  • Soybean (Glycine max (L.) Merr.) seed is the main source of protein and oil for human and animal. The use of soybean protein has been expanded in the food industry due to their excellent nutritional benefits. But, antinutritional and allergenic factors are exist in the raw mature soybean. Lipoxygenase, Kunitz trypsin inhibitor (KTI) protein, and ${\alpha}^{\prime}-subunit$ of 7S globulin are main antinutritional factors in soybean seed. Breeding of a new soybean strain with lacking these components is needed. The objective of this research was to select new soybean line with lipoxygenase-free, KTI-free, and ${\alpha}^{\prime}-subunit$ free (lx1lx1lx2lx2lx3lx3titicgy1cgy1 genotype). Total 434 $F_2$seeds were obtained from the cross of cultivar, "Gaechuck#2" and PI506876. Presence and absence of lipoxygenase, KTI protein, and ${\alpha}^{\prime}-subunit$ of 7S globulin was tested by SDS electrophoresis using a partial seed of each $F_2$seed. Only one $F_2$seed with lacking these three components was selected and was planted to $F_2$plant. Absence of lipoxygenase, KTI, and ${\alpha}^{\prime}-subunit$ protein was confirmed on the $F_3$seeds harvested. Selected line with lx1lx1lx2lx2lx3lx3titicgy1cgy1 genotype might be useful for soybean breeding.

Hydrolytic Patterns of 11S Globulin (Glycinin) by Soymilk-Clotting Enzymes I and II (두유응고효소 I 및 II에 의한 11S 단백질(Glycinin)의 가수분해 패턴)

  • Hydrolytic patterns of 11S globulin (glycinin), storage protein of soybean, by soymilk-clotting enzymes Iand IIfrom Bacillus sp. K-295G-7, which was the first soymilk-clotting enzyme to be found in a bacteria, was investigated. The clotting time of about 4~5 min is revealed by the Enzymes Iand II(0.025 units at 35$^{\circ}C$) on the acidic subunit. In electrophoresis, acidic subunit (A$_3$, M.W. 45,000) disappeared almost completely within 2 min and new products corresponding to the molecular weight of 16,000 and 20,000 were formed by the action of Enzymes I and II. Furthermore, Enzyme II produced a degradation compound having a molecular weight of about 30,000. In contrast, the hydrolytic patterns of basic subunit (M.W. 20,000) by Enzymes I and II were similar, but Enzyme II produced low molecular weight products slower than that of Enzyme I.

  • The changes in protein fractions and its gel electrophoretic pattern of rapeseeds (Brassica napus L) were investigated during germination at $25^{\circ}C$ under dark condition. The major protein fraction was found to be albumin 25.0%) and globulin (24.6%). Both fractions were decreased throughout germination, particularly significant for albumin, while prolamine(2.2%) and glutelin (1.8%) showed an initial decrease followed by a slow increase at the later stage of germination. The initial 5-6 peaks of gel electrophoresis were reduced to a few after 45 hours. The absorption spectrum at the range of 400-700 nm showed a significant increase in absorbance for sodium hexametaphosphate (SHMP) extract of rapeseeds. The protein extractability with SHMP was not significantly affected by germination.

