Genetic Change from Colorectal Carcinoma Patients Using Comparative Genomic Hybridization (비교유전자교잡법을 이용한 대장암환자에서의 유전자변화)

  • Colorectal carcinoma is one of the four major cancers in Korea, and it shows the tendency of increase every year due to economic development and changes to western styles. Accordingly, various diagnostic methods are needed and so comparative genomic hybridization (CGH) was performed. Deletion was detected on 5q (10%), 10q (17%), 17p (40%), 18p (23%), 18q (47%), 22q (23%), and higher deletion loci were 18q (12/30, 47%), 17p (12/30, 40%), and 22q (7/30, 23%). Amplification was shown on chromosomes 6pq (10%), 7p (17%), 7q (33%), 8q (13%), 9pq (10%), 12q (17%), 13q (37%), 20p (23%), and 20q (57%) respectively. The highest amplification was detected on chromosomes 20q (17/30, 57%), 13q (11/30, 37%), and 7q (10/30, 33%). The genetic change pattern with the locus of colorectal carcinoma was shown mean 3.1 (amplification 1.7, deletion 1.4) on the right colorectal carcinoma, while rectal carcinoma appeared high mean 6.3 (amplification 3.7, deletion 2.6) (p<0.001). The genetic change pattern with lymphatic gland metastasis, mean 3.5 (amplification 2.2, deletion 1.3) from "no metastasis" group, while high mean 6.3 (amplification 3.5, deletion 2.8) from metastasis group (p<0.003). The genetic change pattern with disease stages appeared mean 3.5 (amplification 2.1, deletion 1.4) from I-II stages, while high mean 6.0 (amplification 3.4, deletion 2.6) from III-IV stages (p<0.006). No significance was observed in comparing histological classification and serum CEA increased groups.

Water Quality Management of the Youngsan River based on the 7Q10 and Q275 considering Wastewater Treatment Cost (하수처리비용을 감안하고 7Q10과 저수량에 기초한 영산강 수질관리방안 연구)

  • Present condition of the Youngsan River pollution is serious. Concentrations of organic materials and nutrients are high and algal bloom takes place frequently. The pollution is mainly caused by domestic wastewater input from urban areas like Kwangju and Naju City. In this study, 6 times of water quality surveys were done for mainstream and tributaries. Delivery ratios of each tributaries are calculated with the water quality and flow data. With Arc/View GIS, sub-basin are divided and pollution loads are estimated. These data are used for water quality modeling. River quality improvement effects are analysed with 5 scenarios including process upgrade of present WWTPs and construction of new WWTPs. These scenarios are applied for the Youngsan River based on the 7Q10 and Q275. And total wastewater treatment cost in the basin is analysed for each scenario.

Effects of pH and Light Irradiation on Coenzyme Q10 Production Using Rhodobacter sphaeroides

  • To increase the level of $CoQ_{10}$ production in mass culture, the effects of pH and light irradiation on $CoQ_{10}$ production by Rhodobacter sphaeroides were investigated in a 1-L bioreactor. $CoQ_{10}$ production was growth-associated, and the highest production of $CoQ_{10}$ (1.69 mg/g dry cell) was obtained under uncontrolled pH: this production was 1.7 times higher than that obtained at controlled pH 7. Therefore, pH was a key factor affecting $CoQ_{10}$ production. The effect of light irradiation on $CoQ_{10}$ production was negligible. This result offers an advantage for mass production of $CoQ_{10}$.

Litter Production and Nutrient Contents of Litterfall in Oak and Pine Forests at Mt. Worak National Park

