An accelerated sequential sampling for estimating the reliability of N-parallel systems

  • The problem of designing an experiment to estimate the reliability of a system that has N subsystems connected in series where each subsystem n has n $T_n$ components connected in parallel is investigated both theoretically and by simulation. An accelerated sampling sheme is introduced. It is shown that the accelerated sampling scheme is asymptotically optimal as the total number of units goes to infinity. Numerical comparisons for a system that has two subsystems connected in series where each subsystem has two components connected in parallel are also given. They indicate that the accelerated sampling scheme performs better than the batch sequential sampling scheme and is nearly optimal.

Marsh와 Pond 형태의 인공 습지를 이용한 Parathion 농약의 처리에 관한 연구 (A Study on the Treatment of Parathion Pesticide Using Marsh and Pond Type Constructed Wetlands)

  • The microcosm type wetland systems were constructed in order to treat wastewater contaminated with parathion. The microcosm reactor consisted of marsh and pond type. The experiment was carried out using batch (marsh or pond) and continuous (marsh-pond and pond-marsh type) systems. In the batch reactor, marsh-type wetland completely removed parathion in water within 8 days, while pond reactor removed 97% of parathion during the same period. During parathion degradation, the amount of 4-nitrophenol production, one of the metabolites from parathion degradation, was higher in marsh-type batch reactor. In the continuous systems, both marsh-pond and pond-marsh combination systems effectively removed parathion from water, and the production of 4-nitrophenol was also minimal. In the extraction experiment, the parathion and its metabolite were not found in the wetland soil and the plant. In order to achieve both aerobic and anaerobic conditions, the continuous wetland system combining marsh and pond type can be the alternative for the non-point source pollutants such as parathion pesticide.

  • In the study, we Investigated the behavior and removal efficiency of organics, nitrogen. phosphorus with operating conditions in SBRs. Substrate used was synthetic wastewater in which the ratio of $COD_{cr}$. : N : P was 100 : 12 : 2. The cycling the in SBRs was adjusted at 6 hours and 8 hours, and then certainly Included anaerobic and aerobic conditions. Also, for each cycling time. we performed 2 series of experiment simultaneously which was set up 10 days and 20 days as SRT. The removal efficiency of $COD_{cr}$. was over 97% in all operating conditions. In the 6 hours cycling time, the removal efficiency of $PO_4^{3-}-P$ reached almost 100% in steady state. And then we could observe a typical phonemena of phosphorus release and uptake, and the removal efficiency of N was 67%, Residual N source was almost TKN and most of the rest remained as $NO_2-N$. Also the difference in both SRTs was not observed practically. In the 8 hours cycling time, dissolution of sludge appeared. and, $PO_4^{3-}-P was not nearly removed but nitrogen was removed up to 75%, And the residual nitrogen was accumulated as $NO_2^--N$.

Ex-situ Reductive Dechlorination of Carbon Tetrachloride by Iron Sulfide in Batch Reactor

  • Ex-situ reductive dechlorination of carbon tetrachloride (CT) by iron sulfide in a batch reactor was characterized in this study. Reactor scaled-up by 3.5 L was used to investigate the effect of reductant concentration on removal efficiency and process optimization for ex-situ degradation. The experiment was conducted by using both liquid-phase and gas-phase volume at pH 8.5 in anaerobic condition. For 1 mM of initial CT concentration, the removal of the target compound was 98.9% at 6.0 g/L iron sulfide. Process optimization for ex-situ treatment was performed by checking the effect of transition metal and mixing time on synthesizing iron sulfide solution, and by determining of the regeneration time. The effect of Co(II) as transition metal was shown that the reaction rate was slightly improved but the improvement was not that outstanding. The result of determination on the regeneration time indicated that regenerating reductant capacity after $1^{st}$ treatment of target compound was needed. Due to the high removal rates of CT, ex-situ reductive dechlorination in batch reactor can be used for basic treatment for the chlorinated compounds.