  • The concentrations of some components of the serum in a liver function test were determined in samples of 74 apparently healthy dairy cattle, imported from foreign countries in Jeonnam district, during the periods of June to September of 1975 and July to August of 1976. The ranges, mean concentrations and activities of the SGOT, SGPT, SALP, thymol turbidity, total protein, serum albumin, serum globulin, A/G ratio, total bilirubin and total cholesterol were investigated in this work. This results obtained in the survey were summarized as follows: 1. The SGOT activities obtained from Holstein cows ranged from 57 to 129 Sigma Frankel units/ml, with a mean of $96.5{\pm}19.38$ S.F. units/ml. 2. The SGPT activities obtained from Holstein cows ranged from 5 to 49 Sigma Frankel units/ml, with a mean of $21.27{\pm}9.52$ S.F units/ml. 3. The alkaline phosphatase activities of serum obtained from Holstein cows ranged from 0.3 to 3.8 Sigma Frankel units/ml, with a mean of $1.88{\pm}0.94$ S.F. units/ml. 4. The thymol turbidity of serum obtained from Holstein cows ranged from 0.2 to 4.4 Shank Hoagland units/ml, with a mean of $1.69{\pm}0.30$ S.H units/ml. 5. The total serum protein values of Holstein cows ranged from 5.9 to 8.6g/100ml with a mean of $7.17{\pm}0.65g/100ml$. 6. The serum albumin values of Holstein cows ranged from 2.5 to 4.3g/100ml with a mean of $3.24{\pm}0.28g/100ml$. 7. The serum globulin values of Holstein cows ranged from 2.9 to 5.8g/100ml with a mean of $4.02{\pm}0.72g/100ml$. 8. The A/G ratio of serum obtained from Holstein cows ranged from 0.5 to 1.0 with a mean of $0.78{\pm}0.12$. 9. The total bilirubin of serum obtained from Holstein cows ranged from 7.2 to 0.8mg/100ml, with a mean of $0.32{\pm}0.11mg/100ml$. 10. The total cholesterol of serum obtained from Holstein cows ranged from 50.5 to 240.6mg/100ml with a mean of $135.70{\pm}57.44mg/100ml$. 11. There was little difference in the concentrations of the various serum components between cow groups by birth countries and total cow group, except for SGOT activities, serum alkaline phosphatase activities, thymol turbidity of the Holstein cows from New Zealand.

  • Change of protein component in soybean sprout grown at four temperatures was investigated by polyacrylamide gel electrophoresis. Main bands were identified using purified seed globulins. Electrophoretogram showed 5 main bands (a. b, c, d, and p) and 10 minor bands in seed and maximum number (19) of bands (8 main band including 0 and 11 minor) at 4th day after germination in cotyledon. All bands appeared in axis protein but resolution was poor. In cotyledon, a component (most rapidly) and b+c+d component decreased while o+p component and other minor components were increased at 6th day and decreased thereafter. In axis all components increased rapidly, especially in minor components and b+c+d component. High growing temperature accelerated decrease in cotyledon and increase in axis of protein, especially for 11S. The a component was identified as 7S, b+c+d as 11S and o+p as 2S globulin.

Comparison of Effectiveness between Two Different Protocols of Treatment of IV γ-globulin in Idiopathic Thrombocytopenic Purpura (특발성 혈소판 감소성 자반증에서 정주용 감마 글로불린의 투여 방법에 따른 효과 비교)

  • Purpose : Several methods of IV ${\gamma}-globulin$(IVG) infusion are effective in the treatment of autoimmune disease, including idiopathic thrombocytopenic purpura(ITP). But it is not known which method is more effective in the treatment of ITP. The effectiveness of these two methods of IVG infusion was studied in terms of platelet recovery rate, side effects and recurrence rate. Methods : Forty seven patients with acute ITP in the department of pediatrics, Eulji University Hospital from January 1995 to June 2001 were enrolled. We assesssed the treatment effects of 47 patients blindly selected; IV ${\gamma}-globulin$ 2 g/kg/day in one day(treatment group A, n=25), 400 mg/kg/day in five days(treatment group B, n=22). Results : Treatment group A increased platelet count more rapidly on the 2nd, 4th and 6th day of treatment than treatment group B. Side effects like fever, chill and vomiting were more frequent in treatment group A than treatment group B. The platelet count on the sixth day of treatment showed a greater increase in the cases which had side effects than in the cases which did not. There was no difference between the two groups in the recurrence rate. Conclusion : IV ${\gamma}-globulin$ 2 g/kg/day in one day increases platelet count more rapidly than 400 mg/kg/day in five days, and is favorable for the prevention of a severe hemorrhagic episode like early intracranial hemorrhage.

