체로부터 RT-PCR 방법에 의해 획득하였다. 돌돔 SL cDNA의 길이는 1636 bp로서 24개의 아미노산인 signal peptide와 207개 의 aa으로 구성된 mature protein을 암호화하는 696 bp의 open reading frame을 갖고 있다. 또한, 돌돔 SL 아미노산에는 이황화 결합에 관여하는 7개의 시스테인 잔기 $(Cys^{5},\; Cys^{15},\; Cys^{42},\; Cys^{65},\; Cys^{181},\; Cys^{198}\$$Cys^{206})$와 두 개의 potential N-glycosylation site인 $Asn^{121}$$Asn^{153}$을 확인하였다. 돌돔 SL은 goldfish와 channel catfish를 제외한 다른 경골어류 SL에 아미노산 서열은 61.1∼92.6%, 뉴클레오타이드 서열은 63∼92.6%의 일치를 나타낸다. 아미노산 서열 alingment에서 돌돔 SL은 다른 어류 SL에 공통적인 4개의 conserved domain $(A_{SL},\; B_{SL},\; C_{SL}$$D_{SL})$을 갖고 있음을 확인하였다. 이들중 $A_{SL},\; B_{SL}$,과 $D_{SL}$,은 경골어류 growth hormone과 prolactin에도 잘 보존되어 있었다 재조합 돌돔 SL 단백질을 E. coli에서 생산하기 위해 돌돔 SL cDNA를 발현벡터에 클로닝하여 단백질의 발현을 유도하였다 발현된 단백질은 SDS-PAGE에 의해 분자량 약 27 kDa의 재조합 단백질의 발현을 확인하였다.

  • The naval vessel must moves rolling, pitching, yawing by wave when it runs in ocean. Some narrow beam antenna needed position compensation by stabilizer or gimbal for best performance. This paper presents the precision position control for heavy weight(130kg) in roll and pitch direction. Generally it's called for gimbal. This gimbal uses P-I controller, and it's driven by linear actuator and servo motor. This gimbal gets ship's gyro signal and synchro, which have the absolute angle value. Some other similar equipments are driven by huge hydraulic power, but this gimbal is driven by small servo motor. This control loop gets the following procedure repeatedly; reading ship gyro and gimbal synchro, calculating compensated error and control output, driving motor and actuator The performance of gimbal system was satisfied.

Shannon Capacity에 근접하는 고효율의 6-ary Runlength-Limited Code (Very Efficient 6-ary Runlength-Limited Code Approaching Shannon Capacity for Optical Storage Channels)

  • 고효율의 d = 3인 6-ary runlength-limited code를 연구하였다. Rate가 6/7일 경우 98.87%의 효율을 얻을 수 있었고 rate가 13/15일 경우 Shannon capacity에 근접하는 무려 99.95%의 효율을 얻을 수 있었다. 또한 6-ary RLL code를 효과적으로 검출하기 위한 partial response mode에 관하여 고찰하였다.

Baby Lazarus: Listening to the Rebirths in "Lady Lazarus"

  • This paper examines the meaning and significance of the rebirths narrated in Sylvia Plath's "Lady Lazarus." While the previous readings of the poem have regarded the speaker's rebirth as a single event, this paper aims to understand its plurality and the underlying problem of language and sound by listening to the poet's own reading of the poem. I argue first that the sound structure of the poem can be characterized by the poet's unique employment of vowel sounds. Drawing upon Plath's another poem entitled "Morning Song" and Julia Kristeva's concept of the chora, I contend that the poet's vowels signal her desire for regression to the pre-Oedipal space where sound and body are in direct contact without the interference of language. It is my conclusion that the rebirths in "Lady Lazarus" dramatize the poet's ongoing struggle to bypass the symbolic language in order to make her body heard.

