Toxicity of Tomato Spotted Wilt Virus Glycoprotein Signal Peptide and Promoter Activity of th 5' UTR

  • Cloning of the 5'untranslated region (5' UTR) and Nterminus of the glycoprotein precursor (G2G1) open reading frame of tomato spotted wilt virus has been problematic, possibly because of the toxicity of a signal peptide at the beginning of th G2G1 protein precursor. The toxicity of the signal peptide to bacterial growth and the reason for the expression of the peptide gene in Escherichia coli were investigated by cloning the 5' UTR and the signal peptide sequence separately. Cells transformed with the plasmid containing both the first 30 amino acids of the glycoprotein and the 5' UTR showed a severe growth inhibition whereas transformants harboring either the plasmid with the signal sequence or the 5'UTR alone did not show any ingibition. An E. coli promoter-like sequence was found in the 5'UTR and tis promoter acivity was confirmed with a promoter-less GUS gene cloned downstream of the 5'UTR. In the cloning of the Tomato spotted wilt virus (TSWV) glycoprotein G2G1 open reading frame all the recovered plasmids contained stop codons in the signal sequence region. However, clones containing no stop codon were recovered when the signal sequence and the 5'UTR were cloned separately.

Developing a simulator for Super-RENS/ROM disk using finite difference time domain method (Super-ROM/RENS 디스크 구조의 재생신호 해석을 위한 유한차분 시간구역 (FDTD) 방법을 이용한 시뮬레이터 개발)

  • We developed a numerical simulator in order to study the Super-RENS/ROM (Super REsolution Near-Field Structure, Read Only Memory) using 3-dimensional FDTD (finite difference time domain) method. The simulation can be performed by three steps. In the first step, we utilized the vector-diffraction theory to calculate the characteristics of incident laser beam from the object-lens to the surface of the disk. At the second step, we fed the calculated result as an input for the main FDTD simulations on the optical layers in the disk structure. After performed the FDTD simulations, we took near-to-far field transformation for the reflected signal, from the surface of the disk to the detector. Finally, we can get reflected signal at the photo-diode. Using this developed simulator, we were able to study about the reading signal from various disk structures as a function of a laser beam position. We calculated reading signals for various pit sizes for Super-ROM structure, and it is found that the simple optical diffraction theory can not explain the reading mechanism of Super-ROM, and more complicated temperature dependent physics must be involved.

  • In this paper, we investigate the confocal filter for high quality reading signal of micro holographic data storage system. The confocal filter is used to remove crosstalk effects of adjacent recording-layer by multi-layer multiplexing method. To remove a noise of reading signal, we consider size and geometrical structure of filter. From the obtained results, we present optical characteristics including Fresnel and Fraunhofer diffraction effects. Finally, we propose the optimal confocal filter for good image quality and increased reflection power.

Effects of content and formal schema on reading comprehension (내용과 형식 스키마가 독해에 미치는 영향)

  • The purpose of this research was to investigate the effects of content and formal schema on reading comprehension. Five hundred fiftynine subjects from high school were assigned to one of the following levels and treatment conditions : (1) Higher level & Schema Activation, (2) Higher level & Non-schema Activation, (3) Lower level & Schema Activation, and (4) Lower level & Non-schema Activation. To evaluate the effects of schema activation. two experiments were conducted : one was related to the content schema and the other to the formal schema. To evaluate the effects of content schema, three different types of tests were conducted : (1) cloze test, (2) guessing the meanings of nonsense words, and (3) immediate recall test. To evaluate the effects of formal schema instruction, four kinds of tests were conducted : (1) sorting the sentences according to the importance, (2) identifying the signal words, (3) immediate recall test, and (4) identifying the specific information. For content schema condition, results indicated that the subjects given the titles or pictures before reading in "Content Schema Activation" treatment had better grades than those of the other treatment in all types of tests. regardless of their levels. Schema activation helped the subjects to increase the cognitive predictability of missing words and to participate in the tasks more actively with risk-taking. And it was also shown that good readers tend to process the words meaningfully, while poor readers tend to process the words phonetically or morphologically. Formal schema activation through teaching the text organization also had a significant influence on three types of tests: sorting the sentences according to the importance, identifying the signal words, and immediate recall test, but not on identifying the specific information. The implications from this study can be briefly noted as follows : (l) In teaching reading, the student's background knowledge should be activated as a pre-reading activity. (2) In reading, it is more important to emphasize the student's schema than the features of the text. (3) Various educational interventions should be introduced, especially for the lower level students. (4) Teaching text structures can be a powerful method for the top-down processing strategy.