  • Litter production, nutrient contents of each component of litterfall and amount of nutrients returned to forest floor via litterfall were investigated from May 2005 through April 2006 in Quercus mongolica, Quercus variabilis and Pinus densiflora forests at Mt. Worak National Park. Total amount of litterfall during one year in Q. mongolica, Q. variabilis and P. densiflora forests was 542.7, 459.2 and $306.9\;g\;m^{-2}\;yr^{-1}$, respectively. Of the total litterfall, leaf litter, branch and bark, reproductive organ and the others occupied 50.3%, 22.7%, 10.1 % and 16.9% in Q. mongolica forest, 81.9%, 7.2%, 3.1% and 7.9% in Q. variabilis forest, 57.4%, 12.8%, 5.6% and 24.1 % in P. densiflora forest, respectively. Nutrients concentrations in oak litterfall were higher than those in needle litter. N, P, K, Ca and Mg concentration in leaf litterfall were 13.8, 1.1, 7.2, 4.2 and 1.3 mg/g for Q. mongolica forest, 10.5, 0.7, 3.2, 3.7 and 1.6 mg/g for Q. variabilis forest, 5.3, 0.4, 1.2, 2.8 and 0.6mg/g for P. densiflora forest, respectively. The amount of annual input of N, P, K, Ca and Mg to the forest floor via litterfall was 43.36, 2.89, 21.38, 23.31 and $5.62\;kg\;ha^{-1}\;yr^{-1}$ for Q. mongolica forest, 32.28, 2.01, 10.23, 20.29 and $7.78\;kg\;ha^{-1}\;yr^{-1}$ for Q. variabilis forest, 15.80, 1.04, 3.99, 9.70 and $2.10\;kg\;ha^{-1}\;yr^{-1}$ for P. densiflora forest, respectively.

Antioxidant and Cytotoxic Effects of Coenzyme Q10 Derivatives (Coenzyme Q10 유도체들의 항산화 및 세포독성 효과)

  • Coenzyme $Q_{10}$ and six derivatives of coenzyme $Q_n$ were synthesized and tested for their antioxidative effects occurred in proximal tubular epithelial cell (LLC-PK1 cell) and cytotoxicities using in NIH/3T3 cell. As the result, synthetic coenzyme $Q_n$ derivatives showed a potent antioxidative effect compared to coenzyme $Q_{10}$. Among these synthetic compounds, coenzyme $Q_3$-C at ranged 0.04 to 0.4 mmol showed the $107.7{\sim}135.9%$ of cell viability in LLC-PK1 cell. In the test of NIH/3T3, all synthesized coenzyme $Q_n$ derivatives showed the similar effect compared with coenzyme $Q_{10}$. A correlation between isoprene unit number of coenzyme $Q_n$ derivatives and its biological effects, we suggest reduction of isoprene unit number of $Q_n$ derivatives may be related to the increase of antioxidants effects and the reduction of cytotoxicities.

The study of optimization of restraint systems for injuries of Q6 and Q10 child dummies (Q6, Q10 어린이 인체모형 상해치에 대한 안전 구속 시스템 최적화 연구)

  • Occupant protection performance in frontal crashes has been developed and assessed for mainly front seat occupants over many years, and in recent years protection of rear seat occupants has also been extensively discussed. Unlike the front seats, the rear seats are often occupied by children seated in rear-facing or forward - facing child restraint systems, or booster seats. In the ENCAP, child occupant protection assessments using 18-month-old(P1.5) and 3-year-old(P3) test dummies in the rear seat have already been changed to new type of 18-month-old (Q1.5)and 3-year-old(Q3) test dummies. In addition, ENCAP are scheduled with the development and introduction of test dummies of 6-year-old (Q6) and 10.5-year-old children(Q10) starting 2016. In KNCAP, Q6 and Q10 child dummies will be introduced in 2017 as well. Automobile manufacturers need to develop safety performance for new child dummies closely. In this paper, we focused on Q6 and Q10 child dummies sitting in child restraint system. Offset frontal crash tests were conducted using two types of test dummies, Q6 and Q10 child dummies, positioned in the rear seat. Q6 and Q10 were used to compare dummy kinematics in rear seating positions between Q6 behind the driver's seat and Q10 behind the front passenger's seat. The full vehicle sled test results of both dummies were conducted with different restraint systems. It showed that several injury and image data was collected as the result of the full vehicle sled test. Based on the results of these investigations, this paper describes which factor is most important and combination is the best performance when evaluating rear seat occupant protection for Q6 and Q10 child dummies.

  • Recently, Child safety has become one of the issue with Q10 dummy representing large child. The objective of this paper was to evaluate performance of three child restraints system (backless booster, high-back booster and without booster) by changing D-ring location in the rear seat. Sled tests were carried out with a Q 10 in 64km/h frontal impact. Before the dynamic sled tests, we assessed dummy positioning with difference in CRS types and height adjustment positions. Dynamic sled test results indicated that there is different performance of CRS types and belt routing. These test results will use as base line data for development CRS safety performance for Q 10.