  • In microarray technology, many diverse experimental features can cause biases including RNA sources, microarray production or different platforms, diverse sample processing and various experiment protocols. These systematic effects cause a substantial obstacle in the analysis of microarray data. When such data sets derived from different experimental processes were used, the analysis result was almost inconsistent and it is not reliable. Therefore, one of the most pressing challenges in the microarray field is how to combine data that comes from two different groups. As the novel trial to integrate two data sets with batch effect, we simply applied standardization to microarray data before the significant gene selection. In the gene selection step, we used new defined measure that considers the distance between a gene and an ideal gene as well as the between-slide and within-slide variations. Also we discussed the association of biological functions and different expression patterns in selected discriminative gene set. As a result, we could confirm that batch effect was minimized by standardization and the selected genes from the standardized data included various expression pattems and the significant biological functions.

물-암석반응에 따른 물에서의 Sr동위원소의 거동에 대한 예비실험결과 (Preliminary Experimental Result for Clarifying Sr Isotope Behaviour of Water due to Water-Rock Interaction)

  • 물-암석 반응에 따른 물속의 Sr의 농도와 $^{87}Sr/^{86}Sr$ 비의 변화를 조사하기위해 실온에서의 회분식 실험(batch experiment)을 수행하였다. 실험방법은 기원이 서로 다른 2종류 화강암(강화 석모도 흑운모 화강암과 포천 석류석 화강암), 증류수, 지표수를 사용하여 암석을 증류수 및 지표수와 1:1의 비율로 각각 반응시킨 후의 물속의 양이온 및 음이온의 농도변화 및 물속의 $^{87}Sr/^{86}Sr$ 비를 측정하였다. 그리고 암석과 지표수의 경우, 혼합비의 차이에 의한 비교를 위해 암석과 지표수의 비율을 1:10으로 하여 반응시킨 후의 물속의 양이온과 양이온의 농도변화 및 $^{87}Sr/^{86}Sr$ 비를 측정하였다. 연구결과에 의하면, 물속의 용존성분의 함량은 초기 3-4개월간은 증가하였지만, 1년간 방치하여 놓은 시료에서는 함량이 현저하게 줄어들었다. 반면에 물속의 $^{87}Sr/^{86}Sr$ 비는 시간의 경과와 더불어 암석의 $^{87}Sr/^{86}Sr$ 비로 이동해가면서 비교적 안정되는 경향을 보여주었다. 이는 물-암석반응에 의한 물속의 $^{87}Sr/^{86}Sr$ 비는 거의 거의 평형에 도달한 이후에는 Sr 함량이나 기타 용존이온들보다 쉽게 안정됨을 지시해주는 것으로 볼 수 있다. 이 연구결과는 서로 다른 대수층에 분포하는 지하수의 연계성 혹은 혼합비를 계산하는데 있어서 $^{87}Sr/^{86}Sr$ 비가 유용하게 활용되어 질 수 있음을 지시해준다.

Optimization of cell growth and TAPS production by Pichia ciferrii mutant in batch culture

  • Batch culture of mutant derived from Pichia ciferrii ATCC 14091 was investigated for producing the intracellular tetraacetylphytosphingosine (TAPS). Composite experimental design was used to optimize the composition of the culture medium for maximizing the productivity of TAPS. In this experiment, various culture parameters were investigated that were the effects of temperature, the initial culture pH, the carbon-to-nitrogen ratio, the concentration of trace elements, and the concentration of cofactors. The optimal temperature for cell growth and TAPS synthesis appeared to be %25^{\circ}C$. An initial pH value of 7.5 gave the best results. Under the best condition, the maximum TAPS concentration indicated 7.2 g/L and its productivity was 0.06 g/L-hr in a 2.5 L jar

건지황 추출물을 이용한 알콜 발효 수율 증진 (Enhancement of Alcohol Fermentation Yield by Adding the Extract of Dried Rehmannia glutinosa Liboschitz)