  • Lip reading is a field of image processing to assist the process of sound recognition. In some environment, the capture of sound signal usually has significant noise and therefore, the recognition rate of sound signal decreases. Lip reading can be a good feature for the increase of recognition rates. Conventional lip extraction methods have been proposed widely. Maia et. al. proposed a method by the sum of Cr and Cb. However, there are two problems as follows: the point with maximum saturation is not always regarded as lips region and the inner part of lips such as oral cavity and teeth can be classified as lips. To solve these problems, this paper proposes a method which adopts the histogram-based classifier for the extraction of lips region. The proposed method consists of two stages, learning and test. The amount of computation is minimized because this method has no color conversion. The performance of proposed method gives 66.8% of detection rate compared to 28% of conventional ones.

A Study on the Improvement in Performance of Ultrasonic Pulsed Doppler Velocimeter (초음파 펄스 도플러 속도계의 성능 개선에 관한 연구)

  • In the velocimeter, the ultrasonic pulse signal is used for measuring the profile velocity of moving targets distributed in space because of the merits of its high distance resolution and harmless affect to the human body. The velocity reading in conventional ultrasonic pulsed velocimeter depends on the wave pattern reflecting the spatial distribution of scatters and includes observational error due to the signal processing of analyzing pulse signal. In this paper, we evaluate an influence of the received waveform of pulsed signal on the velocity information by setting a model. Subsequently, in order to improve the distance resolution and to obtain precise velocity information without the influence of the spatial distribution of scatterers, we propose a new method for the analysis of Doppler pulsed signal, in which the pulsed signal is transformed into a phase function with local data. Finally, it is confirmed that the performance of the velocimeter is more improved in the proposed method than in the conventional one.

  • Material information for environment may be useful to accomplish mobile robot localization. A procedure to classify a set of indoor materials (glass, steel, wood, aluminum and concrete) with the reflected signal of ultrasonic sensor is proposed in this paper. The main idea is to use material-specific reflection characteristics for the recognition of material type. To achieve the classification task, we modeled reflected signal as a maximum amplitude with respect to distance. In this way, we can generate echo signal models for the given materials and these models are used to compare with the current sensor reading. The experimental results show that the proposed method may give material information during map building task of mobile robot.

Expression Pattern of Acetyl Xylan Esterase of Streptomyces coelicolor A3(2) in Escherichia coli (Escherichia coli에서의 Streptomyces coelicolor A3(2)의 acetyl xylan esterase 발현 양상)

  • We cloned a gene encoding acetyl xylan esterase(axeA) of Streptomyces coelicolor A3(2) and studied its expression pattern in Escherichia coli. The full sequence of axeA was amplified by PCR. Sequence analysis of the PCR product revealed an open reading frame of 1,008 nucleotides encoding a protein consisted of 335 amino acid residues, with a calculated molecular mass of about 38 kDa. The base sequence showed 98% homology to the same gene of Streptomyces lividans. Two different kinds of acetyl xylan esterases were produced in Escherichia coli(pLacI) by IPTG induction; their molecular weights were 38 kDa and 34 kDa, respectively. Of these, 38 kDa protein seemed to be a total protein holding N-terminal signal peptide region, whereas 34 kDa protein seemed to be a matured protein without signal peptide which was produced by peptide bond cleavage between two amino acid residues of alanine 41 and alanine 42.

Cloning of Growth Hormone Complementary DNA from Red-Spotted Grouper (Epinephelus akaara) and Its Expression in E. coli (붉바리(Epinephelus akaara)의 성장호르몬 cDNA의 Cloning과 E. coli에서의 발현)

  • We have cloned and sequenced the cDNA encoding growth hormone (GH) from pituitary poly(A)$^{+}$ RNA of red-spotted grouper (Epinephelus akaara). The cDNA of red-spotted grouper GH is 883 base pairs (bp) consisting of 21 bp of 5'untranslated region (UTR), 615 Up of an open reading frame (ORF) and 247 Up of 3'UTR. The polyadenylation signal, AATAAA, was 20 bp upsteam of polyadenylation site. Based on the nucleotide sequences, the deduced putative polypeptide contains 204 amino acids (aa), representing 17 aa of a signal and 187 aa of a mature polypeptide. The putative GH cDNA encodes a polypeptide with four cysteine residues and only one N-gly- cosylation site. Comparative sequence alignment shows that red-spotted grouper GH exhibits high similarity with its corresponding other Perciformes species GH cDNAs.