  • Bacillus megaterium ATCC 14945의 penicillin G acylase 유전자의 염기배열을 결정하였다. 이 유전자에는 2,406 염기쌍으로 이루어진 하나의 open reading frame이 존재하는데, 개시코돈의 5' 위쪽에서 Shine-Dalgarno 배열과 promoter로 여겨지는 부분을 발견하였으며, 종결코돈의 3' 아래쪽에서 rho-independent한 전사종결체와 dby사한 구조를 발견하였다. 염기배열로부터 폴리펩티드의 아미노산 배열을 유추하였다. 이 폴리펩티드의 분자량은 91,983 Da이었으며, 아미노 말단 부이에 signal sequence가 존재하였다. 이 아미노산 배열을 여러 다른 penicillin G acylase의 아미노산 배열과 비교하고 분리 정제한 효소를 SDS-polyacrylamide gel 전기영동으로 분석한 결과로부터 이 효소는 92kDa의 전구체로 해독된 후 processing 과정을 거쳐 각각 25kDa과 61kDa의 ${\alpha}$-, ${\beta}$-단위체로 구성됨을 알 수 있었다.

집누에로부터 새로운 attactin 유산 항세균성 펩타이드 유전자의 분리 및 발현 (Molecular Cloning and Expression of the Novel Attacin-Like Antibacterial Protein Gene Isolated from the Bombyx mori)

  • 곤충 유전자를 이용한 항세균성 펩타이드 생산 및 응용에 관한 기초 연구로서 비병원성 세균(Escherichia coli K12)으로 면역 반응을 유도한 누에로부터 발현량이 증가하는 유전자 중 Hyalophora cecropia의 attacin과 cDNA 상동성을 나타내는 BmInc6 클론을 분리하고 그 특성을 조사하였다. BmInc6 cDNA의 전체염기서열을 분석한 결과 그 크기는 852 bp이고, 35번째 염기에서 변역이 개시되어 679 bp 위치에서 종결되는 open reading frame을 가지며 812번째 위치에 잠정 전사 종결 신호의 존재가 확인되었다(GenBank, AF005384). BmInc6에 의해 coding되는 아미노산은 214개이며, hydropathy 분석 결과 친수성을 나타내는 단백질이었다. 그리고 BmInc6 유전자에 의해 연역되는 펩타이드를 nuecin으로 명명하였다. Nuecin 유전자를 baculovirus 발현 백터계를 이용하여 곤충세포주에서 발현시킨 결과 전사체의 크기는 약 950 bp였고, 세포내 벌현 펩타이드의 분자량은 약 23 kDa이었다. 세포내에서 발현된 nuecin 전구체로 추정되는 23 kDa 펩타이드는 세포외로 분비되는 과정에서 약 3 kDa의 signal 펩타이드가 제거됨으로서 약 20 kDa의 성숙 nuecin으로 된다는 사실을 단백질 전기영동상으로 확인하였다. Nuecin 단백질의 항세균 활성을 수종의 그람 음성 및 양성 세균에 대해 검정한 결과, 특히 E. coli와 Bacillus subtilis에 높은 활성을 나타내었으며, attacin이 한정된 그람 음성 세균에만 항세균 활성을 나타내는데 비해 nuecin은 그람 음성은 물론 그람 양성에도 항세균 활성을 나타내어, 보다 넓은 항세균 스펙트럼을 가지는 것을 알 수 있었다.

Identification and Characterization of a Conserved Baculoviral Structural Protein ODVP-6E/ODV-E56 from Choristoneura fumiferana Granulovirus

  • A gene that encodes a homologue to baculoviral ODVP-6E/ODV-E56, a baculoviral envelope-associated viral structural protein, has been identified and sequenced on the genome of Choristoneura fumiferana granulovirus (ChfuGV). The ChfuGV odvp-6e/odv-e56 gene was located on an 11-kb BamHI subgenomic fragment using different sets of degenerated primers, which were designed using the results of the protein sequencing of a major 39 kDa structural protein that is associated with the occlusion-derived virus (ODV). The gene has a 1062 nucleotide (nt) open-reading frame (ORF) that encodes a protein with 353 amino acids with a predicated molecular mass of 38.5 kDa. The amino acid sequence data that was derived from the nucleotide sequence in ChfuGV was compared to those of other baculoviruses. ChfuGV ODVP-6E/ODV-E56, along with othe baculoviral ODVP-6E/ODV-E56 proteins, all contained two putative transmembrane domains at their C-terminus. Several putative N-and O-glycosylation, N-myristoylation, and phosphorylation sites were detected in the ChfuGV ODVP-6E/ODV-E56 protein. A similar pattern was detected when a hydrophobicity-plots comparison was performed on ChfuGV ODVP-6E/ODV-E56 with other baculoviral homologue proteins. At the nucleotide level, a late promoter motif (GTAAG) was located at -14 nt upstream to the start codon of the GhfuGV odvp-6e/odv-e56 gene. a slight variant of the polyadenylation signal, AATAAT, was detected at the position +10 nt that is downstream from the termination signal. A phylogenetic tree for baculoviral ODVP-6E/ODV-E56 was constructed using a maximum parsimony analysis. The phylogenetic estimation demonstrated that ChfuGV ODVP-6E/ODV-E56 is most closely related to those of Cydia pomonella granulovirus (CpGV) and Plutella xylostella granulovirus (PxGV).