  • The physical and chemical properties of ${\gamma}-conglycinin$ of soybean (Glycine max) were investigated. The soybean protein extracted from soybean meal using 0.2M NaCl solution at pH 4.5 was passed through a Sephadex G-150 column to isolate 7S globulin. ${\gamma}$-Conglycinin was isolated and purified from the 7S globulin with a DEAE Sephadex A-50 column chromatography. The protein preparation was pure on immunoelectrophoresis, polyacrylamide gel electrophoresis and gel isoelectric fouling. It had an isoelectric point at pH 5.4 and contained 16.12% nitrogen, 4.18% mannose and 1.21% glucosamine. Amino acid composition, in general, shaved that ${\gamma}-conglycinin$ contained higher contents of lysine, dicarboxylic acids and ammonia nitrogen, and lower contents of sulfur-containing amino acids and tryptophan. The subunits of ${\gamma}-conglycinin$ were distributed in the range of pH 4.6-5.5. The subunits located in the pH region of 4.6-5.0 and 5.0-5.5 were glycopeptides (molecular weight of 38,000) and simple peptide (MW of 32,000), respectively.

Effects of Dietary Fatty Acid and Protein Sources on Serum Protein Profiles and Liver Functional Enzyme Activities in Rats with DMBA-Induced Mammary Tumors

  • This study was conducted to examine the effects of dietary sources of fatty acids and protein on serum protein profiles, hepatic functional enzyme activities, mammary tumor incidence and tumor weight in 7, 12-dimethylbenz($\alpha$)anthracene (DMBA)-treated rats. The sources of dietary fatty acids were 18n6 (rich in linoleic acid), 18n3 (rich in linolenic acid) and 22n3 (rich in DHA) : sources of dietary protein were casein (C) and soy protein isolate (S). mammary tumors (MTs) were chemically induced by DMBA (9 mg/100 g body weight) which was gastrically intubated at 7 weeks of age. Each experimental diet was given for the following 25 weeks. Casein-fed rats (group C) exhibited significantly higher levels of weight gain and FER (food efficiency ratio) than did group S. Group C showed higher levels of serum protein and globulin, and higher albumin/globulin (A/G) ratios than group S. Liver functional enzyme activities (GOT, GPT, ALP, LDH, $\gamma$-GT) and LDH/GOT ratios were not influenced by dietary protein. GPT activity was lower in the group given 18n3, and ALP activity was lower in the group given 18n6. The incidence and total number of MTs appeared to be lower in the group given 22n3 than in the group given 18n3 or 18n6, even though the average weight of MTs was highest in the group given 22n3, The average weight of MTs was higher in the C group than in the S group. MT incidence had a positive correlation with LDH activity and LDH/GOT ratio. The average weight of MTs had a negative correlation with serum albumin levels and A/G ratios, and a positive correlation with ALP activity. This research suggests that the measurement of serum protein profiles and liver functional enzyme activities may be utilized to monitor the development of mammary tumors.

  • Gelatin particle agglutination test (GPAT) was optimized for detection of Tobamovirus and contamination of the virus in commercial pepper seeds was evaluated. The optimum concentration of ${\gamma}$-globulin G, specific to tobacco mosaic virus pepper strain, was 100 ug/ml. The sensitivity of GPAT for the detection of Tobamovirus in pepper seeds was as high as enzyme-linked immunosorbent and dot immunoblotting assays. Optimum dilution ranges of the seed extract for GPAT was 5-25 folds. Using the optimized GPAT with above conditions, the rate of Tobamovirus contamination in seeds was turned out to be average of 79.1%.

Characteristics of the Soybean Protein and its Utilization (대두 단백질의 특성과 그 이용)

  • Soybean composition, which is different from those of other beans and grains, gives from 35 to 40 percent protein, 15 to 20 percent oil, and 20 to 25 percent sugar. Soybean has been extensively used as the raw material for traditional foods such as bean curd, soy sauce, soy paste and so on, since ancient times in Korea. Ultracentrifugal components of the soybean proteins represent four major peaks with sedimentation constants of about 2, 7, 11 and 15S. The two major reserve protein of soybean, 7S and 11S globulins, have been isolated and characterized by many works. The curd made with microbial enzyme exhibited minuter structure than those of the metal ion-and acid-treatment. Thus, the curd obtained by enzymatic operation serves as a material for further development of food items, and the procedure may by widely applicable in food processing.