  • 알콜 발효 증진을 위해 5L 배양조에서 혐기적으로 회분 배양 시 건지황 착즙액을 농도별로 첨가하며 알콜 발효 및 균체 생육을 측정한 결과 균체 생육 및 알콜 생산 수율 모두 착즙액 첨가량이 증가함에 따라 일반 발효에 비해 증가했으며, 30% (v/v) 첨가 시 11.8 (g/L)의 최대 균체양, 0.092 (%/hr)의 최대 알콜 생산성과 같은 최대치를 나타냈다. 하지만 40% 이상 첨가 시는 균체 생육 및 알콜 생산 두 경우 다 감소하는 경향을 보였으며 50% 에서는 심각한 생육 저해 현상이 나타났다. 또한 알콜 생산성 증진을 위해 30% 착즙액을 첨가해 유가식 배양을 실시 한 결과 회분 배양보다 월등히 높은 균체 생육 및 알콜 생산성을 보였다. 이 같은 생산성은 일반 알콜 발효에서 얻을 수 있는 수율보다 약 30-40% 이상 높은 것으로 건지황 착즙액이 알콜 발효에 직접적인 영향을 주는 것이 입증되었다. 이는 알콜 발효 시 건지황의 유용 성분의 활용과 함께 알콜 생산 수율 증진에도 이용이 가능한 지황의 이중적 활용도가 있음을 입증한 것으로 평가된다.

Kinetics and Optimization of Dimethyl Carbonate Synthesis by Transesterification using Design of Experiment

  • A comprehensive kinetic study has been conducted on dimethyl carbonate synthesis by transesterification reaction of ethylene carbonate with methanol. An alkali base metal (KOH) was used as catalyst in the synthesis of DMC, and its catalytic ability was investigated in terms of kinetics. The experiment was performed in a batch reactor at atmospheric pressure. The reaction orders, the activation energy and the rate constants were determined for both forward and backward reactions. The reaction order for forward and backward reactions was 0.87 and 2.15, and the activation energy was 12.73 and 29.28 kJ/mol, respectively. Using the general factor analysis in the design of experiments, we analyzed the main effects and interactions according to the MeOH/EC, reaction temperature and KOH concentration. DMC yield with various reaction conditions was presented for all ranges using surface and contour plot. Furthermore, the optimal conditions for DMC yield were determined using response surface method.

평면식 건조기의 적정작업조건 설정에 관한 연구 (Study on The Qptimization of Operating Conditions of batch-type Grain Dryer)

  • Experimental work of batch-type dryer was conducted to develop its optimurm operating conditions by analyzing the major factors which affect the drying performance. A laboratory batch-type dryer was constructed and tested for various levels of heated-air rates, and depths of grain deposit. Tong-il rice variety having the initial moisture content of approximaely 23 per cent in wet basis was used for the experiment. The criteria selected for establishing the optimum operating condition were the drying performance rate, the thermal efficiency, and the operational cost of the dryer. The results of the study are summarized as follows: 1. The performance rate of dryer for a specific operating condition was defined as total amount of material dried per hour when the moisture content of grains in the upperlayer reaches to 16 per cent in wet basis. The optimum operating conditions as viewed in the rate of drying performance could be justified by functional realtionship between the depth of grain deposit and air flow rate. In other words, there was a definite depth of grain deposit for a given air-rate which make the dryer performance maximum. The optimum grain depth for the batch-type dryer with 3.3㎡ loading area and with the attached axial fan was about 35cm. 2. The thermal efficiency for the dryer was evaluated by the ratio of the latent heat required to evaporate the grain moisture to the heat input required to raise the ambient air-temperature to 40 degree centigrade. The optimum operating condition as viewed in term of thermal efficiency analyzed was that grater depth and lower air flow-rate may be desirable. This condition is contracted with the optimum condition as viewed by the dryer performance rate. 3. The annual operating cost of batch-type dryer was analyzed for different annual hour of use and for different operation condition. The optimum condition as viewed in terms of operating cost was almost identical to one as viewed in terms of dryer performance rate. Therefore, the most economical use of batch-type dryer for the same annual operating hours can be obtained when the dryer operated in the condition of maximum dryer performance rate. Increasing the annual operating hour may be desirable to cut down the dryer operation cost, since the annual hour of dryer use is much sensitive to the operating cost than any peractical conditions of dryer operation. 4. The most desirable operational condition as justified by combining all the criteria, dryer performance rate, thermal efficiency and annual operating cost, could be concluded to operate the dryer in the condition of maximum performance rate. The condition in general is identical to the lowest operation cost for a given annual operating hour.