A Novel Glycosyl Hydrolase Family 16 β-Agarase from the Agar-Utilizing Marine Bacterium Gilvimarinus agarilyticus JEA5: the First Molecular and Biochemical Characterization of Agarase in Genus Gilvimarinus

  • The agarase gene gaa16a was identified from a draft genome sequence of Gilvimarinus agarilyticus JEA5, an agar-utilizing marine bacterium. Recently, three agarase-producing bacteria, G. chinensis, G. polysaccharolyticus, and G. agarilyticus, in the genus Gilvimarinus were reported. However, there have been no reports of the molecular characteristics and biochemical properties of these agarases. In this study, we analyzed the molecular characteristics and biochemical properties of agarases in Gilvimarinus. Gaa16A comprised a 1,323-bp open reading frame encoding 441 amino acids. The predicted molecular mass and isoelectric point were 49 kDa and 4.9, respectively. The amino acid sequence of Gaa16A showed features typical of glycosyl hydrolase family 16 (GH16) ${\beta}$-agarases, including a GH16 domain, carbohydrate-binding region (RICIN domain), and signal peptide. Recombinant Gaa16A (excluding the signal peptide and carbohydrate-binding region, rGaa16A) was expressed as a fused protein with maltose-binding protein at its N-terminus in Escherichia coli. rGaa16A had maximum activity at $55^{\circ}C$ and pH 7.0 and 103 U/mg of specific activity in the presence of 2.5 mM $CaCl_2$. The enzyme hydrolyzed agarose to yield neoagarotetraose as the main product. This enzyme may be useful for industrial production of functional neoagaro-oligosaccharides.

Functional Analysis of PepRSH (Pepper relA/spoT homolog) cloned from Capsicum annuum showing Systemic Acquired Resistance against Phytophthora capsici

  • RSH (relA/spoT homolog) has been known to determine the level of guanosine tetraphosphate (ppGpp) and guanosine pentaphosphate (pppGpp), which are the effector nucleotide of the prokaryotic stringent response and also play a role in antibiotic production and differentiation in Streptomyces species but not a little in eukaryotic organism, especially in plant. Salicylic acid (SA), a critical signal molecule of establishing systemic acquired resistance (SAR), could induce SAR in Pepper (Capcicum annuum) against Phytophthora capsici. And the extent of SAR induction was in proportion to the dosage of SA (or BTH). Suppression subtractive hybridization (SSH), a PCR-based method for cDNA subtraction, was carried out between SA-treated and non-SA-treated pepper leaves to isolate genes which may be responsible for defense signaling against pathogens. Early upregulated gene was selected from reverse northern and kinetics of SSH-genes transcripts in SA-treated pepper leaves upon SA treatment. Full-length cDNA of the gene (PepRSH; Pepper RelA / SpoT homolog) had an open reading frame (ORF) of 2166 bp encoding a protein of 722 amino acids and a significant homology with (p)ppGpp phosphohydrolase or synthetase. Genomic DNA gel blot analysis showed that pepper genome has at least single copy of PepRSH. PepRSH transcripts was very low in untreated pepper leaves but strongly induced by SA and methyljasmonic acid (MeJA), indicating that PepRSH may share common SA and MeJA-mediated signal transduction pathway Functional analysis in E. coli showed PepRSH confers phenotypes associated with (p)ppGpp synthesis through a complementation using active site mutagenesis.