Molecular Mapping of the Blast Resistance Loci in the Durable Resistance Japonica Rice Cultivar, Palgong (도열병 내구 저항성 자포니카 벼품종 팔공의 저항성 관련 유전좌위 분석)

  • Rice blast caused by the fungus Magnaporthe grisea (anamorphic: Pyricularia oryzae) is an important disease in rice and development of resistant varieties to blast is one of the most important goals in rice breeding programs. A japonica rice variety, Palgong, has shown resistance to the Korean blast pathogen since it was developed in 1996. Nine blast resistance quantitative trait loci (QTLs) in Palgong alleles were identified on chromosomes 2, 4, 7, and 11. Four QTLs of qBn2.3, qBn4.2, qBn11.1, and qBn11.2 explained 28-56.7% of total phenotypic variation, while five QTLs of qBn2.2, qBn2.4, qBn4.1, qBn7.1, and qBn7.2 explained 9.7-18.8%. In a previous study, one to four resistance genes were located on the loci qBn2.2, qBn2.3, qBn4.2, qBn11.1, and qBn11.2, however, resistance genes were not located on the loci qBn2.4, qBn4.1, and qBn7.1. A major QTL, qBn11.2, explaining 56.7% of total phenotypic variation was related to the durable resistance of Palgong. Additionally, rice stripe virus resistance of Palgong was assumed to be based on the Stvb-i gene, which is located on a major QTL qBn11.2.

QTL Analysis of Germination Rate and Germination Coefficient of Velocity under Low Temperature in Rice (저온에서 벼의 발아율 및 발아속도 관련 양적형질 유전자좌(QTL) 분석)

  • As rice originates from tropical regions, low temperature stress during the germination stage in temperate regions leads to serious problems inhibiting germination and seedling establishment. Identifying and characterizing quantitative trait loci (QTLs) for low-temperature germination (LTG) resistance help accelerate the development of rice cultivars with LTG tolerance. In this study, we identified QTLs for LTG tolerance (qLTG5, qLTG9) and germination coefficient of velocity under optimal conditions (OGCV) (qOGCV7, qOGCV9) using 129 recombinant inbred lines (RILs) derived from the cross between a low-temperature sensitive line Milyang23 and a low-temperature tolerant variety Gihobyeo. qLTG9 and qOGCV9 were detected at the same location on chromosome 9. At both LTG QTLs (qLTG5 and qLTG9), the alleles for LTG tolerance were contributed by the japonica variety Gihobyeo. At qOGCV7 and qOGCV9, the alleles for low temperature tolerance were derived from Milyang23 and Gihobyeo, respectively. The RILs with desirable alleles at two or more QTLs, i.e., GroupVII: qLTG5+qLTG9 (qOGCV9) and GroupVIII: qLTG5+qOGCV7+qLTG9 (qOGCV9), showed stable tolerance under low-temperature stress. Our results are expected to contribute to the improvement of tolerance to low-temperature and anaerobic stress in japonica rice, which would lead to the wide adoption of direct-seeding practices.

Effect of milk and milk products consumption on physical growth and bone mineral density in Korean adolescents

  • This study was conducted to investigate the relationship among the current status of calcium intake from milk and milk products, physical growth and bone mineral density in 664 male and female middle school and high school students aged 15-17 years. In the study, the current status of calcium intake from milk and milk products was analyzed, and the height, body composition, and bone mineral density of the right heel bone (calcaneus) were measured. The daily calcium intake of milk and milk products was calculated as the 'dairy equivalent of calcium', which is the calcium content in 200 mL of white milk. The cutoffs of tertiles of the dairy equivalent of calcium were calculated and then the subjects were categorized into 3 groups according to the tertiles, Q1 group (lower intake group), Q2 group (middle intake group) and Q3 group (upper intake group). The daily calcium intake of milk and milk products in Q1, Q2 and Q3 groups was 16.2 mg, 99.7 mg, and 284.0 mg, respectively, and the ratio of milk and milk product consumption to the daily total calcium intake was 5.4%, 27.4%, and 49.7%, respectively. The ratio of total calcium intake to the daily recommended intake in study subjects was 30.5% in Q1, 42.3% in Q2, and 60.7% in Q3, with significant differences (P < 0.05). Height, body weight, BMI, and % of body fat in three tertile groups (Q1, Q2 and Q3) were not significantly different. However, the T scores for bone mineral density in female students in three tertile groups (Q1, Q2 and Q3) was significantly different (P < 0.05). The study showed that the intake of milk and milk products in adolescents, particularly in girls, can improve the bone mineral density without increasing body weight, and thus confirmed that milk intake is important in adolescence.
